1LRP6 downregulation promotes cardiomyocyte proliferation and heart regeneration显示文摘The adult mammalian heart is thought to be a terminally differentiated organ given the postmitotic nature of cardiomyocytes.Consequently,the potential for cardiac repair through cardiomyocyte proliferation is extremely limited.Low-density lipoprotein receptor-related protein 6(LRP6)is a Wnt co-receptor that is required for embryonic heart development.In this study we investigated the role of LRP6 in heart repair through regulation of cardiomyocyte proliferation.Lrp6 deficiency increased cardiomyocyte cell cycle activity in neonatal,juvenile and adult mice.Cardiomyocyte-specific deletion of Lrp6 in the mouse heart induced a robust regenerative response after myocardial infarction(MI),led to reduced MI area and improvement in left ventricular systolic function.In vivo genetic lineage tracing revealed that the newly formed cardiomyocytes in Lrp6-deficient mouse hearts after MI were mainly derived from resident cardiomyocytes.Furthermore,we found that the pro-proliferative effect of Lrp6 deficiency was mediated by the ING5/P21 signaling pathway.Gene therapy using the adeno-associated virus(AAV)9 miRNAi-Lrp6 construct promoted the repair of heart injury in mice.Lrp6 deficiency also induced the proliferation of human induced pluripotent stem cell-derived cardiomyocytes(iPSC-CMs).Our study identifies LRP6 as a critical regulator of cardiomyocyte proliferation,which may lead to the development of a novel molecular strategy to promote myocardial regeneration and repair.Yahan Wu Liping Zhou Hongyu Liu Ran Duan Huixing Zhou Fulei Zhang Xiaoyu He Dongbo Lu Ke Xiong Maolin Xiong Jinzhu Zhuang Yi Liu Li Li Dandan Liang Yi-Han Chen 2021Cell Research2021,31,4:5

3Hippocampal overexpression of TREM2 ameliorates high fat diet induced cognitive impairment and modulates phenotypic polarization of the microglia显示文摘Type 2 diabetes mellitus(T2DM)and Alzheimer's disease(AD)share several common pathophysiological features.Rare variants of triggering receptor expressed on myeloid cells 2(TREM2)increase the risk of developing AD,suggesting the involvement of TREM2 and innate immunity in AD development.It is still unknown whether TREM2 is related to cognitive impairment in T2DM.Here,we investigated the effects of the hippocampal overexpression of TREM2 on cognitive in long-term high-fat diet(HFD)-fed mice.Male C57BL/6J mice were maintained on HFD for 50 weeks.TREM2 was overexpressed in the hippocampus 36 weeks after HFD feeding using adeno-associated virus vector(AAV)-mediated gene delivery.The results showed that the HFD feeding induced rapid and persistent weight gain,glucose intolerance and significant impairments in learning and memory.Compared with AAV-con,AAV-TREM2 significantly ameliorated cognitive impairment without altering body weight and glucose homeostasis in HFD mice.The overexpression of TREM2 upregulated the synaptic proteins spinophilin,PSD95 and synaptophysin,suggesting the improvement in synaptic transmission.Dendritic complexity and spine density in the CA1 region were rescued after TREM2 overexpression.Furthermore,TREM2 markedly increased the number of iba-1/Arg-1-positive microglia in the hippocampus,suppressed neuroinflammation and microglial activation.In sum,hippocampal TREM2 plays an important role in improving HFD-induced cognitive dysfunction and promoting microglial polarization towards the M2 anti-inflammatory phenotype.Our study also suggests that TREM2 might be a novel target for the intervention of obesity/diabetes-associated cognitive decline.Min Wu Maolin Liao Rongfeng Huang Chunxiu Chen Tian Tian Hongying Wang Jiayu Li Jibin Li Yuxiang Sun Chaodong Wu Qifu Li Xiaoqiu Xiao 2022Genes & Diseases2022,9,2:4
4Typing of biliary tumor thrombus influences the prognoses of patients with hepatocellular carcinoma显示文摘Objective:To establish a new classification of biliary tumor thrombus(BTT).Methods:Overall survival of patients with BTT was first used to determine whether it correlated with current hepatocellular carcinoma staging systems.Univariate and multivariate analyses were used to determine factors affecting the overall survival(OS)to form the basis of our new classification for BTT.Results:All 6 international staging systems showed overlapping survival curves.Univariate followed by multivariate analyses showed that total bilirubin and intrahepatic/extrahepatic BTT were significant risk factors of OS.Based on these data,a new BTT classification was defined as:TypeⅠ:intrahepatic BTT;and TypeⅡ:extrahepatic BTT involving a common bile duct or common hepatic duct.TypeⅠwas further subdivided into type Ia:BTT involving a second-order intrahepatic duct or above,and type Ib:BTT involving a first-order intrahepatic duct.TypeⅡwas further subdivided into typeⅡa and typeⅡb using a cut-off total bilirubin(TB)>300μmol/L.The numbers(percentages)of patients with typesⅠandⅡBTT were 69(34.2%)and 133(65.8%),respectively.The median OS of typeⅠpatients was significantly higher than that of typeⅡpatients(37.5 months vs.23.2 months;P=0.002).Using subgroup analyses,OS outcomes were significantly different between the subgroups of typeⅡb and type IIa,although there was no significant difference between the type Ia and type Ib subgroups(P=0.07).Conclusions:A new BTT classification was established to predict prognoses of HCC patients with BTT who underwent liver resection.Juxian Sun Jiayi Wu Chang Liu Jie Shi Yonggang Wei Jianyin Zhou Zhibo Zhang Wan Yee Lau Maolin Yan Shuqun Cheng 2021Cancer Biology & Medicine2021,18,3:3
5Designing for Long Campaign Life Blast Furnace (2)-The Simulation of Temperature Field of Lining and Cooling Apparatus显示文摘Through the numerical modeling of temperature field for Blast Furnace (BF) lining and stave coolers, it can tell designers how to design a cooler which the hot surface temperature is less than its critical temperature under very high heat flux. Applying low heat re- sistance lining and staves cooler to BF is good for a layer of slag skull frozen on the hot surface of cooling stave. As long as the slag skull can stand, the furnace wall is stable and the heat loss of furnace does not increase. 7Computational and Experimental Study of Cooling Process in Coke Dry Quenching Experimental Shaft显示文摘Owing to its merits in energy savings, environmental protection and coke quality improvement, the Coke Dry Quenching (CDQ) technology is favorite in coke making industry today. In order to validate the mathematical model developed for the cooling process of real CDQ shaft, the one-seventh scale experimental setup was established. With the air as the working fluid and the coke as the packed bed, the coke and gas temperatures and the pressure drop in the cooling shaft were measured. A mathematical model based on the non-Darcian and non-thermal equilibrium model for the cooling process of the CDQ experimental shaft was presented. The computational results were compared to the experimental data. 15Component and simulation of the 4,000-year-old noodles excavated from the archaeological site of Lajia in Qinghai,China显示文摘Noodles are a global food,but the periods when and places where they were made and originated,as well as their ingredients and the cooking methods used to manufacture them,have remained contentious.In the 2005 edition of the journal Nature,we wrote a summary of the millet noodle specimens found in the Late Neolithic Qijia cultural stratum of the Lajia archaeological site in Qinghai Province,China.However,how the ancient people made millet noodles remains controversial.This paper provides a systematic analysis of the remains of noodles found withinan earthenware bowl at the Lajia ‘‘noodle house'' in terms of their plant composition including phytoliths,starch and biomarkers.It provides evidence of how people used millet4,000 years ago and,most specifically,of the principal methods used for producing millet-based noodles.Further,we show how we used traditional hele tools to make hele millet noodles,with especial reference to the gelatinized hydrogel-forming method,to simulate morphology consistent with the composition and form of the unearthed millet noodles.The results of this study provide new evidence and new insights into the cultural characteristics of the prehistoric human diet.Houyuan L Yumei Li Jianping Zhang Xiaoyan Yang Maolin Ye Quan Li Can Wang Naiqin Wu 2014Chinese Science Bulletin2014,59,35:1

17茶多酚对人膀胱癌BIU-87细胞间隙连接蛋白43表达和细胞间隙连接通讯功能的影响(英文)显示文摘Objective:The aim of our study was to investigate the effects of(-)-epigallocatechin-3-gallate(EGCG,the major phytochemistry component in green tea)on the expression of connexin43(Cx43)gene and detect the intercellular communication of the human bladder cancer cell lines BIU-87,and explore its possible mechanisms of prevention and cure for the bladder tumor.Methods:The methyl thiazolyl tetrazolium and Annexin-V/PI double-labeled flow cytometry methods were used to observe the growth inhibitory rate(IR)and apoptosis rate(AR)of BIU-87 cells treated by EGCG at different concentrations(0, 5,10 and 20 mg/L),respectively.The reverse transcription-polymerase chain reaction(RT-PCR)and Western Blotting analy- sis were employed to detect the relative expression levels of the Cx43 mRNA and its protein.The scrape-loading fluorescence dye transfer method was used to assess the gap junction intercellular communication(GJIC)under fluorescence microscope. Results:EGCG at concentrations(10 and 20 mg/L)both could significantly inhibit the proliferation and induce the apoptosis of BIU-87 cells.The IR and AR were(15.67±1.15)%,(18.33±1.53)%and(42.00±4.34)%,(27.33±3.21)%,respectively. 18Beneficial effects of high-pressure homogenization on the dispersion stability of aqueous hydrolysate from Mytilus edulis显示文摘Owing to the formation of aggregation and gelation during storage,certain proteins and peptides exhibit limited applications in aqueous protein food products.The purpose of this study was to investigate the influence of homogenization and xanthan gum addition on the dispersion stability of Mytilus edulis hydrolysate(PHM).High-pressure homogenization(HPH)at 360 and 40 bar in the first and second values,respectively,and adding xanthan gum at a concentration of 1 mg/mL showed significantly improvement on the stability of the PHM solution.PHM-xanthan gum solutions(PHMX)showed the highest polypeptide precipitation rate and turbidity retention rate compared with those of PHM.Moreover,the centrifugal precipitation rate of PHMX without HPH was higher than that of homogeneous PHMX.After HPH treatment at 400 bar,the percentage of smaller particles in PHM and PHMX was increased,the aqueous system became more uniform,and the fluorescence intensity reached its maximum.HPH pretreatment improved the polypeptide dispersion stability and turbidity retention rate of PHM and PHMX and reduced the fluorescence intensity.The interactions of xanthan gum and polypeptide render the network microstructure more uniform under the conditions of homogenization,thus improving the dispersion stability of PHMX solutions.Therefore,under the premise of adding xanthan gum,HPH can better enhance the dispersion stability of the polypeptide in PHM.Fengjiao Mao Maolin Tu Fengjiao Fan Chao Wu Cuiping Yu Ming Du 2020Food Science and Human Wellness2020,9,4:0
19A novel strain of Levilactobacillus brevis PDD-5 isolated from salty vegetables has beneficial effects on hyperuricemia through anti-inflammation and improvement of kidney damage显示文摘Hyperuricemia is a metabolic disorder caused by abnormal purine metabolism,resulting in abnormally high serum uric acid.In this study,a novel Levilactobacillus brevis PDD-5 isolated from salty vegetables was verified with the function of alleviating hyperuricemia.The relevant effects of L.brevis PDD-5 in lowering uric acid were analyzed by in vitro and in vivo experiments.The results showed that the L.brevis PDD-5 has(68.86±15.46)%of inosine uptake capacity and(95.75±3.30)%of guanosine uptake capacity in vitro.Oral administration of L.brevis PDD-5 to hyperuricemia rats reduced uric acid,creatinine,and urea nitrogen in serum,as well as decreased inosine and guanosine levels in the intestinal contents of rats.Analysis of relevant markers in the kidney by ELISA kits revealed that L.brevis PDD-5 alleviated oxidative stress and inflammation.Moreover,the gene expression of uric acid transporter 1(URAT1)and glucose transporter 9(GLUT9)was down-regulated,and the gene expression of organic anion transporter 1(OAT1)was up-regulated after treatment with L.brevis PDD-5.Western blot analysis showed that L.brevis PDD-5 alleviated hyperuricemia-induced kidney injury through the NLRP3 pathway.The se findings suggest that L.brevis PDD-5 can lower uric acid,repair kidney damage,and also has the potential to prevent uric acid nephropathy.Jue Xu Maolin Tu Xiankang Fan Yuxing Guo Tao Zhang Xiaoqun Zeng Zhendong Cai Zhen Wu Daodong Pan 2024Food Science and Human Wellness2024,13,2:0
20Mesopause temperatures and relative densities at midlatitudes observed by the Mengcheng meteor radar显示文摘The atmospheric temperatures and densities in the mesosphere and lower thermosphere(MLT)region are essential for studying the dynamics and climate of the middle and upper atmosphere.In this study,we present more than 9 years of mesopause temperatures and relative densities estimated by using ambipolar diffusion coefficient measurements observed by the Mengcheng meteor radar(33.4°N,116.5°E).The intercomparison between the meteor radar and Thermosphere Ionosphere Mesosphere Energetics and Dynamics/Sounding of the Atmosphere by Broadband Emission Radiometry(TIMED/SABER)and Earth Observing System(EOS)Aura/Microwave Limb Sounder(MLS)observations indicates that the meteor radar temperatures and densities agree well with the simultaneous satellite measurements.Annual variations dominate the mesopause temperatures,with the maximum during winter and the minimum during summer.The mesopause relative densities also show annual variations,with strong maxima near the spring equinox and weak maxima before the winter solstice,and with a minimum during summer.In addition,the mesopause density exhibits a structure similar to that of the zonal wind:as the zonal wind flows eastward(westward),the mesopause density decreases(increases).At the same time,the meridional wind shows a structure similar to that of the mesopause temperature:as the meridional wind shows northward(southward)enhancements,the mesopause temperature increases(decreases).Simultaneous horizontal wind,temperature,and density observations provide multiple mesospheric parameters for investigating mesospheric dynamics and thermodynamic processes and have the potential to improve widely used empirical atmospheric models.Wen Yi XiangHui Xue MaoLin Lu Jie Zeng HaiLun Ye JianFei Wu Chong Wang TingDi Chen 2023Earth and Planetary Physics2023,7,6:0
