1Endothelial PDGF-BB/PDGFR-βsignaling promotes osteoarthritis by enhancing angiogenesis-dependent abnormal subchondral bone formation显示文摘The mechanisms that coordinate the shift from joint homeostasis to osteoarthritis(OA)remain unknown.No pharmacological intervention can currently prevent the progression of osteoarthritis.Accumulating evidence has shown that subchondral bone deterioration is a primary trigger for overlying cartilage degeneration.We previously found that H-type vessels modulate aberrant subchondral bone formation during the pathogenesis of OA.However,the mechanism responsible for the elevation of H-type vessels in OA is still unclear.Here,we found that PDGFR-βexpression,predominantly in the CD31^(hi)Emcn^(hi) endothelium,was substantially elevated in subchondral bones from OA patients and rodent OA models.A mouse model of OA with deletion of PDGFR-βin endothelial cells(ECs)exhibited fewer H-type vessels,ameliorated subchondral bone deterioration and alleviated overlying cartilage degeneration.Endothelial PDGFR-βpromotes angiogenesis through the formation of the PDGFR-β/talin1/FAK complex.Notably,endothelium-specific inhibition of PDGFR-βby local injection of AAV9 in subchondral bone effectively attenuated the pathogenesis of OA compared with that of the vehicle-treated controls.Based on the results from this study,targeting PDGFR-βis a novel and promising approach for the prevention or early treatment of OA.Zhuang Cui Hangtian Wu Ye Xiao Ting Xu Junjie Jia Hancheng Lin Rongmin Lin Kun Chen Yihuang Lin Kaiqun Li Xiaohu Wu Changjun Li Bin Yu 2022Bone Research2022,10,4:4
2Calculation of Maximum Allowable Free Span Length and Safety Assessment of the DF1-1 Submarine Pipeline显示文摘The DF1-1 submarine pipeline was investigated using a dual-frequency side-scan sonar and a swath sounder system.More than a hundred scour pits under the pipeline were found,most of which have caused the span of the pipeline to increase and threatened its safety.The maximum allowable free span length (MAFSL) of the pipeline was determined through the limitations regarding maximum allowable stress under static or quasi-static loads and the onset of Vortex Induced Vibrations (VIV) under different hydrodynamic actions.The results show that the MAFSL under static conditions is 56 m.However,the MAFSLs are 30 m and 20 m under ordinary weather conditions and hurricane-induced currents for the 100-year return period,respectively,to avoid VIV as calculated by using the highest safety class factor.It is suggested that spanning pipelines longer than 20 m should be supported.Additionally,eight successive spans which may also threaten the pipeline were proposed.The most hazardous scour pits are along the pipeline section from KP42 to KP51.XU Jishang LI Guangxue Juan J. 4Characterization of NDM-1-producing carbapenemase in Acinetobacter spp.and E.coli isolates from diseased pigs显示文摘In recent years,the mobile metallo-β-lactamase(MBL)genes have been found to correspond to one of the most important resistance characters identified in Gramnegative bacteria,severely affecting clinical chemotherapy and threatening public health.The prevalence of mobile MBL genes and their flanking regions in Gram-negative bacteria from diseased pigs in China was investigated.A total of 334 lung samples from diseased pigs were screened for Gram-negative bacteria classified as non-susceptible to meropenem(MIC≥4mg·L^(–1)).Six isolates,including three Escherichia coli,two Acinetobacter baumanii and one A.calcoaeticus,exhibitedMBL production and carried the blaNDM-1 gene.S1-PFGE and Southern blot analysis showed that the blaNDM-1 gene was located on the chromosome of one A.baumanii isolate and on plasmids of various sizes in the other five isolates.MIC testing using broth microdilution revealed that all blaNDM-1-carrying isolates and some of their transconjugants exhibited resistance to almost allβ-lactams tested.Whole genome sequencing revealed that the flanking region of the blaNDM-1 gene from all porcine isolates had high levels of similarity with the corresponding regions in human isolates.One porcine E.coli isolate carrying blaNDM-1 was typed as ST48,a common sequence type in human E.coli isolates.These results suggest the possibility of human-tofood animal transfer of blaNDM-1-producing E.coli,highlighting the need for surveillance of carbapenemase producers among bacteria from food animals.In addition,the prudent use of antimicrobial agents to decrease the opportunities for co-selection of carbapenemase genes in food animals is also urgently needed.Rongmin ZHANG Yang WANG Zhihai LIU Jiyun LI Wenjuan YIN Lei LEI Congming WU Jianzhong SHEN 2015Frontiers of Agricultural Science and Engineering2015,2,3:2

8A new cytotoxic salannin-class limonoid alkaloid from seeds of Azadirachta indica A. Juss显示文摘A new cytotoxic salannin-class limonoid alkaloid, azadiramide A(1),was isolated from seeds of Azadirachta indica A. Juss. Its structure was elucidated by extensive analysis of spectroscopic data and quantum chemical calculation. Compound 1 is a rare salannin-class limonoid alkaloid. To our best knowledge, only two compounds belonging to this type were so far found. 12Preparation and antioxidant activity of albumin binding Salen Schiff-base metal complexes显示文摘A kind of novel biopolymer antioxidant (BSA/HOSalenM, M=Co, Mn, Zn) is prepared with conjugation, which increases the antioxidant activity of the bovine serum albumin (BSA). The conjugations have been characterized by IR spectra, UV-Vis spectra, Fluorescence spectra, Circular dichroism (CD) spectra and Native-PAGE. The BSA is used as a biopolymer scaffold, and the insoluble Salen Schiff-base metal complexes HOSalenM make axial coordination with the amino acid residues of the BSA. The structure of the BSA is unchanged when the binding rate of HOSalenCo is less than 10. The HOSalenCo conjugations show an excellent hydroxyl radical (·OH) scavenging activity, and the activity (EC50 ) of BSA/HOSalenCo(10) (BSA : HOSalenCo=1 : 10) is improved by two orders of magnitude compared with the BSA, while the activity of the BSA/HOSalenMn is weak and the BSA/HOSalenZn shows no scavenging activity.LI Gang ZHANG HuiFang WANG RongMin HE YuFeng XIONG YuBing 2013Chinese Science Bulletin2013,58,24:1

16Analysis and Dynamic Prediction of Ecological Environment Development Capacity of Qingshui River Basin显示文摘The Ecological Footprint and Development Capacity Model were used to analyze the ecological environment development capability of the Qingshui River Basin from 2000 to 2015. On this basis,the ecological environment development capacity of the Qingshui River Basin from 2016 to 2020 was dynamically predicted using the GM( 1,1) model. The results showed that the Qingshui River Basin has a strong capacity for sustainable development of the ecological environment,but it is increasingly stressed by the ecological carrying capacity. The potential unsustainability of the ecological environment is facing severe challenges. In the next 5 years,the development capacity of the ecological environment will show a growth trend,but the increase is not large,reflecting the limited development potential.
