1Aberrant translation regulated by METTL1/WDR4-mediated tRNA N7-methylguanosine modification drives head and neck squamous cell carcinoma progression显示文摘Background:Cancer cells selectively promote the translation of oncogenic tran-scripts to stimulate cancer progression.Although growing evidence has revealed that tRNA modifications and related genes participate in this process,their roles in head and neck squamous cell carcinoma(HNSCC)remain largely unchar-acterized.Here,we sought to investigate the function and mechanisms of the transfer RNA(tRNA)N7-methylguanosine(m'G)modification in regulating the occurrence and development of HNSCC.Methods:Cell lost of-function and gain-of function assays,xenograft models,conditional knockout and knockin mouse models were used to study the physi-ological functions of tRNA m'G modification in HNSCC tumorigenesis.tRNA modification and expression profiling,mRNA translation profiling and res-cue assays were performed to uncover the underlying molecular mechanisms.Single-cell RNA sequencing(scRNA seq)was conducted to explore the tumor microenvironment changes.Results:The tRNA.m7G methyltransferase complex components Methyltransferase-like 1(METTL1)/WD repeat domain 4(WDR4)were upregulated in HNSCC and associated with a poor prognosis.Functionally,METTL1/WDR4 promoted HNSCC progression and metastasis in cell-based and transgenic mouse models.Mechanistically,ablation of METTL1 reduced the m'G levels of 16 tRNAS,inhibiting the translation of a subset of oncogenic transcripts,including genes related to the phosphatidylinositol-3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR)signaling pathway.In addition,chemical modulators of the PI3K/Akt/mTOR signaling pathway reversed the effects of Mettll in mouse HNSCC.Furthermore,scRNA-seq results revealed that Mettll knockout in mouse tumor cells altered the immune landscape and cell-cell interaction between the tumor and stromal compartment.Conclusions:The tRNA m?G methyltransferase METTLI was found to promote the development and malignancy of HNSCC through regulating global mRNA translation,including the PI3K/AKT/mTOR signaling pathway,and found to alter immune landscape.METTLI could be a promising treatment target for HNSCC patients.Jie Chen Kang Li Jianwen Chen Xiaochen Wang Rongsong Ling Maosheng Cheng Zhi Chen Fangfang Chen Qianting He Shuai Li Caihua Zhang Yizhou Jiang Qianming Chen Anxun Wang Demeng Chen 2022Cancer Communications2022,42,3:6

3Bi_(2)S_(3) spheres coated with MOF-derived Co_(9)S_(8) and N-doped carbon composite layer for half/full sodium-ion batteries with superior performance显示文摘Bismuth sulfide(Bi_(2)S_(3))has attracted particular interest as a potential anode material for sodium-ion batteries(SIBs).However,the low electrical conductivity and dramatic volumetric change greatly restrict its practical applications.In view of the apparent structural and compositional advantages of metal-organic frameworks(MOFs)derived carbon-based composite,herein,as a proof of concept,Bi_(2)S_(3) spheres coated with the MOF-derived Co_(9)S_(8) and N-doped carbon composite layer(Bi_(2)S_(3)@Co_(9)S_(8)/NC composite spheres)have been rational designed and synthesized.As expected,the core-shell Bi_(2)S_(3)@Co_(9)S_(8)/NC composite spheres exhibit remarkable electrochemical performance in terms of high reversible capacity(597 m Ah g^(-1) after 100 cycles at 0.1 A g^(-1)),good rate capability(341 m Ah g^(-1) at 8 A g^(-1))and long-term cycling stability(458 m Ah g^(-1) after 1000 cycles at 1 A g^(-1))when investigated as anode materials for SIBs.Electrochemical analyses further reveal the favorable reaction kinetics in the Bi_(2)S_(3)@Co_(9)S_(8)/NC composite spheres.In addition,the possible sodium storage mechanism has been studied by ex-situ X-ray diffraction technique.More importantly,a sodium-ion full cell based on Na_(3) V_(2)(PO_(4))_(3)/r GO as cathode and Bi_(2)S_(3)@Co_(9)S_(8)/NC as anode is also fabricated,suggesting their potential for practical applications.It is anticipated that the present work could be extended to construct other advanced electrode materials using MOFs-derived carbon-based composites as surface coating materials for various energy storagerelated applications.Youzhang Huang Xueshuang Zhu Daoping Cai Zhixiang Cui Qianting Wang Hongbing Zhan 2021Journal of Energy Chemistry2021,30,8:2

6Fabrication and Characterization of 3D Graded PDMS Scaffolds Using Vacuum-Assisted Resin Transfer Moulding显示文摘Graded porous scaffold can be applied to study the interactions between cells and scaffold with different pore sizes. Polydimethylsiloxane(PDMS) scaffold with an axial pore size grade was successfully manufactured via vacuum-assisted resin transfer moulding(VARTM) and particle leaching technologies. The properties of graded PDMS scaffolds, including porosity, water absorption, interconnectivity, compression modulus, as well as compression strength were investigated. The results showed that the smaller the size of the NaCl particles is, the higher the porosity and water absorption of graded PDMS scaffolds would be. The graded PDMS scaffold fabricated had a compressive modulus and a compressive strength of 19.69±1.42 kPa and 4.76±0.22 kPa, respectively. Moreover, the graded chitosan(CS)-coated PDMS scaffolds were prepared by using dip-coating technique under low vacuum and their hydrophilicity was examined. 8Laboratory Diagnosis and Monitoring the Viral Shedding of SARS-CoV-2 Infection显示文摘The worldwide epidemic of coronavirus disease 2019(COVID-19)is ongoing.Rapid and accurate detection of the causative virus SARSCoV-2 is vital for the treatment and control of COVID-19.In this study,the comparative sensitivity of different respiratory specimen types were retrospectively analyzed using 3,552 clinical samples from 410 COVID-19 patients confirmed by Guangdong CDC(Center for Disease Control and Prevention).Except for bronchoalveolar lavage fluid(BALF),the sputum possessed the highest positive rate(73.4%–87.5%),followed by nasal swabs(53.1%–85.3%)for both severe and mild cases during the first 14 days after illness onset(d.a.o.).Viral RNA could be detected in all BALF samples collected from the severe group within 14 d.a.o.and lasted up to 46 d.a.o.Moreover,although viral RNA was negative in the upper respiratory samples,it was also positive in BALF samples in most cases from the severe group during treatment.Notably,no viral RNA was detected in BALF samples from the mild group.Despite typical ground-glass opacity observed via computed tomographic scans,no viral RNA was detected in the first three or all upper respiratory tract specimens from some COVID-19 patients.In conclusion,sputum is most sensitive for routine laboratory diagnosis of COVID-19,followed by nasal swabs.Detection of viral RNA in BALF improves diagnostic accuracy in severe COVID-19 patients.Yang Yang Minghui Yang Jing Yuan Fuxiang Wang Zhaoqin Wang Jinxiu Li Mingxia Zhang Jinli Wei Ling Peng Gary Wong Haixia Zheng Weibo Wu Chenguang Shen Mingfeng Liao Kai Feng Jianming Li Qianting Yang Juanjuan Zhao Lei Liu Yingxia Liu 2020The Innovation2020,1,3:1

10Amorphous calcium magnesium phosphate nanocomposites with superior osteogenic activity for bone regeneration显示文摘The seek of bioactive materials for promoting bone regeneration is a challenging and longterm task.Functionalization with inorganic metal ions or drug molecules is considered effective strategies to improve the bioactivity of various existing biomaterials.Herein,amorphous calcium magnesium phosphate(ACMP)nanoparticles and simvastatin(SIM)-loaded ACMP(ACMP/SIM)nanocomposites were developed via a simple co-precipitation strategy.The physiochemical property of ACMP/SIM was explored using transmission electron microscope(TEM),Fourier transform infrared spectroscopy(FTIR),powder X-ray diffraction(XRD)and highperformance liquid chromatograph(HPLC),and the role of Mg^(2+) in the formation of ACMP/SIM was revealed using X-ray absorption near-edge structure(XANES).After that,the transformation process of ACMP/SIM in simulated body fluid(SBF)was also tracked to simulate and explore the in vivo mineralization performance of materials.We find that ACMP/SIM releases ions of Ca^(2+),Mg^(2+)and PO_(4)^(3),when it is immersed in SBF at 37℃,and a phase transformation occurred during which the initially amorphous ACMP turns into self-assembled hydroxyapatite(HAP).Furthermore,ACMP/SIM displays high cytocompatibility and promotes the proliferation and osteogenic differentiation of MC3T3-E1 cells.For the in vivo studies,lamellar ACMP/SIM/Collagen scaffolds with aligned pore structures were prepared and used to repair a rat defect model in calvaria.ACMP/SIM/Collagen scaffolds show a positive effect in promoting the regeneration of calvaria defect after 12weeks.The bioactive ACMP/SIM nanocomposites are promising as bone repair materials.Considering the facile preparation process and superior in vitro/vivo bioactivity,the as-prepared ACMP/SIM would be a potential candidate for bone related biomedical applications.Yingying Jiang Shuo Tan Jianping Hu Xin Chen Feng Chen Qianting Yao Zhi Zhou Xiansong Wang Zifei Zhou Yunshan Fan Junjian Liu Yize Lin Lijia Liu Shisheng He 2021Regenerative Biomaterials2021,8,6:0
11Berberine enhances the anti-hepatocellular carcinoma effect of NK92-MI cells through inhibiting IFN-gamma-mediated PD-L1 expression显示文摘Background and aims:Berberine is one of the most promising clinically tested natural alkaloids,and immunotherapy using natural killer(NK)cells is a potentially effective treatment for hepatocellular carcinoma(HCC).This study aims to elucidate the effect of berberine on the anti-HCC effect of NK92-MI cells.Materials and methods:Human HCC SMMC-7721 and Hep3B cells were co-incubated with NK92-MI cells,berberine(60 or 80 mmol/L),or their combination for 36 h.To evaluate the killing effect of NK92-MI cells on HCC cells,the release of lactate dehydrogenase(LDH)in HCC cells was measured.The anti-tumor effects of berberine,NK92-MI cells,and their combinations were evaluated by MTS,EdU,Tunel,and Western blot assays.A male BALB/c nude mouse subcutaneous tumor model was used to investigate the anti-HCC effect of berberine and NK92-MI cells in vivo.Results:The LDH assay showed that berberine enhanced the cytotoxicity of NK92-MI cells on HCC cells.The combination of berberine and NK92-MI cells demonstrated more obvious anti-HCC effect than did the berberine or NK92-MI single treatment in inhibiting cell proliferation and inducing apoptosis both in vitro and in vivo.Mechanistically,the expression of programmed cell death-ligand 1(PD-L1)in HCC cells was upregulated after co-culture with NK-92MI cells.PD-L1 expression was knocked down,thereby inhibiting the proliferation and promoting apoptosis of HCC cells,and inhibited by berberine that blocked the secretion of interferon gamma(IFN-g),thereby enhancing the anti-tumor effect of NK92-MI cells.Conclusions:Current data show that the immunomodulatory role of berberine is to enhance the cytotoxic effect of NK92-MI cells and inhibit tumor immune escape by reducing the expression of PD-L1.Our study provides a rationale for the clinical application of berberine in combination with NK cells for the treatment of HCC.Kunyuan Wang Chengxin Gu Ganxiang Yu Jiaen Lin Zhilei Wang Qianting Lu Yangzhi Xu Dan Zhao Xiaofeng Jiang Weijian Mai Shiming Liu Hui Yang 2022Liver Research2022,6,3:0
