| 1 | Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable. | Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun | 2020 | Cellular & Molecular Immunology2020,17,6: | 13 |
| 2 | Mitigation of cross-eye jamming using a dual-polarization array显示文摘This paper presents an approach for mitigating the cross-eye jamming using a dual-polarization array. By transmitting a sum beam and a difference beam in two orthogonal polarimetric channels, a synthesized transmitted beam with spatially varying polarization is produced, such that the polarization of the transmitted radar wave varies in azimuth or elevation. Thus, the phases of the signals received on the two antennas of a cross-eye jammer become unequal, and an additional phase difference is introduced to disrupt the 180? phase shifting in the retrodirective loop of the jammer. By means of beam scanning in a small angular range,the optimal beam steering configuration can be found to maximize the phase error for the mitigation of cross-eye jamming. As a result, the jamming performance of the cross-eye jammer degrades largely. | 3 | Full time-domain nonlinear coupled dynamic analysis of a truss spar and its mooring/riser system in irregular wave显示文摘A new full time-domain nonlinear coupled method has been established and then applied to predict the responses of a Truss Spar in irregular wave.For the coupled analysis,a second-order time-domain approach is developed to calculate the wave forces,and a finite element model based on rod theory is established in three dimensions in a global coordinate system.In numerical implementation,the higher-order boundary element method(HOBEM)is employed to solve the velocity potential,and the 4th-order Adams-Bashforth-Moultn scheme is used to update the second-order wave surface.In deriving convergent solutions,the hull displacements and mooring tensions are kept consistent at the fairlead and the motion equations of platform and mooring-lines/risers are solved simultaneously using Newmark-integration scheme including Newton-Raphson iteration.Both the coupled quasi-static analysis and the coupled dynamic analysis are performed.The numerical simulation results are also compared with the model test results,and they coincide very well as a whole.The slow-drift responses can be clearly observed in the time histories of displacements and mooring tensions.Some important characteristics of the coupled responses are concluded. | YANG MinDong TENG Bin XIAO LongFei NING DeZhi SHI ZhongMin QU Yan | 2014 | Science China(Physics,Mechanics & Astronomy)2014,57,1: | 8 |
| 4 | Polarization discrimination between repeater false-target and radar target显示文摘High fidelity repeater false-target badly affects a radar system's detecting, tracking, and data processing. It is an available approach of confronting false-target for radar that discriminates firstly and then eliminates. Whereas for the technique progress about the repeater false-target jam, it is more and more difficult to discriminate this jam in the time-domain, frequency-domain, or space-domain. The technique using polarization information to discriminate the target and false-target is discussed in this paper. With the difference that false-target signal vector's polarization ratio is fixed and target echo signal vector's polarization ratio is variational along with radar transmission signal's polarization, we transform the discrimination problem to beeline distinguish problem in the 2-dim complex space. The distributing characteristic expression of the false-target discrimination statistic is constructed, with which the discrimination ratio of false-target is analyzed. For the target case, the decomposed model of target scattering matrix and the concept of distinguish quantity are proposed. Then, the discrimination ratio of target can be forecasted according to target distinguish quantity. Thus, the performance of discrimination method has been analyzed integrally. | 7 | Purification and molecular weight distribution of a key exopolysaccharide component of Bacillus megaterium TF10显示文摘Extracellular polymeric substances(EPS) are organic metabolic compounds excreted by microorganisms. They largely impact microbial aggregate structures and functions.Extracellular polysaccharides(EP) in EPS are responsible for the formation of microbial aggregates. In this work, we successfully separated and characterized EP from EPS of the bacterium Bacillus megaterium TF10. Extraction of EP from EPS was optimized using Sevag's reagent. Chemical characteristics, functional groups, and molecular weight(MW) distribution of EP were compared with the harvested EPS and soluble microbial products(SMP). We found that the polymers of lower MW and free proteins were successfully removed by Sevag's reagent. The higher MW components of EPS were predominantly polysaccharides,while the polymers of lower MW tended to secrete to the supernatant and were described as SMP. A part of the proteins in the EP was polysaccharide-bonded. | 10 | Single-cell profiling reveals Müller glia coordinate retinal intercellular communication during light/dark adaptation via thyroid hormone signaling显示文摘Light adaptation enables the vertebrate visual system to operate over a wide range of ambient illumination.Regulation of phototransduction in photoreceptors is considered a major mechanism underlying light adaptation.However,various types of neurons and glial cells exist in the retina,and whether and how all retinal cells interact to adapt to light/dark conditions at the cellular and molecular levels requires systematic investigation.Therefore,we utilized single-cell RNA sequencing to dissect retinal cell-type-specific transcriptomes during light/dark adaptation in mice.The results demonstrated that,in addition to photoreceptors,other retinal cell types also showed dynamic molecular changes and specifically enriched signaling pathways under light/dark adaptation.Importantly,Müller glial cells(MGs)were identified as hub cells for intercellular interactions,displaying complex cell‒cell communication with other retinal cells.Furthermore,light increased the transcription of the deiodinase Dio2 in MGs,which converted thyroxine(T4)to active triiodothyronine(T3).Subsequently,light increased T3 levels and regulated mitochondrial respiration in retinal cells in response to light conditions.As cones specifically express the thyroid hormone receptor Thrb,they responded to the increase in T3 by adjusting light responsiveness.Loss of the expression of Dio2 specifically in MGs decreased the light responsive ability of cones.These results suggest that retinal cells display global transcriptional changes under light/dark adaptation and that MGs coordinate intercellular communication during light/dark adaptation via thyroid hormone signaling. | Min Wei Yanping Sun Shouzhen Li Yunuo Chen Longfei Li Minghao Fang Ronghua Shi Dali Tong Jutao Chen Yuqian Ma Kun Qu Mei Zhang Tian Xue | 2023 | Protein & Cell2023,14,8: | 0 |
