1Outline of the Report on Cardiovascular Disease in China,2010显示文摘Major and profound changes have taken place in China over the past 30 years.Rapid socioeconomic progress has exerted a great impact on lifestyle,ranging from food,clothing,working and living conditions,and means of transportation to leisure activities and entertainment.At the same time,new health problems have emerged,and health services are facing new challenges.Presently,cardiovascular diseases (CVD) are among the top health problems of the Chinese people,and pose a serious challenge to all engaged in the prevention and control of these diseases.An epidemic of CVD in China is emerging as a result of lifestyle changes,urbanization and longevity.Both national policy decision-making and medical practice urgently need an authoritative report which comprehensively reflects the trends in the epidemic of CVD and current preventive measures.Since 2005,guided by the Bureau of Disease Prevention of the Ministry of Health of the People's Republic of China and the National Center for Cardiovascular Diseases of China,nationwide experts in the fields of epidemiology,clinical medicine and health economics in the realms of CVD,cerebrovascular disease,diabetes and chronic kidney disease,completed the Report on Cardiovascular Diseases in China every year.The report aims to provide a timely review of the trend of the epidemic and to assess the progress of prevention and control of CVD.In addition,as the report is authoritative,representative and readable,it will become an information platform in the CVD field and an important reference book for government,academic institutes,medical organizations and clinical physicians.This publication is expected to play a positive role in the prevention and control of CVD in China.We present an abstract from the Report on Cardiovascular Diseases in China (2010),including trends in CVD,morbidity and mortality of major CVDs,up-to-date assessment of risk factors,as well as health resources for CVD,and a profile of medical expenditure,with the aim of providing evidence for decision-making in CVD prevention and control programs in China,and of delivering the most authoritative information on CVD prevention and control for all citizens.HUSheng Shou KONG Ling Zhi GAO Run Lin ZHU Man Lu WANG Wen WANG Yong Jun WU Zhao Su CHEN Wei Wei LIU Ming Bo For the editorial board 2012Biomedical and Environmental Sciences2012,25,3:93
2Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. 4Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. 6The geothermal formation mechanism in the Gonghe Basin: Discussion and analysis from the geological background显示文摘The Gonghe Basin,a Cenozoic down-warped basin,is located in the northeastern part of the Qinghai-Xizang (Tibetan) Plateau,and spread over important nodes of the transfer of multiple blocks in the central orogenic belt in the NWW direction.It is also called “Qin Kun Fork” and “Gonghe Gap”.The basin has a high heat flow value and obvious thermal anomaly.The geothermal resources are mainly hot dry rock and underground hot water.In recent years,the mechanism of geothermal formation within the basin has been controversial.On the basis of understanding the knowledge of predecessors,this paper proposes the geothermal formation mechanism of the “heat source–heat transfer–heat reservoir and caprock–thermal system”of the Gonghe Basin from the perspective of a geological background through data integrationintegrated research-expert,discussion-graph,compilation-field verification and other processes: (1) Heat source: geophysical exploration and radioisotope calculations show that the heat source of heat in the basin has both the contribution of mantle and the participation of the earth's crust,but mainly the contribution of the deep mantle.(2) Heat transfer: The petrological properties of the basin and the exposed structure position of the surface hot springs show that one transfer mode is the material of the mantle source upwells and invades from the bottom,directly injecting heat;the other is that the deep fault conducts the deep heat of the basin to the middle and lower parts of the earth's crust,then the secondary fracture transfers the heat to the shallow part.(3) Heat reservoir and caprock: First,the convective strip-shaped heat reservoir exposed by the hot springs on the peripheral fault zone of the basin;second,the underlying hot dry rock layered heat reservoir and the upper new generation heat reservoir and caprock in the basin revealed by drilling data.(4) Thermal system: Based on the characteristics of the “heat source-heat transfer-heat reservoir and caprock”,it is preliminarily believed that the Gonghe Basin belongs to the non-magmatic heat source hydrothermal geothermal system (type II21) and the dry heat geothermal system (type II22).Its favorable structural position and special geological evolutionary history have given birth to a unique environment for the formation of the geothermal system.There may be a cumulative effect of heat accumulation in the eastern part of the basin,which is expected to become a favorable exploration area for hot dry rocks.Yan-fang Feng Xiao-xu Zhang Bo Zhang Jiang-tao Liu Yang-gang Wang De-long Jia Li-rong Hao Zhao-yu Kong 2018China Geology2018,1,3:15
7Melittin induces human gastric cancer cell apoptosis via activation of mitochondrial pathway显示文摘AIM: To investigate the apoptotic effects of melittin on SGC-7901 cells via activation of the mitochondrial signaling pathway in vitro.METHODS: SGC-7901 cells were stimulated by melittin, and its effect on proliferation and apoptosis of was investigated by methyl thiazolyl tetrazolium assay, morphologic structure with transmission electron microscopy, annexin-V/propidium iodide double-staining assay, measuring mitochondrial membrane potential(MMP) levels, and analyzing reactive oxygen species(ROS) concentrations were analyzed by flow cytometry. Cytochrome C(Cyt C), apoptosis-inducing factor(AIF), endonuclease G(Endo G), second mitochondria-derived activator of caspases(Smac)/direct IAP binding protein with low isoelectric point(Diablo), and FAS were analyzed by western blot. The expression of caspase-3 and caspase-8 was measured using activity assay kits.RESULTS: Melittin was incubated at 1.0, 2.0, 4.0, or 6.0 μg/m L for 1, 2, 4, 6, or 8 h and showed a timeand concentration-dependent inhibition of SGC-7901 cell growth. Melittin induced SGC-7901 cell apoptosis, which was confirmed by typical morphological changes. Treatment with 4 μg/m L melittin induced early apoptosis of SGC-7901 cells, and the early apoptosis rates were 39.97% ± 3.19%, 59.27% ± 3.94%, and 71.50% ± 2.87% vs 32.63% ± 2.75% for 1, 2, and 4 h vs 0 h(n = 3, P < 0.05); the ROS levels were 616.53% ± 79.78%, 974.81% ± 102.40%, and 1330.94% ± 93.09% vs 603.74% ± 71.99%(n = 3, P < 0.05); the MMP values were 2.07 ± 0.05, 1.78 ± 0.29, and 1.16 ± 0.25 vs 2.55 ± 0.42(n = 3, P < 0.05); caspase-3 activity was significantly higher compared to the control(5492.3 ± 321.1, 6562.0 ± 381.3, and 8695.7 ± 449.1 vs 2330.0 ± 121.9), but the caspase activity of the non-tumor cell line L-O2 was not different from that of the control. With the addition of the caspase-3 inhibitor(Ac-DEVD-CHO), caspase-3 activity was significantly decreased compared to the control group(1067.0 ± 132.5 U/g vs 8695.7 ± 449.1 U/g). The expression of the Cyt C, Endo G, and AIF proteins in SGC-7901 cells was significantly higher than those in the control(P < 0.05), while the expression of the Smac/Diablo protein was significantly lower than the control group after melittin exposure(P < 0.01). 8Radiofrequency ablation for hepatic hemangiomas: A consensus from a Chinese panel of experts显示文摘Recent studies have shown that radiofrequency(RF) ablation therapy is a safe, feasible, and effective procedure for hepatic hemangiomas, even huge hepatic hemangiomas. RF ablation has the following advantages in the treatment of hepatic hemangiomas: minimal invasiveness, definite efficacy, high safety, fast recovery, relatively simple operation, and wide applicability. 9End-stage liver disease score and future liver remnant volume predict post-hepatectomy liver failure in hepatocellular carcinoma显示文摘BACKGROUND Hepatocellular carcinoma(HCC)is the world’s sixth most common malignant tumor and the third cause of cancer death.Although great progress has been made in hepatectomy,it is still associated with a certain degree of risk of posthepatectomy liver failure(PHLF),which extends the length of hospital stay and remains the leading cause of postoperative death.Studies have shown that assessment of hepatic functional reserve before hepatectomy is beneficial for reducing the incidence of PHLF.AIM To assess the value of model for end-stage liver disease(MELD)score combined with standardized future liver remnant(sFLR)volume in predicting PHLF in patients undergoing hepatectomy for HCC.METHODS This study was attended by 238 patients with HCC who underwent hepatectomy between January 2015 and January 2018.Discrimination of sFLR volume,MELD score,and sFLR/MELD ratio to predict PHLF was evaluated according to the area under the receiver operating characteristic curve.RESULTS The patients were divided into two groups according to whether PHLF occurred after hepatectomy.The incidence of PHLF was 8.4%in our research.The incidence of PHLF increased with the decrease in sFLR volume and the increase in MELD score.Both sFLR volume and MELD score were considered independent predictive factors for PHLF.Moreover,the cut-off value of the sFLR/MELD score to predict PHLF was 0.078(P<0.001).This suggests that an sFLR/MELD≥0.078 indicates a higher incidence of PHLF than an sFLR/MELD<0.078.CONCLUSION MELD combined with sFLR is a reliable and effective PHLF predictor,which is superior to MELD score or sFLR volume alone.Fan-Hua Kong Xiong-Ying Miao Heng Zou Li Xiong Yu Wen Bo Chen Xi Liu Jiang-Jiao Zhou 2019World Journal of Clinical Cases2019,7,22:14

12Cell multiplication, apoptosis and p-Akt protein expression of bone mesenchymal stem cells of rat under hypoxia environment显示文摘Objective: To elucidate whether cell multiplication, apoptosis, glucose intake and p-Akt protein expression of bone Mesenchy-mal Stem Cells(MSCs) of rats is influenced by a hypoxic environment ex vivo. Methods: Passage 3 of bone marrow MSCs takenfrom Wistar rats, were cultured in a culturing chamber with 94%N2, 1%O2, 5%CO2 at 37℃. At different hypoxia time points, 0, 0.5,1, 4 and 8 h, glucose uptake was assayed by using radiation isotope 3H-G, Apoptotic Rate(AR) and dead rate(DR) were analyzedby flow cytometry(FCM) after Annexin V/PI staining, cell multiplication(by MTT methods) and p-Akt protein by immunocyto-chemistry and western blot. Results: Assay for CD29+, CD44+, CD71+, CD34- , Tn T+(after 5-azacytidine agent inducing) andALP+(after bone differentiation agent inducing) suggested these bone-derived cells were MSCs. The 3H-G intaking ratio(CPM/flask value: 157 ±11, 110 ±11, 107 ±13, 103 ±10, 100 ±9 and 98 ±10) of MSCs at different hypoxia time points, significantlydecreased compared to that of normoxia(P < 0.01) and tended to descend slowly with hypoxia time duration, for which there wasno statistical significance(P > 0.05). The AR(0.09 ±2.03%, 12.9 ±1.72%, 13.7 ±2.26%, 13.8 ±3.01%, 14.1 ±2.78% and 14.7 ±4.01% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.01) and DR (0.04 ±1.79%, 0.93 ±1.85%, 3.11 ±2.14%, 4.09 ±2.36%, 4.72 ±2.05% and 4.91 ±3.72% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.05) at different hypoxia time points significantly increasedcompared to those time in normoxia; The AR further went up with time (P < 0.05), however there was no statistical significance(P > 0.05) for the DR. Optical absorption value of MTT methods at different hypoxia time points significantly decreased com-pared to those with a corresponding normoxia time (P < 0.01) and degraded with time (in an hypoxic environment - P < 0.01).IOD of p-Akt protein of MSCs at different hypoxia time points significantly increased (0.367 ±0.031, 0.556 ±0.023, 0.579 ±0.013, 0.660 ±0.024, 0.685 ±0.039 and 0.685 ±0.011, respectively) compared to their equivalents in normoxia (P<0.05), how-ever, there was no statistical significance (P > 0.05) for different hypoxia time points. Hypoxia may result in ultramicrostructurechanges, such as defluvium of Microvilli, apoptotic body, 'margination'and so on and are further aggravated with hypoxia timestretching. 15SIZ1-Mediated SUMOylation of TPR1 Suppresses Plant Immunity in Arabidopsis显示文摘Plant immune responses are tightly regulated to ensure their appropriate deployment. Overexpression of TOPLESS-RELATED 1 (TPR1), a SUPPRESSOR OF npr1-1, CONSTITUTIVE 1 (SNC1)-interacting protein, results in autoimmunity that reduces plant growth and development. However, how TPR1 activity is regulated remains unknown. Loss of function of SIZ1, a (SUMO) E3 ligase, induces an autoimmune response, partially due to elevated SNC1 levels. Here we show that SNC1 expression is upregulated in Arabidopsis thaliana siz1-2 due to positive-feedback regulation by salicylic acid. SIZ1 physically interacts with TPR1 and facilitates its SUMO modification. The K282 and K721 residues in TPR1 serve as critical SUMO attachment sites. Simultaneous introduction of K282R and K721R substitutions in TPR1 blocked its SUMOylation, enhaneed its transcriptional co-repressor activity, and increased its association with HISTONE DEACETYLASE 19 (HDA19), suggesting that SUMOylation of TPR1 represses its transcriptional co-repressor activity and inhibits its interaction with HDA19. In agreement with this finding, the simultaneous introduction of K282R and K721R substitutions enhanced TPR1 mediated immunity, and the tpr1 mutation partially suppressed autoimmunity in siz1-2. 16Mechanisms underlying the promotion of functional recovery by deferoxamine after spinal cord injury in rats显示文摘Deferoxamine, a clinically safe drug used for treating iron overload, also repairs spinal cord injury although the mechanism for this action remains unknown. Here, we determined whether deferoxamine was therapeutic in a rat model of spinal cord injury and explored potential mechanisms for this effect. Spinal cord injury was induced by impacting the spinal cord at the thoracic T10 vertebra level. One group of injured rats received deferoxamine, a second injured group received saline, and a third group was sham operated. Both 2 days and 2 weeks after spinal cord injury, total iron ion levels and protein expression levels of the proinflammatory cytokines tumor necrosis factor-α and interleukin-1β and the pro-apoptotic protein caspase-3 in the spinal cords of the injured deferoxamine-treated rats were significantly lower than those in the injured saline-treated group. The percentage of the area positive for glial fibrillary acidic protein immunoreactivity and the number of terminal deoxynucleotidyl transferase d UTP nick end labeling-positive cells were also significantly decreased both 2 days and 2 weeks post injury, while the number of Neu N-positive cells and the percentage of the area positive for the oligodendrocyte marker CNPase were increased in the injured deferoxamine-treated rats. At 14–56 days post injury, hind limb motor function in the deferoxamine-treated rats was superior to that in the saline-treated rats. 18Developmental changes in intercellular junctions and Kv channels in the intestine of piglets during the suckling and post-weaning periods显示文摘Background: The intestinal epithelium is an important barrier that depends on a complex mixture of proteins and these proteins comprise different intercellular junctions. The purpose of this study was to investigate the postnatal and developmental changes in morphology, intercellular junctions and voltage-gated potassium(Kv) channels in the intestine of piglets during the suckling and post-weaning periods.Results: Samples of the small intestine were obtained from 1-, 7-, 14-, and 21-d-old suckling piglets and piglets on d 1, 3, 5, and 7 after weaning at 14 d of age. The results showed that the percentage of proliferating cell nuclear antigen(PCNA)-positive cells and alkaline phosphatase(AKP) activity, as well as the abundances of E-cadherin,occludin, and Kv1.5 m RNA and claudin-1, claudin-3, and occludin protein in the jejunum were increased from d 1to d 21 during the suckling period(P < 0.05). Weaning induced decreases in the percentage of PCNA-positive cells,AKP activity and the abundances of E-cadherin, occludin and zonula occludens(ZO)-1 m RNA or protein in the jejunum on d 1, 3 and 5 post-weaning(P < 0.05). There were lower abundances of E-cadherin, occludin and ZO-1m RNA as well as claudin-1, claudin-3 and ZO-1 protein in the jejunum of weanling piglets than in 21-d-old suckling piglets(P < 0.05). The abundances of E-cadherin, occludin, ZO-1 and integrin m RNA were positively related to the percentage of PCNA-positive cells.Conclusion: Weaning at 14 d of age induced damage to the intestinal morphology and barrier. 19Advances in statistical mechanics of rock masses and its engineering applications显示文摘To efficiently link the continuum mechanics for rocks with the structural statistics of rock masses,a theoretical and methodological system called the statistical mechanics of rock masses(SMRM)was developed in the past three decades.In SMRM,equivalent continuum models of stressestrain relationship,strength and failure probability for jointed rock masses were established,which were based on the geometric probability models characterising the rock mass structure.This follows the statistical physics,the continuum mechanics,the fracture mechanics and the weakest link hypothesis.A general constitutive model and complete stressestrain models under compressive and shear conditions were also developed as the derivatives of the SMRM theory.An SMRM calculation system was then developed to provide fast and precise solutions for parameter estimations of rock masses,such as full-direction rock quality designation(RQD),elastic modulus,Coulomb compressive strength,rock mass quality rating,and Poisson’s ratio and shear strength.The constitutive equations involved in SMRM were integrated into a FLAC3D based numerical module to apply for engineering rock masses.It is also capable of analysing the complete deformation of rock masses and active reinforcement of engineering rock masses.Examples of engineering applications of SMRM were presented,including a rock mass at QBT hydropower station in northwestern China,a dam slope of Zongo II hydropower station in D.R.Congo,an open-pit mine in Dexing,China,an underground powerhouse of Jinping I hydropower station in southwestern China,and a typical circular tunnel in Lanzhou-Chongqing railway,China.These applications verified the reliability of the SMRM and demonstrated its applicability to broad engineering issues associated with jointed rock masses.Faquan Wu Jie Wu Han Bao Bo Li Zhigang Shan Deheng Kong 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,1:8
