| 4 | Role of calcium mobilization in the regulation of spontaneous transient outward currents in porcine coronary artery myocytes显示文摘The purpose of the present study was to further study the characteristics and regulation of spontaneous transient outward currents (STOCs) in freshly isolated porcine coronary artery smooth muscle cells (ASMCs). STOCs were recorded using the perforated whole-cell patch-clamp configuration. STOCs were voltage-dependent and superimposed stochastically onto whole-cell Ca2+-activated-K+ (BKCa) currents. Charybdotoxin (ChTX, 200 nmol/L), a selective blocker of BKCa channels, completely inhibited STOCs within 10 min. STOCs activity was greatly suppressed when extracellular Ca2+ concentration decreased from 1.8 mmol/L to 200 nmol/L, further removal of Ca2+ abolished STOCs activity. Ca2+ ionophore A23187 (10 μmol/L) increased STOCs activity significantly. Verapamil (20 μmol/L) and CdCl2 (200 μmol/L), two kinds of organic L-type voltage-dependent Ca2+ channels (L-VDCCs) antagonists, had little effect on STOCs. In addition, the ryanodine receptors (RyRs) agonist caffeine (5 mmol/L) significantly activated STOCs. Application of ryanodine (50 μmol/L) to block RyRs abolished STOCs, subsequent washout of ryanodine or application of caffeine failed to reproduce STOCs activity. Inhibition of inositol 1,4,5-trisphosphate receptors (IP3Rs) by 2APB (40 μmol/L) greatly suppressed the activity of STOCs, application of caffeine (5 mmol/L) in the presence of 2APB caused a burst of outward currents followed by inhibition of STOCs. These results suggest that STOCs in porcine coronary ASMCs are mediated by BKCa channels. Extracellular Ca2+ is essential for STOCs activity, while Ca2+ entry through L-VDCCs has little effect on STOCs. C14 in the Intestine of Juvenile Sea Cucumber, Apostichopus japonicus显示文摘Viable cell count was used to determine whether Metschnikowia sp.C14 can colonize the intestine of juvenile sea cucumber Apostichopus japonicus.Sea cucumber individuals were divided into two groups,which were fed the control diet for 38 days or the C14-supplemented diet at 105 cells g−1 diet for 28 days,then the control diet from day 29 to day 38.The number of C14 cells in the intestine of sea cucumber fed the C14-supplemented diet significantly increased from day 7 to day 28,and decreased from day 29 to day 38.Sea cucumber fed with the diet containing C14 showed a significant increase in trypsin activity and lipase activity from day 21 to day 33 compared with the control.Feeding C14 significantly improved the phagocytic activity and respiratory burst in coelomocytes from day 21 to day 35 and from day 14 to day 38,respectively.In addition,there was an obvious enhancement in lysozyme activity(from day 21 to day 38 or day 33),phenoloxidase activity(from day 21 to day 28)and total nitric oxide synthase activity(from day 14 to day 38)in coelomic fluid supernatant and/or coelomocyte cell lysate supernatant compared with the control.There were significant positive correlations between the number of C14 cells colonizing the intestine and trypsin activity of the intestine,lysozyme activity of the coelomic fluid supernatant and coelomocyte lysate supernatant from sea cucumber.These data suggested that the number of C14 cells should be maintained at 105 cfu(colony-forming units)g−1 intestine material for the maximum benefit. | LI Ming BAO Pengyun SONG Jian DING Jianfeng LIU Yubin MA Yuexin | 2020 | Journal of Ocean University of China2020,19,1: | 1 |
| 11 | Whole-genome methylation analysis reveals epigenetic variation between wild-type and nontransgenic cloned,ASMT transgenic cloned dairy goats generated by the somatic cell nuclear transfer显示文摘Background:SCNT(somatic cell nuclear transfer)is of great significance to biological research and also to the livestock breeding.However,the survival rate of the SCNT cloned animals is relatively low compared to other transgenic methods.This indicates the potential epigenetic variations between them.DNA methylation is a key marker of mammalian epigenetics and its alterations will lead to phenotypic differences.In this study,ASMT(acetylserotonin-Omethyltransferase)ovarian overexpression transgenic goat was produced by using SCNT.To investigate whether there are epigenetic differences between cloned and WT(wild type)goats,WGBS(whole-genome bisulfite sequencing)was used to measure the whole-genome methylation of these animals.Results:It is observed that the different m Cp G sites are mainly present in the intergenic and intronic regions between cloned and WT animals,and their CG-type methylation sites are strongly correlated.DMR(differentially methylated region)lengths are located around 1000 bp,mainly distributed in the exonic,intergenic and intronic functional domains.A total of 56 and 36 DMGs(differentially methylated genes)were identified by GO and KEGG databases,respectively.Functional annotation showed that DMGs were enriched in biological-process,cellularcomponent,molecular-function and other signaling pathways.A total of 10 identical genes related to growth and development were identified in GO and KEGG databases.Conclusion:The differences in methylation genes among the tested animals have been identified.A total of 10 DMGs associated with growth and development were identified between cloned and WT animals.The results indicate that the differential patterns of DNA methylation between the cloned and WT goats are probably caused by the SCNT.These novel observations will help us to further identify the unveiled mechanisms of somatic cell cloning technology,particularly in goats. | Hao Wu Wendi Zhou Haijun Liu Xudai Cui Wenkui Ma Haixin Wu Guangdong Li Likai Wang Jinlong Zhang Xiaosheng Zhang Pengyun Ji Zhengxing Lian Guoshi Liu | 2023 | Journal of Animal Science and Biotechnology2023,14,1: | 0 |
| 12 | An Evaluation on the Ratio of Plant to Animal Protein in the Diet of Juvenile Sea Cucumber(Apostichopus japonicus):Growth,Nutrient Digestibility and Nonspecific Immunity显示文摘This study was conducted to evaluate the effects of plant/animal(P/A) protein ratios(viz.1:4, 1:3, 1:2, 1:1,2:1, 3:1, 4:1) on growth performance, body composition, apparent digestibility of diets, and nonspecific immunity of juvenile sea cucumber(Apostichopus japonicus). Sea cucumbers were divided into 21 plastic tanks, and each tank was stocked with 15 individuals(initial weight: about 23.73 g). Each feed was allocated to three replicates of sea cucumbers. The feeding experiment lasted for 50 days. Results indicated that weight gain rate(WGR) and body wall weight(BWW) significantly increased as dietary ratio of P/A increased from 1:4 to 3:1, and then decreased significantly with further increase of this ratio(P < 0.05). The body wall coefficient(BWC) showed a similar tendency to WGR and BWW, but no significance was detected among dietary treatments(P > 0.05). The apparent digestibility of dry matter, protein and lipid increased with ratio of P/A increasing from 1:4 to 2:1(P < 0.05), and then decreased with further increase of this ratio. Correspondingly, activities of trypsin and amylase were significantly increased as P/A increased from 1:4 to 2:1(P < 0.05). The activities of SOD and CAT showed a similar trend with WGR, with the highest value observed in the ratio of 1:2 and 1:1, respectively. Results above showed that moderate or relatively higher ratio of P/A protein(1:1-3:1) significantly increased the growth performance, apparent digestibility, and nonspecific immunity of sea cucumber. | 13 | Realizing the Long Lifespan of Molybdenum Trioxide in Aqueous Aluminum Ion Batteries Through Potential Regulation显示文摘MoO_(3) is one of the most promising anode materials for aqueous aluminum batteries due to its high theoretical capacity and suitable aluminum insertion/de-insertion potential.However,the inferior cycling stability limits its further application,and the failure mechanism is still unclear.In this article,we provide a straightforward potential regulation technique to manage phase evolution during the charge/discharge process,which ultimately results in a markedly enhanced MoO_(3) electrode cycling stability.The failure mechanism study reveals that the excessive oxidation of the electrode during charge/discharge generates the H_(0.34)MoO_(3) phase,which has high solubility and is the primary cause of MoO_(3) deactivation.Although the dissolved Mo species will be deposited onto the electrode sheet again,the deposition is not electrochemically active and cannot contribute to the capacitance.Controlling the cutoff potential prevented the production of H_(0.34)MoO_(3),resulting in excellent cycling performance(80.1% capacity retention after 4000 cycles).The as-assembled α-MoO_(3)//MnO_(2) full battery exhibits high discharge plateaus(1.4 and 0.9 V),large specific capacity(200 mAhg^(-1) at 2 Ag^(-1)),and ultra-high coulombic efficiency(99%).The research presented here may contribute to the development of highly stable electrode materials for aqueous batteries. | Haodong Fan Xuejin Li Jie Zhou Xiaoning Wang Xiuli Gao Haoyu Hu Li Zhou Tonghui Cai Yongpeng Cui Pengyun Liu Qingzhong Xue Zifeng Yan Wei Xing | 2023 | Renewables2023,1,4: | 0 |
| 14 | Structured road-oriented motion planning and tracking framework for active collision avoidance of autonomous vehicles显示文摘This paper proposes a novel motion planning and tracking framework based on improved artificial potential fields(APFs) and a lane change strategy to enhance the performance of the active collision avoidance systems of autonomous vehicles on structured roads. First, an improved APF-based hazard evaluation module, which is inspired by discrete optimization, is established to describe driving hazards in the Frenet-Serret coordinate. Next, a strategy for changing lane is developed in accordance with the characteristics of the gradient descent method(GDM). On the basis of the potential energy distribution of the target obstacle and road boundaries, GDM is utilized to generate the path for changing lane. In consideration of the safety threats of traffic participants, the effects of other obstacles on safety are taken as additional safety constraints when the lane-changing speed profile for ego vehicles is designed. Then, after being mapped into the Cartesian coordinate, the feasible trajectory is sent to the tracking layer, where a proportional-integral control and model predictive control(PI-MPC) based coordinated controller is applied. Lastly, several cases composed of different road geometrics and obstacles are tested to validate the effectiveness of the proposed algorithm. | 15 | Discovery of novel exceptionally potent and orally active c-MET PROTACs for the treatment of tumors with MET alterations显示文摘Various c-mesenchymal-to-epithelial transition(c-MET) inhibitors are effective in the treatment of non-small cell lung cancer;however, the inevitable drug resistance remains a challenge, limiting their clinical efficacy. Therefore, novel strategies targeting c-MET are urgently required. Herein, through rational structure optimization, we obtained novel exceptionally potent and orally active c-MET proteolysis targeting chimeras(PROTACs) namely D10 and D15 based on thalidomide and tepotinib. D10 and D15 inhibited cell growth with low nanomolar IC_(50) values and achieved picomolar DC_(50) values and>99% of maximum degradation(D_(max)) in EBC-1 and Hs746T cells. Mechanistically, D10 and D15dramatically induced cell apoptosis, G1 cell cycle arrest and inhibited cell migration and invasion.Notably, intraperitoneal administration of D10 and D15 significantly inhibited tumor growth in the EBC-1 xenograft model and oral administration of D15 induced approximately complete tumor suppression in the Hs746T xenograft model with well-tolerated dose-schedules. Furthermore, D10 and D15 exerted significant anti-tumor effect in cells with c-MET^(Y1230H) and c-MET^(D1228N) mutations, which are resistant to tepotinib in clinic. The concentration of IFN-α in serum and that inthe supernatant of peripheral blood mononuclear cells (PBMCs)cultured after stimulation with CpG ODN2216 were examinedboth in CML patients and in the healthy controlsRESULTS There was significant reduction in the numberof circulating pDCs, serum concentration of IFN-α and thecapacity of IFN-α producing PBMCs in CML patients comparedwith those in healthy control individuals (P < 0.001). After theactive treatment with IFN-α and hydroxyurea, the quantity andfunction of pDCs were increased in stabilized patients, especiallythe function of pDCs in 2 patients achieving major cytogeneticresponse (MCR). The proportion and function of pDCs and theserum levels of IFN were inversely correlated with both WBC andage of the patients with CML, and positively correlated with thestate of the illness.CONCLUSION CML patients had a reduced number anddysfunction of circulating pDCs. | 17 | Construction of Frame-Structured Flexible MXene Film Electrode to Achieve High Areal Capacitance Micro-Supercapacitor显示文摘Two-dimensional MXene-based film materials as flexible electrodes have been widely studied in wearable microsupercapacitors(MSCs).However,the existence of strong van derWaals interactions leads to serious self-stacking ofMXene layers,resulting in poor ionic dynamics and loss of active sites,which causes MXene film electrodes to exhibit low capacitance and poor rate performance in practical studies.To solve this,a frame-structured hybrid film(labeled as CN-MX hybrid film)is constructed by introducing intercalating agents(nanometer g-C_(3)N_(4))into MXene layers.In this unique hybrid film,the g-C_(3)N_(4)nanoparticles rationally occupy the interspace between MXene layers so as to alleviate layer stacking,thus effectively expanding the electrochemically active surface and promoting proton transfer.Synergistic pseudocapacitance inducted by g-C_(3)N_(4)surface groups,consequently,the CN-MX hybrid film electrode achieves an enhanced capacitive capability.In the three-electrode system,this frame-structured film electrode exhibits an ultra-high areal capacitance of 1932.8 mF cm^(−2).The assembled symmetry flexible MSC device based on CN-MX hybrid film can achieve an energy density of 2.28μWh cm^(−2)at 0.075 mW cm^(−2),as well as a superior cyclic stability with 90.4%retention after 700 cycles in alternating 90o bending and releasing states,revealing its potential in practical applications. | Yesheng Wang Yongpeng Cui Dongqing Kong Xiaoning Wang Wenting Feng Pengyun Liu Tonghui Cai Xuejin Li Lianming Zhao Debin Kong Linjie Zhi Qingzhong Xue Zifeng Yan Wei Xing | 2023 | Renewables2023,1,2: | 0 |
