| 3 | Contemporary crustal movement of continental China obtained by global positioning system(GPS) measurements显示文摘We obtain a unified horizontal velocity field of continental China and vicinity through measurements at 81 GPS stations provided by the National Key Infrastructure Project, Crustal Movement Observation Network of China. The velocity field delineates patterns of movements and deformation of active crustal blocks in continental China under the Eurasia-fixed reference frame. The space-based geodesy also clearly shows for the first time the horizontal movementand deformation of continental China induced by indentation of the Indian plate. | 5 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 14 | Science Letters:IGFBP7 plays a potential tumor suppressor role against colorectal carcinogenesis with its expression associated with DNA hypomethylation of exon 1显示文摘Insulin-like growth factor binding-protein-7 (IGFBP7) was obtained from our previous colonic adenocarcinoma (CRC) and normal mucosa suppression subtraction hybridization (SSH) cDNA libraries. By RT-PCR and immunohistochemistry, we found that IGFBP7 was overexpressed in CRC tissue compared to normal tissue. However, our in vitro experiments performed in 10 CRC cell lines showed that IGFBP7 expressed only in SW480 and Caco2 cell lines, which implied an underlying reversible regulatory mechanism. Using methylation-specific PCR (MSP) and bisulfite sodium PCR (BSP), we found that its expression was associated with DNA hypomethylation of exon1. This was further supported by the in vitro study which showed restored IGFBP7 expression after demethylation agent 5-aza-2’-deoxycytidine treatment. Correlation analysis between IGFBP7 expression and prognosis indicated that overexpression of IGFBP7 in CRC tissue correlated with favourable survival. Investigation of the func-tional role of IGFBP7 through transfection studies showed that IGFBP7 protein could inhibit growth rate, decrease colony for-mation activity, and induce apoptosis in RKO and SW620 cells, suggesting it a potential tumor suppressor protein in colorectal carcinogenesis. | 15 | Origin and evolution of oilfield brines from Tertiary strata in western Qaidam Basin:Constraints from ^(87)Sr/^(86)Sr,δD,δ^(18)O,δ^(34)S and water chemistry显示文摘Chemistry of major and minor elements,87Sr/86Sr,δD,δ18O and δ34S of brines were measured from Tertiary strata and Quaternary salt lakes in the western Qaidam Basin.The water chemistry data show that all oilfield brines are CaCl2 type.They were enriched in Ca2+,B3+,Li+,Sr2+,Br-,and were depleted in Mg2+,SO42-,which indicated that these brines had the characteristics of deeply circulated water.The relationship between δD and δ18O shows that all data of these brines decline towards the Global Meteoric Water Line(GWL) and Qaidam Meteoric Water Line(QWL),and that the intersection between oilfield brines and Meteoric Water Lines was close to the local spring and fresh water in the piedmont in the western Qaidam Basin.The results suggest that oilfield brines has initially originated from meteoric water,and then might be affected by water-rock metamorphose,because most oilfield brines distribute in the range of metamorphosing water.The 87Sr/86Sr values of most oilfield brines range from 0.71121 to 0.71194,and was less than that in salt lake water(>0.712),but close to that of halite in the study area.These imply that salt dissolution occurred in the process of migration.In addition,all oilfield brines have obviously much positive δ34S values(ranging from 26.46‰ to 54.57‰) than that of salt lake brines,which was caused by bacterial sulfate reduction resulting in positive shift of δ34S value and depleteed SO42-in oilfield brines.Combined with water chemical data and δD,δ18O,87Sr/86Sr,δ34S values,we concluded that oilfield brines mainly originate from the deeply circulated meteoric waters,and then are affected by salt dissolution,water-rock metamorphose,sulfate reduction and dolomitization during the process of migration.These processes alter the chemical compositions of oilfield brines and accumulate rich elements(such as B,Li,Sr,Br,K and so on) for sustainable utilization of salt lake resources in the Qaidam Basin. | FAN Qishun MA Haizhou LAI Zhongping TAN Hongbing LI Tingwei | 2010 | Chinese Journal Of Geochemistry2010,29,4: | 10 |
| 16 | Prevention and management of hepatitis B virus reactivation in patients with hematological malignancies treated with anticancer therapy显示文摘Hepatitis due to hepatitis B virus(HBV) reactivation can be severe and potentially fatal, but is preventable. HBV reactivation is most commonly reported in patients receiving cancer chemotherapy, especially rituximabcontaining therapy for hematological malignancies and those receiving stem cell transplantation. All patients with hematological malignancies receiving anticancer therapy should be screened for active or resolved HBV infection by blood tests for hepatitis B surface antigen(HBs Ag) and antibody to hepatitis B core antigen(antiHBc). Patients found to be positive for HBs Ag should be given prophylactic antiviral therapy to prevent HBV reactivation. For patients with resolved HBV infection, no standard strategy has yet been established to prevent HBV reactivation. There are usually two options. One is pre-emptive therapy guided by serial HBV DNA monitoring, whereby antiviral therapy is given as soon as HBV DNA becomes detectable. However, there is little evidence regarding the optimal interval and period of monitoring. An alternative approach is prophylactic antiviral therapy, especially for patients receiving highrisk therapy such as rituximab, newer generation of anti-CD20 monoclonal antibody, obinutuzumab or hematopoietic stem cell transplantation. This strategy may effectively prevent HBV reactivation and avoid the inconvenience of repeated HBV DNA monitoring. Entecavir or tenofovir are preferred over lamivudine as prophylactic therapy. Although there is no well-defined guideline on the optimal duration of prophylactic therapy, there is growing evidence to recommend continuing prophylactic antiviral therapy for at least 12 mo after cessation of chemotherapy, and even longer for those who receive rituximab or who had high serum HBV DNA levels before the start of immunosuppressive therapy. Many novel agents have recently become available for the treatment of hematological malignancies, and these agents may be associated with HBV reactivation. Although there is currently limited evidence to guide the optimal preventive measures, we recommend antiviral prophylaxis in HBs Ag-positive patients receiving novel treatments, especially the Bruton tyrosine kinase inhibitors and the phosphatidylinositol 3-kinase inhibitors, which are B-cell receptor signaling modulators and reduce proliferation of malignant B-cells. | 17 | BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD. | Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du | 2018 | Cellular & Molecular Immunology2018,15,12: | 9 |
| 18 | Early changes of hepatic hemodynamics measured by functional CT perfusion in a rabbit model of liver tumor显示文摘BACKGROUND:Early detection and treatment of hepatocellular carcinoma is crucial to improving the patients' survival.The hemodynamic changes caused by tumors can be serially measured using CT perfusion.In this study,we used a CT perfusion technique to demonstrate the changes of hepatic hemodynamics in early tumor growth,as a proof-of-concept study for human early hepatocellular carcinoma.METHODS:VX2 tumors were implanted in the liver of ten New Zealand rabbits.CT perfusion scans were made 1 week(early) and 2 weeks(late) after tumor implantation.Ten normal rabbits served as controls.CT perfusion parameters were obtained at the tumor rim,normal tissue surrounding the tumor,and control liver;the parameters were hepatic blood flow,hepatic blood volume,mean transit time,permeability of capillary vessel surface,hepatic arterial index,hepatic arterial perfusion and hepatic portal perfusion.Microvessel density and vascular endothelial growth factor were correlated.RESULTS:At the tumor rim,compared to the controls,hepatic blood flow,hepatic blood volume,permeability of capillary vessel surface,hepatic arterial index,and hepatic arterial perfusion increased,while mean transit time and hepatic portal perfusion decreased on both early and late scans(P<0.05).Hepatic arterial index increased(135%,P<0.05),combined with a sharp increase in hepatic arterial perfusion(182%,P<0.05) and a marked decrease in hepatic portal perfusion(-76%,P<0.05) at 2 weeks rather than at 1 week(P<0.05).Microvessel density and vascular endothelial growth factor showed significant linear correlations with hepatic blood flow,permeability of capillary vessel surface and hepatic arterial index,but not with hepatic blood volume or mean transit time.CONCLUSION:The CT perfusion technique demonstrated early changes of hepatic hemodynamics in this tumor model as proof-of-concept for early hepatocellular carcinoma detection in humans. | Guo-Lin Ma,Rong-Jie Bai,Hui-Jie Jiang,Xue-Jia Hao,Xu-Peng Dong,Da-Qing Li,Xin-Ding Liu and Lai Wei Department of Radiology,China-Japan Friendship Hospital,Beijing 100029,China Department of Radiology,Beijing Jishuitan Hospital,Beijing 100035,China Department of Radiology,Second Affiliated Hospital,Harbin Medical University,Harbin 150086,China | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,4: | 8 |
| 19 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 20 | Heavy metal contamination of urban topsoil in a petrochemical industrial city in Xinjiang, China显示文摘Heavy metal pollution is a widespread phenomenon in many countries of the world.In this study,we conducted a field investigation to assess the status of heavy metal pollution in urban soils of Dushanzi,a district of Karamay city in Xinjiang,China.A total of 56 soil samples in the topsoil layer of 0–15 cm were collected within the urban area and seven elements(Cu,Zn,Cd,Pb,Cr,As and Ni) were analyzed.The mean concentrations of these metals were all higher than their corresponding background values of soils in Xinjiang.We used the pollution index and ecological risk index to assess the degree of heavy metal pollution and the potential ecological risk of urban soils.The pollution index values of Cu,Zn,Cd,Pb,Cr,As and Ni were 1.81,1.35,4.64,1.27,1.80,1.39 and 1.22,respectively;and the potential ecological risk index values for them were 12.03,1.79,185.05,8.39,4.78,18.44 and 1.79,respectively.These results indicated that urban soils in Dushanzi were polluted by heavy metals to some extent and demonstrated a high ecological risk,as influenced by industrial activities.Cd was the key element for the metal pollution of urban soils in the study area.Correlation analyses,principal component analysis coupled with the spatial distribution maps of element concentrations further revealed that heavy metal pollution of urban soils can be mainly attributed to petrochemical industry,coal chemical industry,traffic and commercial activities. | WANG Wei LAI Yingshuai MA Yuanyuan LIU Zilong WANG Shufen HONG Chenglin | 2016 | Journal of Arid Land2016,8,6: | 7 |
