| 1 | Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. | 4 | Toxicity of Multi-Walled Carbon Nanotubes,Graphene Oxide,and Reduced Graphene Oxide to Zebrafish Embryos显示文摘Objective This study was aimed to investigate the toxic effects of 3 nanomaterials, i.e. multi-walled carbon nanotubes(MWCNTs), graphene oxide(GO), and reduced graphene oxide(RGO), on zebrafish embryos. Methods The 2-h post-fertilization(hpf) zebrafish embryos were exposed to MWCNTs, GO, and RGO at different concentrations(1, 5, 10, 50, 100 mg/L) for 96 h. Afterwards, the effects of the 3 nanomateria on spontaneous movement, heart rate, hatching rate, length of larvae, mortality, and malformations ls were evaluated. Results Statistical analysis indicated that RGO significantly inhibited the hatching of zebrafish embryos. Furthermore, RGO and MWCNTs decreased the length of the hatched larvae at 96 hpf. No obvious morphological malformation or mortality was observed in the zebrafish embryos after exposure to the three nanomaterials. Conclusion MWCNTs, GO, and RGO were all toxic to zebrafish embryos to influence embryos hatching and larvae length. | 8 | A New Method for Blood NT-proBNP Determination Based on a Near-infrared Point of Care Testing Device with High Sensitivity and Wide Scope显示文摘Objective To develop a rapid, highly sensitive, and quantitative method for the detection of NT-proBNP levels based on a near-infrared point-of-care diagnostic(POCT) device with wide scope. Methods The lateral flow assay(LFA) strip of NT-proBNP was first prepared to achieve rapid detection. Then, the antibody pairs for NT-proBNP were screened and labeled with the near-infrared fluorescent dye Dylight-800. The capture antibody was fixed on a nitrocellulose membrane by a scribing device. Serial dilutions of serum samples were prepared using NT-proBNP-free serum series. The prepared test strips, combined with a near-infrared POCT device, were validated by known concentrations of clinical samples. The POCT device gave the output of the ratio of the intensity of the fluorescence signal of the detection line to that of the quality control line. The relationship between the ratio value and the concentration of the specimen was plotted as a work curve. The results of 62 clinical specimens obtained from our method were compared in parallel with those obtained from the Roche E411 kit. Results Based on the log-log plot, the new method demonstrated that there was a good linear relationship between the ratio value and NT-proBNP concentrations ranging from 20 pg/mL to 10 ng/mL. The results of the 62 clinical specimens measured by our method showed a good linear correlation with those measured by the Roche E411 kit. | 9 | Evidence-based intervention on postoperative fear,compliance,and self-efficacy in elderly patients with hip fracture显示文摘BACKGROUND Elderly patients tend to have poor self-efficacy and poor confidence in postoperative rehabilitation for hip fractures,and are prone to negative emotions,which affect treatment compliance.AIM To evaluate the effects of evidence-based intervention on postoperative fear,compliance,and self-efficacy in elderly patients with hip fractures.METHODS A total of 120 patients with hip fracture surgically treated from June 2018 to June 2020 at the orthopedic department of our hospital were selected and divided into intervention and routine groups(n=60 each)according to different nursing methods.The basic rehabilitation methods of the two groups were consistent,but patients in the intervention group received evidence-based nursing interventions at the same time.Differences between groups in the scores of motion phobia,pain fear,rehabilitation training compliance,self-efficacy,nursing satisfaction,and hip joint function were compared before and after the intervention.RESULTS Before the intervention,there were no statistically significant differences in motion phobia and pain fear scores between the groups(all P>0.05).However,motion phobia scores at 1 wk after intervention initiation(P<0.05),and pain fear scores at 1 wk and 2 wk after intervention initiation(all P<0.05),were significantly lower in the intervention group than in the routine group.On the first day of intervention,there was no significant difference in rehabilitation treatment compliance between the groups(P>0.05);however,at 2 wk after intervention initiation,rehabilitation compliance was significantly better in the intervention group than in the routine group(P<0.05).Before the intervention,there were no statistically significant differences in the scores for the two self-efficacy dimensions(overcoming difficulties and rehabilitation exercise self-efficacy)and the total self-efficacy score between the groups(all P>0.05).After 2 wk of intervention,the scores for these two dimensions of self-efficacy and the total self-efficacy score were significantly higher in the intervention group than in the routine group(all P<0.05).At 3 and 6 mo after surgery,hip function as evaluated by the Harris hip score,was significantly better in the intervention group than in the routine group(P<0.05).Additionally,overall nursing satisfaction was significantly higher in the intervention group than in the routine group(P<0.05).CONCLUSION Evidence-based nursing intervention can alleviate fear of postoperative rehabilitation in elderly patients who underwent hip fracture surgery,and improve rehabilitation treatment compliance and patient self-efficacy,which promote hip function recovery. | Ying Fu Li-Juan Zhu Da-Cheng Li Jing-Lei Yan Hai-Ting Zhang Yu-Hong Xuan Chun-Ling Meng Yan-Hong Sun | 2022 | World Journal of Clinical Cases2022,10,10: | 4 |

| 15 | Sod gene of Curvularia lunata is associated with the virulence in maize leaf显示文摘Curvularia leaf spot,caused mainly by Curvularia lunata,is a widespread plant disease in China. In the recent years,directional host selection by the pathogen,which likely results in the virulence differentiation in pathogens,is widely reported. Among the hallmarks potentially associated to pathogen variation in virulence,superoxide dismutase gene Sod has been found to be closely related to the enhancement of virulence. In the present study,the full-length of Sod was obtained via Blastn alignment against Gen Bank and the whole genome of C. lunata. In order to understand the role of Sod in the virulence variation in C. lunata,targeted gene disruption was performed to construct Sod mutants. The cell wall degrading enzyme(CWDE)activities and toxin production of ΔSod were not distinctly different from wild-type strain CX-3 and its complon. However,at an early stage of infection,ΔSod virulence appeared to be lower than CX-3 and the complon,while at a later stage,its virulence gradually returned to the level of CX-3 and the complon. Furthermore,the melanin production of ΔSod was significantly reduced compared to CX-3 and the complon,suggesting that Sod gene influences the virulence by regulating melanin production at an early stage of infection but is not essential for pathogenicity. However,the disruption of Sod did not significantly affect the transcriptional expression of the melanin biosynthesis-associated genes,brn1 and scd. The competitive adsorption mechanisms between thiophene and 1-hexene were studied by in-situ Fourier transform infrared spectroscopy(in-situ FTIR) and temperature-programmed desorption(TPD). CsY adsorbents exhibited high selectivity for thiophene even when a large amount of olefins exist.In-situ FTIR spectra of thiophene and 1-hexene adsorption indicated that both thiophene and 1-hexene were mainly adsorbed on CsY via π-complexation.
