1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Garcinol inhibits esophageal cancer metastasis by suppressing the p300 and TGF-β1 signaling pathways显示文摘Metastasis causes the main lethality in esophageal cancer patient.Garcinol,a natural compound extracted from Gambogic genera,is a histone acetyltransferase(HAT)inhibitor that has shown anticancer activities such as cell cycle arrest and apoptosis induction.In this study,we investigated the effects of garcinol on the metastasis of esophageal cancer in vitro and in vivo.We found that garcinol(5-15μM)dose-dependently inhibited the migration and invasion of human esophageal cancer cell lines KYSE150 and KYSE450 in wound healing,transwell migration,and Matrigel invasion assays.Furthermore,garcinol treatment dose-dependently decreased the protein levels of p300/CBP(transcriptional cofactors and HATs)and p-Smad2/3 expression in the nucleus,thus impeding tumor cell proliferation and metastasis.Knockdown of p300 could inhibit cell metastasis,but CBP knockdown did not affect the cell mobility.It has been reported that TGF-β1 stimulated the phosphorylation of Smad2/3,which directly interact with p300/CBP in the nucleus,and upregulating HAT activity of p300.We showed that garcinol treatment dose-dependently suppressed TGF-β1-activated Smad and non-Smad pathway,inhibiting esophageal cancer cell metastasis.In a tail vein injection pulmonary metastasis mouse model,intraperitoneal administration of garcinol(20 mg/kg)or 5-FU(20 mg/kg)significantly decreased the number of lung tumor nodules and the expression levels of Ki-67,p300,and p-Smad2/3 in lung tissues.In conclusion,our study demonstrates that garcinol inhibits esophageal cancer metastasis in vitro and in vivo,which might be related to the suppression of p300 and TGF-β1 signaling pathways,suggesting the therapeutic potential of Garcinol for metastatic tumors.Jing Wang Man Wu Dan Zheng Hong Zhang Yue Lv Li Zhang Hong-sheng Tan Hua Zhou Yuan-zhi Lao Hong-xi Xu 2020Acta Pharmacologica Sinica2020,41,1:12

5A Crucial Role of GA-Regulated Flavonol Biosynthesis in Root Growth of Arabidopsis显示文摘Flavonols have been demonstrated to play many important roles in plant growth, development, and communication with other organisms. Flavonol biosynthesis is spatiotemporally regulated by the subgroup 7 R2R3-MYB (SG7 MYB) transcription factors including MYB11/MYB12/MYB111. However, whether SG7- MYB activity is subject to post-translational regulation remains unclear. Here, we show that gibberellic acid (GA) inhibits flavonol biosynthesis via DELLA proteins in Arabidopsis. Protein-protein interaction analyses revealed that DELLAs (RGA and GAI) interacted with SG7 MYBs (MYB12 and MYB111) both in vitro and in vivo, leading to enhanced affinity of MYB binding to the promoter regions of key genes for flavonol biosynthesis and thus increasing their transcriptional levels. We observed that the level of auxin in the root tip was negatively correlated with root flavonol content. Furthermore, genetic assays showed that loss-of-function mutations in MYB 12, which is predominantly expressed in roots, partially rescued the short-root phenotype of the GA-deficient mutant ga1-3 by increasing root meristem size and mature cell size. Consistent with these observations, exogenous application of the flavonol quercetin restored the root meristem size of myb12 ga1-3 to that of ga1-3. 6Garciesculenxanthone B induces PINK1-Parkin-mediated mitophagy and prevents ischemia-reperfusion brain injury in mice显示文摘Mitophagy is a selective form of autophagy involving the removal of damaged mitochondria via the autophagy-lysosome pathway.PINK1-Parkin-mediated mitophagy is one of the most important mechanisms in cardiovascular disease,cerebral ischemia-reperfusion(I/R)injury,and neurodegenerative diseases.In this study we conducted an image-based screening in YFP-Parkin HeLa cells to discover new mitophagy regulators from natural xanthone compounds.We found that garciesculenxanthone B(GeB),a new xanthone compound from Garcinia esculenta,induced the formation of YFP-Parkin puncta,a well known mitophagy marker.Furthermore,treatment with GeB dose-dependently promoted the degradation of mitochondrial proteins Tom20,Tim23,and MFN1 in YFP-Parkin HeLa cells and SH-SY5Y cells.We revealed that GeB stabilized PINK1 and triggered Parkin translocation to the impaired mitochondria to induce mitophagy,and these effects were abolished by knockdown of PINK1.Finally,in vivo experiments demonstrated that GeB partially rescued ischemia-reperfusion-induced brain injury in mice.Taken together,our findings demonstrate that the natural compound GeB can promote the PINK1-Parkin-mediated mitophagy pathway,which may be implicated in protection against I/R brain injury.Man Wu Guang Lu Yuan-zhi Lao Hong Zhang Dan Zheng Zhao-qing Zheng Juan Yi Qian Xiang Li-ming Wang Hong-sheng Tan Hua Zhou Han-ming Shen Hong-xi Xu 2021Acta Pharmacologica Sinica2021,42,2:7
7Cultivation of microalgae for biodiesel production:A review on upstream and downstream processing显示文摘Fluctuating market price of fossil fuel and overwhelming emission of greenhouse gases to the atmosphere have resulted in climate change and have been a global concern in this decade. Hence, biodiesel has become an alternative option to fossil diesel as it is renewable and environmentally friendly. Nevertheless, this alternative fuel that is usually derived from terrestrial oil crops will cause shortage in food supply and deforestation if mass production is realized. In recent years, cultivation of aquatic microorganism(particularly microalgae) to produce biodiesel is considered as a practical solution due to their high growth rate and ability to synthesize large quantity of lipid within their cell. However, the development of energy and cost-efficiency of microalgae cultivation system are the main issues in producing renewable microalgae biodiesel. Of late, wastewater or organic compost has been used as the cultivation medium as it can provide sufficient nutrients to sustain microalgae growth.Microalgae cultivation method and system are vitally important as these factors undoubtedly affect the final microalgae biomass and lipid yield. In this review, the cultivation system of microalgae, nutrients demanded for microalgae production, cell harvesting and drying, microalgae oil extraction, and utilization of microalgae biomass for biodiesel production are introduced and discussed. 9Porous V_2O_5-SnO_2 /CNTs composites as high performance cathode materials for lithium-ion batteries显示文摘Vanadium pentoxide (V2O5 ) exhibits high theoretical capacities when used as a cathode in lithium ion batteries (LIBs), but its application is limited by its structural instability as well as its low lithium and electronic conductivities. A porous composite of V2O5 -SnO2 /carbon nanotubes (CNTs) was prepared by a hydrothermal method and followed by thermal treatment. The small particles of V2O5 , their porous structure and the coexistence of SnO2 and CNTs can all facilitate the diffusion rates of the electrons and lithium ions. Electrochemical impedance spectra indicated higher ionic and electric conductivities, as compared to commercial V2O5 . 10Oblongifolin C suppresses lysosomal function independently of TFEB nuclear translocation显示文摘Lysosomes are the terminal organelles of the autophagic-endocytic pathway and play a key role in the degradation of autophagic contents. We previously reported that a natural compound oblongifolin C (OC) increased the number of autophagosomes and impaired the degradation of P62, most likely via suppression of lysosomal function and blockage of autophagosome-lysosome fusion. However, the precise mechanisms of how OC inhibits the lysosome-autophagy pathway remain unclear. In the present study, we investigated the effect of OC on transcription factor EB (TFEB), a master regulator of lysosomal biogenesis, lysosomal function and autophagy. We showed that treatment with OC (15 μM) markedly enhanced the nuclear translocation of TFEB in HeLa cells, concomitantly reduced the interaction of TFEB with 14-3-3 proteins. We further demonstrated that OC caused signi?cant inhibition of mTORC1 along with TFEB nuclear translocation, and OC-mediated TFEB nuclear translocation was dependent on mTORC1 suppression. Intriguingly, this increased nuclear TFEB was accompanied by reduced TFEB luciferase activity, increased lysosomal pH and impaired cathepsin enzyme activities. In HeLa cells, treatment with OC (7.5 μM) resulted in about 30% of cell death, whereas treatment with hydroxycitrate, a caloric restriction mimetic (20 μM) did not affect the cell viability. However, cotreatment with OC and hydroxycitrate caused signi?cantly great cytotoxicity (>50%). 11Ultrathin, flexible, and high-strength Ni/Cu/metallic glass/Cu/Ni composite with alternate magneto-electric structures for electromagnetic shielding显示文摘Electromagnetic interference(EMI)shielding materials with ultrathin,flexible,superior mechanical and thermal management properties are highly desirable for smart and wearable electronics.Here,ultrathin and flexible Ni/Cu/metallic glass/Cu/Ni(Ni/Cu/MG)multilayer composite with alternate magnetic and electrical structures was designed via facial electroless plating of Cu and Ni on an Fe-based metallic glass.The resultant 0.02 mm-thick Ni/Cu/MG composite displays a superior EMI shielding effectiveness(EMI SE)of 35 dB and a great EMI SE/t of 1750 dB/mm,which is greater than those of composites with monotonous multilayer or homogeneous structures.The improved EMI SE originates from the massive ohmic losses,the enhanced internal reflection/absorption,and the abundant interfacial polarization loss.Particularly,Ni/Cu/MG exhibits a high tensile strength of up to 1.2 GPa and outstanding mechanical stability,enabling the EMI SE remains unchanged after 10,000 times of bending.Moreover,Ni/Cu/MG has excellent Joule heating characteristics and thermal stability,which is very suitable for heating components of wearable hyperthermia devices.Zenan Ma Jiawei Li Jijun Zhang Aina He Yaqiang Dong Guoguo Tan Mingqiang Ning Qikui Man Xincai Liu 2021Journal of Materials Science & Technology2021,,22:3
12Vulnerability and adaptation of an oasis social–ecological system affected by glacier change in an arid region of northwestern China显示文摘The Hexi Inland River Basin in an arid region of northwestern China was chosen as the study area for this research. The authors define the vulnerability of an oasis social-ecological system to glacier change; select 16 indicators from natural and socioeconomic systems according to exposure, sensitivity, and adaptive capacity; and construct a vulnerability-assessment indicator system aimed at an inland river basin in the arid region of Northwestern China. Vulnerability of the oasis socialecological system affected by glacier change in the study area is evaluated by Spatial Principal Component Analysis(SPCA) under the circumstance of glacier change. The key factors affecting the vulnerability are analyzed. The vulnerability of the oasis social-ecological system in the Hexi Inland River Basin affected by glacier change is of more than medium grade, accounting for about 48.0% of the total number of counties in the study area. In terms of the spatial pattern of the vulnerability, the oasis economic belt is the most vulnerable. With the rapid development of the area's society and economy, the exposure of the system to glacial changes is significantly increased; and an increase in glacial meltwater is not enough to overcome the impact of increased exposure, which is the main reason for the high vulnerability. 14Syntheses, Crystal Structures and Biological Activity of the Diorganotin 2-(2-(4-Methoxybenzoyl)hydrazono)-3-phenylpropanoic Carboxylate Complexes显示文摘Diorganotin 2-(2-(4-methoxybenzoyl)hydrazono)-3-phenylpropanoic carboxylate complexes Ⅰ{[p-CH3 O-C6 H4 O(O)C=N-N=C(Ph CH2)COO](n-Bu)2 Sn(CH3 OH)}2 and Ⅱ {[p-CH3 O-C6 H4 O(O)C=NN=C(Ph CH2)COO](Ph CH2)2 Sn(CH3 OH)}2 have been synthesized. The complexes have been characterized by IR, 1 H, 13 C and 119 Sn NMR spectra, HRMS, elemental analysis, thermal stability analysis and the crystal structures have been determined by X-ray diffraction. The crystal of complex I belongs to triclinic system, space group P1 with a = 10.437(2), b = 11.576(3), c = 13.085(3) ?, α = 66.272(2)°, β = 86.920(3)°, γ = 73.367(3)°, Z = 2, V = 1383.3(5) ?3, Dc = 1.381 Mg·m-3, m(Mo Kα) = 0.959 mm-1, F(000) = 592, R = 0.0233 and w R = 0.0644. The crystal of complex Ⅱ is of triclinic system, space group P1 with a = 10.446(1), b = 11.720(1), c = 13.063(2) ?, α = 84.190(1)°, β = 71.610(1)°, γ = 72.624(1)°, Z = 1, V = 1448.3(3) ?3, Dc = 1.475 Mg·m-3, m(Mo Kα) = 0.925 mm-1, F(000) = 656, R = 0.0169 and w R = 0.0432. In vitro antitumor activities of both complexes were evaluated by MTT against three human cancer cell lines(NCI-H460, Hep G2 and MCF7), and two complexes exhibited good antitumor activity. 16Establish a normal fetal lung gestational age grading model and explore the potential value of deep learning algorithms in fetal lung maturity evaluation显示文摘Background:Prenatal evaluation of fetal lung maturity(FLM)is a challenge,and an effective non-invasive method for prenatal assessment of FLM is needed.The study aimed to establish a normal fetal lung gestational age(GA)grading model based on deep learning(DL)algorithms,validate the effectiveness of the model,and explore the potential value of DL algorithms in assessing FLM.Methods:A total of 7013 ultrasound images obtained from 1023 normal pregnancies between 20 and 41+6 weeks were analyzed in this study.There were no pregnancy-related complications that affected fetal lung development,and all infants were born without neonatal respiratory diseases.The images were divided into three classes based on the gestational week:class I:20 to 29+6 weeks,class II:30 to 36+6 weeks,and class III:37 to 41+6 weeks.There were 3323,2142,and 1548 images in each class,respectively.First,we performed a pre-processing algorithm to remove irrelevant information from each image.Then,a convolutional neural network was designed to identify different categories of fetal lung ultrasound images.Finally,we used ten-fold cross-validation to validate the performance of our model.This new machine learning algorithm automatically extracted and classified lung ultrasound image information related to GA.This was used to establish a grading model.The performance of the grading model was assessed using accuracy,sensitivity,specificity,and receiver operating characteristic curves.Results:A normal fetal lung GA grading model was established and validated.The sensitivity of each class in the independent test set was 91.7%,69.8%,and 86.4%,respectively.The specificity of each class in the independent test set was 76.8%,90.0%,and 83.1%,respectively.The total accuracy was 83.8%.The area under the curve(AUC)of each class was 0.982,0.907,and 0.960,respectively.The micro-average AUC was 0.957,and the macro-average AUC was 0.949.Conclusions:The normal fetal lung GA grading model could accurately identify ultrasound images of the fetal lung at different GAs,which can be used to identify cases of abnormal lung development due to gestational diseases and evaluate lung maturity after antenatal corticosteroid therapy.The results indicate that DL algorithms can be used as a non-invasive method to predict FLM.Tai-Hui Xia Man Tan Jing-Hua Li Jing-Jing Wang Qing-Qing Wu De-Xing Kong 2021Chinese Medical Journal2021,,15:2
17Scoring the collective effects of SNPs: association of minor alleles with complex traits in model organisms显示文摘It has long been assumed that most parts of a genome and most genetic variations or SNPs are non-functional with regard to reproductive fitness.However,the collective effects of SNPs have yet to be examined by experimental science.We here developed a novel approach to examine the relationship between traits and the total amount of SNPs in panels of genetic reference populations.We identified the minor alleles(MAs)in each panel and the MA content(MAC)that each inbred strain carried for a set of SNPs with genotypes determined in these panels.MAC was nearly linearly linked to quantitative variations in numerous traits in model organisms,including life span,tumor susceptibility,learning and memory,sensitivity to alcohol and anti-psychotic drugs,and two correlated traits poor reproductive fitness and strong immunity.These results suggest that the collective effects of SNPs are functional and do affect reproductive fitness.YUAN DeJian ZHU ZuoBin TAN XiaoHua LIANG Jie ZENG Chen ZHANG JieGen CHEN Jun MA Long DOGAN Ayca BROCKMANN Gudrun GOLDMANN Oliver MEDINA Eva RICE Amanda D.
