2Sodium Stress in the Halophyte Thellungiella halophila and Transcriptional Changes in a thsos1-RNA Interference Line显示文摘The plasma membrane Na^+/H^+-antiporter salt overly sensitive1(SOS1)from the halophytic Arabidopsis-relative Thel-lungiella halophila(ThSOS1)shows conserved sequence and domain structure with the orthologous genes fromArabidopsis thaliana and other plants.When expression of ThSOS1 was reduced by RNA interference(RNAi),pronouncedcharacteristics of salt-sensitivity were observed.We were interested in monitoring altered transcriptional responsesbetween Thellungiella wild type and thsosl-4,a representative RNAi line with particular emphasis on root responses to saltstress at 350 mmol/L NaCl,a concentration that is only moderately stressful for mature wild type plants.Transcript profilingrevealed several functional categories of genes that were differently affected in wild-type and RNAi plants.Down-regulationof SOS1 resulted in different gene expression even in the absence of stress.The pattern of gene induction in the RNAi plantunder salt stress was similar to that of glycophytic Arabidopsis rather than that of wild type Thellungiella.The RNAi plantsfailed to down-regulate functions that are normally reduced in wild type Thellungiella upon stress and did not up-regulatefunctions that characterize the Thellungiella salt stress response.Metabolite changes observed in wild type Thellungiellaafter salt stress were less pronounced or absent in RNAi plants.Transcript and metabolite behavior suggested SOS1functions including but also extending its established function as a sodium transporter.The down-regulation of ThSOS1converted the halophyte Thellungiella into a salt-sensitive plant.Dong-Ha Oh Qingqiu Gong Alex Ulanov Quan Zhang Youzhi Li Wenying Ma Dae-Jin Yun Ray A. 3Numerical investigation into the effect of backfilling on coal pillar strength in highwall mining显示文摘This paper attempts to quantify the effect of backfilling on pillar strength in highwall mining using numerical modelling. Calibration against the new empirical strength formula for highwall mining was conducted to obtain the material parameters used in the numerical modelling. With the obtained coal strength parameters, three sets of backfill properties were investigated. The results reveal that the behavior of pillars varies with the type and amount of backfill as well as the pillar width to mining height ratio(w/h). In case of cohesive backfill, generally 75% backfill shows a significant increase in peak strength, and the increase in peak strength is more pronounced for the pillars having lower w/h ratios. In case of noncohesive backfill, the changes in both the peak and residual strengths with up to 92% backfill are negligible while the residual strength constantly increases after reaching the peak strength only when 100%backfill is placed. Based on the modelling results, different backfilling strategies should be considered on a case by case basis depending on the type of backfill available and desired pillar dimension.Mo S. 9Environmental Characterization of Periphyton Community显示文摘The present paper deals with the behavior of the Attached Microbial Community (AMC) for water self-purification at different riverbeds in a typical local river. The study quantitatively investigated the problem starting with in-situ sampling. It was found that more biomass of AMC was at riffles with wider distribution than in pools. High current velocity (HCV) plays a negative role at the initial stage of attachment on the riverbed, but HCV aids the community proliferation after stable attachment. External disturbances such as rainfalls and discharges from dams or reservoirs would detach the periphyton depending on the intensity of turbulence in water. However, it was discovered that the flock of periphyton could be restored very quickly because it was not completely removed. Thus, in order to enhance self-purification by periphyton, a suitable configuration of the riverbed must be constructed, and occasional appropriate repair along the channels would improve the decontamination of the river.Y.
