| 3 | Severe burn injury alters intestinal microbiota composition and impairs intestinal barrier in mice显示文摘Background:The intestinal barrier integrity is crucial for maintaining intestinal homeostasis,and the mechanisms of intestinal barrier disruption induced by burn injury remain obscure.This study was aimed to investigate the changes of intestinal microbiota and barrier function in burned mice to further comprehend the mechanisms of burn-induced intestinal barrier dysfunction.Methods:Samples were from mice inflicted with 30%total body surface area(TBSA)full-thickness burns.The intestinal permeability,tight junction proteins expressions,zonula occludens-1(ZO-1)localization,inflammatory cytokines expressions,and short-chain fatty acids(SCFAs)contents were determined.The microbial community was assessed via 16S rDNA Illumina sequencing.Results:The intestinal permeability was increased after severe burn injury,peaking at 6 h post-burn,with approximately 20-folds of the control(p<0.001).The expression of tight junction proteins(ZO-1,occludin,claudin-1,and claudin-2)was significantly altered(p<0.05).The ZO-1 morphology was dramatically changed following burn injury.The fecal SCFAs’contents(acetate,propionate,butyrate,isobutyrate,and isovalerate)were noticeably declined after burn injury(p<0.05).The expressions of pro-inflammatory cytokines(interleukin(IL)-1βand IL-6)in ileal mucosa were increased,whereas the expressions of anti-inflammatory cytokines(IL-4 and IL-13)were decreased following burn injury(p<0.05).In addition,burned mice showed an alteration of intestinal microbial community,such as decreased diversity,reduced Bacteroidetes abundance,and increased Firmicutes abundance.Conclusions:The severe burn-induced intestinal barrier dysfunction is along with the alterations of microbial community. | Yanhai Feng Yalan Huang Yu Wang Pei Wang Fengjun Wang | 2019 | Burns & Trauma2019,7,1: | 11 |

| 6 | Inactivation of TFEB and NF-kB by marchantin M alleviates the chemotherapy-driven pro-tumorigenic senescent secretion显示文摘It is critical to regulate the senescence-associated secretory phenotype(SASP) due to its effect on promoting malignant phenotypes and limiting the efficiency of cancer therapy. In this study, we demonstrated that marchantin M(Mar-M, a naturally occurring bisbibenzyl) suppressed proinflammatory SASP components which were elevated in chemotherapy-resistant cells. Mar-M treatment attenuated the pro-tumorigenic effects of SASP and enhanced survival in drug-resistant mouse models.No toxicity was detected on normal fibroblast cells or in animals following this treatment. Inactivation of transcription factor EB(TFEB) and nuclear factor-k B(NF-k B) by Mar-M significantly accounted for its suppression on the components of SASP. Furthermore, inhibition of SASP by Mar-M contributed to a synergistic effect during co-treatment with doxorubicin to lower toxicity and enhance antitumor efficacy. Thus, chemotherapy-driven pro-inflammatory activity, seen to contribute to drug-resistance, is an important target for Mar-M. | 18 | Simulation of directional propagation of hydraulic fractures induced by slotting based on discrete element method显示文摘Hydraulic fracturing(HF)technology can safely and efficiently increase the permeability of coal seam,which is conducive to CBM exploration and prevent coal and gas outburst.However,conventional HF fractures tend to expand in the direction of maximum principal stress,which may be inconsistent with the direction of fracturing required by the project.Therefore,the increased direction of coal seam permeability is different from that expected.To solve these problems,PFC2D software simulation is used to study directional hydraulic fracturing(DHF),that is the combination of slotting and hydraulic fracturing.The effects of different slotting angles(θ),different horizontal stress difference coefficients(K)and different injection pressures on DHF fracture propagation are analyzed.The results show that the DHF method can overcome the dominant effect of initial in-situ stress on the propagation direction of hydraulic fractures and control the propagation of fractures along and perpendicular to the slotting direction when θ,K and liquid injection pressure are small.When the DHF fracture is connected with manual slotting,the pressure will shake violently,and the fracturing curve presents a multi-peak type.The increase and decrease of particle pressure around the fracturing hole reflect the process of pressure accumulation and fracture propagation at the fracture tip respectively.Compared with conventional HF,DHF can not only shorten the fracturing time but also make the fracture network more complex,which is more conducive to gas flow.Under the action of in-situ stress,the stress between slots will increase to exceed the maximum horizontal principal stress.Moreover,with the change in fracturing time,the local stress of the model will also change.Hydraulic fractures are always expanding to the area with large local stress.The research results could provide certain help for DHF theoretical research and engineering application. | Kai Wang Guodong Zhang Feng Du Yanhai Wang Liangping Yi Jianquan Zhang | 2023 | Petroleum2023,9,4: | 0 |
