2A Study of the Kunlun-Qilian-Qinling Suture System显示文摘The Late Neoproterozoic-Early Palaeozoic suture zones within theCentral Orogenic Belt of China can be considered a system-the Kunlun-Qilian-Qinling suture system (KQQ suture system). It is basically divided into the western, central and eastern sectors. The western sector consists of the Küda-Subashi suture zone, the Mazar Kangxiwar suture and the central West Kunlun microblock. The central sector covers the following five suture zones: the Altun, North Qilian, Qaidam northern marginal, Qimantag-Wutuo and East Kunlun southern marginal sutures, in addition to some microblocks such as the mid-south Qilian, Qaidam, Ayakkum and Maqen microblocks. The eastern sector is composed of the Shangdan suture zone, Mianlüe suture zone and central Qinling-Dabie microblock. The KQQ suture system experienced (1) extending-rifting of the Rodinia super-continent in the early-middle Sinian (~780-600 Ma); (2) the formation and evolution of the KQQ archipelagic ocean during the Late Sinian-Ordovician (~600-440 Ma), and (3) closing of the KQQ ocean and forming of the KQQ suture system during the Late Ordovician-early Silurian (~440-400 Ma). 3Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. 4One-step base editing in multiple genes by direct embryo injection for pig trait improvement显示文摘The precise and simultaneous acquisition of multiple beneficial alleles in the genome is in great demand for the development of elite pig breeders. Cytidine base editors(CBEs) that convert C:G to T:A have emerged as powerful tools for single-nucleotide replacement. Whether CBEs can effectively mediate C-to-T substitution at multiple sites/loci for trait improvement by direct zygote injection has not been verified in large animals. Here, we determined the editing efficiency of four CBE variants in porcine embryonic fibroblast cells and embryos. The findings showed that hA3A-BE3-Y130F and hA3A-eBE-Y130F consistently resulted in increased base-editing efficiency and low toxic effects in embryonic development. Further, we verified that using a one-step approach, direct zygote microinjection of the CBE system can generate pigs harboring multiple point mutations.Our process resulted in a stop codon in CD163 and myostatin(MSTN) and introduced a beneficial allele in insulin-like growth factor-2(IGF2). The pigs showed disrupted expression of CD163 and MSTN and increased expression of IGF2, which significantly improved growth performance and infectious disease resistance. 5Discovery of the Jiawengmen Stromatolite Assemblage in the Southern Belt of Eastern Kunlun, NW China and Its Significance显示文摘This paper reports a newly discovered Late Mesoproterozoic–Early Neoproterozoic stromatolite assemblage, named here the “Jiawengmen stromatolite assemblage”, represented by a Conophyton-Baicalia association in the Jiawenmen area in the southern belt of the Eastern Kunlun. This stromatolite assemblage is dominated by large-scale conical stromatolites and related elements, i.e., Conophyton garganicus var. inkeni, C. cf. ressoti Menchikov, Jacutophyton cf., Conicodomenia f., which commonly co-exist with elements of the group of Baicalia. This assemblage can be correlated with that of the middle Jixian-middle Qingbaikou System in North and Northwest China, but is different from that in South China. Correlation can also be made with that in the upper horizon of the Middle Riphean-lower horizon of the Upper Riphean in the South Ural Mountains and Siberia of Russia, in North Africa, and in the Alaskan Peninsula of North America. These facts suggest that the Jiawengmen stromatolite assemblage probably colonized during 1300–850 Ma ago. Accordingly, the stromatolite-bearing carbonate rocks are then proposed to correspond to the middle Jixian System-middle Qingbaikou System or the upper Middle Riphean-lower Upper Riphean. Our stromatolite data further suggest that a Precambrian microblock, named the Xialawen microblock here, occurred in the southern belt of Eastern Kunlun, the western part of the Maqên microblock. Similar stromatolite assemblages in the Maqên microblock and those blocks that occurred in North China, Siberia and North Africa point to similar paleogeographic and paleoenvironmental conditions. These microblock and blocks were probably located at low latitudes and on the continental margins of the Rodinian supercontinent, where warm epicontinental seas were favorable to widespread colonization of stromatolites during the Late Mesoproterozoic-Early Neoproterozoic. 7EB病毒与原发性鼻咽部B细胞性淋巴瘤有关与继发性者无关显示文摘目的:对19 例鼻咽部B细胞性恶性淋巴瘤与EB 病毒(EpsteinBarr virus,EBV) 感染的相关性进行研究。方法:利用免疫组织化学及原位杂交方法对EBV 进行检测,并用免疫组化及原位杂交双染法标记肿瘤细胞,以鉴定EBV阳性的细胞为B淋巴瘤细胞。结果:EBV 编码的小m RNA探针(EBER) 原位杂交显示,8 例原发性鼻咽部B细胞性恶性淋巴瘤中3 例绝大多数肿瘤细胞呈阳性表达,1 例潜在性膜蛋白1(latent membrane protein 1,LMP1)阳性。而11 例继发性鼻咽部B细胞性恶性淋巴瘤EBER全部为阴性。全部病例进行了LMP1 检测,除1 例原发者,全部阴性。利用EBERISH(EBER原位杂交) 和免疫组化CD 标记物进行双标记染色证实,EBER 和LMP1阳性细胞为CD20 阳性,CD45RO 阴性。鼻咽部原发性B细胞性恶性淋巴瘤EBV 表达4/8 ,而继发者为0/11。结论:EBV 与鼻咽部原发性B细胞恶性淋巴瘤有较高的相关性,而与继发性者无关。高子芬 FaithC.S.Ho AlexC.L.Chan QianTao RamondLiang GopeshSrivastava 1999北京医科大学学报1999,31,5:3

18A harlequin ichthyosis pig model with a novel ABCA12 mutation can be rescued by acitretin treatment显示文摘Harlequin ichthyosis (HI) is a severe genetic skin disorder and caused by mutation in the ATP-binding cassette A12 (ABCA12) gene. The retinoid administration has dramatically improved long-term survival of HI, but improvements are still needed. However, the ABCA12 null mice failed to respond to retinoid treatment, which impedes the development of novel cure strategies for HI. Here we generated an ethylnitrosourea mutagenic HI pig model (named Z9), which carries a novel deep intronic mutation IVS49-727 A>G in the ABCA12 gene, resulting in abnormal mRNA splicing and truncated protein production. Z9 pigs exhibit significant clinical symptom as human patients with HI. Most importantly, systemic retinoid treatment significantly prolonged the life span of the mutant pigs via improving epidermal maturation, decreasing epidermal apoptosis, and triggering the expression of ABCA6.
