| 2 | Metabolic engineering of Streptomyces coelicolor for enhanced prodigiosins (RED) production显示文摘Bacterial prodigiosins are red-colored secondary metabolites with multiple activities,such as anticancer,antimalarial and immunosuppressive,which hold great potential for medical applications.In this study,dramatically enhanced prodigiosins(RED) production in Streptomyces coelicolor was achieved by combinatorial metabolic engineering,including inactivation of the repressor gene ohkA,deletion of the actinorhodin(ACT) and calcium-dependent antibiotic(CDA) biosynthetic gene clusters(BGCs) and multi-copy chromosomal integration of the RED BGC.The results showed that ohkA deletion led to a 1-fold increase of RED production over the wild-type strain M145.Then,the ACT and CDA BGCs were deleted successively based on the AohkA mutant(SBJ101).To achieve multi-copy RED BGC integration,artificial ΦC31 attB site(s) were inserted simultaneously at the position where the ACT and CDA BGCs were deleted.The resulting strains SBJ102(with a single deletion of the ACT BGC and insertion of one artificial attB site) and SBJ103(with the deletion of both BGCs and insertion of two artificial attB sites) produced 1.9-and 6-fold higher RED titers than M145,respectively.Finally,the entire RED BGC was introduced into mutants from SBJ101 to SBJ103,generating three mutants(from SBJ104 to SBJ106) with chromosomal integration of one to three copies of the RED BGC.The highest RED yield was from SBJ106,which produced a maximum level of 96.8 mg g^(-1) cell dry weight,showing a 12-fold increase relative to M145.Collectively,the metabolic engineering strategies employed in this study are very efficient for the construction of high prodigiosin-producing strains. | Panpan Liu Hong Zhu Guosong Zheng Weihong Jiang Yinhua Lu | 2017 | Science China(Life Sciences)2017,60,9: | 7 |
| 3 | Mapping stripe rust resistance genes by BSR-Seq:YrMM58 and YrHY1 on chromosome 2AS in Chinese wheat lines Mengmai 58 and Huaiyang 1 are Yr17显示文摘Stripe rust(yellow rust), caused by Puccinia striiformis f. sp. tritici(PST),is one of the most devastating fungal diseases in common wheat(Triticum aestivum L.) in China and worldwide. Resistance breeding is the most effective strategy to control diseases in crop plants. Chinese wheat lines Mengmai 58 and Huaiyang 1 are highly resistant to PST race CYR34(V26) at the adult plant stage. To genetically map the underlying resistance genes we developed segregating populations by crossing Mengmai 58 and Huaiyang 1 with the susceptible cultivar Nongda 399. The stripe rust resistances in Mengmai 58 and Huaiyang 1 were both controlled by single dominant genes, provisionally designated YrMM58 and YrHY1, respectively. Bulked segregant RNA-Seq(BSR-Seq) analysis showed that YrMM58 and YrHY1 were located in the same distal ~16 Mb region on chromosome 2 AS.Comparative genomics analysis with the physical map of Aegilops tauschii proved useful for developing additional markers to saturate the genetic linkage map. YrMM58 and YrHY1 were mapped to the distal end of chromosome arm 2 AS, with the closest marker WGGB148 being 7.7 cM and 3.8 cM from the resistance gene, which was considered to be Yr17. | 4 | Autophagy receptor CCDC50 tunes the STING-mediated interferon response in viral infections and autoimmune diseases显示文摘DNA sensing and timely activation of interferon(IFN)-mediated innate immunity are crucial for the defense against DNA virus infections and the clearance of abnormal cells.However,overactivation of immune responses may lead to tissue damage and autoimmune diseases;therefore,these processes must be intricately regulated.STING is the key adaptor protein,which is activated by cyclic GMP-AMP,the second messenger derived from cGAS-mediated DNA sensing.Here,we report that CCDC50,a newly identified autophagy receptor,tunes STING-directed type I IFN signaling activity by delivering K63-polyubiquitinated STING to autolysosomes for degradation.Knockout of CCDC50 significantly increases herpes simplex virus 1(HSV-1)-or DNA ligand-induced production of type I IFN and proinflammatory cytokines.Ccdc50-deficient mice show increased production of IFN,decreased viral replication,reduced cell infiltration,and improved survival rates compared with their wild-type littermates when challenged with HSV-1.Remarkably,the expression of CCDC50 is downregulated in systemic lupus erythematosus(SLE),a chronic autoimmune disease.CCDC50 levels are negatively correlated with IFN signaling pathway activation and disease severity in human SLE patients.CCDC50 deficiency potentiates the cGAS-STING-mediated immune response triggered by SLE serum.Thus,our findings reveal the critical role of CCDC50 in the immune regulation of viral infections and autoimmune diseases and provide insights into the therapeutic implications of CCDC50 manipulation. | Panpan Hou Yuxin Lin Zibo Li Ruiqing Lu Yicheng Wang Tian Tian Penghui Jia Xi Zhang Liu Cao Zhongwei Zhou Chunmei Li Jieruo Gu Deyin Guo | 2021 | Cellular & Molecular Immunology2021,18,10: | 3 |
| 5 | Mapping a leaf senescence gene els1 by BSR-Seq in common wheat显示文摘Leaf senescence is normally the last stage of plant development. Early senescence of functional leaves significantly reduces the photosynthetic time and efficiency, seriously affecting grain yield and quality in wheat. Discovering genes responsible for early leaf senescence(els) are necessary for developing novel germplasms and cultivars with delayed leaf-senescence through molecular manipulation and marker assisted selection. In this study, we identified an early leaf senescence line M114 in a derivative of a wheat breeding population. Genetic analysis indicated that early leaf senescence in M114 is controlled by a single recessive gene, provisionally designated els1. By applying bulked segregant analysis and RNA-Seq(BSR-Seq), seven polymorphic markers linked to els1 were developed and the gene was located on chromosome arm 2 BS in a 1.5 c M genetic interval between markers WGGB303 and WGGB305. | 10 | Effect of mechanical degradation of laminated elastomeric bearings and shear keys upon seismic behaviors of small-to-medium-span highway bridges in transverse direction显示文摘Laminated elastomeric bearings have been widely used for small-to-medium-span highway bridges in China, in which concrete shear keys are set transversely to prohibit large girder displacement. To evaluate bridge seismic responses more accurately, proper analytical models of bearings and shear keys should be developed. Based on a series of cyclic loading experiments and analyses, rational analytical models of laminated elastomeric bearings and shear keys, which can consider mechanical degradation, were developed. The effect of the mechanical degradation was investigated by examining the seismic response of a small-to-medium-span bridge in the transverse direction under a wide range of peak ground accelerations(PGA). The damage mechanism for small-to-medium-span highway bridges was determined, which can explain the seismic damage investigation during earthquakes in recent years. The experimental results show that the mechanical properties of laminated elastomeric bearings will degrade due to friction sliding, but the degree of decrease is dependent upon the influencing parameters. It can be concluded that the mechanical degradation of laminated elastomeric bearings and shear keys play an important role in the seismic response of bridges. | 11 | Design and experiment of seed furrow cleaning device based on throwing and sliding for no-till maize seeding显示文摘In order to solve the serious problems of seeds are covered by residual film and overhead by straw during no-till seeding,a seed furrow cleaning device for no-till maize seeding was developed,which adopted a collaborative cleaning method of rotating spring teeth and curved sliding shovel.The movement process and motion trajectory of throwing residual film and straw were constructed.The maximum distance of throwing to one side in horizontal and maximum height in vertical were obtained.The motion trajectory of adjacent spring teeth was analyzed by Matlab,the motion trajectories of adjacent spring teeth at different speeds of 120 r/min,150 r/min and 180 r/min were achieved,the theoretical analysis results showed that the area of omitted area decreased with the increase of rotation speed.Based on theoretical and simulation analysis of critical parameters,the forward speed of machine,rotation speed of spring teeth,and dip angle between spring teeth and rotary disc were selected as the influencing factor.Straw cleaning rate(SCR)and residual film cleaning rate(RFCR)were selected as the response values for three factors and three levels of orthogonal experiment design.The optimal combination of the selected parameters was obtained,and the field test verification was also conducted.The results showed that the rotation speed of spring teeth,forward speed and dip angle of spring teeth significantly affect SCR and RFCR were in decreasing order.The field test results indicated that when forward speed was 6 km/h,rotation speed of spring teeth was 180 r/min and dip angle of spring teeth was 40°,SCR and RFCR were 88.27%and 84.31%,respectively.This study provides a reference for the development of no-till seeder in Xinjiang and the northwestern regions of China. | Panpan Yuan Hongwen Li Caiyun Lu Qingjie Wang Jin He Shenghai Huang Dandan Cui | 2022 | International Journal of Agricultural and Biological Engineering2022,15,4: | 1 |
| 12 | A computational model of topological and geometric recovery for visual curve completion显示文摘Visual curve completion is a fundamental problem in understanding the principles of the human visual system. This problem is usually divided into two problems: a grouping problem and a shape problem.On one hand, though perception of the visually completed curve is clearly a global task(for example,a human perceives the Kanizsa triangle only when seeing all three black objects), conventional methods for solving the grouping problem are generally based on local Gestalt laws. On the other hand, the shape of the visually completed curve is usually recovered by minimizing shape energy in existing methods.However, not only do these methods lack mechanisms to adjust the shape of the recovered visual curve using perceptual, psychophysical, and neurophysiological knowledge, but it is also difficult to calculate an explicit representation of the visually completed curve. In this paper, we present a systematic computational model for generating a visually completed curve. Firstly, based on recent studies of perception, psychophysics, and neurophysiology, we formulate a grouping procedure based on the human visual system by seeking a minimum Hamiltonian cycle in a graph, solving the grouping problem in a global manner. Secondly, we employ a B′ezier curve-based model to represent the visually completed curve. Not only is an explicit representation deduced, but we also present a means to integrate knowledge from related areas, such as perception, psychophysics, and neurophysiology, and so on. The material was characterized by XRD, FT-IR, UV-Vis and N2-adsorption. The Cosalen@MIL-100(Cr) modified glassy carbon electrode exhibits a well-defined reduction peak at the potential of –0.21 V toward the oxygen reduction reaction(ORR) by cyclic voltammetry(CV) in pH = 6.84 phosphate buffer. Almost 400 mV positive shift of potential at Cosalen@MIL-100(Cr) modified electrode for ORR compared with that at bare glassy carbon, indicates that Cosalen@MIL-100(Cr) possesses excellent electrocatalytic activity. The transferred number of electrons for ORR was determined by chronocoulometry. The result suggests that the introduction of Co(II)-salen complex into MOF increases the electrocatalytic activity via a four-electron reduction pathway. According to the experimental results,under low phosphorus treatment,four phosphorus-efficient soybean genotypes exhibited significantly high phosphorus uptake in mature period,especially for D31 and D37; however,three phosphorus-efficient genotypes showed no advantages in adaptability of phosphorus utilization efficiency,while only D31 exhibited high phosphorus utilization and absorption capacity. Correlation analysis and path analysis showed that phosphorus deficiency of soybean was mainly determined by phosphorus absorption capacity,and phosphorus deficiency under(- P) treatment was significantly higher than( + P) treatment. Phosphorus uptake and phosphorus utilization efficiency posed great direct effects on phosphorus efficiency,and phosphorus uptake exhibited a greater contribution; in addition,these two factors both posed small indirect effects. In mature period at reproductive growth stage,phosphorus absorption efficiency( phosphorus uptake) was the main variation source of phosphorus efficiency of various soybean genotypes in mature period.
