| 2 | Surveillance, epidemiology, and pathogen spectrum of hand, foot, and mouth disease in mainland of China from 2008 to 2017显示文摘Hand,foot and mouth disease(HFMD)was reported in May 2,2008 to be the 38th legally notifiable disease in China's National Notifiable Disease Reporting and Surveillance System.In order to solve the infection,an extensive three-level HFMD surveillance laboratory network was established.In this study,the framework of that network is assessed and the incidence of HFMD in China from 2008 to 2017 is reported using a descriptive epidemiologic method.During these 10 years,a series of techniques have been widely applied in all the network laboratories.Using information and material obtained from the network,a virus bank and database containing 18,238 viruses were established.Nationally,18,184,834 HFMD cases,including 152,436 severe cases and 3633 fatal cases,were reported in mainland of China.The average annual incidence in the population was 133.99/100,000 people,with a maximum incidence of 205.06/100,000 people in 2014.The incidence and mortality rates of HFMD were the highest in children aged 1–2 years.The numbers of reported cases fluctuated,with a high incidence observed every 2 years.An overall increase in the number of reported cases was also observed throughout the study period.Despite this,the incidence of severe cases and the mortality rate have been decreasing.High-risk regions are located in southern,eastern,and central China.Two peaks of HFMD infection cases were observed annually except for Northeast China.Different proportions of enterovirus serotypes were observed during the studied years.The predominant enterovirus varies from year to year,but the disease severity is always closely related to the specific serotype.EV-A71 is the dominant serotype associated with severe and fatal cases,with constituent ratios of 70.03%and 92.23%,respectively.The studied highly sensitive and efficient surveillance network provides information that is critical for prevention and control of the disease.It is extremely necessary and important to continuously conduct extensive virological surveillance for HFMD. | Tianjiao Ji Taoli Han Xiaojuan Tan Shuangli Zhu Dongmei Yan Qian Yang Yang Song Aili Cui Yan Zhang Naiying Mao Songtao Xu Zhen Zhu Dandan Niu Yong Zhang Wenbo Xu | 2019 | Biosafety and Health2019,1,1: | 26 |
| 3 | Promoter methylation and mRNA expression of DKK-3 and WIF-1 in hepatocellular carcinoma显示文摘AIM:To investigate the promoter methylation status and mRNA expression of DKK-3 and WIF-1 gene in hepatocellular carcinoma(HCC).METHODS:DKK-3 and WIF-1 acted as Wnt-antagonists and tumor suppressors,but hypermethylation of the gene promoter and low mRNA expression activated Wnt signaling aberrantly and induced the development of HCC.Methylation status of the DKK-3 and WIF-1 gene promoter was investigated using methylation specific polymerase chain reaction(PCR) in tumor and adjacent non-cancerous tissues from 33 HCC patients and 20 normal liver tissues served as control.The expression of DKK-3 and WIF-1 mRNA was also determined by real-time quantitative reverse transcriptase PCR.The relationship between methylation,mRNA expression,and clinical data,as well as methylation and mRNA expression of the two genes were analyzed.RESULTS:The methylation of DKK-3 and WIF-1 genes in HCC increased significantly compared with adjacent non-cancerous tissues and normal control tissues(χ2 =7.79,P < 0.05;χ2 = 4.89,P < 0.05),and no significant difference in methylation between adjacent non-cancerous tissues and normal control tissues was observed.In HCC tissues,significant differences in the DKK-3 promoter methylation were observed in age and cirrhosis,and significant differences of the WIF-1 promoter methylation were observed in HBsAg and cirrhosis.The average expression of DKK-3 mRNA in HCC and adjacent non-cancerous tissues was increased significantly compared with normal control tissues.The average expression of WIF-1 mRNA showed no significant difference among the three tissues.The mRNA expression of DKK-3 gene in HCC was decreased as the pathological grade increased.CONCLUSION:The aberrant promoter methylation and decreased expression of DKK-3 and WIF-1 may be an important mechanism in HCC,and may be a far-reaching significance in early diagnosis and therapy of HCC. | Zhen Ding Ye-Ben Qian Li-Xin Zhu Qi-Ru Xiong | 2009 | World Journal of Gastroenterology2009,15,21: | 22 |

| 5 | p53 upregulated by HIF-la promotes hypoxiainduced G2/M arrest and renal fibrosis in vitro and in vivo显示文摘Hypoxia plays an important role in the genesis and progression of renal fibrosis.The underlying mechanisms, however, have not been sufficiently elucidated. We examined the role of p53 in hypoxia-induced renal fibrosis in cell culture (human and rat renal tubular epithelial cells) and a mouse unilateral ureteral obstruction (UUO) model. Cell cycle of tubular cells was determined by flow cytometry, and the expression of profibrogenic factors was determined by RT-PCR, immunohistochemistry, and western blotting. Chromatin immunoprecipitation and luciferase reporter experiments were performed to explore the effect of HIF-lα on p53 expression. We showed that, in hypoxic tubular cells, p53 upregulation suppressed the expression of CDK1 and cyclins Bl and DI, leading to cell cycle (G2/M) arrest (or delay) and higher expression of TGF-β, CTGF, collagens, and fibronectin. p53 suppression by siRNA or by a specific p53 inhibitor (PIF-α) triggered opposite effects preventing the G2/M arrest and profibrotic changes. In vivo experiments in the UUO model revealed similar antifibrotic results following intraperitoneal administration of PIF-α(2.2 mg/kg). Using gain-of-function, loss-of-function, and luciferase assays, we further identified an HRE3 region on the p53 promoter as the HIF-lα-binding site. The HIF-la-HRE3 binding resulted in a sharp transcriptional activation of p53. Collectively, we show the presence of a hypoxia-activated, p53-responsive profibrogenic pathway in the kidney. | 11 | Prevalence and characteristics of MAFLD in Chinese adults aged 40 years or older:A community-based study显示文摘Background: Nonalcoholic fatty liver disease(NAFLD) was recently proposed to be renamed metabolic dysfunction-associated fatty liver disease(MAFLD) with the diagnostic criteria revised. We investigated the similarities and differences in the prevalence and clinical characteristics of MAFLD and NAFLD in Chinese adults. Methods: A cross-sectional study of 9980 Chinese individuals aged 40 years or older was performed between 2011 and 2012 using randomized, stratifed cluster sampling in Shanghai, China. A detailed questionnaire and the results of abdominal ultrasonography, a standardized 2-h 75-g oral glucose tolerance test and blood biochemical examinations were collected. Results: A total of 9927 subjects were included in this study. The prevalence of MAFLD(40.3%) was significantly higher than that of NAFLD(36.9%)( P < 0.05). MAFLD was highly prevalent in type 2 diabetes mellitus(T2DM)(53.8%), impaired fasting glucose(35.7%) and impaired glucose tolerance(40.9%). High risk of advanced fbrosis based on fbrosis-4 was highly prevalent(14.7%) in lean MAFLD with T2DM. Among 9927 subjects, 3481(35.1%) fulflled the diagnostic criteria for MAFLD and NAFLD(MAFLD + NAFLD +), 521(5.2%) MAFLD + NAFLD-, and 181(1.8%) MAFLD-NAFLD +. The MAFLD + NAFLD-group had more signifcant metabolic disorders than those in the MAFLD + NAFLD + group(all P < 0.05). Among MAFLD-NAFLD + subjects, 82.9% had metabolic disorders. Conclusions: The new defnition of MAFLD may better reflect the pathogenesis related to metabolism. | 13 | Expressions of chromogranin A and cathepsin D in human primary hepatocellular carcinoma显示文摘AIM To determine the expression and clinicalsignificance of chromogranin A and cathepsin Din hepatocellular carcinoma(HCC).METHODS Double immunofluorescence stain-ing techniques combined with laser confocalscanning microscopy(LSCM)was used toinvestigate chromogranin A and cathepsin Dexpressions in 85 HCC patients.RESULTS Cathepsin D was expressed in :3normal liver tissues,while in HCC the stainingshowed regional variation and the fraction ofstrongly stained cells increased as the tumorsbecame less differentiated and usually clinicallymore malignant.Cells which showed strongpositivity for cathepsin D were present in 71/85(83.5%)cases.Strong expression of cathepsinD in cancer cells was related tohistopathological features.They were morecommon in grade 3-4(26/28,92.9%)and grade2(46/53,86.8%)tumors than in grade 1 tumors(1/4,25.0%)(P<0.01).No significantcorrelation was found between age andcathepsin D expression.In patients with positivecathepsin D reaction,the mean age was 52.1±2.8 years(range 32-68 years)and in the groupwith negative reaction,the mean age was 51.3±4.5 years(range 28-71 years).No obvious relationship was observed between CgAexpression in cancer cells and thehistopathological features.The CgA positiverate was 75.0%(3/4)in grade 1,71.7%(38/53)in grade 2,and 71.4%(20/28)in grade 3-4(P>0.05)tumors.The coexpression of CgA andcathepsin D was found by double labeledimmunofluorescence staining techniques.Theprocessing of cathepsin D was disturbed in HCCcells and accumulated in the cells.Cathepsin Dhad proteolytic activity and autocrine mitogeniceffect,suggesting their functions in invasion.These findings demonstrated that the expressionof cathepsin D in HCC had prognostic value.CONCLUSION Chromogranin A and cathepsin Dare expressed in a high proportion of HCC andthe existence of cathepsin D in HCC might berelated to processing of CgA.This is clearly asubject for further studies because of itspotential clinical applications. | Xiao Feng Huang Chun Mei Wang Xiao Wen Dai Zhen Jiang Li Bo Rong Pan Li Bin Yu Bin Qian Li Fang | 2000 | World Journal of Gastroenterology2000,6,5: | 12 |
| 14 | Timing and splitting of nitrogen fertilizer supply to increase crop yield and efficiency of nitrogen utilization in a wheat–peanut relay intercropping system in China显示文摘Agronomically optimizing the timing and rates of nitrogen(N) fertilizer application can increase crop yield and decrease N loss to the environment. Wheat(Triticum aestivum L.)–peanut(Arachis hypogaea L.) relay intercropping systems are a mainstay of economic and food security in China. We performed a field experiment to investigate the effects of N fertilizer on N recovery efficiency, crop yield, and N loss rate in wheat–peanut relay intercropping systems in the Huang-Huai-Hai Plain, China during 2015–2017. The N was applied on the day before sowing, the jointing stage(G30) or the booting stage(G40) of winter wheat, and the anthesis stage(R1) of peanut in the following percentage splits: 50-50-0-0(N1), 35-35-0-30(N2), and 35-0-35-30(N3), using 300 kg N ha-1, with 0 kg N ha-1(N0) as control. ^(15)N-labeled(20.14 atom %) urea was used to trace the fate of N in microplots. The yields of wheat and peanut increased by 12.4% and 15.4% under the N2 and N3 treatments, relative to those under the N1 treatment. The ^(15)N recovery efficiencies( ^(15)NRE) were 64.9% and 58.1% for treatments N2 and N3, significantly greater than that for the N1 treatment(45.3%). The potential N loss rates for the treatments N2 and N3 were23.7% and 7.0%, significantly lower than that for treatment N1(30.1%). Withholding N supply until the booting stage(N3) did not reduce the wheat grain yield; however, it increased the N content derived from ^(15)N-labeled urea in peanuts, promoted the distribution of ^(15)N to pods, and ultimately increased pod yields in comparison with those obtained by topdressing N at jointing stage(N2). In comparison with N2, the N uptake and N recovery efficiency(NRE) of N3 was increased by 12.0% and 24.1%,respectively, while the apparent N loss decreased by 16.7%. In conclusion, applying N fertilizer with three splits and delaying topdressing fertilization until G40 of winter wheat increased total grain yields and NRE and reduced N loss. LEK1 and CMF1, which are its murine and chicken orthologs, however, are implicated in muscle differentiation and reportedly not distributed at kinetochores.We therefore conducted several assays to clarify this issue. The typical centromere staining patterns were observed in mitotic cells from both human primary culture and murine, canine, and mink cell lines. A C-terminal portion of LEK1 also conferred centromere localization. Our analysis further suggests conserved kinetochore localization of mammalian mitosin orthologs. Moreover, mitosin was associated preferentially with kinetochores of unaligned chromosomes. It was also constantly transported from kinetochores to spindle poles by cytoplasmic dynein. These properties resemble those of other kinetochore proteins important for the spindle checkpoint, thus implying a role of mitosin in this checkpoint. | 19 | Outcomes of the national programme on prevention of mother-to-child transmission of hepatitis B virus in China,2016-2017显示文摘Background:In addition to providing free hepatitis B vaccine(HBvacc)series to all infants in China since 2005,the national programme on prevention of mother-to-child transmission(PMTCT)of hepatitis B virus(HBV)started providing free hepatitis B immunoglobulin for all new-borns born to hepatitis B surface-antigen(HBsAg)positive mothers in 2010.However,few studies have evaluated the effectiveness of the PMTCT programme.Therefore,we aimed to investigate the outcomes of the programme and identify associated factors.Method:Using a cross-sectional study design,we collected data on 4112 pairs of HBsAg-positive mothers and their children aged 7-22 months in four representative provinces through interviews and medical record review.We tested HBsAg and hepatitis B surface antibody(anti-HBs)of children by enzyme-linked immunosorbent assay at designated maternal and child hospital laboratories.We used logistic regression to analyse factors associated with child HBsAg and anti-HBs positivity.Results:Thirty-five children were HBsAg positive,indicating the mother-to-child transmission(MTCT)rate was 0.9%(0.6-1.1%).The anti-HBs positive rate was 96.8%(96.3-97.4%).Children receiving HBvacc between 12 and 24h of birth were 2.9 times more likely to be infected than those vaccinated in less than 12 h(adjusted odds ratio[aOR]=2.9,95%confidence interval[CI]:1.4-63,P=0.01).Maternal hepatitis B e-antigen(HBeAg)positivity was associated with higher MTCT rate(aOR=79.1,95%Cl:10.8-580.2,P<0.001)and lower anti-HBs positive rate(aOR=0.4,95%CI:03-0.6,P<0.001).Children with low birth weight(LBW)were 60%less likely to be anti-HBs positive than those with normal birth weight(aOR=0A,95%CI:02-0.8,P=0.01).Conclusions:The MTCT rate was lower than the 2030 WHO elimination goal,which implies the programme is on track to achieve this target.As earlier HBvacc birth dose(HBvcc-BD)was associated with lower MTCT rate,we suggest that the PMTCT programme work with the Expanded Programme on Immunization(EPI)to modify the current recommendation for early HBvcc-BD to a requirement Our finding that LBW was associated with lower anti-HBs positivity points to the need for further studies to understand factors associated with these risks and opportunities for program strengthening.The programme needs to ensure providing essential test to identify HBeAg-positive mothers and their infants and provide them with appropriate medical care and follow-up. | Ya-Ping Qiao Min Su Yao Song Xiao-Yan Wang Zhen Li Yan-Lin Li Li-Xia Dou Qian Wang Katrina Hann Guo-Min Zhang Xiao-Na Huang Yu-Ning Yang Xi Jin Ai-Ling Wang | 2019 | Infectious Diseases of Poverty2019,8,4: | 10 |
| 20 | Metformin attenuates motility,contraction,and fibrogenic response of hepatic stellate cells in vivo and in vitro by activating AMP-activated protein kinase显示文摘AIM To investigate the effect of metformin on activated hepatic stellate cells(HSCs) and the possible signaling pathways involved. METHODS A fibrotic mouse model was generated by intraperitoneal injection of carbon tetrachloride(CCl_4) and subsequent treatment with or without metformin. The level of fibrosis was detected by hematoxylin-eosin staining, Sirius Red staining, and immunohistochemistry. The HSC cell line LX-2 was used for in vitro studies. The effect of metformin on cell proliferation(CCK8 assay),motility(scratch test and Transwell assay), contraction(collagen gel contraction assay), extracellular matrix(ECM) secretion(Western blot), and angiogenesis(ELISA and tube formation assay) was investigated. We also analyzed the possible signaling pathways involved by Western blot analysis.RESULTS Mice developed marked liver fibrosis after intraperitoneal injection with CCl_4 for 6 wk. Metformin decreased the activation of HSCs, reduced the deposition of ECM, and inhibited angiogenesis in CCl_4-treated mice. Platelet-derived growth factor(PDGF) promoted the fibrogenic response of HSCs in vitro, while metformin inhibited the activation, proliferation, migration, and contraction of HSCs, and reduced the secretion of ECM. Metformin decreased the expression of vascular endothelial growth factor(VEGF) in HSCs through inhibition of hypoxia inducible factor(HIF)-1α in both PDGF-BB treatment and hypoxic conditions, and it down-regulated VEGF secretion by HSCs and inhibited HSC-based angiogenesis in hypoxic conditions in vitro.
