Quantitative evaluation methods for waterflooded layers of conglomerate reservoir based on well logging data显示文摘The rapid changing near source, multi-stream depositional environment of conglomerate reservoirs leads to severe heterogeneity, complex lithology and physical properties, and large changes of oil layer resistivity. Quantitative evaluation of water-flooded layers has become an important but difficult focus for secondary development of oilfields. In this paper, based on the analysis of current problems in quantitative evaluation of water-flooded layers, the Kexia Group conglomerate reservoir of the Sixth District in the Karamay Oilfield was studied. Eight types of conglomerate reservoir lithology were identified effectively by a data mining method combined with the data from sealed coring wells, and then a multi-parameter model for quantitative evaluation of the water-flooded layers of the main oil-bearing lithology was developed. Water production rate, oil saturation and oil productivity index were selected as the characteristic parameters for quantitative evaluation of water-flooded layers of conglomerate reservoirs. Finally, quantitative evaluation criteria and identification rules for water-flooded layers of main oil-bearing lithology formed by integration of the three characteristic parameters of water-flooded layer and undisturbed formation resistivity. This method has been used in evaluation of the water-flooded layers of a conglomerate reservoir in the Karamay Oilfield and achieved good results, improving the interpretation accuracy and compliance rate. It will provide technical support for avoiding perforation of high water-bearing layers and for adjustment of developmental programs.Tan Fengqi Li Hongqi Xu Changfu Li Qingyuan Peng Shouchang 2010Petroleum Science2010,7,4:21
Ubiquitination of Rheb governs growth factor-induced mTORC1 activation显示文摘Mechanistic target of rapamycin mTOR complex 1 (mTORC1)plays a key role in the integration of various environmental signals to regulate cell growth and metabolism,mTORC1 is recruited to the lysosome where it is activated by its interaction with GTP-bound Rheb GTPase. However, the regulatory mechanism of Rheb activity remains largely unknown. Here,we show that ubiquitination governs the nucleotide-bound status of Rheb.Lysosome-anchored E3 ligase RNF152 catalyzes Rheb ubiquitination and promotes its binding to the TSC complex.EGF enhances the deubiquitination of Rheb through AKT-dependent USP4 phosphorylation, leading to the release of Rheb from the TSC complex.Functionally, ubiquitination of Rheb is linked to mTORC1-mediated signaling and consequently regulates tumor growth. Thus,we propose a mechanistic model whereby Rheb-mediated mTORC1 activation is dictated by a dynamic opposing act between Rheb ubiquitination and deubiquitination that are catalyzed by RNF152 and USP4 respectively.Lu Deng Lei Chen Linlin Zhao Yan Xu Xiaoping Peng Xinbo Wang Lin Ding Jiali Jin Hongqi Teng Yanming Wang Weijuan Pan Fei Yu Lujian Liao Li Li Xin Ge Ping Wang 2019Cell Research2019,29,2:16
Inversion of Array Induction Logs and Its Application显示文摘With the help of the modified geometrical factor theory, the Marquardt method was used to calculate the true electrical parameters of the formation from array induction logs. The inversion results derived from the assumed model and some practical cases show that the rebuilt formation profile determined by 2-ft resolution array induction logs is reasonable when the formation thickness is greater than 1 m, which thus indicates that the inversion method is reliable and can provide quantitative information for the discrimination of oil/gas or water zone.Gao Jie Zhao Aibin Peng Fei Li Hongqi 2007Petroleum Science2007,4,3:7
Comparison of nonhuman primates identified the suitable model for COVID-19显示文摘Identification of a suitable nonhuman primate(NHP)model of COVID-19 remains challenging. Here,we characterized severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection in three NHP species:Old World monkeys Macaca mulatta(M.mulatta)and Macaca fascicularis(M.fascicularis)and New World monkey Callithrix jacchus(C.jacchus).Infected M.mulatta and M.fascicularis showed abnormal chest radiographs,an increased body temperature and a decreased body weight.Viral genomes were detected in swab and blood samples from all animals.Viral load was detected in the pulmonary tissues of M.mulatta and M.fascicularis but not C.jacchus.Furthermore,among the three animal species,M.mulatta showed the strongest response to SARS-CoV-2,including increased inflammatory cytokine expression and pathological changes in the pulmonary tissues.Collectively, these data revealed the different susceptibilities of Old World and New World monkeys to SARS-CoV-2 and identified M.mulatta as the most suitable for modeling COVID-19.Shuaiyao Lu Yuan Zhao Wenhai Yu Yun Yang Jiahong Gao Junbin Wang Dexuan Kuang Mengli Yang Jing Yang Chunxia Ma Jingwen Xu Xingli Qian Haiyan Li Siwen Zhao Jingmei Li Haixuan Wang Haiting Long Jingxian Zhou Fangyu Luo Kaiyun Ding Daoju Wu Yong Zhang Yinliang Dong Yuqin Liu Yinqiu Zheng Xiaochen Lin Li Jiao Huanying Zheng Qing Dai Qiangming Sun Yunzhang Hu Changwen Ke Hongqi Liu Xiaozhong Peng 2020Signal Transduction and Targeted Therapy2020,5,1:5

Effective treatment of SARS-CoV-2-infected rhesus macaques by attenuating inflammation显示文摘Dear Editor,The COVID-19 pandemic caused by the SARS-CoV-2 virus has caused a significant public health crisis worldwide.Recent studies show that excessive inflammatory response is critical for SARS-CoV-2 pathogenesis and COVID-19 severity,'which can lead to acute lung injury(ALI)and acute respiratory distress syndrome(ARDS).2 One major factor for acute inflammation in SARS-CoV-2 infection is the inflammatory macrophages, which has been considered important for the production of large amounts of proinflammatory cytokines.'Autopsy identifhed an intense infltra-tion of macrophages in the lung tissues of fatal COVID-19 patients.Shuaiyao Lu Jingjing Zhao Jiebin Dong Hongqi Liu Yinhua Zhu Honggang Li Liping Liu Yun Yang Shicheng Sun Yifan Song Yuan Zhao Ruiping She Tuoping Luo Hongkui Deng Xiaozhong Peng 2021Cell Research2021,31,2:1

The olfactory route is a potential way for SARS-CoV-2 to invade the central nervous system of rhesus monkeys显示文摘Neurological manifestations are frequently reported in the COVID-19 patients.Neuromechanism of SARS-CoV-2 remains to be elucidated. In this study,we explored the mechanisms of SARS-CoV-2 neurotropism via our established non-human primate model of COVID-19.In rhesus monkey,SARS-CoV-2 invades the CNS primarily via the olfactory bulb.Thereafter, viruses rapidly spread to functional areas of the central nervous system, such as hippocampus, thalamus, and medulla oblongata. The infection of SARS-CoV-2 induces the inflammation possibly by targeting neurons, microglia, and astrocytes in the CNS.Consistently,SARS-CoV-2 infects neuro-derived SK-N-SH,glial-derived U251,and brain microvascular endothelial cells in vitro. To our knowledge, this is the first experimental evidence of SARS-CoV-2 neuroinvasion in the NHP model, which provides important insights into the CNS-related pathogenesis of SARS-CoV-2.Li Jiao Yun Yang Wenhai Yu Yuan Zhao Haiting Long Jiahong Gao Kaiyun Ding Chunxia Ma Jingmei Li Siwen Zhao Haixuan Wang Haiyan Li Mengli Yang Jingwen Xu Junbin Wang Jing Yang Dexuan Kuang Fangyu Luo Xingli Qian Longjiang Xu Bin Yin Wei Liu Hongqi Liu Shuaiyao Lu Xiaozhong Peng 2021Signal Transduction and Targeted Therapy2021,6,5:0

Roles of light transmission aggregometry and CYP2C19 genotype in predicting ischaemic complications during interventional therapy for intracranial aneurysms显示文摘Background and purpose Light transmission aggregometry(LTA)and CYP2C19 genotype analysis are commonly used to evaluate the antiplatelet effects of clopidogrel during the interventional treatment of intracranial aneurysms. The aim of this study was to determine which test can predict ischaemic events during these treatments. Methods Patient demographic information, imaging data, laboratory data and ischaemic complications were recorded.LTA and CYP2C19 genotype results were compared, and multiple linear regression was performed to examine factors related to platelet reactivity.Multivariate regression analysis was performed to determine whether LTA and CYP2C19 could predict ischaemic complications and to identify other clinical risk factors.Receiver operating characteristic curve analysis was conducted to calculate the cut-off value for predicting ischaemic complications.A subgroup analysis was also performed for different CYP2C19 genotype metabolisers,as well as for patients with flow diverters and traditional stents. Results A total of 379 patients were included,of which 22 developed ischaemic events.Maximum platelet aggregation induced by ADP(ADP-MPA)could predict ischaemic events(p<0.001;area under the curve,0.752(95%CI 0.663 to 0.842)),and its cut-off value was 41.5%.ADP-MPA(p=0.001)and hypertension duration>10 years(p=0.022)were independent risk factors for ischaemic events, while the CYP2C19 genotype was not associated with ischaemic events. In the subgroup analysis,ADP-MPA could predict ischaemic events in fast metabolisers(p=0.004)and intermediate metabolisers(p=0.003).The cut-off value for ischaemic events was lower in patients with flow diverters(ADP-MPA=36.4%)than in patients with traditional stents(ADP-MPA=42.9%).Conclusions ADP-MPA can predict ischaemic complications during endovascular treatment of intracranial aneurysms.Patients with flow diverters require stronger antiplatelet medication than patients with traditional stents.Yangyang Zhou Wenqiang Li Chao Wang Ruhang Xie Yongnan Zhu Qichen Peng Limin Zhang Hongqi Zhang Yuxiang Gu Shiqing Mu Jian Liu Xinjian Yang 2023Stroke & Vascular Neurology2023,8,4:0
Reverse genetics in virology:A double edged sword显示文摘Reverse genetics via targeted modification of gene sequences to obtain a phenotype and the inference of a gene's function or regulatory mechanism is widely used as a potent tool in viral biology and application. However, while reverse genetics has contributed significantly to our understanding of molecular biology and the pathogenesis of viruses, its accessibility(operation)and openness(data)have raised many concerns regarding biosafety and biosecurity. In this review,we retrospectively examine the development of reverse genetics and its applications in virology, then emphasize global biosafety and biosecurity concerns regarding reverse genetics, and summarize global regulations, governance, and laws on reverse genetics. This review seeks to enhance our understanding and rational application of reverse genetics technology for the benefit of humankind.Hongyu Chen Hongqi Liu Xiaozhong Peng 2022Biosafety and Health2022,4,5:0
