| 1 | Rebamipide suppresses diclofenac-induced intestinal permeability via mitochondrial protection in mice显示文摘AIM: To investigate the protective effect and mechanism of rebamipide on small intestinal permeability induced by diclofenac in mice. METHODS: Diclofenac (2.5 mg/kg) was administered once daily for 3 d orally. A control group received the vehicle by gavage. Rebamipide (100 mg/kg, 200 mg/kg, 400 mg/kg) was administered intragastrically once a day for 3 d 4 h after diclofenac administration. Intestinal permeability was evaluated by Evans blue and the FITC-dextran method. The ultrastructure of the mucosal barrier was evaluated by transmission electron microscopy (TEM). Mitochondrial function including mitochondrial swelling, mitochondrial membrane potential, mitochondrial nicotinamide adenine dinucleotide-reduced (NADH) levels, succinate dehydrogenase (SDH) and ATPase activities were measured. Small intestinal mucosa was collected for assessment of malondialdehyde (MDA) content and myeloperoxidase (MPO) activity. RESULTS: Compared with the control group, intestinal permeability was significantly increased in the diclofenac group, which was accompanied by broken tight junctions, and significant increases in MDA content and MPO activity. Rebamipide significantly reduced intestinal permeability, improved inter-cellular tight junctions, and was associated with decreases in intestinal MDA content and MPO activity. At the mitochondrial level, rebamipide increased SDH and ATPase activities, NADH level and decreased mitochondrial swelling. | 3 | Construction of the recombinant adenovirus vector carrying antisense multidrug resistance gene显示文摘BACKGROUND: Multidrug resistance proteins serve as transporters for chemical drugs in human malignancies. The objective of this study was to construct a homologous recombinant adenovirus carrying a reversal fragment of multidrug resistance gene 1 (mdr1) gene cDNA sequence. METHODS: The fragment of the mdr1 gene from the plasmid pHaMDRI-1 carrying the whole human mdr1 cDNA sequence was inserted reversely into the shuttle plasmid pAdTrack-CMV of adenoviral vector system AdEasy. The homologous recombination process was taken place in E. coli BJ5183 with the backbone plasmid pAdEasy-1. After packaging in 293 cells, recombinant adenoviral plasmid was generated. The recombinant adenoviral plasmid was identified by polymerase chain reaction (PCR), restriction endonucleases digest, DNA sequence analysis and fluorescence microscopic photograph, respectively. RESULTS: The recombinant adenovirus pAdEasy-GFPASmdr1 was successfully constructed and identified by PCR, restriction digest, and sequencing with strong green fluorescence expression in fluorescence microscopic photograph. | 5 | Culturable plant pathogenic fungi associated with sugarcane in southern China显示文摘Sugarcane(Saccharum officinarum)is one of the oldest crops cultivated by mankind.Numerous fungal taxa have been reported from this host,although most were not identified beyond genus level.In this study,we explored 31 sampling sites in Guangxi and Guangdong Provinces(China),and collected 370 diseased samples from leaves and roots of sugarcane,from which 762 strains were isolated.Our preliminary analysis based on internal transcribed spacer(ITS)sequences suggested that these isolates belonged to 143 species in 51 genera,but we could not assign 129 strains.Bipolaris,Chaetomium,Curvularia,Phoma and Nigrospora represented the top five most common genera identified,while 27 rare genera comprised only one identified species.In this study,we chose above mentioned common genera for in-depth morphological observations and multi-locus analyses,in order to identify these strains to species level.In this paper,we described one new genus,32 new species,and reported 19 new records for China and the asexual morph of Chaetomium olivaceum.Hitherto,this is the most comprehensive study with molecular identification and illustration of fungi associated with sugarcane,which greatly improves our understanding of culturable mycota associated with this host. | Mubashar Raza Zhi-Feng Zhang Kevin D.Hyde Yong-Zhao Diao Lei Cai | 2019 | Fungal Diversity2019,,6: | 4 |

| 7 | Numerical Analysis on Nanoparticles-laden Gas Film Thrust Bearing显示文摘Nanoparticles can be taken as additives and added into various fluids to improve their lubricating performances. At present, researches in this area are mainly concentrated on the improvement effects of nanoparticles on the lubricating performances of liquid such as oil and water. Nanoparticles will also affect gas lubrication, but few related studies have been reported. Nanoparticles-laden gas film (NLGF) is formed when adding nanoparticles into gas bearing. Then, the lubricating performances of gas bearing including pressure distribution and load-carrying capacity will change. The variations of pressure distribution and load-carrying capacity in nanoparticles-laden gas film thrust bearing are investigated by numerical method. Taking account of the compressibility of gas and the interactions between gas and nanoparticles, a computational fluid dynamics model based on Navier-Stokes equations is applied to simulate the NLGF flow. The effects of inlet nanoparticles volume fraction and orifice radius on film pressure distribution and load-carrying capacity of the NLGF are calculated. The numerical calculation results show that both of the film land pressure and the maximum film pressure both increase when the nanoparticles are added into gas bearing, and the film pressures increase with the rising of the inlet nanoparticles volume fraction. The nanoparticles have an enhancement effect on load-carrying capacity of the studied bearing, and the enhancement effect becomes greater as the film thickness decrease. Therefore, nanoparticles can effectively improve the lubricating performance of gas bearing. | 10 | A potent PGK1 antagonist reveals PGK1 regulates the production of IL-1βand IL-6显示文摘Glycolytic metabolism enzymes have been implicated in the immunometabolism field through changes in metabolic status. PGK1 is a catalytic enzyme in the glycolytic pathway. Here, we set up a high-throughput screen platform to identify PGK1 inhibitors. DC-PGKI is an ATPcompetitive inhibitor of PGK1 with an affinity of Kd= 99.08 nmol/L. DC-PGKI stabilizes PGK1in vitro and in vivo, and suppresses both glycolytic activity and the kinase function of PGK1. In addition,DC-PGKI unveils that PGK1 regulates production of IL-1β and IL-6 in LPS-stimulated macrophages.Mechanistically, inhibition of PGK1 with DC-PGKI results in NRF2(nuclear factor-erythroid factor 2-related factor 2, NFE2L2) accumulation, then NRF2 translocates to the nucleus and binds to the proximity region of Il-1β and Il-6 genes, and inhibits LPS-induced expression of these genes. DC-PGKI ameliorates colitis in the dextran sulfate sodium(DSS)-induced colitis mouse model. Based on the structural optimization design of a folded biconcave cavity using the 808 nm pulsed laser diode(LD) side-pumped ceramic Nd: YAG rod, output pulses with maximum pulse energy and peak power up to39.6 m J and 9.73 MW were obtained, corresponding to 100 Hz repetition rate and 4.07 ns pulse width. The instabilities of pulse width and pulse energy were ±1.55% and ±2.06%, respectively. At the highest repetition rate of 1 kHz, the pulse energy, pulse width, and peak power were 11.3 mJ, 5.05 ns, and 2.24 MW, respectively.
