| 1 | Involvement of carbon monoxide produced by heme oxygenase in ABA-induced stomatal closure in Vicia faba and its proposed signal transduction pathway显示文摘Carbon monoxide (CO) has recently proven to be an important bioactive or signaling molecule in mammalian cells. Its effects are mainly mediated by nitric oxide (NO) and cyclic GMP (cGMP). In Vicia faba leaves, CO production and heme oxygenase (HO) activity, an important CO synthetic enzyme, are first reported to increase in response to ABA treatment, which could result in stomatal closure. Inter- estingly, ABA-induced stomatal closure in V. faba guard cells is partially blocked when the synthetic CO inhibitor ZnPP, or the CO/NO scavenger Hb is added. Furthermore, we show that, exogenously applied CO donor, hematin, and CO aqueous solution not only result in the enhancement of CO release, but also time-dependently induce stomatal closure, and the latter is mimicked by the application of an NO donor SNP. The above-mentioned stomatal closure effects are differentially reversed by the addition of tungstate, a potent inhibitor of NO synthetic enzyme nitrate reductase (NR), the specific NO scavenger cPTIO, ZnPP, or Hb. During treatment for 4 h, SNP, 0.01% CO aqueous solution or hematin significantly triggers NO synthesis, whereas cPTIO, or tungstate approximately fully inhibits NO fluorescence. Ad- ditionally, application of the GC inhibitor ODQ blocks CO-induced stomatal closure. This inhibition could be reversed when 8-Br-cGMP is added. | 3 | Prevalence of liver injury among patients with acquired immunodeficiency syndrome treated with highly active antiretroviral therapy in China显示文摘OBJECTIVE: To estimate the prevalence of liver injury among patients with acquired immunodeficiency syndrome(AIDS) who received highly active antiretroviral therapy(HAART) in rural Henan Province in China, and to explore whether Traditional Chinese Medicine(TCM) treatment based on HAART would increase this risk.METHODS: This was a retrospective cross-sectional study. We collected medical information on patients with AIDS from two treatment databases in2014. Criteria established by the AIDS Clinical Trials Group in 1996 were used for grading liver injury,classified based on the limit of normal(ULN) for alanine transaminase and aspartate aminotransferase:grade 1(1.25-2.5 × ULN); grade 2(2.6-5 × ULN);grade 3(5.1-10 × ULN); and grade 4(> 10 × ULN).Factors associated with liver injury were evaluated using a logistic regression model.RESULTS: A total 6953 patients with AIDS(3324 male and 3629 female patients) were enrolled into this study. The prevalence of liver injury was 22.0%(18.0% grade 1, 3.1% grade 2, 0.9% grade 3). In multivariate analysis, patients aged 34-45 years were more likely to have liver injury than patients in other age groups [adjusted odds ratio(AOR), 1.39; 95%CI, 1.01-1.91)]. Other factors associated with liver injury included male sex(AOR, 1.64; 95% CI,1.46-1.85), HIV infection via blood(AOR, 1.47; 95%CI, 1.19-1.82), hepatitis B virus antibody positive(AOR, 1.07; 95% CI, 0.85-1.36), and hepatitis C virus(HCV) antibody positive(AOR, 2.76; 95% CI,2.28-3.34).CONCLUSION: The prevalence of liver injury was relatively high among HAART-experienced patients. Several factors associated with liver injury included male sex, age 35-45 years old, HIV infection through blood, and concurrent HCV infection. | 7 | Determinants of self-management behaviors among pulmonary tuberculosis patients: a path analysis显示文摘Background Tuberculosis(TB)is one of the top 10 causes of death in the world.Since Directly Observed Therapy(DOT)as a core strategy for the global TB control are not applicable to all types of TB patients,and self-management of TB patients(SMTP)as a patient-centered supervision type is a supplement to DOT and can improve TB case management.However,the factors related to SMTP are complex and need more study.This study aimed at identifying the determinants of SMTP and examining the direct/indirect effects of these determinants.Methods The purposive sampling technique was used to select study sites and participants were recruited from the study sites by the consecutive sampling method.The PRECEDE model was used as the framework to analyze the determinants of SMTP.The responses of TB patients were acquired via a questionnaire survey for data collection.A Pearson correlation analysis was used to define the relationship between the predisposing,enabling,reinforcing factors with SMTP behaviors.A regression-based path analysis was used to determine the action paths of the predisposing,enabling,and reinforcing factors on SMTP behaviors.Results The predisposing(TB knowledge),enabling[health education and healthcare workers(HCWs)support],reinforcing factors(family support)had significant positive correlations with SMTP behaviors(P<0.05).The predisposing,enabling,reinforcing factors were positively correlated with each other(r=0.123‒0.918,P<0.05),except for family support and HCWs support.The predisposing factors(TB knowledge,β=0.330)and the enabling factors(HCWs support,β=0.437)had direct effects on SMTP behaviors.The enabling factors(health education and HCWs support)and the reinforcing factors(family support)had indirect effects on SMTP behaviors.Conclusions This study revealed the effects and action path of TB knowledge,health education,HCWs support,and family support on SMTP behaviors via a path analysis.Assessing patient’s needs for SMTP along with promoting effective TB health education and providing firm support from HCWs and family members are potential strategies to promote SMTP behaviors. | Jin Li Jie Pu Jiaqing Liu Qingya Wang Rui Zhang Ting Zhang Jiani Zhou Wei Xing Shengxiang Liang Daiyu Hu Ying Li | 2021 | Infectious Diseases of Poverty2021,10,4: | 2 |

| 10 | Cytosolic TGM2 promotes malignant progression in gastric cancer by suppressing the TRIM21-mediated ubiquitination/degradation of STAT1 in a GTP binding-dependent modality显示文摘Background:Previous studies have revealed the critical role of transglutaminase 2(TGM2)as a potential therapeutic target in cancers,but the oncogenic roles and underlying mechanisms of TGM2 in gastric cancer(GC)are not fully understood.In this study,we examined the role and potential mechanism of TGM2 in GC.Methods:Western blotting,immunohistochemistry,CCK8,colony formation and transwell assays were used to measure TGM2 expression in the GC cells and tissues and to examine the in vitro role of TGM2 in GC.Xenograft and in vivo metastasis experiments were performed to examine the in vivo role of TGM2 in GC.Gene set enrichment analysis,quantitative PCR and western blotting were conducted to screen for potential TGM2 targets involved in GC.Gain/loss-offunction and rescue experiments were conducted to detect the biological roles of STAT1 in GC cells in the context of TGM2.Co-immunoprecipitation,mass spectrometry,quantitative PCR and western blotting were conducted to identify STAT1-interacting proteins and elucidate their regulatory mechanisms.Mutations in TGM2 and two molecules(ZM39923 and A23187)were used to identify the enzymatic activity of TGM2 involved in the malignant progression of GC and elucidate the underlying mechanism.Results:In this study,we demonstrated elevated TGM2 expression in the GC tissues,which closely related to pathological grade,and predicted poor survival in patients with GC.TGM2 overexpression or knockdown promoted(and inhibited)cell proliferation,migration,and invasion,which were reversed by STAT1 knockdown or overexpression.Further studies showed that TGM2 promoted GC progression by inhibiting STAT1 ubiquitination/degradation.Then,tripartite motif-containing protein 21(TRIM21)was identified as a ubiquitin E3 ligase of STAT1 in GC.TGM2 maintained STAT1 stability by facilitating the dissociation of TRIM21 and STAT1 with GTP-binding enzymatic activity.A23187 abolished the role of TGM2 in STAT1 and reversed the pro-tumor role of TGM2 in vitro and in vivo.Conclusions:This study revealed a critical role and regulatory mechanism of TGM2 on STAT1 in GC and highlighted the potential of TGM2 as a therapeutic target,which elucidates the development of medicine or strategies by regulating the GTP-binding activity of TGM2 in GC. | Lu Zhang Qingya Li Jing Yang Penghui Xu Zhe Xuan Jianghao Xu Zekuan Xu | 2023 | Cancer Communications2023,43,1: | 1 |

| 19 | A-π-A structured non-fullerene acceptors for stable organic solar cells with efficiency over 17%显示文摘With the development of photovoltaic materials, especially the small molecule acceptors(SMAs), organic solar cells(OSCs)have made breakthroughs in power conversion efficiencies(PCEs). However, the stability of high-performance OSCs remains a critical challenge for future technological applications. To tackle the inherent instability of SMA materials under the ambient conditions, much effort has been made to improve OSCs stability, including device modification and new materials design. Here we proposed a new electron acceptor design strategy and developed a “quasi-macromolecule”(QM) with an A-π-A structure,where the functionalized π-bridge is used as a linker between two SMAs(A), to improve the long-term stability without deteriorating device efficiencies. Such type of QMs enables excellent synthetic flexibility to modulate their optical/electrochemical properties, crystallization and aggregation behaviors by changing the A and π units. Moreover, QMs possess a unique long conjugated backbone combining high molecular weight over 3.5 k Da with high purity. Compared with the corresponding SMA BTP-4F-OD(Y6-OD), the devices based on newly synthesized A-π-A type acceptors QM1 and QM2 could exhibit better device stability and more promising PCEs of 17.05% and 16.36%, respectively.
