1TaCML36, a wheat calmodulin-like protein,positively participates in an immune response to Rhizoctonia cerealis显示文摘Sharp eyespot,mainly caused by the soil-borne fungus Rhizoctonia cerealis,affects wheat(Triticum aestivum L.)production worldwide.In this study,we isolated TaCML36 gene encoding a wheat calmodulin-like protein,and studied its defense role in protection against R.cerealis.Transcription of TaCML36 was significantly elevated by both R.cerealis infection and exogenous ethylene treatment.Transcription was higher in resistant wheat lines than in susceptible ones.There were copies of TaCML36 on chromosomes 5A,5B,and 5D.The TaCML36 protein is composed of 183 amino acids and contains two calcium-binding EFhand domains.Subcellular localization assays in wheat indicated that TaCML36 localizes in both the cytoplasm and nucleus.Virus-induced gene silencing and disease assessment indicated that compared to the controls,TaCML36-silenced wheat plants displayed significantly reduced resistance to R.cerealis and had greater fungal biomass,suggesting that knockdown of TaCML36 impaired host resistance.Knockdown of TaCML36 also significantly repressed expression of pathogenesis-related genes such as Chitinase 1,PDF35,and PR17C,the ethylene response factor-encoding gene TaPIE1,and ethylene biosynthesis gene ACO2.Collectively,our results suggest that TaCML36 positively participates in the innate immune response to R.cerealis infection by modulating expression of defense-associated genes possibly in the ethylene signaling pathway.Lin Lu Wei Rong Ronghua Zhou Naxin Huo Zengyan Zhang 2019The Crop Journal2019,7,5:3

3Gene expression profiling related to powdery mildew resistance in wheat with the method of suppression subtractive hybridization显示文摘'Bainong 3217 × Mardler' BC5F4 wheat line at the initial stage of inoculation with powdery mildew pathogen (Erysiphe graminis DC) was used to construct a suppression subtractive hybridization (SSH) cDNA library. Totally 760 ESTs were obtained through sequencing. Similarity analysis of ESTs based on BLASTn and BLASTx with the sequences in GenBank, in combination with macroarray differential screening, revealed that 199 ESTs of 65 kinds were known to be functionally disease resistance related. Based on the gene expression profiling in the present study, it is postulated that salicylic acid (SA) and MAP-related signal transduction pathways were involved in powdery mildew resistance in wheat. System acquired resistance genes were predominant in terms of kinds and quantity. With the initiation of cell defense reaction, the genes conferring anti-oxidation substances were largely expressed and thus cell protection mechanism was activated. 4Structural characteristics and deep-water hydrocarbon accumulation model of the Scotian Basin, Eastern Canada显示文摘Commercial hydrocarbon reservoirs have been discovered in shallow-water areas of the Scotian Basin, Eastern Canada. However, knowledge about the structure and hydrocarbon accumulation characteristics of the basin is still insufficient, which constrains the oil and gas exploration in deep-water areas. Based on comprehensive data of magnetic anomalies, seismic survey, and drilling, this study determines the structure characteristics of the Scotian Basin and its hydrocarbon accumulation conditions in deep waters and evaluates the deep-water hydrocarbon exploration potential. The transform faults and basement structures in the northern basin control the sedimentary framework showing thick strata in east and thin strata in west of the basin. The bowl-shaped depression formed by thermal subsidence during the transitional phase and the confined environment (micro basins) caused by salt tectonics provide favorable conditions for the development of source rocks during the depression stage (also referred to as the depression period sequence) of the basin. The progradation of large shelf-margin deltas during the drift phase and steep continental slope provide favorable conditions for the deposition of slope-floor fans on continental margins of the basin. Moreover, the source-reservoir assemblage comprising the source rocks within the depression stage and the turbidite sandstones on the continental margin in the deep waters may form large deep-water turbidite sandstone reservoirs. 19Impact of the slope angle of hummocks on the ride-up of finger ice in the Liaohe Estuary显示文摘The ride-up and pile-up of sea ice have effects on the ice load of marine structure. The ride-up angle is an important parameter to study the process of ride-up and to determine the possibility of the ride-up to occur. Some conclusions about the ride-up angle are drawn based on field-survey data in this paper. Thirty hummocks with full structure, formed by 0.4-1.6 cm ice layers, were investigated in the Liaohe Estuary of the north of Liaodong Bay. After analyzing the cross-sections of the pile-up body, some conclusions on the ride-up angle are reached. The results indicate that whether the ride-up occurs or not is associated with the slope angle of the hummock. If the slope angle is greater than 10.31 , the ride-up can take place. With the development of riding-up and piling-up, if the slope angle increases to 40.0 , the climbing-up process will stop and the drifting ice begins to accumulate in front of the hummock. The climbing process does not continue until a new slope angle, which is less than 28.0 , is formed.
