1p53 upregulated by HIF-la promotes hypoxiainduced G2/M arrest and renal fibrosis in vitro and in vivo显示文摘Hypoxia plays an important role in the genesis and progression of renal fibrosis.The underlying mechanisms, however, have not been sufficiently elucidated. We examined the role of p53 in hypoxia-induced renal fibrosis in cell culture (human and rat renal tubular epithelial cells) and a mouse unilateral ureteral obstruction (UUO) model. Cell cycle of tubular cells was determined by flow cytometry, and the expression of profibrogenic factors was determined by RT-PCR, immunohistochemistry, and western blotting. Chromatin immunoprecipitation and luciferase reporter experiments were performed to explore the effect of HIF-lα on p53 expression. We showed that, in hypoxic tubular cells, p53 upregulation suppressed the expression of CDK1 and cyclins Bl and DI, leading to cell cycle (G2/M) arrest (or delay) and higher expression of TGF-β, CTGF, collagens, and fibronectin. p53 suppression by siRNA or by a specific p53 inhibitor (PIF-α) triggered opposite effects preventing the G2/M arrest and profibrotic changes. In vivo experiments in the UUO model revealed similar antifibrotic results following intraperitoneal administration of PIF-α(2.2 mg/kg). Using gain-of-function, loss-of-function, and luciferase assays, we further identified an HRE3 region on the p53 promoter as the HIF-lα-binding site. The HIF-la-HRE3 binding resulted in a sharp transcriptional activation of p53. Collectively, we show the presence of a hypoxia-activated, p53-responsive profibrogenic pathway in the kidney. 2Regulatory effect of gut microbes on blood pressure显示文摘Hypertension is an important global public health issue because of its high morbidity as well as the increased risk of other diseases.Recent studies have indicated that the development of hypertension is related to the dysbiosis of the gut microbiota in both animals and humans.In this review,we outline the interaction between gut microbiota and hypertension,including gut microbial changes in hypertension,the effect of microbial dysbiosis on blood pressure(BP),indicators of gut microbial dysbiosis in hypertension,and the microbial genera that affect BP at the taxonomic level.For example,increases in Lactobacillus,Roseburia,Coprococcus,Akkermansia,and Bifidobacterium are associated with reduced BP,while increases in Streptococcus,Blautia,and Prevotella are associated with elevated BP.Furthermore,we describe the potential mechanisms involved in the regulation between gut microbiota and hypertension.Finally,we summarize the commonly used treatments of hypertension that are based on gut microbes,including fecal microbiota transfer,probiotics and prebiotics,antibiotics,and dietary supplements.This review aims to find novel potential genera for improving hypertension and give a direction for future studies on gut microbiota in hypertension.Dong Yan Ye Sun Xiaoyue Zhou Wenhao Si Jieyu Liu Min Li Minna Wu 2022Animal Models and Experimental Medicine2022,5,6:1

4The relative and combined effects of herbivore assemblage and soil nitrogen on plant diversity显示文摘Plant diversity can be affected by both herbivore grazing and soil resources. However, it is unclear if the joint effects of herbivores and soil resources might vary with components of plant diversity. Here, we evaluated the relative and combined effects of herbivore assemblage and soil nitrogen(N) quantity and heterogeneity on the α and β components of plant diversity in a grassland that was subjected to four years of grazing under differing herbivore assemblages(no grazing, cattle grazing, sheep grazing, and mixed grazing). We found that herbivore assemblage combined with soil N quantity explained 41% of the variation in plant α-diversity, while herbivore assemblage combined with soil N heterogeneity explained 15% of the variation in plant β-diversity. The independent effects of herbivore assemblage explained more than those of soil N for both α-and β-diversity(α-diversity: 12% vs. 4%;β-diversity: 18% vs. 16%).
