| 2 | Rapid quantification of semen hepatitis B virus DNA by real-time polymerase chain reaction显示文摘AIM: To examine the sensitivity and accuracy of real-time polymerase chain reaction (PCR) for the quantification of hepatitis B virus (HBV) DNA in semen.METHODS: Hepatitis B viral DNA was isolated from HBV carriers' semen and sera using phenol extraction method and QTAamp DNA blood mini kit (Qiagen, Germany). HBV DNA was detected by conventional PCR and quantified by TaqMan technology-based real-time PCR (quantitative polymerase chain reaction (qPCR)). The detection threshold was 200 copies of HBV DNA for conventional PCR and 10 copies of HBV DNA for real time PCR per reaction.RESULTS: Both methods of phenol extraction and QIAamp DNA blood mini kit were suitable for isolating HBV DNA from semen. The value of the detection thresholds was 500 copies of HBV DNA per mL in the semen. The viral loads were 7.5×107 and 1.67×107 copies of HBV DNA per mL in two HBV infected patients' sera, while 2.L4×105 and 3.02×105 copies of HBV DNA per mL in the semen.CONCLUSION: Real-time PCR is a more sensitive and accurate method to detect and quantify HBV DNA in the semen. | Wei-Ping Qian Yue-Qiu Tan Ying Chen Ying Peng Zhi Li Guang-Xiu Lu Made C. | 5 | Topographic differentiations of biological soil crusts and hydraulic properties in fixed sand dunes, Tengger Desert显示文摘Biological soil crusts(BSCs) play an important role in surface soil hydrology. Soils dominated with moss BSCs may have higher infiltration rates than those dominated with cyanobacteria or algal BSCs. However, it is unnown whether improved infiltration in moss BSCs is accompanied by an increase in soil hydraulic conductivity or water retention capacity. We investigated this question in the Tengger Desert, where a 43-year-old revegetation program has promoted the formation of two distinct types of BSCs along topographic positions, i.e. the moss-dominated BSCs on the interdune land and windward slopes of the fixed sand dunes, and the algal-dominated BSCs on the crest and leeward slopes. Soil water retention capacity and hydraulic conductivity were measured using an indoor evaporation method and a field infiltration method. And the results were fitted to the van Genuchten–Mualem model. Unsaturated hydraulic conductivities under greater water pressure(<–0.01 MPa) and water retention capacities in the entire pressure head range were higher for both crust types than for bare sand. However, saturated and unsaturated hydraulic conductivities in the near-saturation range(>–0.01 MPa) showed decreasing trends from bare sand to moss crusts and to algal crusts. | 6 | Spatiotemporal patterns of the fish assemblages downstream of the Gezhouba Dam on the Yangtze River显示文摘An explicit demonstration of the changes in fish assemblages is required to reveal the influence of damming on fish species.However,information from which to draw general conclusions regarding changes in fish assemblages is insufficient because of the limitations of available approaches.We used a combination of acoustic surveys,gillnet sampling,and geostatistical simulations to document the spatiotemporal variations in the fish assemblages downstream of the Gezhouba Dam,before and after the third impoundment of Three Gorges Reservoir(TGR).To conduct a hydroacoustic identification of individual species,we matched the size distributions of the fishes captured by gillnet with those of the acoustic surveys.An optimum threshold of target strength of 50 dB re 1 m 2 was defined,and acoustic surveys were purposefully extended to the selected fish assemblages(i.e.,endemic Coreius species) that was acquired by the size and species selectivity of the gillnet sampling.The relative proportion of fish species in acoustic surveys was allocated based on the composition(%) of the harvest in the gillnet surveys.Geostatistical simulations were likewise used to generate spatial patterns of fish distribution,and to determine the absolute abundance of the selected fish assemblages.We observed both the species composition and the spatial distribution of the selected fish assemblages changed significantly after implementation of new flow regulation in the TGR,wherein an immediate sharp population decline in the Coreius occurred.Our results strongly suggested that the new flow regulation in the TGR impoundment adversely affected downstream fish species,particularly the endemic Coreius species.To determine the factors responsible for the decline,we associated the variation in the fish assemblage patterns with changes in the environment and determined that substrate erosion resulting from trapping practices in the TGR likely played a key role. | TAO JiangPing GONG YuTian TAN XiChang YANG Zhi CHANG JianBo | 2012 | Science China(Life Sciences)2012,55,7: | 12 |

| 8 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 9 | Higher PD-1 expression concurrent with exhausted CD8+ T cells in patients with de novo acute myeloid leukemia显示文摘Objective: To investigate the association between the T cell inhibitory receptor programmed death 1(PD-1)and T cell exhaustion status in T cells from patients with de novo acute myeloid leukemia(AML) and AML in complete remission(CR).Methods:Surface expression of PD-1 and the exhaustion and immunosenescence markers CD244 and CD57 on CD3+,CD4+ and CD8+ T cells from peripheral blood samples from 20 newly diagnosed,untreated AML patients and 10 cases with AML in CR was analyzed by flow cytometry.Twenty-three healthy individuals served as control.Results:A significantly higher percentage of PD-1+ cells were found for CD3+ T cells in the de novo AML group compared with healthy controls.In addition,an increased level of PD-1+ CD8+ T cells,but not PD-1+ CD4+,was found for CD3+ T cells in the de novo AML and AML-CR samples.A higher percentage of CD244+ CD4+,CD244+ CD8+,CD57+ CD4+ and CD57+ CD8+ T cells was found in CD3+ T cells in samples from those with de novo AML compared with those from healthy controls.Strong increased PD-1+ CD244+ and PD-1+ CD57+ coexpression was found for CD4+ and CD8+ T cells in the de novo AML group compared with healthy controls.Conclusions:We characterized the major T cell defects,including co-expression of PD-1 and CD244,CD57-exhausted T cells in patients with de novo AML,and found a particular influence on CD8+ T cells,suggesting a poor anti-leukemia immune response in these patients. | Jiaxiong Tan Shaohua Chen Yuhong Lu Danlin Yao Ling Xu Yikai Zhang Lijian Yang Jie Chen Jing Lai Zhi Yu Kanger Zhu Yangqiu Li | 2017 | Chinese Journal of Cancer Research2017,29,5: | 10 |

We study the impact of controlling persons with foreign residency rights on corporate audit perspective. We find that firms whose controlling persons have foreign residency rights are more likely to use high-quality auditing services, and that this behavior is more obvious in regions with lower marketization and in firms with higher separation of ownership and control. | 12 | Diabetes inhibits corneal epithelial cell migration and tight junction formation in mice and human via increasing ROS and impairing Akt signaling显示文摘Corneal wounds usually heal quickly;but diabetic patients have more fragile corneas and experience delayed and painful healing. In the present study, we compared the healing capacity of corneal epithelial cells (CECs) between normal and diabetic conditions and the potential mechanisms. Primary murine CEC derived from wild-type and diabetic (db/db) mice, as well as primary human CEC were prepared. Human CEC were exposed to high glucose (30 mM) to mimic diabetic conditions. Cell migration and proliferation were assessed using Scratch test and MTT assays, respectively. Reactive oxygen species (ROS) production in the cells was measured using dichlorofluorescein reagent. Western blot was used to evaluate the expression levels of Akt. Transepithelial electrical resistance (TEER) and zonula occludens-1 (ZO-1) expression were used to determine tight junction integrity. We found that the diabetic CEC displayed significantly slower cell proliferation and migration compared with the normal CEC from both mice and humans. Furthermore, ROS production was markedly increased in CEC grown under diabetic conditions. Treatment with an antioxidant N-acetyl cysteine (NAC, 100 μM) significantly decreased ROS production and increased wound healing in diabetic CEC. Barrier function was significantly reduced in both diabetic mouse and human CEC, while NAC treatment mitigated these effects. We further showed that Akt signaling was impaired in diabetic CEC, which was partially improved by NAC treatment. These results show that diabetic conditions lead to delayed wound-healing capacity of CEC and impaired tight junction formation in both mice and human. | 13 | Behavioral Abnormality along with NMDAR-related CREB Suppression in Rat Hippocampus after Shortwave Exposure显示文摘Objective To estimate the detrimental effects of shortwave exposure on rat hippocampal structure and function and explore the underlying mechanisms. Methods One hundred Wistar rats were randomly divided into four groups(25 rats per group) and exposed to 27 MHz continuous shortwave at a power density of 5, 10, or 30 m W/cm^2 for 6 min once only or underwent sham exposure for the control. The spatial learning and memory, electroencephalogram(EEG), hippocampal structure and Nissl bodies were analysed. Furthermore, the expressions of N-methyl-D-aspartate receptor(NMDAR) subunits(NR1, NR2 A, and NR2 B), c AMP responsive element-binding protein(CREB) and phosphorylated CREB(p-CREB) in hippocampal tissue were analysed on 1, 7, and 14 days after exposure. Results The rats in the 10 and 30 m W/cm^2 groups had poor learning and memory, disrupted EEG oscillations, and injured hippocampal structures, including hippocampal neurons degeneration, mitochondria cavitation and blood capillaries swelling. The Nissl body content was also reduced in the exposure groups. Moreover, the hippocampal tissue in the 30 m W/cm^2 group had increased expressions of NR2 A and NR2 B and decreased levels of CREB and p-CREB. Conclusion Shortwave exposure(27 MHz, with an average power density of 10 and 30 m W/cm^2) impaired rats' spatial learning and memory and caused a series of dose-dependent pathophysiological changes. | 16 | Real-time Microwave Exposure Induces Calcium Efflux in Primary Hippocampal Neurons and Primary Cardiomyocytes显示文摘Objective To detect the effects of microwave on calcium levels in primary hippocampal neurons and primary cardiomyocytes by the real-time microwave exposure combined with laser scanning confocal microscopy. Methods The primary hippocampal neurons and primary cardiomyocytes were cultured and labeled with probes, including Fluo-4 AM, Mag-Fluo-AM, and Rhod-2, to reflect the levels of whole calcium [Ca^(2+)], endoplasmic reticulum calcium [Ca^(2+)]ER, and mitochondrial calcium [Ca^(2+)]MIT, respectively. Then, the cells were exposed to a pulsed microwave of 2.856 GHz with specific absorption rate(SAR) values of 0, 4, and 40 W/kg for 6 min to observe the changes in calcium levels. Results The results showed that the 4 and 40 W/kg microwave radiation caused a significant decrease in the levels of [Ca^(2+)], [Ca^(2+)]ER, and [Ca^(2+)]MIT in primary hippocampal neurons. In the primary cardiomyocytes, only the 40 W/kg microwave radiation caused the decrease in the levels of [Ca^(2+)], [Ca^(2+)]ER, and [Ca^(2+)]MIT. Primary hippocampal neurons were more sensitive to microwave exposure than primary cardiomyocytes. The mitochondria were more sensitive to microwave exposure than the endoplasmic reticulum. Conclusion The calcium efflux was occurred during microwave exposure in primary hippocampal neurons and primary cardiomyocytes. | 19 | Bare sclera resection followed by mitomycin C and/or autograft limbus conjunctiva in the surgery for pterygium:a Meta-analysis显示文摘AIM: To evaluate the recurrence and complications after bare sclera resection(BSR) combined with mitomycin C(MMC) treatment and/or autograft limbus conjunctiva(ALC) in the surgery for pterygium.·METHODS: Meta-analysis was used to evaluate the differences in patient outcomes between BSR of pterygium with or without MMC and/or ALC. All included studies were randomized trials of patients with pterygium who received BSR followed by MMC and/or ALC in the surgery. The recurrence of pterygium and other complications resulting from different treatments were extracted for analysis.·RESULTS: Thirteen studies met the inclusion criteria.The recurrence of pterygium with intraoperative(IO) MMC was higher than that with ALC(OR=2.38,95% confidence interval 1.45-3.91, I2=29%). Postoperative MMC resulted in an incidence of recurrence similar to that of ALC(OR=0.66, 95% confidence interval 0.30-1.42, I2=0%), and IO MMC treatment in combination with ALC produced similar patient outcomes to ALC alone(OR =0.41, 95%confidence interval 0.16-1.01, I2=16%). Other complications such as punctate epitheliopathy, scleral thinning and ischemia, irritation and persistent epithelium defect, were more common in patients in the MMC group as compared to those treated with ALC.·CONCLUSION: The recurrence of pterygium with BSR followed by ALC is lower than that of BSR followed by MMC, and the incidence of other complications is lower.While ALC is a more effective strategy for treating pterygium, the quality of the ALC transplant should be considered when the patient has a history of glaucoma. | Tan Long Zhi Li | 2015 | International Journal of Ophthalmology(English edition)2015,8,5: | 7 |
| 20 | Overexpression of kallikrein gene 10 is a biomarker for predicting poor prognosis in gastric cancer显示文摘AIM:To analyze the expression of kallikrein gene 10(KLK10)in gastric cancer and to determine whether KLK10 has independent prognostic value in gastric cancer.METHODS:We studied KLK10 expression in 80 histologically confirmed gastric cancer samples using realtime quantitative reverse transcription-PCR and hK10expression using immunohistochemistry.Correlations with clinicopathological variables(lymph node metastasis,depth of invasion and histology)and with outcomes(disease-free survival and overall survival)during a median follow-up period of 31 mo were assessed.Gastric cancer tissues were then classified as KLK10 positive or negative.RESULTS:KLK10 was found to be highly expressed in 57/80(70%)of gastric cancer samples,while its expression was very low in normal gastric tissues.Positive relationships between KLK10 expression and lymph node metastasis(P=0.048),depth of invasion(P=0.034)and histology(P=0.015)were observed.Univariate survival analysis revealed that gastric cancer patients with positive KLK10 expression had an increased risk for relapse/metastasis and death(P=0.005 and0.002,respectively).Cox multivariate analysis indicated that KLK10 was an independent prognostic indicator of disease-free survival and overall survival in patients with gastric cancer.CONCLUSION:KLK10 expression is an independent biomarker of unfavorable prognosis in patients with gastric cancer. | Xin Jiao Hong-Jun Lu Mi-Mi Zhai Zhi-Jun Tan Hai-Ning Zhi Xiao-Man Liu Chen-Hao Liu Da-Peng Zhang | 2013 | World Journal of Gastroenterology2013,19,48: | 7 |
