| 1 | Tomato SlAN11 regulates flavonoid biosynthesis and seed dormancy by interaction with bHLH proteins but not with MYB proteins显示文摘The flavonoid compounds are important secondary metabolites with versatile human nutritive benefits and fulfill a multitude of functions during plant growth and development.The abundance of different flavonoid compounds are finely tuned with species-specific pattern by a ternary MBW complex,which consists of a MYB,a bHLH,and a WD40 protein,but the essential role of SlAN11,which is a WD40 protein,is not fully understood in tomato until now.In this study,a tomato WD40 protein named as SlAN11 was characterized as an effective transcription regulator to promote plant anthocyanin and seed proanthocyanidin(PA)contents,with late flavonoid biosynthetic genes activated in 35S::SlAN11 transgenic lines,while the dihydroflavonol flow to the accumulation of flavonols or their glycosylated derivatives was reduced by repressing the expression of SlFLS in this SlAN11-overexpressed lines.The above changes were reversed in 35S::SlAN11-RNAi transgenic lines except remained levels of flavonol compounds and SlFLS expression.Interestingly,our data revealed that SlAN11 gene could affect seed dormancy by regulating the expressions of abscisic acid(ABA)signaling-related genes SlABI3 and SlABI5,and the sensitivity to ABA treatment in seed germination is conversely changed by SlAN11-overexpressed or-downregulated lines.Yeast two-hybrid assays demonstrated that SlAN11 interacted with bHLH but not with MYB proteins in the ternary MBW complex,whereas bHLH interacted with MYB in tomato.Our results indicated that low level of anthocyanins in tomato fruits,with low expression of bHLH(SlTT8)and MYB(SlANT1 and SlAN2)genes,remain unchanged upon modification of SlAN11 gene alone in the transgenic lines.These results suggest that the tomato WD40 protein SlAN11,coordinating with bHLH and MYB proteins,plays a crucial role in the fine adjustment of the flavonoid biosynthesis and seed dormancy in tomato. | Yongfeng Gao Jikai Liu Yongfu Chen Hai Tang Yang Wang Yongmei He Yongbin Ou Xiaochun Sun Songhu Wang Yinan Yao | 2018 | Horticulture Research2018,5,1: | 13 |

| 3 | Role of AMPK in atherosclerosis via autophagy regulation显示文摘Atherosclerosis is characterized by the accumulation of lipids and deposition of fibrous elements in the vascular wall, which is the primary cause of cardiovascular diseases. Adenosine monophosphate-activated protein kinase(AMPK) is a metabolic sensor of energy metabolism that regulates multiple physiological processes, including lipid and glucose metabolism and the normalization of energy imbalances. Overwhelming evidence indicates that AMPK activation markedly attenuates atherosclerosis development. Autophagy inhibits cell apoptosis and inflammation and promotes cholesterol efflux and efferocytosis. Physiological autophagy is essential for maintaining normal cardiovascular function. Increasing evidence demonstrates that autophagy occurs in developing atherosclerotic plaques. Emerging evidence indicates that AMPK regulates autophagy via a downstream signaling pathway. The complex relationship between AMPK and autophagy has attracted the attention of many researchers because of this close relationship to atherosclerosis development. This review demonstrates the role of AMPK and autophagy in atherosclerosis. | 7 | Effects of climate change on potential habitats of the cold temperate coniferous forest in Yunnan province, southwestern China显示文摘We built a classification tree(CT) model to estimate climatic factors controlling the cold temperate coniferous forest(CTCF) distributions in Yunnan province and to predict its potential habitats under the current and future climates, using seven climate change scenarios, projected over the years of 2070-2099. The accurate CT model on CTCFs showed that minimum temperature of coldest month(TMW) was the overwhelmingly potent factor among the six climate variables. The areas of TMW<-4.05 were suitable habitats of CTCF, and the areas of -1.35 < TMW were non-habitats, where temperate conifer and broad-leaved mixed forests(TCBLFs) were distribute in lower elevation, bordering on the CTCF. Dominant species of Abies, Picea, and Larix in the CTCFs, are more tolerant to winter coldness than Tsuga and broad-leaved trees including deciduous broad-leaved Acer and Betula, evergreen broadleaved Cyclobalanopsis and Lithocarpus in TCBLFs. Winter coldness may actually limit the cool-side distributions of TCBLFs in the areas between -1.35°C and -4.05°C, and the warm-side distributions of CTCFs may be controlled by competition to the species of TCBLFs. Under future climate scenarios, the vulnerable area, where current potential(suitable + marginal) habitats(80,749 km^2) shift to nonhabitats, was predicted to decrease to 55.91%(45,053 km^2) of the current area. Inferring from the current vegetation distribution pattern, TCBLFs will replace declining CTCFs. | 9 | Liver X receptor agonist T0901317 reduces atherosclerotic lesions in apoE^(-/-) mice by up-regulating NPC1 expression显示文摘In this study, we studied the effect of liver X receptor (LXR) agonist T0901317 on Niemann-Pick C1 protein (NPC1) expression in apoE-/- mice. Male apoE-/- mice were randomized into 4 groups, baseline group (n=10), control group (n=14), treatment group (n=14) and prevention group (n=14). All of the mice were fed with a high-fat/high-cholesterol (HFHC) diet containing 15% fat and 0.25% cholesterol. The baseline group treated with vehicle was sacrificed after 8 weeks of the diet. The control group and the prevention group were treated with either vehicle or T0901317 daily by oral gavage for 14 weeks. The treatment group was treated with vehicle for 8 weeks, and then was treated with the agonist T0901317 for additional 6 weeks. Gene and protein expression was analyzed by real-time quantitative PCR, immunohistochemistry and Western blotting, respectively. Plasma lipid concentrations were measured by commercially enzymatic methods. We used RNA interference technology to silence NPC1 gene expression in THP-1 macrophage-derived foam cells and then detected the effect of LXR agonist T0901317 on cholesterol efflux. Plasma triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and apoA-I concentrations were markedly increased in T0901317-treated groups. T0901317 treatment reduced the aortic atherosclerotic lesion area by 64.2% in the prevention group and 58.3% in the treatment group. LXR agonist treatment increased NPC1 mRNA expression and protein levels in the small intestine, liver and aorta of apoE-/- mice. Compared with the normal cells, cholesterol efflux of siRNA THP-1 macrophage-derived foam cells was significantly decreased, whereas cholesterol efflux of LXR agonist T0901317-treated THP-1 macrophage-derived foam cells was significantly increased. | 11 | Copper transportion of WD protein in hepatocytes from Wilson disease patients in vitro显示文摘AIM: To study the effect of copper transporting P-type ATPase in copper metabolism of hepatocyte and pathogenesis of Wilson disease (WD).METHODS: WD copper transporting properties in some organelles of the cultured hepatocytes were studied from WD patients and normal controls. These cultured hepatocytes were incubated in the media of copper 15mg.L-1 only, copper 15 mg. L-1 with vincristine (agonist of P-type ArPase) 0.5mg. L-1, or copper 15 mg. L-1 withvanadate (antagonist of P-type ATPase) 18.39 mg. L-1separately. Microsome (endoplasmic reticulum and Golgi apparatus), lysosome, mitochondria, and cytosol were isolated by differential centrifugation. Copper contents in these organelles were measured with atomic absorption spectrophotometer, and the influence in copper transportion of these organelles by vanadate and vincristine were comparatively analyzed between WD patients and controls.WD copper transporting P-type ATPase was detected by SDS-PAGE in conjunction with Western blot in liver samples of WD patients and controls.RESULTS: The specific WD proteins (Mr155 000 lanes) were expressed in human hepatocytes, including the control and WD patients. After incubation with medium containing copper for 2 h or 24 h, the microsome copper concentration in WD patients was obviously lower than that of controls,and the addtion of vanadate or vincristine would change the copper transporting of microsomes obviously. When incubated with vincristine, levels of copper in microsome were significantly increased, while incubated with vanadate,the copper concentrations in microsome were obviously decreased. The results indicated that there were Wdproteins, the copper transportion P-type ATPase in the microsome of hepatocytes. | 13 | Effects of octreotide on glucose transporter type 2expression in obese rat small intestine显示文摘AIM:To investigate the effects of the somatostatin analogue,octreotide,on maltose and sucrase activities and expression of glucose transporter type 2(GLUT2) in obese rat intestinal mucosa.METHODS:We divided 49 Sprague-Dawley rats into a group of 31 high fat diet-induced obese rats and a group of 18 normal controls.The obese rats were separated into an octreotide treated group of 16 rats and an obese group of 15.The intervention group was injected with octreotide at 40μg/kg body weight every 12 h for 8 d.Rat body weight was measured weekly to calculate Lee's index.After euthanization,maltase and sucrase activities in the small intestine were measuredby activity assays,and the fasting plasma glucose level was measured.The expression of GLUT2 in small intestinal mucosa was analyzed by immunohistochem-istry,reverse transcriptase polymerase chain reaction and Western blotting assays.RESULTS:Body weight,Lee's index,fasting plasma glucose level,maltase activity in small intestinal mucosa,mucosa and apical GLUT2,GLUT2 mRNA and protein expression levels were all significantly higher in the obese group than in the normal control group(605.61±141.00 vs 378.54±111.75,337.61 ±10.82 vs 318.73±20.10,8.60±1.38 vs 7.33± 0.70,156.01±58.81 vs 50.43±30.49,390 744.2 ±62 469.21 vs 170 546.50±50 646.14,26 740.18 ±3809.60 vs 354.98±57.19,0.26±0.11 vs 0.07 ±0.02,and 2.08±0.59 vs 1.27±0.38,respectively,all P<0.01).Sucrase activity did not differ between the two groups.Octreotide intervention significantly decreased the body weight and fasting plasma glucose level of obese rats(508.27±94.39 vs 605.61±141.00,7.58±1.51 vs 8.60±1.38,respectively,all P<0.05).The intestinal mucosa and apical GLUT2,expression of GLUT2 mRNA and protein were also significantly lower in the octreotide intervention group than in the obese group(269 975.2 ±53 730.94 vs 390 744.2±62 469.21,3758.06± 364.51 vs 26 740.18±3809.60,0.08±0.02 vs 0.26± 0.11,and 1.31±0.27 vs 2.08±0.59,respectively,all P<0.01).CONCLUSION:High fat dietinduced obesity is associated with elevated intestinal maltase activity,GLUT2 expression,and permanent apical GLUT2 in the small intestinal mucosa of rats.Octreotide can inhibit these effects. | Na Wei Rui Liu Yan Ou Xian Li Ou Qiang Wei Guo Cheng-Wei Tang | 2011 | World Journal of Gastroenterology2011,17,39: | 4 |
| 14 | Intensifying Gaseous Reduction of High Phosphorus Iron Ore Fines by Microwave Pretreatment显示文摘Gaseous reduction kinetics of the high phosphorus iron ore fines from Hubei in China and effect of microwave pretreatment on the gaseous reduction behavior were studied. Gaseous reduction kinetics were investigated by TG (Thermogravimetric) methods using LINSEIS STA PT 1600 thermal analysis equipment. Microwave pretreatments to the ore fines with four power levels were performed using a high temperature microwave reactor. Its effect was examined by TG methods and its mechanism was analyzed by SEM (scanning electron microscope) and EDS (energy dispersive spectrometer). Gaseous reduction tests were carried out using a tubular furnace. Results of kinetic study indicate that controlling step of the gaseous reduction of the ore fines is a mixing control of gas internal diffusion and interface chemical reaction when reduction fraction is less than 0.8 and is solid state diffusion when reduction fraction is more than 0.8. Microwave pretreatment of the ore fines could change the pore structure of the oolitic unit to generate cracks, fissures and loose zones, which promotes reduction in the early stage and delays the occurrence of sintering. Gaseous reduction tests show in the condition that the ore fines are pretreated with a microwave power of 450 W for 4 min and reduced under temperature of 1 273 K, the gaseous reduction of the ore fines could be apparently intensified. | 16 | A novel secretagogin/ATF4 pathway is involved in oxidized LDL-induced endoplasmic reticulum stress and islet β-cell apoptosis显示文摘Excessive accumulation of cholesterol inβcells initiates endoplasmic reticulum(ER)stress and associated apoptosis.We have reported that excessive uptake of cholesterol by MIN6 cells decreases the expression of secretagogin(SCGN)and then attenuates insulin secretion.Here,we aimed to determine whether cholesterol-induced SCGN decrease is involved in the modulation of ER stress and apoptosis in pancreaticβcells.In this study,MIN6 cells were treated with oxidized low-density lipoprotein(ox-LDL)for 24 h,and then intracellular lipid droplets and cell apoptosis were quantified,and SCGN and ER stress markers were identified by western blot analysis.Furthermore,small interfer RNA(siRNA)-mediated SCGN knockdown and recombinant plasmid-mediated SCGN restoration experiments were performed to confirm the role of SCGN in ER stress and associated cell apoptosis.Finally,the interaction of SCGN with ATF4 was computationally predicted and then validated by a co-immunoprecipitation assay.We found that ox-LDL treatment increased the levels of ER stress markers,such as phosphorylated protein kinase-like endoplasmic reticulum kinase,phosphorylated eukaryotic initiation factor 2 alpha,activating transcription factor 4(ATF4),and transcription factor CCAAT-enhancer-binding protein homologous protein,and promoted MIN6 cell apoptosis;in addition,the expression of SCGN was downregulated.siRNA-mediated SCGN knockdown exacerbatedβ-cell ER stress by increasing ATF4 expression.Pretreatment of MIN6 cells with the recombinant SCGN partly antagonized ox-LDL-induced ER stress and apoptosis.Furthermore,a co-immunoprecipitation assay revealed an interaction between SCGN and ATF4 in MIN6 cells.Taken together,these results demonstrated that pancreaticβ-cell apoptosis induced by ox-LDL treatment can be attributed,in part,to an SCGN/ATF4-dependent ER stress response. | Li Wu Yuncheng Lv Ying Lv Sunmin Xiang Zhibo Zhao Ziqing Tang Linling Ou Bin Yan Xinhua Xiao Gebo Wen Renxian Cao Jing Yang | 2021 | Acta Biochimica et Biophysica Sinica2021,53,1: | 2 |
