| 1 | USP29 maintains the stability of cGAS and promotes cellular antiviral responses and autoimmunity显示文摘Cyclic GMP-AMP synthase(cGAS)is an essential sensor of cytosolic DNA and critically mediates innate immune responses and autoimmunity.Modulating the activity and stability of cGAS provides potential strategies for treating viral or autoimmune diseases.Here,we report that ubiquitin-specific protease 29(USP29)deubiquitinates and stabilizes cGAS and promotes cellular antiviral responses and autoimmunity.Knockdown or knockout of USP29 severely impairs Herpes simplex virus 1(HSV-1)-or cytosolic DNA-induced expression of type I interferons(IFNs)and proinflammatory cytokines.Consistently,Usp29m/m mice produce decreased type I IFNs and proinflammatory cytokines after HSV-1 infection and are hypersensitive to HSV-1 infection compared to the wild-type littermates.In addition,genetic ablation of USP29 in Trex1−/−mice eliminated the detectable pathological and molecular autoimmune phenotypes.Mechanistically,USP29 constitutively interacts with cGAS,deconjugates K48-linked polyubiquitin chains from cGAS and stabilizes cGAS in uninfected cells or after HSV-1 infection.Reconstitution of cGAS into Usp29−/−cells fully rescues type I IFN induction and cellular antiviral responses after HSV-1 infection.Our findings thus reveal a critical role of USP29 in the innate antiviral responses against DNA viruses and autoimmune diseases and provide insight into the regulation of cGAS. | Qiang Zhang Zhen Tang Ran An Liya Ye Bo Zhong | 2020 | Cell Research2020,30,10: | 5 |
| 2 | Modulation of Beta Oscillations for Implicit Motor Timing in Primate Sensorimotor Cortex during Movement Preparation显示文摘Motor timing is an important part of sensorimotor control. Previous studies have shown that beta oscillations embody the process of temporal perception in explicit timing tasks. In contrast, studies focusing on beta oscillations in implicit timing tasks are lacking. In this study, we set up an implicit motor timing task and found a modulation pattern of beta oscillations with temporal perception during movement preparation. We trained two macaques in a repetitive visually-guided reach-to-grasp task with different holding intervals. Spikes and local field potentials were recorded from microelectrode arrays in the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. We analyzed the association between beta oscillations and temporal interval in fixedduration experiments(500 ms as the Short Group and1500 ms as the Long Group) and random-duration experiments(500 ms to 1500 ms). The results showed that the peak beta frequencies in both experiments ranged from15 Hz to 25 Hz. The beta power was higher during the hold period than the movement(reach and grasp) period.Further, in the fixed-duration experiments, the mean poweras well as the maximum rate of change of beta power in the first 300 ms were higher in the Short Group than in the Long Group when aligned with the Center Hit event. In contrast, in the random-duration experiments, the corresponding values showed no statistical differences among groups. The peak latency of beta power was shorter in the Short Group than in the Long Group in the fixed-duration experiments, while no consistent modulation pattern was found in the random-duration experiments. These results indicate that beta oscillations can modulate with temporal interval in their power mode. | 11 | Single-cell analysis reveals an Angpt4-initiated EPDC-EC-CM cellular coordination cascade during heart regeneration显示文摘Mammals exhibit limited heart regeneration ability,which can lead to heart failure after myocardial infarction.In contrast,zebrafish exhibit remarkable cardiac regeneration capacity.Several cell types and signaling pathways have been reported to participate in this process.However,a comprehensive analysis of how different cells and signals interact and coordinate to regulate cardiac regeneration is unavailable.We collected major cardiac cell types from zebrafish and performed high-precision single-cell transcriptome analyses during both development and post-injury regeneration.We revealed the cellular heterogeneity as well as the molecular progress of cardiomyocytes during these processes,and identified a subtype of atrial cardiomyocyte exhibiting a stem-like state which may transdifferentiate into ventricular cardiomyocytes during regeneration.Furthermore,we identified a regeneration-induced cell(RIC)population in the epicardium-derived cells(EPDC),and demonstrated Angiopoietin 4(Angpt4)as a specific regulator of heart regeneration.angpt4 expression is specifically and transiently activated in RIC,which initiates a signaling cascade from EPDC to endocardium through the Tie2-MAPK pathway,and further induces activation of cathepsin K in cardiomyocytes through RA signaling.Loss of angpt4 leads to defects in scar tissue resolution and cardiomyocyte proliferation,while overexpression of angpt4 accelerates regeneration.Furthermore,we found that ANGPT4 could enhance proliferation of neonatal rat cardiomyocytes,and promote cardiac repair in mice after myocardial infarction,indicating that the function of Angpt4 is conserved in mammals.Our study provides a mechanistic understanding of heart regeneration at single-cell precision,identifies Angpt4 as a key regulator of cardiomyocyte proliferation and regeneration,and offers a novel therapeutic target for improved recovery after human heart injuries. | Zekai Wu Yuan Shi Yueli Cui Xin Xing Liya Zhang Da Liu Yutian Zhang Ji Dong Li Jin Meijun Pang Rui-Ping Xiao Zuoyan Zhu Jing-Wei Xiong Xiangjun Tong Yan Zhang Shiqiang Wang Fuchou Tang Bo Zhang | 2023 | Protein & Cell2023,14,5: | 0 |
| 12 | A novel and effective hyperthermia method for Schistosomiasis japonica prevention and treatment显示文摘The emerging resistance to schistosoma has been paid much attention and is in urgent need for a novel strategy to control the prevalent parasitic zoonosis. In this study, the efficiency of hyperthermia therapy was investigated by the skin hyperthermia device. The survival rate of cercariae decreased from 68.15 %(37 °C, 5 min) to 0(49 °C, 10 min) with the thermal dosages increased, which proved the preventing effect of hyperthermia therapy(P \ 0.05). Therapeutic effects were assessed in Schistosoma japonicum-infected BALB/c mice. When the cercarial contact region of the skin was treated at 45–49 °C for5 min within 8 h of infection, worm reduction rate(WRR)reached 74 %–83 %(P \ 0.01). The sensitivity of adultschistosoma to heat was also investigated using microwave intraperitoneal hyperthermia(thermal dosages 42–43 °C,20 min). The WRR, hepatic shift rates and egg reduction rates were 23.7 %, 40 % and 30 %, respectively, comparing with 80.2 %, 59.6 % and 53.9 % of praziquantel(PZQ)-treated group. Encouraging results have been obtained that hyperthermia can effectively kill schistosomula, especially with the appearance of cercarial dermatitis, while PZQ lacks efficacy against the cercariae.
