| 1 | Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically. | Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang | 2020 | Cell Research2020,30,1: | 11 |

| 3 | Diversity of root-associated fungi of Vaccinium mandarinorum along a human disturbance gradient in subtropical forests,China显示文摘Aims Ericaceous plant species can host diverse fungi in their roots,including ericoid mycorrhizal fungi(ERMF),endophytes,pathogens and some species with unknown functions.However,how this diversity of fungi responds to different human disturbances is not well understood.Methods In this study,we examined the effects of different human disturbance on fungal diversity in hair roots of Vaccinium mandarinorum,an ericaceous plant.Fungal DNA was extracted from hair roots of V.mandarinorum and high-throughput sequencing was applied to detect the diversity of root-associated fungi along a human disturbance gradient in subtropical forests in Gutianshan National Nature Reserve(GNNR)in East China.The four forest types with different disturbance regime were:old growth forest(OGF),secondary forest with once cut(SEC I),secondary forest with twice cut(SEC II)and Cunninghamia lanceolata plantation(PLF).Important Findings The results showed that:(i)diverse fungal operational units(OTUs)were detected in hair roots of V.mandarinorum in the four types of forests,covering fungal phyla of Ascomycota,Basidiomycota,Chytridiomycota,Glomeromycota and Zygomycota;(ii)Community composition of root-associated fungi of V.mandarinorum in PLF was distinct from those in the other three forest types,and two types of secondary forests had similar fungal community composition;(iii)Different fungal families respond differently to human disturbances:fungal families with significant preference to OGF were ectomycorrhizal or saprophytic fungi while fungal families with higher relative abundance in PLF were plant pathogenic or saprophytic fungi;(iv)The first principal component(PC1)of plant community had a significant effect on composition of root-associated fungal community,while edaphic parameters showed no significant effect on fungal community composition in roots of V.mandarinorum.Our results help to better understand the responses of both ericaceous plants and their fungal partners to human disturbances and forest managements. | Yanhua Zhang Jian Ni Fangping Tang Lifen Jiang Tianrong Guo Kequan Pei Lifu Sun Yu Liang | 2017 | Journal of Plant Ecology2017,10,1: | 6 |
| 4 | METTL5 stabilizes c-Myc by facilitating USP5 translation to reprogram glucose metabolism and promote hepatocellular carcinoma progression显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most prevalent cancers in the world,with a high likelihood of metastasis and a dismal prognosis.The reprogramming of glucosemetabolism is critical in the development ofHCC.TheWarburg effect has recently been confirmed to occur in a variety of cancers,including HCC.However,little is known about the molecular biological mechanisms underlying the Warburg effect in HCC cells.In this study,we sought to better understand how methyltransferase 5,N6-adenosine(METTL5)controls the development of HCC and theWarburg effect.Methods:In the current study,quantitative real-time polymerase chain reaction and Western blotting were used to detect the expression of METTL5 in HCC tissues and cell lines.Several different cell models and animal models were established to determine the role of METTL5 in glucose metabolism reprogramming and the underlying molecularmechanism of HCC.Glutathione-S-transferase pulldown,coimmunoprecipitation,RNA sequencing,non-targeted metabolomics,polysome profiling,and luciferase reporter assays were performed to investigate the molecular mechanisms of METTL5 in HCC cells.Results:We discovered that METTL5 drove glucose metabolic reprogramming to promote the proliferation and metastasis of HCC.Mechanistically,upregulation of METTL5 promoted c-Myc stability and thus activated its downstream glycolytic genes lactate dehydrogenase A(LDHA),enolase 1(ENO1),triosephosphate isomerase 1(TPI1),solute carrier family 2 member 1(SLC2A1),and pyruvate kinase M2(PKM2).The c-Box and ubiquitin binding domain(UBA)regions of ubiquitin specific peptidase 5(USP5)binded to c-Myc protein and inhibited K48-linked polyubiquitination of c-Myc.Further study revealed that METTL5 controled the USP5 translation process,which in turn regulated the ubiquitination of c-Myc.Furthermore,we identified cAMP responsive element binding protein 1(CREB1)/P300 as a critical transcriptional regulator ofMETTL5 that promoted the transcription of METTL5 in HCC.In patient-derived tumor xenograft(PDX)models,adenovirus-mediated knockout of METTL5 had a good antitumor effect and prolonged the survival of PDX-bearing mice.Conclusions:These findings point to a novel mechanism by which CREB1/P300-METTL5-USP5-c-Myc controls abnormal glucose metabolism and promotes tumor growth,suggesting that METTL5 is a potential therapeutic target and prognostic biomarker for HCC. | Peng Xia Hao Zhang Haofeng Lu Kequan Xu Xiang Jiang Yuke Jiang Xiangdong Gongye Zhang Chen Jie Liu Xi Chen Weijie Ma Zhonglin Zhang Yufeng Yuan | 2023 | Cancer Communications2023,43,3: | 3 |
| 5 | Optical Properties and Source Analysis of Aerosols over a Desert Area in Dunhuang,Northwest China显示文摘Aerosol observational data for 2012 obtained from Dunhuang Station of CARE-China(Campaign on Atmospheric Aerosol Research Network of China) were analyzed to achieve in-depth knowledge of aerosol optical properties over Dunhuang region. The results showed that the annual average aerosol optical depth(AOD) at 500 nm was 0.32 ± 0.06, and the ?ngstr?m exponent(α) was 0.73 ± 0.27. Aerosol optical properties revealed significant seasonal characteristics. Frequent sandstorms in MAM(March–April–May) resulted in the seasonal maximum AOD, 0.41 ± 0.04, and a relatively smaller αvalue, 0.44 ± 0.04. The tourism seasons, JJA(June–July–August) and SON(September–October–November) coincide with serious emissions of small anthropogenic aerosols. While in DJF(December–January–February), the composition of the atmosphere was a mixture of dust particles and polluted aerosols released by domestic heating; the average AOD and αwere 0.29 ± 0.02 and 0.66 ± 0.17, respectively. Different air masses exhibited different degrees of influence on the aerosol concentration over Dunhuang in different seasons. During MAM, ranges of AOD(0.11–1.18) and α(0.06–0.82) were the largest under the dust influence of northwest-short-distance air mass in the four trajectories. Urban aerosols transported by northwest-short-distance air mass accounted for a very large proportion in JJA and the mixed aerosols observed in SON were mainly conveyed by air masses from the west. | 11 | Evaluating the Ozone Valley over the Tibetan Plateau in CMIP6 Models显示文摘Total column ozone(TCO)over the Tibetan Plateau(TP)is lower than that over other regions at the same latitude,particularly in summer.This feature is known as the“TP ozone valley”.This study evaluates long-term changes in TCO and the ozone valley over the TP from 1984 to 2100 using Coupled Model Intercomparison Project Phase 6(CMIP6).The TP ozone valley consists of two low centers,one is located in the upper troposphere and lower stratosphere(UTLS),and the other is in the middle and upper stratosphere.Overall,the CMIP6 models simulate the low ozone center in the UTLS well and capture the spatial characteristics and seasonal cycle of the TP ozone valley,with spatial correlation coefficients between the modeled TCO and the Multi Sensor Reanalysis version 2(MSR2)TCO observations greater than 0.8 for all CMIP6 models.Further analysis reveals that models which use fully coupled and online stratospheric chemistry schemes simulate the anticorrelation between the 150 hPa geopotential height and zonal anomaly of TCO over the TP better than models without interactive chemistry schemes.This suggests that coupled chemical-radiative-dynamical processes play a key role in the simulation of the TP ozone valley.Most CMIP6 models underestimate the low center in the middle and upper stratosphere when compared with the Microwave Limb Sounder(MLS)observations.However,the bias in the middle and upper stratospheric ozone simulations has a marginal effect on the simulation of the TP ozone valley.Most CMIP6 models predict the TP ozone valley in summer will deepen in the future. | Kequan ZHANG Jiakang DUAN Siyi ZHAO Jiankai ZHANG James KEEBLE Hongwen LIU | 2022 | Advances in Atmospheric Sciences2022,39,7: | 1 |
