1Recently duplicated sesterterpene(C25) gene clusters in Arabidopsis thaliana modulate root microbiota显示文摘Land plants co-speciate with a diversity of continually expanding plant specialized metabolites(PSMs) and root microbial communities(microbiota).Homeostatic interactions between plants and root microbiota are essential for plant survival in natural environments.A growing appreciation of microbiota for plant health is fuelling rapid advances in genetic mechanisms of controlling microbiota by host plants.PSMs have long been proposed to mediate plant and single microbe interactions.However,the effects of PSMs,especially those evolutionarily new PSMs,on root microbiota at community level remain to be elucidated.Here,we discovered sesterterpenes in Arabidopsis thaliana,produced by recently duplicated prenyltransferase-terpene synthase(PT-TPS) gene clusters,with neo-functionalization.A single-residue substitution played a critical role in the acquisition of sesterterpene synthase(sesterTPS) activity in Brassicaceae plants.Moreover,we found that the absence of two root-specific sesterterpenoids,with similar chemical structure,significantly affected root microbiota assembly in similar patterns.Our results not only demonstrate the sensitivity of plant microbiota to PSMs but also establish a complete framework of host plants to control root microbiota composition through evolutionarily dynamic PSMs.Qingwen Chen Ting Jiang Yong-Xin Liu Haili Liu Tao Zhao Zhixi Liu Xiangchao Gan Asis Hallab Xuemei Wang Juan He Yihua Ma Fengxia Zhang Tao Jin M. 3Influence of aerodynamic configuration of a streamline box girder on bridge flutter and vortex-induced vibration显示文摘Streamline box girders are widely applied in the design and construction of long-span bridges all over the world.In order to study the influence of modifications of aerodynamic configuration and accessory components on flutter and vortex-induced vibration(VIV),more than 60 cases were tested through a 1?50 scale section model.The test results indicates that the aerodynamic configuration and accessory components of streamline box girders can signifi-cantly affect the wind-induced vibration of bridge,which is in good agreement with the experience of past researchers.From the tests carried out,it is observed that if the horizontal angle of the inclined web of the streamline box girder is below 16°,the critical flutter wind speed of bridge will increase remarkably,and the VIV will diminish.The test results also show that the 15° inclined web can restrain the formation of vortex near the tail,and consequently improve the performance of aerodynamic stability of long-span bridges.Finally,a new streamline box girder with 15° inclined web was presented and strongly recommended in the aerodynamic configuration design of long-span bridges.Qi WANG Haili LIAO Mingshui LI Cunming MA 2011Journal of Modern Transportation2011,19,4:7
44-Formyl-2-(2H-benzotriazol-2-yl)-phenol: an ESIPT chromophore显示文摘In the present work, excited state intramolecular proton transfer(ESIPT) emission properties of a new benzotriazole derivative 4-formyl-2-(2H-benzotriazol-2-yl)-phenol(C1) were studied. 4-Formyl-2-(2H-benzotriazol-2-yl)-methoxy-benzene(C2), 4-formyl-2-(2H-benzotriazol-2-yl)-phenyl acetate(C3) and 4-methyl-2-(2H-benzotriazol-2-yl)-phenol(C4) were used as the reference molecules.1H NMR chemical shift of hydroxy group in C1 was located at more down field than that of C4 or p-hydroxy-benzaldehyde(C5), respectively. C1 showed two absorption bands in the range of 260–400 nm zones in various solvents, while C2 and C3 exhibited single absorption band. The equal molar mixtures of C2/C5 or C3/C5 showed single absorption band.C2 and C3 displayed single fluorescence emission band in various solvents, while C1 exhibited dual emission bands in some strong polar solvents. Furthermore, the second emission band in these strong polar solvents showed the large Stokes shift. The results show that the second emission band of C1 was produced by ESIPT. C2 and C3 could not undergo ESIPT due to no hydroxy group. 51.06 μm high-power InGaAs/GaAsP quantum well lasers显示文摘The high power and low internal loss 1.06 μm InGaAs/GaAsP quantum well lasers with asymmetric waveguide structure were designed and fabricated. For a 4000 μm cavity length and 100 μm stripe width device,the maximum output power and conversion efficiency of the device are 7.13 W and 56.4%, respectively. 12The molecular tumor burden index as a response evaluation criterion in breast cancer显示文摘Circulating tumor DNA(ctDNA)is a potential biomarker of prognosis and therapeutic response.We conducted this study to explore the role of the molecular tumor burden index(mTBI)in ctDNA as a therapeutic response and prognostic biomarker in a larger cohort prospective phase III randomized multicenter study.We collected 291 plasma samples from 125 metastatic breast cancer patients from the CAMELLIA study(NCT01917279).Target-capture deep sequencing of 1021 genes was performed to detect somatic variants in ctDNA from the plasma samples.The pretreatment mTBI value was correlated with tumor burden(P=0.025).Patients with high-level pretreatment mTBI had shorter overall survival than patients with low-level pretreatment mTBI,and the median overall survival was 40.9 months and 68.4 months,respectively(P=0.011).Patients with mTBI decrease to less than 0.02%at the first tumor evaluation had longer progression-free survival and overall survival(P<0.001 and P=0.007,respectively).The mTBI has good sensitivity to identify complete response/partial response and progressive disease based on computed tomography scans(88.5%and 87.5%,respectively).The patients classified as molecular responders had longer progression-free survival and overall survival than the nonmolecular responders in the overall cohort(P<0.001 and P=0.036,respectively),as well as in the cohort in which computed tomography scans were defined as representing stable disease(P=0.027 and P=0.015,respectively).The mTBI in ctDNA detected in liquid biopsies is a potential biomarker of therapeutic response and prognosis in patients with metastatic breast cancer.Zongbi Yi Fei Ma Guohua Rong Binliang Liu Yanfang Guan Jin Li Xiaoying Sun Wenna Wang Xiuwen Guan Hongnan Mo Jiani Wang Haili Qian Binghe Xu 2021Signal Transduction and Targeted Therapy2021,6,8:1

16Clinical value of next-generation sequencing in guiding decisions regarding endocrine therapy for advanced HR-positive/HER-2-negative breast cancer显示文摘Objective: The mechanism of acquired gene mutation plays a major role in resistance to endocrine therapy in hormone receptor(HR)-positive advanced breast cancer. Circulating tumor DNA(ctDNA) has been allowed for the assessment of the genomic profiles of patients with advanced cancer. We performed this study to search for molecular markers of endocrine therapy efficacy and to explore the clinical value of ctDNA to guide precise endocrine therapy for HR-positive/human epidermal growth factor receptor-2(HER-2)-negative metastatic breast cancer patients.Methods: In this open-label, multicohort, prospective study, patients were assigned to four parallel cohorts and matched according to mutations identified in ctDNA: 1) activation of the phosphatidylinositol-3-kinase(PI3K)/AKT/mammalian target of rapamycin(mTOR) signaling pathway preferred mTOR inhibitor combined with endocrine therapy;2) estrogen receptor 1(ESR1) mutation preferred fulvestrant;3) HER-2 mutations preferred pyrotinib;and 4) no actionable mutations received treatment according to the clinical situation. In all cohorts, patients were divided into compliance group and violation group. The primary outcome measure was progression-free survival(PFS), and the secondary outcome measure was overall survival(OS).Results: In all cohorts, the combined median PFS was 4.9 months, and median PFS for the compliance and violation groups was 6.0 and 3.0 months, respectively [P=0.022, hazard ratio(HR)=0.57]. Multivariate Cox regression model showed the risk of disease progression was lower in compliance group than in violation group(P=0.023, HR=0.55). Among the patients with HER-2 mutations, the median PFS was 11.1 months in the compliance group and 2.2 months in the violation group(P=0.011, HR=0.20). There was no significant difference in the median PFS between patients who did and did not comply with the treatment protocol in patients with activation of the PI3K/AKT/mTOR or ESR1 mutation.Conclusions: The results suggest that ctDNA may help to guide the optimal endocrine therapy strategy for metastatic breast cancer patients and to achieve a better PFS. 17A chromosome-level genome assembly reveals that tandem-duplicated CYP706V oxidase genes control oridonin biosynthesis in the shoot apex of Isodon rubescens显示文摘The ent-kaurenoids(e.g.,oridonin and enmein)from the Isodon genus(Lamiaceae)are one class of diterpenoids with rich structural diversity and intriguing pharmaceutical activity.In contrast to the well-established gibberellin pathway,oxidative modifications diversifying the ent-kaurene skeleton in Isodon have remained undetermined for half a century.Here we report a chromosome-level genome assembly of I.rubescens,a well-recognized oridonin producer long favored by Asian people as a traditional herb with antitumor effects.The shoot apex was confirmed to be the actual region actively producing ent-kaurene diterpenoids.Through comparative genomics and phylogenetic analyses,we discovered a cluster of tandem-duplicated CYP706V oxygenase-encoding genes located on an ancient genomic block widely distributed in eudicots,whereas almost exclusively emerged in Isodon plants.In the shoot apex,IrCYP706V2 and IrCYP706V7 oxidized the ent-kaurene core in the initial stage of oridonin biosynthesis.Loss of CYP706Vs in other Lamiaceae plants offered an explanation for the specific kaurenoid production in Isodon plants.Moreover,we found that the Isodon genomes encode multiple diterpenoid synthases that are potentially involved in generating diterpenoid diversity.These findings provided new insights into the evolution of the lineage-specific diterpenoid pathway and laid a foundation for improving production of bioactive ent-kaurene-type diterpenoids by molecular breeding and synthetic biology approaches.Yuwei Sun Jie Shao Haili Liu Hua Wang Guangyi Wang Jianhua Li Yaping Mao Zhuo Chen Ke Ma Lin Xu Yong Wang 2023Molecular Plant2023,16,3:0
18Molecular detection of porcine circovirus type 3 in Shanxi Province,China显示文摘Porcine circovirus type 3(PCV3),which was first detected in the United States of America in 2015,is a potential threat to the swine industry.However,the prevalence of PCV3 in Shanxi Province,China,is unclear.In this research,the prevalence and genetic diversity of PCV3 were investigated in above area.Lung tissue samples(n=491)from 19 pig slaughterhouses across 11 cities throughout Shanxi Province were analyzed for PCV3 infection by PCR in 2019.The results showed that PCV3 positive rates in slaughterhouses and individuals were 100%(19/19)and 86.76%(426/491),respectively.PCV2 and PCV3 double-positive rates in slaughterhouses and individuals were 100%(19/19)and 59.27%(291/491),respectively.PCR positive samples were further sequenced and 8 PCV3 isolates were identified.The nucleotide homology of these isolates with other PCV3 isolates in NCBI database was 97.45-99.90%.A phylogenetic analysis,based on the complete genomic sequence and ORF2,divided these PCV3 strains into 2 major groups.Based on A24A/and R27/K amino acid mutations of capsid protein,the 8 identified PCV3 strains were separated to 2 clades.This was the first detailed investigation into the epidemiology of PCV3 in Shanxi Province.Our findings enabled us to assess the possibility of widespread transmission from this region.Thus,current findings establish a basis for further studies of genetic variations in PCV3 strains circulating in China.Weidong Yue Yihui Liu Xinrong Zhang Haili Ma Junping He 2021Animal Diseases2021,1,2:0
19Molecular Basis for Sesterterpene Diversity Produced by Plant Terpene Synthases显示文摘Class I terpene synthase(TPS)generates bioactive terpenoids with diverse backbones.Sesterterpene synthase(sester-TPS,C25),a branch of class I TPSs,was recently identified in Brassicaceae.However,the catalytic mechanisms of sester-TPSs are not fully understood.Here,we first identified three nonclustered functional sester-TPSs(AtTPS06,AtTPS22,and AtTPS29)in Arabidopsis thaliana.AtTPS06 utilizes a type-B cyclization mechanism,whereas most other sester-TPSs produce various sesterterpene backbones via a type-A cyclization mechanism.We then determined the crystal structure of the AtTPS18–FSPP complex to explore the cyclization mechanism of plant sester-TPSs.We used structural comparisons and site-directed mutagenesis to further elucidate the mechanism:(1)mainly due to the outward shift of helix G,plant sester-TPSs have a larger catalytic pocket than do mono-,sesqui-,and di-TPSs to accommodate GFPP;(2)type-A sester-TPSs have more aromatic residues(five or six)in their catalytic pocket than classic TPSs(two or three),which also determines whether the type-A or type-B cyclization mechanism is active;and(3)the other residues responsible for product fidelity are determined by interconversion of AtTPS18 and its close homologs.Altogether,this study improves our understanding of the catalytic mechanism of plant sester-TPS,which ultimately enables the rational engineering of sesterterpenoids for future applications.Qingwen Chen Jianxu Li Zhixi Liu Takaaki Mitsuhashi Yuting Zhang Haili Liu Yihua Ma Juan He Tetsuro Shinada Tsutomu Sato Yong Wang Hongwei Liu Ikuro Abe Peng Zhang Guodong Wang 2020Plant Communications2020,1,5:0
