1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29

3Oroxindin inhibits macrophage NLRP3 inflammasome activation in DSS-induced ulcerative colitis in mice via suppressing TXNIP-dependent NF-κB pathway显示文摘Oroxindin is a flavonoid isolated from the traditional Chinese medicine Huang-Qin,which has shown various pharmacological activities including anti-inflammatory,antitumor,antioxidant,etc.Thus far,the effect of oroxindin on colonic inflammation and the underlying mechanism remain unknown.In this study,we investigated the tissue distribution of oroxindin and its therapeutic effects on ulcerative colitis(UC)as well as the underlying mechanisms.UC model was established in mice by administrating dextran sulfate sodium(DSS)in drinking water for 7 d.We first showed that oroxindin was largely absorbed by the colon as an active ingredient after normal mice received Huang-Qin-Tang,a traditional Chinese medicine decoction.UC mice were then treated with oroxindin(12.5,25,50 mg-kg^-1·d^-1,i.g.)for 10 d.We found that oroxindin treatment greatly suppressed massive macrophages infiltration and attenuated pathological changes in colonic tissue.Furthermore,oroxindin treatment significantly inhibited the generation of IL-13 and IL-18 in the colon via inhibiting the nucleotide-binding oligomerization domain-like receptor 3(NLRP3)inflammasome formation and activation.In cultured macrophages,LPS induced NLRp3 inflammasome formation and caspase-1 activation,which were suppressed by oroxindin(12.5-50μM).In LPS-treated macrophages,oroxindin dose-dependently restored the expression of TXNIP protein,leading to suppressing TXNIP-dependent NF-κB activation.In conclusion,these results demonstrate that oroxindin could be absorbed by the colon and attenuate inflammatory responses via inhibiting NLRP3 inflammasome formation and activation,which is related to the inhibitory effect on TXNIP-dependent NF-κB-signaling pathway.Hence,oroxindin has the potential of becoming an effective drug for treating UC.Qi Liu Rui Zuo Kai Wang Fei-fei Nong Ya-jun Fu Shao-wei Huang Zeng-feng Pan Yi Zhang Xia Luo Xiang-liang Deng Xiao-xue Zhang Lian Zhou Yang Chen 2020Acta Pharmacologica Sinica2020,41,6:15

5Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s显示文摘Ulcerative colitis(UC)is a chronic inflammatory disease of the gastrointestinal tract,which is closely related to gut barrier dysfunction.Emerging evidence shows that interleukin-22(IL-22)derived from group 3 innate lymphoid cells(ILC3s)confers benefits on intestinal barrier,and IL-22 expression is controlled by aryl hydrocarbon receptor(AhR).Previous studies show that baicalein protects the colon from inflammatory damage.In this study we elucidated the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice.Mice were administered baicalein(10,20,40 mg·kg^(-1)·d^(-1),i.g.)for 10 days;the mice freely drank 3%dextran sulfate sodium(DSS)on D1-D7 to induce colitis.We showed that baicalein administration simultaneously ameliorated gut inflammation,decreased intestinal permeability,restored tight junctions of colons possibly via promoting AhR/IL-22 pathway.Co-administration of AhR antagonist CH223191(10 mg/kg,i.p.)partially blocked the therapeutic effects of baicalein in colitis mice,whereas AhR agonist FICZ(1μg,i.p.)ameliorated symptoms and gut barrier function in colitis mice.In a murine lymphocyte line MNK-3,baicalein(5-20μM)dose-dependently increased the expression of AhR downstream target protein CYP1A1,and enhanced IL-22 production through facilitating AhR nuclear translocation,these effects were greatly diminished in shAhR-MNK3 cells,suggesting that baicalein induced IL-22 production in AhR-dependent manner.To further clarify that,we constructed an in vitro system consisting of MNK-3 and Caco-2 cells,in which MNK-3 cell supernatant treated with baicalein could decrease FITC-dextran permeability and promoted the expression of tight junction proteins ZO-1 and occluding in Caco-2 cells.In conclusion,this study demonstrates that baicalein ameliorates colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s,thus providing a potential therapy for UC.Yan-yang Li Xiao-jing Wang Yu-lin Su Qing Wang Shao-wei Huang Zeng-feng Pan Yan-ping Chen Jun-jie Liang Mei-ling Zhang Xue-qian Xie Zhi-yun Wu Jin-yan Chen Lian Zhou Xia Luo 2022Acta Pharmacologica Sinica2022,43,6:14

7Mutations in components of the Wnt signaling pathway in gastric cancer显示文摘AIM: To explore the contribution of AXIN1, AXIN2 and beta-catenin, components of Wnt signaling pathway, to the carcinogenesis of gastric cancer (GC), we examined AXIN1, AXIN2 exon7 and CTNNB1 (encoding beta- catenin) exon3 mutations in 70 GCs. METHODS: The presence of mutations was identified by polymerase chain reaction (PCR)-based denaturing high-performance liquid chromatography and direct DNA sequencing. Beta-catenin expression was detected by immunohistochemical analysis. RESULTS: Among the 70 GCs, 5 (7.1%) had mutations in one or two of these three components. A frameshift mutation (1 bp deletion) in exon7 of AXIN2 was found in one case. Four cases, including the case with a mutation in AXIN2, had frameshift mutations and missense mutations in AXIN1. Five single nucleotide polymorphisms (SNPs), 334 C>T, 874 C>T, 1396 G>A, 1690 C>T and 1942 T>G, were identified in AXIN1. A frameshift mutation (27 bp deletion) spanning exon3 of CTNNB1 was observed in one case. All four cases with mutations in AXIN1 and AXIN2 showed nuclear beta- catenin expression. 9Cardioprotective effects of ghrelin and des-octanoyl ghrelin on myocardial injury induced by isoproterenol in rats显示文摘<正> Aim:To determine the cardioprotective action of ghrelin and des-octanoyl ghrelinin rats with isoproterenol-induced myocardial injury.Methods:Rats were subcu-taneously injected with isoproterenol (ISO;20,10,and 5 mg/kg) on d 1,2 and 3,respectively,and then 3 mg/kg for the next 7d with or without ghrelin or des-octanoyl-ghrelin (100μg/kg,twice daily).Plasma ghrelin and growth hormonelevels were assayed using radioimmunoassay methods.Growth hormone secreta-gogue receptor (GHSR) and ghrelin mRNA were determined using RT-PCR.Themaximal binding capacity and the affinity for [~3H]ghrelin were determined by re-ceptor binding assays.Results:Compared with controls,ISO-treated rats showedsevere myocardial injury,cardiomegaly,infarction-like necrosis and massive fi-brosis with increases in irradiated-ghrelin (ir-ghrelin) content in plasma by 67%and myocardia by 66% and in the mRNA level in the myocardia by 93% (P<0.01).ISO-treated rats had 95% (P<0.01) higher GHSR mRNA levels in the myocardia.The maximal binding capacity of [~3H]ghrelin for myocardial sarcolemma was higherin ISO-treated rats than in controls.Ghrelin administration improved cardiac func-tion and ameliorated cardiomegaly and attenuated myocardial lipid peroxidationinjury and relieved cardiac fibrosis as compared with ISO treatment alone.Admin-istration of des-octanoyl ghrelin effectively antagonized ISO-induced myocardialinjury and improved all parameters measured.However,the therapeutic effect ofdes-octanoyl ghrelin was significantly weaker than that of ghrelin.The plasmagrowth hormone level increased markedly,by 1.5-fold (P<0.01),with ghrelinadministration as compared with that in controls,but was unaltered in the des-octanoyl ghrelin group.Conclusion:Myocardial ghrelin and GHSR were up-regulated during ISO-induced myocardial injury.The protective effect of ghrelinagainst ISO-induced cardiac function injury and fibrosis was more potent thanthat of des-octanoyl ghrelin,which suggests that ghrelin could be an endog-enous cardioprotective factor in ischemic heart disease,and that its effects in-clude growth hormone-dependent and-independent pathways.Lian LI Li-ke ZHANG Yong-zheng PANG Chun-shui PAN Yong-fen QI Li CHEN Xian WANG Chao-shu TANG Jing ZHANG 2006Acta Pharmacologica Sinica2006,27,5:9
10Micro-trichome as a class I homeodomain-leucine zipper gene regulates multicellular trichome development in Cucumis sativus显示文摘Plant trichomes serve as a highly suitable model for investigating cell differentiation at the single-cell level.The regulatory genes involved in unicellular trichome development in Arabidopsis thaliana have been intensively studied,but genes regulating multicellular trichome development in plants remain unclear.Here,we characterized Cucumis sativus(cucumber) trichomes as representative multicellular and unbranched structures,and identified Micro-trichome(Mict),using map-based cloning in an F_2 segregating population of7,936 individuals generated from a spontaneous mict mutant.In mict plants,trichomes in both leaves and fruits,are small,poorly developed,and denser than in the wild type.Sequence analysis revealed that a 2,649-bp genomic deletion,spanning the first and second exons,occurred in a plant-specific class I homeodomain-leucine zipper gene.Tissue-specific expression analysis indicated that Mict is strongly expressed in the trichome cells.Transcriptome profiling identified potential targets of Mict including putative homologs of genes known in other systems to regulate trichome development,meristem determinacy,and hormone responsiveness.Phylogenic analysis charted the relationships among putative homologs in angiosperms.Our paper represents initial steps toward understanding the development of multicellular trichomes.Jun-Long Zhao Jun-Song Pan Yuan Guan Wei-Wei Zhang Bei-Bei Bie Yun-Li Wang Huan-Le He Hong-Li Lian Run Cai 2015Journal of Integrative Plant Biology2015,57,11:9

12Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9

14Shifts in soil bacterial communities induced by the controlled-release fertilizer coatings显示文摘Coated controlled-release fertilizers(CRFs) have been widely applied in agriculture due to their increased efficiency. However, the widespread and a lot of coated CRFs application may leave undesired coating residues in the soil due to their slow degradation. Limited information is available on the effects of substantial residual coatings on the soil bacterial community. By adding 0, 5, 10, 20, and 50 times quantities of residual coating from conventional application amount of resin and water-soluble coated CRFs, we studied the responses of soil properties and bacterial community composition to these two residual coatings in black soil. The results showed that the resin and water-soluble coatings did not essentially alter the properties of black soil or cause dramatic changes to bacterial diversity within the test concentration range. The residual resin and water-soluble coatings also did not distinctly alter the relative abundance of the top ten bacteria at phylum level. Heatmap results suggested that the treatments were basically clustered into two groups by concentration rather than types of coating material. Pearson correlation analysis showed that the Simpson's diversity index of the bacterial community was significantly correlated with microbial biomass carbon(MBC, r=0.394, P<0.05), and the richness index abundance-based coverage estimator(ACE) of the bacterial community was significantly correlated with microbial biomass nitrogen(MBN, r=0.407, P<0.05). Methods Surveillance data from 1997 to 2012 were extracted from the Public Health Surveillance System of The Three Gorges established in 1997. Temporal changes in the incidences of hemorrhagic fever with renal syndrome(HFRS) and leptospirosis, rodent density, pathogen-carrying rates, and their correlations were analyzed. Results The average indoor and outdoor rodent densities decreased overall from 1997 to 2012. The average densities decreased by 47.72%(from 4.38% to 2.29%) and 39.68%(from 4.41% to 2.66%), respectively, after impoundment(2003-2012) compared with before impoundment(1997-2002). The average annual incidence rates of HFRS and leptospirosis were 0.29/100,000 and 0.52/100,000, respectively, and decreased by 85.74%(from 0.68/100,000 to 0.10/100,000) and 95.73%(from 1.47/100,000 to 0.065/100,000), respectively, after impoundment compared with before impoundment. Incidences of HFRS and leptospirosis appear to be positively correlated with rodent density in the reservoir area. Conclusion This study demonstrated that rodent density and incidences of rodent-borne diseases decreased and were maintained at low levels during construction of the Three Gorges dam. 19Photoelectrocatalytic reduction of CO_2 to methanol over a photosystem Ⅱ-enhanced Cu foam/Si-nanowire system显示文摘A solar-light double illumination photoelectrocatalytic cell(SLDIPEC) was fabricated for autonomous CO_2 reduction and O_2 evolution with the aid of photosystem II(PS-II, an efficient light-driven water-oxidized enzyme from nature) and utilized in a photoanode solution. The proposed SLPEC system was composed of Cu foam as the photoanode and p-Si nanowires(Si-NW) as the photocathode. Under solar irradiation, it exhibited a super-photoelectrocatalytic performance for CO_2 conversion to methanol, with a high evolution rate(41.94 mmol/hr), owing to fast electron transfer from PS-II to Cu foam.Electrons were subsequently trapped by Si-NW through an external circuit via bias voltage(0.5 V), and a suitable conduction band potential of Si(-0.6 e V) allowed CO_2 to be easily reduced to CH_3 OH at the photocathode. The constructed Z-scheme between Cu foam and Si-NW can allow the SLDIPEC system to reduce CO_2(8.03 mmol/hr) in the absence of bias voltage.
