| 3 | Epidemiology,clinical and laboratory characteristics of currently alive HIV-1 infected former blood donors naive to antiretroviral therapy in Anhui Province,China显示文摘Background Unregulated commercial blood/plasma collection among farmers occurred between 1992 and 1995 in central China and caused the second major epidemic of human immunodeficiency virus type 1 (HIV-1) infection in China. It is important to characterize HIV-1-infected former blood donors and to study characteristics associated with disease progression for future clinical intervention and vaccine development. Methods A cross-sectional study was performed on HIV-1-infected former blood donors (FBDs) and age-matched HIV-seronegative local residents. Demographic, epidemiologic, clinical and key laboratory data were collected from all study participants. Both unadjusted and adjusted multivariate linear regressions were employed to analyze the association of the decrease of CD4+ T-cell counts with other characteristics. Results Two hundred and ninety-four HIV-1-infected FBDs and 59 age-matched HIV-seronegative local residents were enrolled in this study. The unregulated blood/plasma collection occurred more than a decade (10.8 -12.8 years) ago, which caused the rapid spread of HIV-1 infection and the high prevalence of co-infection with hepatitis C virus (HCV, 89.5%); hepatitis B virus (HBV) co-infection was observed in only 11 HIV+participants (3.7%). Deterioration in both clinical manifestation and laboratory parameters and increase of viral loads were observed in parallel with the decrease of CD4+ T-cell counts. The decrease of total lymphocyte counts (P<0.001) and hemoglobin levels (P<0.001) and the appearance of dermatosis (P=0.03) were observed in parallel with the decrease of CD4+ T-cell counts whereas viral loads (P<0.001) and CD8+ T-cell counts (P=0.01) were inverselyassociated with CD4+ T-cell counts. Conclusions Co-infection with HCV but not HBV is highly prevalent among HIV-1-infected FBDs. CD4+ T-cell counts is a reliable indicator for disease progression among FBDs. | 4 | Strengths, weaknesses, opportunities and threats (SWOT) analysis of reinitiation into methamphetamine abusers: qualitative findings from an exploration of methamphetamine abusers in Shanghai, China显示文摘Background Despite high relapse rate among methamphetamine (MA) abusers, there still have been little empirical data to date detecting the risk factors related to craving and relapse from the perspective of MA abusers. Therefore, the aim of this study was to use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors that could facilitate or impede individuals' abstinence in a real-life context from the perspectives of MA abusers. Aim To use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors relating to individuals' abstinence in a real-life context. Method Semistructured interviews were conducted with 32 MA abusers recruited in Shanghai guided by open-ended questions on narrating the real-life catalysts and inhibitors related to craving, initiation, relapse and abstinence. All data were transcribed verbatim and analysed using a strengths, weaknesses, opportunities and threats (SWOT) analysis. Results The results of the SWOT analysis revealed that contextual factors including peer influence, prevalence and availability of MA, familiar venue of MA use, discrimination, sexual behaviours, alcohol, emotional states and their attitudes towards smoking MA were important factors that contribute to reinitiation and relapse. Surveillance systems, antidrug social workers, vocational skills trainings, moving to another city and family responsibility might serve as counter measures targeti叩 those mentioned weaknesses and threats above. Conclusion This SWOT analysis highlights the complex nature of relapse. | 5 | Optimization of biotransformation from phytosterol to androstenedione by a mutant Mycobacterium neoaurum ZJUVN-08显示文摘Biotransformation of phytosterol(PS) by a newly isolated mutant Mycobacterium neoaurum ZJUVN-08 to produce androstenedione has been investigated in this paper.The parameters of the biotransformation process were optimized using fractional factorial design and response surface methodology.Androstenedione was the sole product in the fermentation broth catalyzed by the mutant M.neoaurum ZJUVN-08 strain.Results showed that molar ratio of hydroxypropyl-β-cyclodextrin(HP-β-CD) to PS and substrate concentrations were the two most significant factors affecting androstenedione production.By analyzing the statistical model of three-dimensional surface plot,the optimal process conditions were observed at 0.1 g/L inducer,pH 7.0,molar ratio of HP-β-CD to PS 1.92:1,8.98 g/L PS,and at 120 h of incubation time.Under these conditions,the maximum androstenedione yield was 5.96 g/L and nearly the same with the non-optimized(5.99 g/L),while the maximum PS conversion rate was 94.69% which increased by 10.66% compared with the non-optimized(84.03%).The predicted optimum conditions from the mathematical model were in agreement with the verification experimental results.It is considered that response surface methodology was a powerful and efficient method to optimize the parameters of PS biotransformation process. | Xiao-yan ZHANG Yong PENG Zhong-rui SU Qi-he CHEN Hui RUAN Guo-qing HE | 2013 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2013,14,2: | 7 |
| 6 | Stromal cell-derived factor-1α promotes recruitment and differentiation of nucleus pulposus-derived stem cells显示文摘BACKGROUND Intervertebral disc(IVD) degeneration is a condition characterized by a reduction in the water and extracellular matrix content of the nucleus pulposus(NP) and is considered as one of the dominating contributing factors to low back pain. Recent evidence suggests that stromal cell-derived factor 1α(SDF-1α) and its receptor CX-C chemokine receptor type 4(CXCR4) direct the migration of stem cells associated with injury repair in different musculoskeletal tissues.AIM To investigate the effects of SDF-1α on recruitment and chondrogenic differentiation of nucleus pulposus-derived stem cells(NPSCs).METHODS We performed real-time RT-PCR and enzyme-linked immunosorbent assay to examine the expression of SDF-1α in nucleus pulposus cells after treatment with pro-inflammatory cytokines in vitro. An animal model of IVD degeneration was established using annular fibrosus puncture in rat coccygeal discs. Tissue samples were collected from normal control and degeneration groups.Differences in the expression of SDF-1α between the normal and degenerative IVDs were analyzed by immunohistochemistry. The migration capacity of NPSCs induced by SDF-1α was evaluated using wound healing and transwell migration assays. To determine the effect of SDF-1α on chondrogenic differentiation of NPSCs, we conducted cell micromass culture and examined the expression levels of Sox-9, aggrecan, and collagen II. Moreover, the roles of SDF-1/CXCR4 axis in the migration and chondrogenesis differentiation of NPSCs were analyzed by immunofluorescence, immunoblotting, and real-time RT-PCR.RESULTS SDF-1α was significantly upregulated in the native IVD cells cultured in vitro with pro-inflammatory cytokines, such as interleukin-1β and tumor necrosis factor-α, mimicking the degenerative settings. Immunohistochemical staining showed that the level of SDF-1α was also significantly higher in the degenerative group than in the normal group. SDF-1α enhanced the migration capacity of NPSCs in a dose-dependent manner. In addition, SDF-1α induced chondrogenic differentiation of NPSCs, as evidenced by the increased expression of chondrogenic markers using histological and immunoblotting analyses. Realtime RT-PCR, immunoblotting, and immunofluorescence showed that SDF-1αnot only increased CXCR4 expression but also stimulated translocation of CXCR4 from the cytoplasm to membrane, accompanied by cytoskeletal rearrangement.Furthermore, blocking CXCR4 with AMD3100 effectively suppressed the SDF-1α-induced migration and differentiation capacities of NPSCs.CONCLUSION These findings demonstrate that SDF-1α has the potential to enhance recruitment and chondrogenic differentiation of NPSCs via SDF-1/CXCR4 chemotaxis signals that contribute to IVD regeneration. | Jin-Wei Ying Tian-Yong Wen Shi-Shen Pei Ling-Hao Su Di-Ke Ruan | 2019 | World Journal of Stem Cells2019,11,3: | 6 |
| 7 | Pressure from the lips and the tongue in children with class III malocclusion显示文摘Objective:To discuss possible relationships between class III malocclusion and perioral forces by measuring the pressure from the lips and the tongue of children with class III malocclusion. Methods:Thirty-one children with class III malocclusion were investigated and their perioral forces were measured at rest and during swallowing under natural head position by a custom-made miniperioral force computer measuring system. Results:The resting pressures exerted on the labial side and palatine side of the upper left incisor, as well as the labial side and lingual side of the lower left incisor, were 0 g/cm2, 0 g/cm2, 0.57 g/cm2 and 0.23 g/cm2, respectively. Correspondingly, the swallowing forces were 2.87 g/cm2, 5.97 g/cm2, 4.09 g/cm2 and 7.89 g/cm2, respectively. No statistical difference between muscular pressure and gender existed. During swallowing, the lingual forces were significantly higher than the labial forces (P<0.01), however, at rest there was no significantly different force between these two sides. Compared to the normal occlusion patients, children with class III malocclusion had lower perioral forces. The upper labial resting forces (P<0.01), the lower labial resting forces (P<0.05) and all the swallowing pressures from the lips and the tongue (P<0.01) showed statistical differences between the two different occlusion conditions. Meanwhile, no significant difference was found for the resting pressure from the tongue between class III malocclusion and normal occlusion. Conclusion:Patients with class III malocclusion have lower perioral forces and this muscle hypofunction may be secondary to the spatial relations of the jaws. | 11 | Characteristics of rifampicin-resistant tuberculosis detection in China,2015–2019显示文摘Background The very high burden of rifampicin resistance tuberculosis(RR-TB)and the very low detection of RR-TB cases are a major challenge that China has been facing.This study analyzed the characteristics of RR-TB detection in China after the change of RR-TB detection strategy since 2015,aiming to provide reference and evidence for the development of more precise national drug resistance tuberculosis prevention and control policy.Methods We extracted data related to rifampicin resistance screening from the national Tuberculosis Information Management System(TBIMS)from 2015 to 2019,and used descriptive research methods to analyze the screening rate of presumptive RR-TB,the number and duration of RR-TB patients detected and drug resistance testing methods in each year.Chi-square test was used to compare the differences in component ratio or rate between years,and Kruskal Wallis test was used to compare the differences in median days for detection of RR-TB patients in each year.Results A total of 68,200 RR-TB cases were detected during 2015–2019,of which 48.1%were new cases.The number and detection rate of RR-TB cases increased year by year,from 10019 and 14.3%in 2015 to 18623 and 28.7%in 2019,respectively.Of the bacteriologically confirmed TB cases,81.9%were tested for RR in 2019,a considerable increase from 29.5%in 2015.In 2019,only 41.0%of RR-TB cases had fluoroquinolones(FQs)susceptibility testing performed,and this proportion has been declining year by year since 2016.The proportion of application of rapid molecular tools increased from 24.0%in 2015 to 67.1%in 2019,and the median days to obtain RR results was significantly shortened.In 2019,76.0%of RR-TB cases were diagnosed as presumptive RR-TB in county-level hospitals.Conclusions After China modified the RR-TB detection strategy,the screening rate of RR and the number of RR-TB cases increased significantly.The RR testing methods now predominantly utilize rapid molecular tools.However,comprehensive measures should be implemented to close the gap in the detection of RR-TB cases.It is imperative to take FQs susceptibility testing seriously and effectively strengthen the laboratory capacity of county-level hospitals. | Wei Su Yun-zhou Ruan Tao Li Xin Du Jia-wen Jiang Ren-zhong Li | 2021 | Infectious Diseases of Poverty2021,10,4: | 5 |

| 17 | A mathematical modeling study of the HIV epidemics at two rural townships in the Liangshan Prefecture of the Sichuan Province of China显示文摘Background:As a response to a severe HIV epidemic in the Liangshan Prefecture,one of the worst in China,population based HIV interventions,including two population-wide HIV screening,have been carried out since 2005 at two townships in a remote mountainous region of Liangshan.The objective of our mathematical modeling study is to assess the temporal dynamics of the HIV epidemic in the two townships based on the data collected in the study area during the period 2005e2010.Methods:A mathematical model was set up to describe the population dynamics of HIV transmission in study area.The model was calibrated by fitting it to the HIV testing and treatment data from 2005 to 2008.Validation of the model was done by comparing its predicted value of HIV prevalence in 2010 to the prevalence data obtained in the 2010 population wide HIV testing.The validated model was used to produce estimation of HIV incidence,prevalence and death.Results:Our model estimations show that population-based HIV interventions have significantly slowed down the rise of the HIV epidemic in the two townships.Over the five-year period from 2005 to 2010,the year-over-year rate of increase in HIV incidence,prevalence,and death has declined by 91.5%,28.7%,and 52.3%,respectively.Conclusion:Mathematical models,when integrated with epidemiological and surveillance data,can be an effective tool for predicting the temporal dynamics of HIV and assessing the impacts of HIV interventions. | Zhimin Su Caiting Dong Ping Li Hongxia Deng Yuhan Gong Shiyong Zhong Min Wu Yuhua Ruan Guangming Qin Wen Yang Yiming Shao Michael Li | 2016 | Infectious Disease Modelling2016,1,1: | 2 |
