| 3 | Methylprednisolone promotes recovery of neurological function after spinal cord injury: association with Wnt/β-catenin signaling pathway activation显示文摘Some studies have indicated that the Wnt/β-catenin signaling pathway is activated following spinal cord injury,and expression levels of specific proteins,including low-density lipoprotein receptor related protein-6 phosphorylation,β-catenin,and glycogen synthase kinase-3β,are significantly altered.We hypothesized that methylprednisolone treatment contributes to functional recovery after spinal cord injury by inhibiting apoptosis and activating the Wnt/β-catenin signaling pathway.In the current study,30 mg/kg methylprednisolone was injected into rats with spinal cord injury immediately post-injury and at 1 and 2 days post-injury.Basso,Beattie,and Bresnahan scores showed that methylprednisolone treatment significantly promoted locomotor functional recovery between 2 and 6 weeks post-injury.The number of surviving motor neurons increased,whereas the lesion size significantly decreased following methylprednisolone treatment at 7 days post-injury.Additionally,caspase-3,caspase-9,and Bax protein expression levels and the number of apoptotic cells were reduced at 3 and 7days post-injury,while Bcl-2 levels at 7 days post-injury were higher in methylprednisolone-treated rats compared with saline-treated rats.At 3 and 7 days post-injury,methylprednisolone up-regulated expression and activation of the Wnt/β-catenin signaling pathway,including low-density lipoprotein receptor related protein-6 phosphorylation,β-catenin,and glycogen synthase kinase-3β phosphorylation.These results indicate that methylprednisolone-induced neuroprotection may correlate with activation of the Wnt/β-catenin signaling pathway. | Gong-biao Lu Fu-wen Niu Ying-chun Zhang Lin DU Zhi-yuan Liang Yuan Gao Ting-zhen Yan Zhi-kui Nie Kai Gao | 2016 | Neural Regeneration Research2016,11,11: | 11 |
| 4 | TaqMan Real-time RT-PCR Assay for Detecting and Differentiating Japanese Encephalitis Virus显示文摘Objective To detect Japanese encephalitis virus(JEV) rapidly and distinguish its genotypes, a TaqMan-based reverse transcriptase quantitative polymerase chain reaction(RT-PCR) detection system was developed.Methods By aligning the full-length sequences of JEV(G1-G5), six sets of highly specific TaqMan real-time RT-PCR primers and probes were designed based on the highly conserved NS1, NS2, and M genes of JEV, which included one set for non-specific JEV detection and five sets for the detection of specific JEV genotypes. Twenty batches of mosquito samples were used to evaluate our quantitative PCR assay.Results With the specific assay, no other flavivirus were detected. The lower limits of detection of the system were 1 pfu/mL for JEV titers and 100 RNA copies/μL. The coefficients of variation of this real-time RT-PCR were all < 2.8%. | 5 | Risk factors of intra-abdominal bacterial infection after liver transplantation in patients with hepatocellular carcinoma显示文摘Objective: To explore the risk factors of intra-abdominal bacterial infection(IAI) after liver transplantation(LT) in patients with hepatocellular carcinoma(HCC). Methods: A series of 82 HCC patients who received LT surgeries in our department between March 2004 and April 2010 was recruited in this study. Then we collected and analyzed the clinical data retrospectively. Statistical analysis system(SPSS) software was adopted to perform statistical analysis. Chi-square test, t-test and Wilcoxon rank sum test were used to analyze the clinical data and compute the significance of the incidences of early-stage IAI after LT for HCC patients. Binary logistic regression was performed to screen out the risk factors, and multiple logistic regression analyses were performed to compute the independent risk factors. Results: A series of 13 patients(13/82, 15.9%) had postoperative IAI. The independent risk factors of postoperative intra-abdominal bacterial infections after LT for HCC patients were preoperative anemia [Hemoglobin(HGB) <90 g/L] and postoperative abdominal hemorrhage(72 hours >400 mL), with the odds ratios at 8.121(95% CI, 1.417 to 46.550, P=0.019) and 5.911(95% CI, 1.112 to 31.432, P=0.037). Conclusions: Postoperative IAI after LT in patients with HCC was a common complication. | 8 | An Improved Barcoded Oligonucleotide Primers-based Next-generation Sequencing Approach for Direct Identification of Viral Pathogens in Clinical Specimens显示文摘Objective To provide a feasible and cost-effective next-generation sequencing(NGS) method for accurate identification of viral pathogens in clinical specimens, because enormous limitations impede the clinical use of common NGS, such as high cost, complicated procedures, tremendous data analysis, and high background noise in clinical samples. Methods Viruses from cell culture materials or clinical specimens were identified following an improved NGS procedure: reduction of background noise by sample preprocessing, viral enrichment by barcoded oligonucleotide(random hexamer or non-ribosomal hexanucleotide) primer-based amplification, fragmentation-free library construction and sequencing of one-tube mixtures, as well as rapid data analysis using an in-house pipeline. Results NGS data demonstrated that both barcoded primer sets were useful to simultaneously capture multiple viral pathogens in cell culture materials or clinical specimens and verified that hexanucleotide primers captured as many viral sequences as hexamers did. Moreover, direct testing of clinical specimens using this improved hexanucleotide primer-based NGS approach provided further detailed genotypes of enteroviruses causing hand, foot, and mouth disease(HFMD) and identified other potential viruses or differentiated misdiagnosis events. | 10 | A highly porous animal bone-derived char with a superiority of promoting nZVI for Cr(Ⅵ)sequestration in agricultural soils显示文摘Paddy soil and irrigation water are commonly contaminated with hexavalent chromium[Cr(Ⅵ)]near urban industrial areas,thereby threatening the safety of agricultural products and human health.In this study,we develop a porous and high specific area bone char(BC)to support nanoscale zero-valent iron(n ZVI)and apply it to remediate Cr(Ⅵ)pollution in water and paddy soil under anaerobic conditions.The batch experiments reveal that BC/n ZVI exhibits a higher removal capacity of 516.7 mg/(g·n ZVI)for Cr(Ⅵ)than n ZVI when normalized to the actual n ZVI content,which is 2.8 times that of n ZVI;moreover,the highest n ZVI utilization is the n ZVI loading of 15%(BC/n ZVI15).The Cr(Ⅵ)removal efficiency of BC/n ZVI15 decreases with increasing p H(4–10).Coexisting ions(phosphate and carbonate)and humic acid can inhibit the removal of Cr(Ⅵ)with BC/n ZVI15.Additionally,BC exhibits a strong advantage in promoting Cr(Ⅵ)removal by n ZVI compared to the widely used biochar and activated carbon.Our results demonstrate that reduction and coprecipitation are the dominant Cr(Ⅵ)removal mechanisms.Furthermore,BC/n ZVI15 shows a significantly higher reduction and removal efficiency as well as a strong anti-interference ability for Cr(Ⅵ)in paddy soil,as compared to n ZVI.These findings provide a new effective material for remediating Cr(Ⅵ)pollution from water and soil. | Kai Liu Fangbai Li Qingwen Tian Chengrong Nie Yibing Ma Zhenlong Zhu Liping Fang Yuanying Huang Siwen Liu | 2021 | Journal of Environmental Sciences2021,33,6: | 7 |
| 11 | Risk Factors for Strangulated Ovarian Hernia in Female Infants:the Role of Ovarian Volume显示文摘The risk factors associated with strangulated ovarian hernia (SOH)in female patients (<1 year old)were identified.A retrospective analysis was conducted regarding the data from 2006 to 2017.The patients were divided into 2groups:SOH group (n=9)and non-SOH group (n=23). Aberrant HH signaling activation may accelerate the growth of gastrointestinal tumors and lead to tumor immune tolerance and drug resistance.The interaction between HH signaling and the TME is intimately involved in these processes, for example, tumor growth, tumor immune tolerance, inflammation, and drug resistance. Evidence indicates that inflammatory factors in the TME, such as interleukin 6(IL-6) and interferon-g(IFN-g), macrophages, and T cell-dependent immune responses, play a vital role in tumor growth by affecting the HH signaling pathway. Moreover, inhibition of proliferating cancer-associated fibroblasts(CAFs) and inflammatory factors can normalize the TME by suppressing HH signaling. Furthermore, aberrant HH signaling activation is favorable to both the proliferation of cancer stem cells(CSCs) and the drug resistance of gastrointestinal tumors. | 14 | Salidroside Protects Against 6-Hydroxydopamine-Induced Cytotoxicity by Attenuating ER Stress显示文摘Parkinson's disease(PD) is a neurodegenerative disease characterized by a persistent decline of dopaminergic(DA) neurons in the substantia nigra pars compacta. Despite its frequency, effective therapeutic strategies that halt the neurodegenerative processes are lacking, reinforcing the need to better understand the molecular drivers of this disease. Importantly, increasing evidence suggests that the endoplasmic reticulum(ER)stress-induced unfolded protein response is likely involved in DA neuronal death. Salidroside, a major compound isolated from Rhodiola rosea L., possesses potent antioxidative stress properties and protects against DA neuronal death. However, the underlying mechanisms are not well understood. In the present study, we demonstrate that salidroside prevents 6-hydroxydopamine(6-OHDA)-induced cytotoxicity by attenuating ER stress. Furthermore, treatment of a DA neuronal cell line(SN4741) and primary cortical neurons with salidroside significantly reduced neurotoxin-induced increases in cytoplasmic reactive oxygen species and calcium, both of which cause ER stress, and cleaved caspase-12, which is responsible for ER stress-induced cell death. Together, these results suggest that salidroside protects SN4741 cells and primary cortical neurons from 6-OHDA-induced neurotoxicity by attenuating ER stress. | 17 | Design and Optimization of a Single Stage Centrifugal Compressor for a Solar Dish-Brayton System显示文摘According to the requirements of a solar dish-Brayton system,a centrifugal compressor stage with a minimum total pressure ratio of 5,an adiabatic efficiency above 75% and a surge margin more than 12% needs to be designed.A single stage,which consists of impeller,radial vaned diffuser,90° crossover and two rows of axial stators,was chosen to satisfy this system.To achieve the stage performance,an impeller with a 6:1 total pressure ratio and an adiabatic efficiency of 90% was designed and its preliminary geometry came from an in-house one-dimensional program.Radial vaned diffuser was applied downstream of the impeller.Two rows of axial stators after 90° crossover were added to guide the flow into axial direction.Since jet-wake flow,shockwave and boundary layer separation coexisted in the impeller-diffuser region,optimization on the radius ratio of radial diffuser vane inlet to impeller exit,diffuser vane inlet blade angle and number of diffuser vanes was carried out at design point.Finally,an optimized centrifugal compressor stage fulfilled the high expectations and presented proper performance.Numerical simulation showed that at design point the stage adiabatic efficiency was 79.93% and the total pressure ratio was 5.6.The surge margin was 15%.The performance map including 80%,90% and 100% design speed was also presented. | Yongsheng Wang Kai Wang Zhiting Tong Feng Lin Chaoqun Nie Abraham Engeda | 2013 | Journal of Thermal Science2013,22,5: | 5 |

| 19 | Critical function of DNA methyltransferase 1 in tomato development and regulation of the DNA methylome and transcriptome显示文摘DNA methylation confers epigenetic regulation on gene expression and thereby on various biological processes.Tomato has emerged as an excellent system to study the function of DNA methylation in plant development.To date,regulation and function of DNA methylation maintenance remains unclear in tomato plants.Here,we report the critical function of tomato(Solanum lycopersicum)Methyltransferase 1(Sl MET1)in plant development and DNA methylome and transcriptome regulation.Using CRISPR-Cas9 gene editing,we generated slmet1 mutants and observed severe developmental defects with a frame-shift mutation,including small and curly leaves,defective inflorescence,and parthenocarpy.In leaf tissues,mutations in Sl MET1 caused CG hypomethylation and CHH hypermethylationon a whole-genome scale,leading to a disturbed transcriptome including ectopic expression of many RIN target genes such as ACC2 in leaf tissues,which are normally expressed in fruits.Neither the CG hypomethylation nor CHH hypermethylation in the slmet1 mutants is related to tissue culture.Meanwhile,tissue culture induces non-CG hypomethylation,which occurs more frequently at gene regions than at TE regions.Our results depict Sl MET1-and tissue culture-dependent tomato DNA methylomes,and that Sl MET1 is required for maintaining a normal transcriptome and normal development of tomato. | Yu Yang Kai Tang Tatsiana U Datsenka Wenshan Liu Suhui Lv Zhaobo Lang Xingang Wang Jinghui Gao Wei Wang Wenfeng Nie Zhaoqing Chu Heng Zhang Avtar K HANDa Jian-Kang Zhu Huiming Zhang | 2019 | Journal of Integrative Plant Biology2019,61,12: | 3 |
| 20 | The LRXs-RALFs-FER module controls plant growth and salt stress responses by modulating multiple plant hormones显示文摘Salt stress is a major environmental factor limiting plant growth and productivity.We recently discovered an important new salt tolerance pathway,where the cell wall leucine-rich repeat extensins LRX3/4/5,the RAPID ALKALINIZATION FACTOR(RALF)peptides RALF22/23 and receptor-like kinase FERONIA(FER)function as a module to simultaneously regulate plant growth and salt stress tolerance.However,the intracellular signaling pathways that are regulated by the extracellular LRX3/4/5-RALF22/23-FER module to coordinate growth,cell wall integrity and salt stress responses are still unknown.Here,we report that the LRX3/4/5-RALF22/23-FER module negatively regulates the levels of jasmonic acid(JA),salicylic acid(SA)and abscisic acid(ABA).Blocking JA pathway rescues the dwarf phenotype of the lrx345 and fer-4mutants,while disruption of ABA biosynthesis suppresses the salt-hypersensitivity of these mutants.Many salt stress-responsive genes display abnormal expression patterns in the lrx345 and fer-4 mutants,as well as in the wild type plants treated with epigallocatechin gallate(EGCG),an inhibitor of pectin methylesterases,suggesting cell wall integrity as a critical factor that determines the expression pattern of stress-responsive genes.Production of reactive oxygen species(ROS)is constitutively increased in the lrx345 and fer-4mutants,and inhibition of ROS accumulation suppresses the salt-hypersensitivity of these mutants.Together,our work provides strong evidence that the LRX3/4/5-RALF22/23-FER module controls plant growth and salt stress responses by regulating hormonal homeostasis and ROS accumulation. | Chunzhao Zhao Wei Jiang Omar Zayed Xin Liu Kai Tang Wenfeng Nie Yali Li Shaojun Xie Yuan Li Tiandan Long Linlin Liu Yingfang Zhu Yang Zhao Jian-Kang Zhu | 2021 | National Science Review2021,8,1: | 3 |
