| 1 | Protein trans-splicing based dual-vector delivery of the coagulation factor Ⅷ gene显示文摘A dual-vector system was explored for the delivery of the coagulation factor VIII gene,using intein-mediated protein trans-splicing as a means to produce intact functional factor VIII post-translationally.A pair of eukaryotic expression vectors,expressing Ssp DnaB intein-fused heavy and light chain genes of B-domain deleted factor VIII (BDD-FVIII),was constructed.With transient co-transfection of the two vectors into 293 and COS-7 cells,the culture supernatants contained (137±23) and (109±22) ng mL–1 spliced BDD-FVIII antigen with an activity of (1.05±0.16) and (0.79±0.23) IU mL–1 for 293 and COS-7 cells,respectively.The spliced BDD-FVIII was also detected in supernatants from a mixture of cells transfected with inteinfused heavy and light chain genes.The spliced BDD-FVIII protein bands from cell lysates were visualized by Western blotting.The data demonstrated that intein could be used to transfer the split factor VIII gene and provided valuable information on factor VIII gene delivery by dual-adeno-associated virus in hemophilia A gene therapy. | ZHU FuXiang,LIU ZeLong,CHI XiaoYan & QU HuiGe Life Science College of Ludong University,Yantai 264025,China | 2010 | Science China(Life Sciences)2010,53,6: | 27 |
| 2 | Circular RNA PIP5K1A promotes colon cancer development through inhibiting mi R-1273a显示文摘BACKGROUND Circular RNAs (circRNAs) are considered to be highly stable due to the closed structure, which are predominately correlated with the development and progression of a wide variety of cancers. Colon cancer is one of the most common malignancies worldwide. A recent study demonstrated the upregulated expression of circPIP5K1A in non-small cell lung cancer. However, few studies have investigated the relationship between circ_0014130 level and colon cancer. Therefore, elucidating the underlying mechanisms of circPIP5K1A’s role may help with the identification of novel diagnostic and therapeutic targets for colon cancer. AIM To investigate the status of circPIP5K1A in colon cancers and its effects on the modulation of cancer development. METHODS The expression level of circPIP5K1A in tissue and serum samples from colon cancer patients, as well as human colonic cancer cell lines was detected by realtime quantitative reverse transcription-polymerase chain reaction. Following the transfection of specifically synthesized small interfering RNA (siRNA) into colon cell lines, we used Hoechst staining assay to measure the ratio of cell death in the absence of circPIP5K1A. Moreover, we also used the Transwell assay to assess the migratory function of colon cells overexpressing circPIP5K1A. Additionally, we employed a series of bioinformatics prediction programs to predict the potential of circPIP5K1A-targeted miRNAs and mRNAs. The miR-1273a vector was constructed, and then transfected with or without circPIP5K1A vector into colon cancer cells. Afterwards, the expression of activator protein 1 (AP-1), interferon regulating factor 4 (IRF-4), caudal type homeobox 2 (CDX-2), and zinc finger of the cerebellum 1 (Zic-1) was detected by western blotting. RESULTS CircPIP5K1A was significantly upregulated in colon cancer tissue relative to their adjacent normal tissues. Knockdown of circPIP5K1A in colon cancer cells impaired cell viability and suppressed cell invasion and migration, while enforced expression of circPIP5K1A exhibited the opposite effects on cell migration. Bioinformatics prediction program predicted that the association of circPIP5K1A with miR-1273a, as well as AP-1, IRF-4, CDX-2, and Zic-1. Subsequent studies showed that overexpression of circPIP5K1A augmented the expression of AP-1 but attenuated the expression of IRF-4, CDX-2, and Zic-1. Reciprocally, overexpression of miR-1273a abrogated the oncogenic function of circPIP5K1A in colon cancers. | 3 | Synthesis and Properties of High Performance Functional Polyimides Containing Rigid Nonplanar Conjugated Fluorene Moieties显示文摘A diamine(WuFDA) containing vertical rigid non-planar conjugated fluorene moiety and low polarizability group(C―F)was designed and synthesized through three steps of reactions(halogenated reaction, Suzuki coupling reaction, and reduction reaction).Four kinds of high performance functional polyimides(WuFPI-6 F, WuFPI-BP, WuFPI-BT, and WuFPI-PM) were thus prepared by the condensation polymerization of WuFDA with four commercial dianhydride 6 FDA, BPDA, BTDA, and PMDA, respectively. The polyimides exhibited low dielectric constant, excellent thermal stability, outstanding solubility, good film-forming property, and mechanical properties. The dielectric constants of the polyimides were in the range of 2.28-2.88(f = 10~4 Hz). The 5% weight-loss temperatures(Td 5%)in nitrogen were in the range of 555-584 °C, and the glass transition temperatures(T_g) were in the range of 408-448 °C. The weight loss of WuFPI-BP maintaining at 450 and 500 °C for half an hour was only 0.33% and 1.26%, respectively. All the WuFPIs could be dissolved in almost all organic solvents, even chloroform. The tensile strength and tensile modulus of these films were in the ranges of 78.6-85.7 MPa and 3.1-3.2 GPa, respectively. In addition, the polyimides displayed light color with special fluorescent and resistive switching(ON-OFF) characteristics; the maximum fluorescence emission was observed at 422-424 nm in NMP solution and at 470-548 nm in film state. The memory devices with the configuration of indium tin oxide/WuFPIs/aluminum(ITO/WuFPIs/Al) exhibited distinct volatile memory characteristics of static random access memory(SRAM), with an ON/OFF current ratio of 10~5-10~6. | 4 | The effect of a secretion-enhanced heavy chain on improving intein-based dual-vector co-delivery of a full-length factor Ⅷ gene显示文摘Treatment of hemophilia A by gene therapy is adversely affected by inefficient FVIII secretion and the large FVIII gene,which is difficult to package in the promising adeno-associated virus (AAV) vectors.Inhibited secretion of FVIII is caused mainly by inefficient secretion of its heavy chain.Previously,we have employed a protein splicing-based dual-vector to co-transfer a B-domain-deleted FVIII (BDD-FVIII) gene,suggesting that the light chain,covalently ligated to a co-expressed heavy chain can improve the secretion of spliced BDD-FVIII.However,its level of secretion was affected by inefficient secretion the heavy chain.Here,we studied the effect of a mutant heavy chain with L303E/F309S substitutions,which enhance FVIII secretion on the heavy chain itself and spliced FVIII when using a protein splicing-based split-delivery of a full-length FVIII gene.Eukaryotic vectors expressing Ssp DnaB intein-fused mutant heavy and light chains were transiently co-transfected into cultured COS-7 cells.A spliced FVIII protein was seen in co-transfected cells by Western blot analysis.The heavy chain was secreted by cells transfected with the mutant heavy chain gene alone at (39±11) ng/mL and this secretion increased to (123±13) ng/mL when cells were co-transfected with the light chain gene,which was greater than the secretion of wild-type heavy chain.The amount of spliced FVIII in the culture supernatant of co-transfected cells was (86±14) ng/mL,with an activity of (0.61±0.08) IU/mL,which was greater than that of wild-type FVIII co-transfected cells.Spliced FVIII and bioactivity were also detected in the combined culture supernatant of cells individually transfected with mutant heavy and light chain gene at a higher level than that of combined wild-type heavy and light chain transfections.This suggested that the heavy chain with improved secretion markedly increased the efficacy of protein splicing-based split delivery of the full-length FVIII gene using a dual-vector.These results encourage the transfer of this technology to an animal model using a dual-AAV vector. | ZHU FuXiang YANG ShuDe LIU ZeLong MIAO Jing QU HuiGe CHI XiaoYan | 2011 | Chinese Science Bulletin2011,56,2: | 8 |
| 5 | IL-6 and INF-γ levels in patients with brucellosis in severe epidemic region,Xinjiang,China显示文摘Background:The incidence of brucellosis,which is caused by the Brucella species of bacteria,is rapidly rising worldwide;however,few studies have investigated the immune response to this pathogen and clinical biochemical features.In this paper,we examined the levels of various cytokines and inflammatory factors as well as clinical course characteristics in patients with brucellosis,in order to provide evidence for the diagnosis,assessment,and prognosis of this infectious disease.Methods:A total of 191 brucellosis inpatients(50 acute cases and 141 chronic cases),as well as 60 healthy control subjects,were included in the analysis.We investigated changes in the levels of six cytokines(IL-4,IL-6,IL-10,IL-17,TNF-α,INF-γ)and related clinical biochemical markers in patients with acute and chronic brucellosis in Xinjiang,China.Possible factors were statistically analyzed using the t test,χ2 test,z test and a multivariate logistic stepwise regression test.Results:We found that IL-4,IL-6,IL-10,IL-17,IFN-γ,and TNF-αlevels were higher in those with brucellosis than in controls(P<0.05).With regard to disease progression,procalcitonin(PCT)and C-reactive protein(CRP)levels were significantly higher in those with an acute infection compared to chronic cases(P<0.05).We found that the expression of all six cytokines tested was closely related to the degree of brucellosis using univariate logistic regression;however,only IL-6 and INF-γlevels were independent factors associated with the severity of brucellosis.Conclusions:Assessing cytokine levels in patients with acute and chronic brucellosis is not only useful for detecting the immune response,but can also be indicative of the severity of brucellosis.In particular,we propose IL-6 and INF-γlevels may be useful independent predictive factors in the clinical evaluation and diagnosis of brucellosis. | Zhi-Qiang Lin Guo-Yue Lin Wen-Wen He Chi Zhang Rui Zhang Yuan-Da Li Fan Wang Ying Qin Li Duan Dou-Dou Zhao Xiao-Juan Qu Hui Gao Hai Jiang | 2020 | Infectious Diseases of Poverty2020,9,3: | 7 |

| 10 | Enhanced plasma factor Ⅷ activity in mice via cysteine mutation using dual vectors显示文摘Hemophilia A is caused by a genetic mutation in coagulation factor Ⅷ(FⅧ) gene and gene therapy is considered to be a promising strategy for its treatment.We recently demonstrated that co-delivery of two vectors expressing M662C mutated heavy and D1828C mutated light chain genes of B-domain-deleted coagulation factor Ⅷ(BDD-FⅧ) leads to inter-chain disulfide cross-linking and improved heavy chain secretion in vitro.In this study,co-injection of both M662C and D1828C mutated BDD-FⅧ gene expression vectors into mice resulted in increased heavy chain secretion and coagulation activity in plasma in vivo.Approximately(239±56) ng mL 1 above endogenous levels of transgenic FⅧ heavy chain was found in mouse plasma using a chain-specific ELISA.For FⅧ coagulation activity,approximately(1.09±0.25) IU mL 1 above endogenous levels were detected in co-injected transgenic mouse plasma using a chromogenic assay.These data demonstrate that inter-chain disulfide bonds likely increase heavy chain secretion and coagulation activity in the plasma of transgenic mice with an improved efficacy of a dual-vector delivery of BDD-FⅧ gene.These findings support our ongoing efforts to develop a gene therapy for hemophilia A treatment using dual-AAV vectors. | ZHU FuXiang LIU ZeLong MIAO Jing QU HuiGe CHI XiaoYan | 2012 | Science China(Life Sciences)2012,55,6: | 2 |

| 12 | Spatiotemporal patterns of the forage-livestock balance in the Xilin Gol steppe,China:implications for sustainably utilizing grassland-ecosystem services显示文摘Understanding the spatiotemporal patterns of the forage-livestock balance is imperative for regionally arranging animal husbandry production while ensuring sustainable grassland-ecosystem service use.The Xilin Gol steppe is an important native grassland resource in Inner Mongolia Autonomous Region,China.This study aimed to elucidate the dynamics of the forage-livestock balance in the Xilin Gol steppe during the period 2000–2015.We evaluated the forage production and corresponding livestock carrying capacity(LCC)in the growing seasons of 2000–2015 using remote sensing data and field surveys.The spatiotemporal patterns of the forage-livestock balance were then assessed at regional,city(including city,county and banner),and village scales using statistical and household survey data.The results showed that both forage production and LCC decreased in the Xilin Gol steppe from east to west.During the period 2000–2015,the regional average forage production and corresponding LCC fluctuated without following a distinct trend,but were consistent with the variations in precipitation.The forage-livestock balance varied with time,space,and scale.At the regional scale,steppes were overgrazed in the early 2000s,but a forage-livestock balance or even grazing potential was achieved in other years.At the city scale,approximately half of the region exhibited a'forage-livestock balance'since 2000.However,about half of the region still experienced overgrazing,which mainly located in the southwest sandy zones.Such changes may have been affected by the variations in grassland quality,forage production,compensation payment,and so on.We suggest a location-specific management scheme for grazing constraints,ecological compensation payment,and industry development to aid in harmonizing animal husbandry and environmental restoration,while promoting sustainable development goals by 2030. | QU Yingbo ZHAO Yuanyuan DING Guodong CHI Wenfeng GAO Guanglei | 2021 | Journal of Arid Land2021,13,2: | 2 |

| 17 | Transcriptome characterization of Ishige okamurae(Phaeophyceae)shows strong environmental acclimation显示文摘Ishige okamurae,with leathery branched narrow fronds consisting of cylindrical hairs,is the typical species of the genus Ishige,which is considered as one of the most basal genera in the phylogeny of the Phaeophyceae.Apart from great public interest from the evolutionary respect,more attention has been brought on the abundant bioactive compounds in I.okamurae for therapeutic or economic considerations,such as diphlorethohydroxycarmalol and ishigoside.Yet little is known about related key genes or metabolic pathways involved in I.okamurae,which calls upon us to carry out global analyses of transcriptome by next generation sequencing.Altogether,we obtained 78 583 assembled scaffolds with N50 of 1 709 nucleotides,and 25 357 unigenes with significant BLAST matches(E-value cutoff of 10-5).In terms of characterization of the transcriptome of I.okamurae,we focused on anti-stress metabolic pathways and synthetic routes of bioactive compounds in an attempt to obtain a better understanding of the interactive organism-environment regulatory networks.Pathway-based analysis helped us to deepen our comprehension of the interaction between I.okamurae and its surroundings,with MAPK signal pathway as an example.Furthermore,we discovered a wide range of novel putative functional proteins that could be of wide application,such as Rab family,using sequence-based transcriptome.In conclusion,transcriptome characterization of I.okamurae(Phaeophyceae) shows strong environmental acclimation. | QU Jieqiong WANG Xumin CHI Shan WU Shuangxiu SUN Jing LIU Cui CHEN Shengping YU Jun LIU Tao | 2014 | Acta Oceanologica Sinica2014,33,2: | 1 |
