1Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. 2Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. 3Drug targeting through platelet membrane-coated nanoparticles for the treatment of rheumatoid arthritis显示文摘The effective drug treatment of rheumatoid arthritis (RA) is hindered by poordelivery efficiency to the diseased site and the serious side effects caused bywide-spread drug distribution. Traditional drug-targeting strategies, such asligand modification, are complex, laborious, and inefficient. Inspired by theintrinsic relationship between platelets and RA, platelet-mimetic nanoparticles(PNPs) were developed for targeted drug delivery in RA. Through plateletreceptor-mediated adhesion, an intact platelet membrane was coated onto poly(lactic-co-glycolic add) nanopartides, endowing the resulting PNPs with variousfunctional receptors. By coating with platelet membranes, the nanoparticleswere stabilized and had a better circulation profile, providing a benefit forpassive targeting. In vitro binding of PNPs to inflamed endothelium, and in vivoaccumulation in joints of a collagen-induced arthritis (CIA) mouse model of RAwere significantly improved via P-selectin and GVPI recognition, indicating thatthe PNPs could effectively target to RA tissues through multiple mechanisms,similar to natural platelets. Moreover, FK506, a model drug, was loaded into thePNPs and used to treat RA. Pharmacodynamic studies demonstrated that theFK506-PNPs had a notable anti-arthritic effect in CIA mice. 4THE TYPHOON DISASTERS AND RELATED EFFECTS IN CHINA显示文摘China is one of the countries which were affected very seriously by tropical cyclones. In this paper, the active features of tropical cyclones which affected and landfell over China, the distinguishing characteristics of typhoon disasters and related formation laws have been studied, and the impact of trphoon disasters on China’s socio-economic development have been discussed. The study reveals that typhoon disasters which affected China are characterized by high freqency in occurrence, violent in sudden occurrence, remarkable effects in China,widescope in impact and severe intensities in disasters, and this type of disaster was caused by huge gales, rainstorms, storm surges and reatal chain-disasters.Typhoon disasters have not only caused great casualties but also exerted severe effects on every economic departments in China. 5Genome-wide detection of additional fetal chromosomal abnormalities by cell-free DNA testing of 15,626 consecutive pregnant women显示文摘Cell-free DNA(cfDNA) testing for common fetal trisomies(T21, T18, T13) is highly effective. However, the usefulness of cfDNA testing in detecting other chromosomal abnormalities is unclear. We evaluated the performance of cfDNA testing for genome-wide abnormalities, and analyzed the incremental yield by reporting extra abnormalities. We performed genome-wide cfDNA testing in 15,626 consecutive pregnancies prospectively enrolled in this study. cfDNA testing results were reported and counseling was given depending on the presence of extra chromosomal abnormalities. cfDNA testing identified 190 cases(1.2%)of chromosomal abnormalities including 100 common trisomies and 90 additional abnormalities. By expanding the cfDNA reporting range to genome-wide abnormalities, the false positive rate increased to 0.39%(P<0.001) and positive predictive value(PPV) was reduced to 65.58%(P=0.42). However, the detection yield increased from 0.44% to 0.65%(P=0.014), and cfDNA testing detected 38.61%(39/101) additional abnormalities with no ultrasound and biochemical screening findings. cfDNA testing outperformed biochemical screening by showing 60 times higher true positive rate and fewer false negative results.Genome-wide cfDNA testing significantly increased the diagnostic yield by detecting extra abnormalities, especially those without diagnostic indications. 6Cloning and characterization of vernalization-related gene (ver203F) cDNA 3'end显示文摘Based on the cDNA fragment sequence of vernalization-related gene verc203 cloned by differential screening in our lab, the 5′primer has been designed. The cDNA 3′end of ver203 gene (1 197 bp) has been cloned by the RACE method. And it is identified by Northern blotting that its expression is special in vernalization treatment. After comparing the sequence in the nucleotide sequence databases of Genbank, EMBL and DDBJ, the gene has homology with Hordeum vulgare jesmonate-induced protein gene. 11Advances of nanoparticles as drug delivery systems for disease diagnosis and treatment显示文摘Decades have passed since the first nanoparticles-base medicine was approved for human cancer treatment, and the research and development of nanoparticles for drug delivery are always undergoing.Nowadays, the significant advances complicate nanoparticles’ branches, including liposomes, solid lipid nanoparticles, inorganic nanoparticles, micelles, nanovaccines and nano-antibodies, etc. These nanoparticles show numerous capabilities in treatment and diagnosis of stubborn diseases like cancer and neurodegenerative diseases, emerging as novel drug carriers or therapeutic agents in future. In this review, the complicated branches of nanoparticles are classified and summarized, with their property and functions concluded. Besides, there are also some delivery strategies that make nanoparticles smarter and more efficient in drug delivery, and frontiers in these strategies are also summarized in this review.
