3Therapeutic effect of Qingyi decoction in severe acute pancreatitis-induced intestinal barrier injury显示文摘AIM: To investigate the effect of Qingyi decoction onthe expression of secreted phospholipase A2(s PLA2) in intestinal barrier injury.METHODS: Fifty healthy Sprague-Dawley rats were randomly divided into control, severe acute pancreatitis(SAP), Qingyi decoction-treated(QYT), dexamethasonetreated(DEX), and verapamil-treated(VER) groups. The SAP model was induced by retrograde infusion of 1.5% sodium deoxycholate into the biliopancreatic duct of the rats. All rats were sacrificed 24 h post-SAP induction. Arterial blood, intestine, and pancreas from each rat were harvested for investigations. The levels of serum amylase(AMY) and diamine oxidase(DAO) were determined using biochemical methods, and serum tumor necrosis factor(TNF)-α level was measured by an enzyme linked immunosorbent assay. Pathologic changes in the harvested tissues were investigated by microscopic examination of hematoxylin and eosinstained tissue sections. The expressions of s PLA2 at m RNA and protein levels were detected by reverse transcriptase PCR and Western blot, respectively. A terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay was used to investigate apoptosis of epithelial cells in the intestinal tissues. RESULTS: Compared to the control group, the expression of s PLA2 at both the m RNA and protein levels increased significantly in the SAP group(0.36 ± 0.13 vs 0.90 ± 0.38, and 0.16 ± 0.05 vs 0.64 ± 0.05, respectively; P s < 0.01). The levels of AMY, TNF-α and DAO in serum were also significantly increased(917 ± 62 U/L vs 6870 ± 810 U/L, 59.7 ± 14.3 ng/L vs 180.5 ± 20.1 ng/L, and 10.37 ± 2.44 U/L vs 37.89 ± 5.86 U/L, respectively; P s < 0.01). The apoptosis index of intestinal epithelial cells also differed significantly between the SAP and control rats(0.05 ± 0.02 vs 0.26 ± 0.06; P < 0.01). The serum levels of DAO and TNF-α, and the intestinal apoptosis index significantly correlated with s PLA2 expression in the intestine(r = 0.895, 0.893 and 0.926, respectively; Ps < 0.05). Thelevels of s PLA2, AMY, TNF-α, and DAO in the QYT, VER, and DEX groups were all decreased compared with the SAP group, but not the control group. Qingyi decoction intervention, however, gave the most therapeutic effect against intestinal barrier damage, although the onset of its therapeutic effect was slower. CONCLUSION: Qingyi decoction ameliorates acute pancreatitis-induced intestinal barrier injury by inhibiting the overexpression of intestinal s PLA2. 5Single-cell transcriptomic analysis defines the interplay between tumor cells,viral infection,and the microenvironment in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an Epstein-Barr virus(EBV)-associated malignancy with a complex tumor ecosystem.How the interplay between tumor cells,EBV,and the microenvironment contributes to NPC progression and immune evasion remains unclear.Here we performed single-cell RNA sequencing on^104,000 cells from 19 EBV^+NPCs and 7 nonmalignant nasopharyngeal biopsies,simultaneously profiling the transcriptomes of malignant cells,EBV,stromal and immune cells.Overall,we identified global upregulation of interferon responses in the multicellular ecosystem of NPC.Notably,an epithelial–immune dual feature of malignant cells was discovered and associated with poor prognosis.Functional experiments revealed that tumor cells with this dual feature exhibited a higher capacity for tumorigenesis.Further characterization of the cellular components of the tumor microenvironment(TME)and their interactions with tumor cells revealed that the dual feature of tumor cells was positively correlated with the expression of co-inhibitory receptors on CD8^+tumor-infiltrating T cells.In addition,tumor cells with the dual feature were found to repress IFN-γproduction by T cells,demonstrating their capacity for immune suppression.Our results provide new insights into the multicellular ecosystem of NPC and offer important clinical implications.Shanzhao Jin Ruoyan Li Ming-Yuan Chen Chao Yu Lin-Quan Tang Yan-Min Liu Jiang-Ping Li Yi-Na Liu Yi-Ling Luo Yifan Zhao Yu Zhang Tian-Liang Xia Shang-Xin Liu Qi Liu Guan-Nan Wang Rui You Jing-Yun Peng Jiang Li Feng Han Jianwei Wang Qiu-Yan Chen Li Zhang Hai-Qiang Mai Benjamin E.Gewurz Bo Zhao Lawrence S.Young Qian Zhong Fan Bai Mu-Sheng Zeng 2020Cell Research2020,30,11:16
6Clinical features and treatment outcomes of endogenous Klebsiella endophthalmitis:a 12-year review显示文摘AIM:To identify the clinical features and treatment outcomes of endogenous Klebsiella pneumoniae endophthalmitis and investigate prognostic factors of poor visual outcome.METHODS:The clinical records of all patients diagnosed with endogenous Klebsiella endophthalmitis between January 2007 to December 2018 in Prince of Wales Hospital,Hong Kong,China were retrospectively reviewed.Thorough ophthalmological examination findings were recorded in the case note,including visual acuity testing,slit-lamp examination,indirect ophthalmoscopy and B-scan ultrasonography if media opacity precluded fundus viewing.RESULTS:A total of 18 eyes in 14 patients were identified.Bilateral involvement was noted in 4 patients(28.6%).Hepatobiliary sepsis was the source in 9 patients(64.3%).Culture of intraocular fluid was positive in 5 out of 18 eyes(27.8%).Mortality was noted in 2 patients(14.3%).Mean final visual acuity was 20/1500.Six out of 16 eyes had total loss of sight(37.5%)and 3 eyes required evisceration(18.8%).Multivariate linear regression revealed poor presenting visual acuity(P=0.031)and lack of fundus view due to vitritis(P=0.02)as prognostic factors of poor visual outcome.CONCLUSION:Visual outcome of endogenous Klebsiella endophthalmitis is poor.Poor presenting visual acuity and lack of fundus view predict poor visual outcome.High index of suspicion for endophthalmitis is important in Klebsiella sepsis patients with complaints of ocular symptoms.Ophthalmological screening is recommended in noncommunicable patients with Klebsiella sepsis.Chun Yue Mak Mary Ho Lawrence Pui-leung Iu Helena Pui-yee Sin Li Jia Chen Grace Lui Marten Erik Brelen Alvin Lerrmann Young 2020International Journal of Ophthalmology(English edition)2020,13,12:5

10A convergence analysis for a class of practical variance-reduction stochastic gradient MCMC显示文摘Stochastic gradient Markov chain Monte Carlo(SG-MCMC) has been developed as a flexible family of scalable Bayesian sampling algorithms. However, there has been little theoretical analysis of the impact of minibatch size to the algorithm's convergence rate. In this paper, we prove that at the beginning of an SG-MCMC algorithm, i.e., under limited computational budget/time, a larger minibatch size leads to a faster decrease of the mean squared error bound. The reason for this is due to the prominent noise in small minibatches when calculating stochastic gradients, motivating the necessity of variance reduction in SG-MCMC for practical use. By borrowing ideas from stochastic optimization, we propose a simple and practical variance-reduction technique for SG-MCMC, that is efficient in both computation and storage.More importantly, we develop the theory to prove that our algorithm induces a faster convergence rate than standard SG-MCMC. 11Growing Operational Use of FY-3 Data in the ECMWF System显示文摘This paper reviews the data quality and impact of observations from the FY-3 satellite series used operationally in the ECMWF system. This includes data from the passive microwave radiometers MWHS-1, MWHS-2 and MWRI, as well as observations from the radio occultation receiver GNOS. Evaluations against background equivalents show that the quality of the observations is broadly comparable to that of similar instruments on other polar-orbiting satellites, even though biases for the passive microwave observations can be somewhat larger and more complex for some channels. An observing system experiment shows that the FY-3 instruments jointly contribute significantly to the forecast skill in the ECMWF system. 12Dye-sensitized solar cells based on functionalized truxene structure显示文摘Three new metal-free organic dyes(TX1, TX2 and TX3) based on truxene core structure, with triphenylamine as the electron donor, thiophene as the p spacers, and cyanoacetic acid or rhodanine-3-acetic acid as the electron acceptor are designed and synthesized. Their UV–vis absorption spectra,electrochemical and photovoltaic properties were investigated. The cyanoacrylic acid is verified to be a better acceptor unit(meanwhile the anchoring group) compared to the rhodanine-3-acetic acid. And also, two anchoring groups in TX2 could provide stronger adsorption ability on the Ti O2 surface. In addition, the EIS results indicate a slower charge recombination processes for TX2.
