| 1 | miR-182参与调节胶质母细胞瘤的凋亡、生长和分化(英文)显示文摘Glioblastoma multiforme(GBM)is a lethal,therapy-resistant brain cancer consisting of numerous tumor cell subpopulations,including stem-like glioma-initiating cells(GICs),which contribute to tumor recurrence following initial response to therapy.Here,we identified miR-182 as a regulator of apoptosis,growth,and differentiation programs whose expression level is correlated with GBM patient survival.Repression of Bcl2-like12(Bcl2L12),c-Met,and hypoxia-inducible factor 2α(HIF2A)is of central importance to miR-182 anti-tumor activity,as it results in enhanced therapy susceptibility,decreased GIC sphere size,expansion,and stemness in vitro.To evaluate the tumor-suppressive function of miR-182 in vivo,we synthesized miR-182-based spherical nucleic acids(182-SNAs);i.e.,gold nanoparticles covalently functionalized with mature miR-182 duplexes.Intravenously administered 182-SNAs penetrated the bloodbrain/blood-tumor barriers(BBB/BTB)in orthotopic GBM xenografts and selectively disseminated throughout extravascular glioma parenchyma,causing reduced tumor burden and increased animal survival.Our results indicate that harnessing the anti-tumor activities of miR-182 via safe and robust delivery of 182-SNAs represents a novel strategy for therapeutic intervention in GBM. | Kouri FM Hurley LA Daniel WL Day ES Hua Y Hao L Peng CY Merkel TJ Queisser MA Ritner C Zhang H James CD Sznajder JI Chin L Giljohann DA Kessler JA Peter ME Mirkin CA Stegh AH | 2015 | 中华神经外科疾病研究杂志2015,14,2: | 14 |
| 2 | Supplementation with sodium butyrate improves growth and antioxidant function in dairy calves before weaning显示文摘Background: There is increasing research interest in using short-chain fatty acids(SCFAs) including butyrate as potential alternatives to antibiotic growth promoters in animal production. This study was conducted to evaluate the effects of supplementation of sodium butyrate(SB) in liquid feeds(milk, milk replacer, and the mixture of both)on the growth performance, rumen fermentation, and serum antioxidant capacity and immunoglobins in dairy calves before weaning. Forty healthy female Holstein calves(4-day-old, 40 ± 5 kg of body weight) were housed in individual hutches and randomly allocated to 1 of 4 treatment groups(n = 10 per group) using the RAND function in Excel. The control group was fed no SB(SB0), while the other three groups were supplemented with 15(SB15),30(SB30), or 45(SB45) g/d of SB mixed into liquid feeds offered. The calves were initially fed milk only(days 2 to 20), then a mixture of milk and milk replacer(days 21 to 23), and finally milk replacer only(days 24 to 60).Results: The SB supplementation enhanced growth and improved feed conversion into body weight gain compared with the SB0 group, and the average daily gain increased quadratically with increasing SB supplementation. No significant effect on rumen pH;concentrations of NH_3-N, individual and total VFAs;or acetate:propionate(A:P) ratio was found during the whole experimental period. Serum glutathione peroxidase activity increased linearly with the increased SB supplementation, while the serum concentration of maleic dialdehyde linearly decreased. Serum concentrations of immunoglobulin A, immunoglobulin G, or immunoglobulin M were not affected by the SB supplementation during the whole experimental period.Conclusions: Under the conditions of this study, SB supplementation improved growth performance and antioxidant function in pre-weaned dairy calves. | 3 | Characteristics of deep drainage and soil water in the mobile sandy lands of Inner Mongolia, northern China显示文摘Quantification of deep drainage and the response of soil water content to rainfall patterns are critical for an effective management strategy of soil water conservation and groundwater utilization. However, there has been little information on how rainfall characteristics influence soil water dynamics and deep drainage in mobile sandy lands. We used an underground chamber to examine the responses of deep drainage and soil water content in mobile sandy lands to rainfall characteristics in Inner Mongolia during the growing seasons of 2010, 2011 and 2012. Results showed that rainfall in this area was dominated by small events(≤5 mm), which increased soil water content in the surface soil layers(0–40 cm), but did not increase soil water content in the deeper soil layers(greater than 40 cm). Soil water content at the 0–100 cm depth increased significantly when the total amount of rain was >20 mm. Rainfall amount, intensity and the duration of dry intervals were significantly related to the soil water content in different soil layers. Deep drainage was significantly correlated with rainfall amount and intensity, but not with the duration of dry intervals. The coefficients of deep drainage in the mobile sandy lands ranged from 61.30% to 67.94% during the growing seasons. | 4 | The Tibetan Plateau Surface–Atmosphere Coupling System and Its Weather and Climate Effects: The Third Tibetan Plateau Atmospheric Science Experiment显示文摘The Tibetan Plateau(TP) is a key area affecting forecasts of weather and climate in China and occurrences of extreme weather and climate events over the world. The China Meteorological Administration, the National Natural Science Foundation of China, and the Chinese Academy of Sciences jointly initiated the Third Tibetan Plateau Atmospheric Science Experiment(TIPEX-Ⅲ) in 2013, with an 8–10-yr implementation plan. Since its preliminary field measurements conducted in 2013, routine automatic sounding systems have been deployed at Shiquanhe, Gaize, and Shenzha stations in western TP, where no routine sounding observations were available previously. The observational networks for soil temperature and soil moisture in the central and western TP have also been established. Meanwhile, the plateau-scale and regional-scale boundary layer observations, cloud–precipitation microphysical observations with multiple radars and aircraft campaigns, and tropospheric–stratospheric air composition observations at multiple sites, were performed. The results so far show that the turbulent heat exchange coefficient and sensible heat flux are remarkably lower than the earlier estimations at grassland, meadow, and bare soil surfaces of the central and western TP. Climatologically, cumulus clouds over the main body of the TP might develop locally instead of originating from the cumulus clouds that propagate northward from South Asia. The TIPEX-Ⅲ observations up to now also reveal diurnal variations, macro-and microphysical characteristics, and water-phase transition mechanisms, of cumulus clouds at Naqu station. Moreover, TIPEX-Ⅲ related studies have proposed a maintenance mechanism responsible for the Asian 'atmospheric water tower' and demonstrated the effects of the TP heating anomalies on African, Asian, and North American climates. Additionally, numerical modeling studies show that the Γ distribution of raindrop size is more suitable for depicting the TP raindrop characteristics compared to the M–P distribution, the overestimation of sensible heat flux can be reduced via modifying the heat transfer parameterization over the TP, and considering climatic signals in some key areas of the TP can improve the skill for rainfall forecast in the central and eastern parts of China. | 5 | Synergistic effect and different toxicities of adjuvant components of Realgar–Indigo Naturalis formula显示文摘Objective:Realgar–Indigo Naturalis formula(RIF)is a well-known arsenic-containing preparation that is used to treat acute promyelocytic leukemia(APL)in China.In recent multicenter clinical trials,complete remission rates in APL patients have ranged from 96.08%to 100%.RIF has a satisfactory therapeutic effect,but its safety is a widespread concern,since the preparation contains arsenic,a wide-ranging and naturally occurring toxicant.In this study,in order to determine the toxic potential of RIF,acute toxicity and sub chronic toxicity assays were performed to evaluate the toxic potential of Realgar and the adjuvant components of RIF in addition to Realgar’s synergy with these adjuvant components.Methods:KM mice and Wistar rats were selected for these experiments.To evaluate acute toxicity,the toxic effects of a single dose of a gradient of concentrations of Realgar were firstly determined.Then,the toxic effects of combinations of gradient doses of Realgar and fixed doses of Indigo naturalis and Salvia miltiorrhiza were evaluated.Results:The results showed that when Realgar was used alone,the2756.73 mg/kg(equivalent to 23.6 mg/kg).However,theto 936.90 mg/kg when Realgar was used with I.naturalis.By contrast,theto 7538.86 mg/kg when Realgar was used with S.miltiorrhiza.Hence,I.naturalis strengthened the toxicity of Realgar,whereas S.miltiorrhiza displayed the opposite effect.The sub chronic toxicity assessment results revealed a trend that was consistent with acute toxicity.Changes in the levels of different valence states of arsenic were also taken into account.The test results of the effects of in vitro combinations of Realgar and adjuvant components on soluble arsenic dissolution showed that I.naturalis increased the level of soluble arsenic in Realgar extracts and I.naturalis suspensions when the Realgar/I.naturalis ratio was 2,1.5,and 1.0.However,S.miltiorrhiza did not affect it.Conclusion:Based on the collective experimental results presented here,it can be concluded that the toxicity of RIF is the result of the soluble arsenic in Realgar and that the I.naturalis and S.miltiorrhiza in the RIF exert completely opposite effects on the toxicity of Realgar.This maybe an intelligent explanation for the compatibility of this formula,and this RIF study may therefore be viewed as a classic case of traditional Chinese medicine research on compatibility. | Huan-Hua Xu Zeng-Chun Ma Qiao-Li Shi Shi-Han Yang La Jiang Xiang-Mei Chen Yue Gao | 2018 | Chinese Herbal Medicines2018,10,2: | 3 |

| 9 | Correlation of DNA methylation patterns to the phenotypic features of Tibetan elite alpinists in extreme hypoxia显示文摘High altitude is an extreme environment that imposes hypoxic pressure on physiological processes,and natives living at high altitudes are more adaptive in certain physiological processes.So far,epigenetic modifications under extreme changes in hypoxic pressures are relatively less understood.Here,we recruit 32 Tibetan elite alpinists(TEAs),who have successfully mounted Everest(8848 m)at least five times.Blood samples and physiological phenotypes of TEAs and 32 matched non-alpinist Tibetan volunteers(non-TEAs)are collected for analysis.Genome-wide DNA methylation analysis identifies 23,202 differentially methylated CpGs(P_(adj)<0.05,|β|>0.1)between the two groups.Some differentially methylated CpGs are in hypoxia-related genes such as PPP1R13L,MAP3K7CL,SEPTI-9,and CUL2.In addition,Gene ontology enrichment analysis reveals several inflammation-related pathways.Phenotypic analysis indicates that 12 phenotypes are significantly different between the two groups.In particular,TEAs exhibit higher blood oxygen saturation levels and lower neutrophil count,platelet count,and heart rate.For DNA methylation association analysis,we find that two CpGs(cg16687447,cg06947206)upstream of PTEN were associated with platelet count.In conclusion,extreme hypoxia exposure leads to epigenetic modifications and phenotypic alterations of TEA,providing us clues for exploring the molecular mechanism underlying changes under extreme hypoxia conditions. | Zhuoma Basang Shixuan Zhang La Yang Deji Quzong Yi Li Yanyun Ma Meng Hao WeiLin Pu Xiaoyu Liu Hongjun Xie Meng Liang Jiucun Wang Qiangba Danzeng | 2021 | Journal of Genetics and Genomics2021,48,10: | 2 |
