| 1 | Y-chromosome evidence for no independent origin of modern human in China显示文摘East Asia is one of the few regions in the world where a large number of human fossils have been unearthed. The continuity of hominid fossils in East Asia, particularly in China has been presented as strong evidence supporting an independent origin of modern humans in this area. To search for such evidence of a possible independent origin of modern humans in China, a total of 9988 male individuals were sampled across China. Three Y-chromosome biallelic markers (M89, M130 and YAP), which were located at the non-re-combinant region of Y-chromosome, were typed among the samples. Our result showed that all the individuals carry a mutation at one of the three loci. The three mutations (M89T, M130T, YAP+) coalesce to another mutation (M168T), which was originally derived from Africa about 31000 to 79000 years ago. In other words, all Y-chromosome samples from China, with no exception, were originally derived from a lineage of African origin. | 2 | Effects of CIITA antisense RNA on the expression of HLA class Ⅱ molecules显示文摘To study the effect of the major histocompatibility complex class Ⅱ (MHC Ⅱ ) transactivator (CIITA) antisense RNA on the expression of the human leukemia (HLA) class Ⅱ molecules, 5’ end cDNA sequence of CIITA gene was cloned, and antisense RNA expression vector pcDNA-Ⅱ was constructed. HeLa cells transfected with pcDNA-Ⅱ and pcDNA3 were induced by IFN-γ for 3 d. The expression of HLA class Ⅱ molecules on HeLa/pcDNA-Ⅱ cells was significantly decreased, while it has no effect on the expression of HLA class Ⅰ molecules. This result suggests that the CIITA antisense RNA can inhibit the expression of HLA class Ⅱ molecules in HeLa cells. | 3 | Efficient transfer and expression of human clotting factor IX cDNA in neonatal hemophilia B mice mediated by VSV-G pseudotyped retrovi-rus显示文摘The feasibility of in vivo gene therapy for hemophilia B by VSV-G pseudotyped retroviral vector was introduced. The novel packaging cell line 293GPG was used to produce VSV-G/G1NaBAIX pseudotyped virus with the highest liters up to 8.5×108cfu .mL-1. In contrast to the conventional retrovirus, VSV-G pseudotyped virus was more resistant to inactivation by serum complements (P<0.001). Our results also demonstrated that VSV-G pseudotyped virus was more stable in neonatal mice serum than in adult mice serum (P<0.01). After intraperitoneal injection of different doses of virus, hFIX antigen was detected and lasted for more than 120 d, the highest level reached (72.5+6.1) ng- mL11. Moreover, the functional activity was improved to some extent in all hFIX-treated mice, the most remarkable improvement was observed in the mice treated with higher dose of virus whose clotting activity increased to (3.4±1.5)% and APTT (activated partial thromboplastin time) reduced to (43.2±7.2) s. | 4 | Expression of biologically active human clotting factor Ⅸ(hFⅨ) in the mammary gland of transgenic mice显示文摘The DNA of human factor Ⅸ (hFⅨ) gene vector pMCⅨm, which had been proven to be able to express in in vitro and living cells, was introduced into 586 zygotes of Kunming White Mice by positive pressure microinjection technique with manual operation. The 499 survival embryos after microinjection were then transferred into pseudopregnant recipient mice and 216 F 0 pups were born. The analysis of PCR and Southern blot hybridization showed that, of the 216, 6 (2 females and 4 males) were integrated with foreign DNA in their genomes, giving an integration frequency of 3% (6/216). Two F\-0 female transgenic mice could express hFⅨ protein in their milk and the content was over 100 ng/mL as measured with ELISA. | 17 | miR-1205/DNAJB1 reverses docetaxel chemoresistance in human triple negative breast carcinoma cells via regulation of mutp53/TAp63 signaling显示文摘Chemoresistance is the major cause of therapeutic failure in human triple negative breast carcinoma(TNBC).Docetaxel(DOC),a first-line therapeutic drug in TNBC treatment,is limited for long-term use due to the development of chemoresistance.Thus,overcoming chemoresistance of DOC remains an important challenge to improve patient’s outcome of TNBC.In this study,we aimed to investigate the molecular mechanism behind DOC chemoresistance and the possible therapeutic effects of miRNAs.Utilizing qRT-PCR analysis,we discovered that miR-1205 is gradually downregulated in human triple negative breast carcinoma MDA-MB-231 and docetaxel-resistant MDA-MB-231(MDA-MB-231/DOC)cells compared with Hs 578Bst normal human breast fibroblasts.Cell viability,cell cycle and apoptosis assays in MDA-MB-231/DOC cells indicated that miR-1205 overexpression enhances docetaxel sensitivity by reducing cell viability as well as inducing G2/M cell cycle arrest and cell apoptosis.Western blot analysis,dual-luciferase reporter assay,co-immunoprecipitation assay and chromatin immunoprecipitation assay revealed that miR-1205 overexpression disrupts the stable complex formation of DNAJB1,mutp53 and TAp63 by directly reducing DNAJB1 expression,which abates the sequestrating effect of mutp53 on TAp63,thereby leading to the enhanced DOC sensitivity in MDA-MB-231/DOC cells.Our findings demonstrate the role of the miR-1205/DNAJB1 axis in the docetaxel resistance of TNBC,which may offer a promising therapeutic approach to resolve docetaxel resistance in TNBC. | Yongxiang Yin Jinqiu Zhang Tao Ma Daozhen Chen Daru Lu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,1: | 1 |
