2Morphological responses to water-level fluctuations of two submerged macrophytes, Myriophyllum spicatum and Hydrilla verticillata显示文摘Aims Myriophyllum spicatum and Hydrilla verticillata are common submerged macrophytes in the Yangtze River basin.To investigate their tolerances and adaptations to water-level fluctuations,an experiment was conducted in a pond.Methods We designed five different amplitudes of water-level fluctuations as static,615,630,660 and 690 cm during the 74 days of the experiment.In each amplitude treatment,two cultivation methods were examined as monoculture and mixed culture.Important Findings The results showed that M.spicatum had greater morphological responses to water-level fluctuations than H.verticillata.Fluctuating amplitude had significant effects on branch number,shoot length and root dry weight(DW)of M.spicatum,whereas it only had significant effect on branch number of H.verticillata.Both fluctuating amplitude and cultivation method had significant effects on total DW of M.spicatum,which was higher in monoculture than mixed culture.The total DW of H.verticillata was affected by fluctuating amplitude only,and the largest biomass was in the amplitude of 630 cm.Fruit DW of M.spicatum was largest in the amplitude of 630 cm,high amplitude of water-level fluctuations would inhibit flowering and seed production.Root DW proportion was significantly affected by fluctuating amplitude and cultivation method in both species,and the root DW proportion of M.spicatum was significantly higher in the amplitude-of 690 cm.We conclude that moderate amplitude of water-level fluctuations can promote the distribution and growth of both species,and in order to accelerate the restoration of both species in natural habitats,the optimum amplitude should be keep at 615 to 630 cm.Xiaoke Zhang Xueqin Liu Qingzhang Ding 2013Journal of Plant Ecology2013,6,1:10
3Expression and function of leptin and its receptor in mouse mammary gland显示文摘Leptin is an autocrine and paracrine factor which affects the development of duct, formation of gland alveolus, expression of milk protein gene and onset involution of mammary gland. In order to know the function and mechanism of leptin in mammary gland, the protein expression and localization of leptin and its long form receptor (OB-Rb) were detected by a confocal laser scanning microscope. To study the impacts of leptin on mammary gland and leptin signal transduction pathway in pregnancy-, lacta-tion-and involution-stage mammary gland, explants were cultured and Western blotting was used. The results showed that in the whole development cycle of mammary gland, the expression of leptin and OB-Rb was in positive correlation. In virgin the leptin expression was the highest and then decreased in pregnancy. In lactation the expression of leptin was low and upgraded in involution, and recovered to the original level about virgin on involution 13 d. The localization of leptin and OB-Rb revealed that leptin induced the expression of OB-Rb specifically and controlled the development and physiological function of the mammary gland by binding to OB-Rb. In pregnancy stage, leptin stimulated proliferation and differentiation of ductal epithelial cells by JAK-MAPK signal pathway. 4A novel xeno-free and feeder-cell-free system for human pluripotent stem cell culture显示文摘While human induced pluripotent stem cells(hiPSCs)have promising applications in regenerative medicine,most of the hiPSC lines available today are not suitable for clinical applications due to contamination with nonhuman materials,such as sialic acid,and potential pathogens from animal-product-containing cell culture systems.Although several xeno-free cell culture systems have been established recently,their use of human fibroblasts as feeders reduces the clinical potential of hiPSCs due to batch-to-batch variation in the feeders and time-consuming preparation processes.In this study,we have developed a xeno-free and feeder-cell-free human embryonic stem cell(hESC)/hiPSC culture system using human plasma and human placenta extracts.The system maintains the self-renewing capacity and pluripotency of hESCs for more than 40 passages.Human iPSCs were also derived from human dermal fibroblasts using this culture system by overexpressing three transcription factors—Oct4,Sox2 and Nanog.The culture system developed here is inexpensive and suitable for large scale production.Qihui Wang Xiaoning Mou Henghua Cao Qingzhang Meng Yanni Ma Pengcheng Han Junjie Jiang Hao Zhang Yue Ma 2012Protein & Cell2012,3,1:6

6Current status and trends in forest genomics显示文摘Forests are not only the most predominant of the Earth's terrestrial ecosystems,but are also the core supply for essential products for human use.However,global climate change and ongoing population explosion severely threatens the health of the forest ecosystem and aggravtes the deforestation and forest degradation.Forest genomics has great potential of increasing forest productivity and adaptation to the changing climate.In the last two decades,the field of forest genomics has advanced quickly owing to the advent of multiple high-throughput sequencing technologies,single cell RNA-seq,clustered regularly interspaced short palindromic repeats(CRISPR)-mediated genome editing,and spatial transcriptomes,as well as bioinformatics analysis technologies,which have led to the generation of multidimensional,multilayered,and spatiotemporal gene expression data.These technologies,together with basic technologies routinely used in plant biotechnology,enable us to tackle many important or unique issues in forest biology,and provide a panoramic view and an integrative elucidation of molecular regulatory mechanisms underlying phenotypic changes and variations.In this review,we recapitulated the advancement and current status of 12 research branches of forest genomics,and then provided future research directions and focuses for each area.Evidently,a shift from simple biotechnology-based research to advanced and integrative genomics research,and a setup for investigation and interpretation of many spatiotemporal development and differentiation issues in forest genomics have just begun to emerge.Dulal Borthakur Victor Busov Xuan Hieu Cao Qingzhang Du Oliver Gailing Fikret Isik Jae-Heung Ko Chenghao Li Quanzi Li Shihui Niu Guanzheng Qu Thi Ha Giang Vu Xiao-Ru Wang Zhigang Wei Lin Zhang Hairong Wei 2022Forestry Research2022,2,1:2
