12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 3Oxidative Damage to DNA and Its Relationship With Diabetic Complications显示文摘Objective To detect the oxidative DNA damage in diabetic patients and to investigate the relationship of oxidative DNA damage with diabetes and diabetic nephropathy. Methods Single cell gel electrophoresis(SCGE) was used to detect the DNA strand breaks in peripheral blood lymphocytes,and oxidative DNA damage product and serum 8-OHdG were determined by a competitive ELISA in 47 cases,including 25 patients without diabetic complications,22 patients with diabetic nephropathy and 25 normal control subjects. Results Diabetic patients showed greater oxidative damage to DNA. The percentage of comet cells and the length of DNA migration(comet tail length) of peripheral blood lymphocytes were significantly increased in patients with diabetes,and significantly higher in patients with diabetic nephropathy than in diabetic patients without vascular complications(P<0.05). There was a significant increase in serum 8-OHdG in diabetic patients compared with normal subjects(P<0.05). Moreover,serum 8-OHdG was much higher in patients with diabetic nephropathy than in diabetic patients without vascular complications(P<0.05). Conclusion There is severe oxidative DNA damage in diabetic patients. 4The role of KDR in the interactions between human gastric carcinoma cell and vascular endothelial cell显示文摘AIM:To study the interactions between human gastriccarcinoma cell (HGCC) and human vascular endothelialcell (HVEC), and the role of KDR in these interactions.METHODS:Antisense oligodexynucleotide(ASODN)specific to KDR gene was devised and added to theculture medium of HGCC and HVEC. After the action ofASODN, the proliferation of two cells was measured byMTT method.The role of KDR in regulating theproliferation of two kinds of cells was known throughobserving the effect of ASODN on them. The conditionedmediums (CMs) of HGCC and HVEC were prepared. TheCM of one kind of cell was added acting on the otherkind of cell, then the cell proliferation was measuredby MTT. After the action of ASODN or CM, the cellularexpression of KDR gene was detected with in situhybridization(ISH) for mRNA level and withimmunohistochemical staining for protein level. ABC-ELISA was used to detect/NEGF in the CMs of two cells.RESULTS: KDR ASODN could specifically inhibit theproliferation of HGCC and HVEC significantiy. The growthinhibitory rate amounted to 55.35 % and 54.83 %,respectively (P<0.01). HGCC and HVEC could secret acertain level of hVEGF(92.06±1.69 ng/L, 77.70±8.04ng/L). The CM of HGCC could significantiy stimulate thegrowth(2.70±0.01 times) and KDR gene expression ofHVEC(P<0.01) while the CM of HVEC could significantiyinhibit the growth(52.97±0.01%) and KDR geneexpression of HGCC (P <0.01).CONCLUSION: KDR plays a key role in regulating theproliferation of HGCC and HVEC. 7Efficient production of omega-3 fatty acid desaturase (sFat-1)-transgenic pigs by somatic cell nuclear transfer显示文摘Omega-3(ω-3) fatty acid desaturase transgenic pigs may improve carcass fatty acid composition. The use of transgenic pigs is also an excellent large animal model for studying the role of ω-3 fatty acids in the prevention and treatment of coronary heart disease and cancer. Transgenic pigs carrying synthesized fatty acid desaturase-1 gene (sFat-1) from Caenorhabditis briggsae by somatic cell nuclear transfer (SCNT) were produced for the first time in China. Porcine fetal fibroblast cells were transfected with a sFat-1 expression cassette by the liposome-mediated method. Transgenic embryos were reconstructed by nuclear transfer of positive cells into enucleated in vitro matured oocytes. A total of 1889 reconstructed embryos were transferred into 10 naturally cycling gilts. Nine early pregnancies were established, 7 of which went to term. Twenty-one piglets were born. The cloning efficiency was 1.1% (born piglets/transferred embryos). The integration of the sFat-1 gene was confirmed in 15 live cloned piglets by PCR and Southern blot except for 2 piglets. Expression of the sFat-1 gene in 12 of 13 piglets was detected with RT-PCR. 8Effect of porosities of bilayered porous scaffolds on spontaneous osteochondral repair in cartilage tissue engineering显示文摘Poly(lactide-co-glycolide)-bilayered scaffolds with the same porosity or different ones on the two layers were fabricated,and the porosity effect on in vivo repairing of the osteochondral defect was examined in a comparative way for the first time.The constructs of scaffolds and bone marrow-derived mesenchymal stem cells were implanted into pre-created osteochondral defects in the femoral condyle of New Zealand white rabbits.After 12 weeks,all experimental groups exhibited good cartilage repairing according to macroscopic appearance,cross-section view,haematoxylin and eosin staining,toluidine blue staining,immunohistochemical staining and real-time polymerase chain reaction of characteristic genes.The group of 92%porosity in the cartilage layer and 77%porosity in the bone layer resulted in the best efficacy,which was understood by more biomechanical mimicking of the natural cartilage and subchondral bone.This study illustrates unambiguously that cartilage tissue engineering allows for a wide range of scaffold porosity,yet some porosity group is optimal.It is also revealed that the biomechanical matching with the natural composite tissue should be taken into consideration in the design of practical biomaterials,which is especially important for porosities of a multi-compartment scaffold concerning connected tissues.Zhen Pan Pingguo Duan Xiangnan Liu Huiren Wang Lu Cao Yao He Jian Dong Jiandong Ding 2015Regenerative Biomaterials2015,2,1:20
9Identification of cytokines involved in hepatic differentiation of mBM-MSCs under liver-injury conditions显示文摘AIM: To identify the key cytokines involved in hepatic differentiation of mouse bone marrow mesenchymal stem cells (mBM-MSCs) under liver-injury conditions. METHODS: Abdominal injection of CCl4 was adopted to duplicate a mouse acute liver injury model. Global gene expression analysis was performed to evaluate the potential genes involved in hepatic commitment under liver-injury conditions. The cytokines involved in hepatic differentiation of mBM-MSCs was function-ally examined by depletion experiment using specifi c antibodies, followed by rescue experiment and direct inducing assay. The hepatic differentiation was characterized by the expression of hepatic lineage genes and proteins, as well as functional features. RESULTS: Cytokines potentially participating in hepatic fate commitment under liver-injury conditions were initially measured by microarray. Among the up-regulated genes determined, 18 cytokines known to closely relate to liver growth, repair and development, were selected for further identif ication. The f ibroblast growth factor-4 (FGF-4), hepatocyte growth factor (HGF) and oncostatin M (OSM) were fi nally found to be involved in hepatic differentiation of mBM-MSCs under liver-injury conditions. Hepatic differentiation could be dramatically decreased after removing FGF-4, HGF and OSM from the liver-injury conditioned medium, and could be rescued by supplementing these cytokines. The FGF-4, HGF and OSM play different roles in the hepatic differentiation of mBM-MSCs, in which FGF-4 and HGF are essential for the initiation of hepatic differentiation, while OSM is critical for the maturation of hepatocytes. 11YAP promotes osteogenesis and suppresses adipogenic differentiation by regulatingβ-catenin signaling显示文摘YAP(yes-associated protein) is a transcriptional factor that is negatively regulated by Hippo pathway, a conserved pathway for the development and size control of multiple organs. The exact function of YAP in bone homeostasis remains controversial. Here we provide evidence for YAP's function in promoting osteogenesis, suppressing adipogenesis, and thus maintaining bone homeostasis.YAP is selectively expressed in osteoblast(OB)-lineage cells. Conditionally knocking out Yap in the OB lineage in mice reduces cell proliferation and OB differentiation and increases adipocyte formation, resulting in a trabecular bone loss. Mechanistically, YAP interacts with β-catenin and is necessary for maintenance of nuclear β-catenin level and Wnt/β-catenin signaling. Expression of β-catenin in YAP-deficient BMSCs(bone marrow stromal cells) diminishes the osteogenesis deficit. 14Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. 15Laparoscopic cystogastrostomy for the treatment of pancreatic pseudocysts: A case report显示文摘Pancreatic pseudocysts(PPs) are collections o pancreatic secretions that are lined by fibrous tissue and may contain necrotic debris or blood. The interventions including percutaneous, endoscopic o surgical approaches are based on the size, location symptoms and complications of a pseudocyst With the availability of advanced imaging system and cameras, better hemostatic equipments and excellent laparoscopic techniques, most pseudocyst can be found and managed by laparoscopy. We describe a case of a 30-year-old male patient with a pancreatic pseudocyst amenable to laparoscopi cystogastrostomy. An incision was made through the anterior gastric wall to expose the posterior gastri wall in close contact with the pseudocyst using an ultrasonically activated scalpel. Then, another incision was made for cystogastrostomy to obtain complete and unobstructed drainage. 16Evaluation of Oxidative Stress in Colorectal Cancer Patients显示文摘Objective To evaluate the oxidative stress in patients with colorectal cancer and to investigate the relationship between oxidative stress and colorectal cancer. Methods Seventy-six subjects were divided into two groups (36 colorectal cancer patients as the study group and 40 normal healthy individuals as the control group). Their protein oxidation, DNA damage, lipid peroxidation and antioxidants, vitamin C, vitamin E, glutathione (GSH), and antioxidative enzymes in serum were detected. Results The levels of protein carbonyl and advanced oxidation protein products (AOPP) were significantly higher in the study group than in the control group (P<0.01). Serum 8-OHdG was significantly increased in the study group compared to the control group (P<0.01). However, the mean serum level of MDA and conjugated diene was lower in the study group than in the control group (P<0.01). The activity of antioxidative enzymes was significantly decreased in the study group compared to the control group (P<0.01). Serum vitamins C and E concentrations were significantly reduced in the study group compared to the control group (P<0.01). 17A newly discovered Gigantopithecus fauna from Sanhe Cave, Chongzuo, Guangxi, South China显示文摘Among the most important faunas in the Late Cenozoic, the Gigantopithecus faunas have received a good deal of attention. The Gigantopithecus fauna recently discovered in Sanhe Cave consists of more than 80 mammal species, including cf. Hominidae, Pongo sp., Hylobates sp., Sinomastodon yangzien-sis, Stegodon preorientalis, Cervavitus fenqii, Dicoryphochoerus ultimus and Sus xiaozhu. It is the southernmost Gigantopithecus fauna found so far in China. Its geological age is estimated to be Early Pleistocene based on the fauna and stratigraphic correlation. The significant increase in the estimated body sizes of Ailuropoda, Gigantopithecus and Tapirus shows that the Sanhe fauna is middle Early Pleistocene, later than those from Wushan and Liucheng (early Early Pleistocene). Paleomagnetic dating of the fossil-bearing strata in Sanhe Cave gives an age of approximately 1.2 Ma. The fauna is characterized by tropical-subtropical forest types, including Pongo sp., Tupaia sp., Ia sp., Typhlomys intermedius, etc., and it lacks Palaearctic types. It is a typical tropical forest fauna, suggesting an en-vironment with a lush forest and a warm and humid climate. 20Evaluation of mercury speciation and removal through air pollution control devices of a 190 MW boiler显示文摘Air pollution control devices(APCDs) are installed at coal-fired power plants for air pollutant regulation.Selective catalytic reduction(SCR) and wet flue gas desulfurization(FGD) systems have the co-benefits of air pollutant and mercury removal.Configuration and operational conditions of APCDs and mercury speciation affect mercury removal efficiently at coal-fired utilities.The Ontario Hydro Method(OHM) recommended by the U.S.Environmental Protection Agency(EPA) was used to determine mercury speciation simultaneously at five sampling locations through SCR-ESP-FGD at a 190 MW unit.Chlorine in coal had been suggested as a factor affecting the mercury speciation in flue gas;and low-chlorine coal was purported to produce less oxidized mercury(Hg2+) and more elemental mercury(Hg0 ) at the SCR inlet compared to higher chlorine coal.SCR could oxidize elemental mercury into oxidized mercury when SCR was in service,and oxidation efficiency reached 71.0%.Therefore,oxidized mercury removal efficiency was enhanced through a wet FGD system.In the non-ozone season,about 89.5%-96.8% of oxidized mercury was controlled,but only 54.9%-68.8% of the total mercury was captured through wet FGD.Oxidized mercury removal efficiency was 95.9%-98.0%,and there was a big difference in the total mercury removal efficiencies from 78.0% to 90.2% in the ozone season.Mercury mass balance was evaluated to validate reliability of OHM testing data,and the ratio of mercury input in the coal to mercury output at the stack was from 0.84 to 1.08.Chengli Wu Yan Cao Zhongbing Dong Chinmin Cheng Hanxu Li Weiping Pan 2010Journal of Environmental Sciences2010,22,2:13
