| 1 | Effects of Xinfeng capsule on pulmonary function based on treg-mediated notch pathway in a rat model of adjuvant arthritis显示文摘OBJECTIVE:To observe the impact of xinfeng xapsule(XFC) on pulmonary function in a rat model of adjuvant arthritis(AA) and to investigate the mechanism of action.METHODS:Forty rats were randomly divided into four groups of ten:normal control(NC);model control(MC);tripterygium glycosides tablet(TPT);and xinfeng capsule(XFC).Except for the NC group,AA was induced in all rats by intracutaneous injection of 0.1 mL Freund's complete adjuvant in the right paw on the 19th day.NC and MC groups were given(0.9%) physiological saline.The TPT and XFC groups were given TPT(10 mg/kg) and XFC(1.2 g/kg),respectively.Thirty days after administration,changes in paw edema(E),the arthritis index(AI),pulmonary function,levels of regulatory T-cells(Treg),ultrastructure of lung tissue,and expression of Notch receptors and ligands in lung tissue were observed.RESULTS:In the MC group,E and the AI were increased and pulmonary function significantly decreased;the structure of alveolar type-II cells was damaged;ratios of Treg in peripheral blood were reduced;and expression of Notch receptors such as Notch3 and Notch4 and ligands such as Delta1 in lung tissue were significantly increased whereas expression of Notch1,Jagged1 and Jagged2 were significantly decreased.After intervention with XFC,E and the AI were decreased;pulmonary function was enhanced;the structure of alveolar type-II cells was improved;and expression of Treg,Notch1,Jagged1,Jagged2 was elevated,whereas that of Notch3,Notch4 and Delta1 was reduced.CONCLUSION:XFC can not only inhibit E and the AI and improve joint symptoms,it can also improve pulmonary function and reduce inflammation in lung tissue.These actions could be carried out through increases in the expression of Treg,Notch receptors(Notch1) and ligands(Jagged1,Jagged2),and reductions in the expression of Notch3,Notch4 and Delta1.These phenomena would reduce the deposition of immune complexes and the inflammatory response in lung tissue,thereby improving joint symptoms and pulmonary function. | Lei Wan Jian Liu Chuanbing Huang Yuan Wang Xi Shen Wandong Zhang Guizheng Wang Haixia Fan Yao Ge Ruilian Chen Yunxiang Cao Ruikai Zong Li Lei | 2012 | Journal of Traditional Chinese Medicine2012,32,3: | 17 |
| 2 | Genetic analysis of tolerance to photo-oxidative stress induced by high light in winter wheat (Triticum aestivum L.)显示文摘High light induced photooxidation (HLIP) usually leads to leaf premature senescence and causes great yield loss in winter wheat. In order to explore the genetic control of wheat tolerance to HLIP stress, a quantitative trait loci (QTL) analysis was conducted on a set of doubled haploid population, derived from two winter wheat cultivars. Actual values of chlorophyll content (Chl), minimum fluorescence level (Fo), maximum fluorescence level (Fm), and the maximum quantum efficiency of photosystem II (Fv/Fm) under both HLIP and non-stress conditions as well as the ratios of HLIP to non-stress were evaluated. HLIP considerably reduced Chl, Fm, and Fv/Fm, but in-creased Fo, compared with that under non-stress condition. A total of 27, 16, and 28 QTLs were associated with the investigated traits under HLIP and non-stress and the ratios of HLIP to non-stress, respectively. Most of the QTLs for the ratios of HLIP to non-stress collocated or nearly linked with those detected under HLIP condition. HLIP-induced QTLs were mapped on 15 chromosomes, involving in 1A, 1B, 1D, 2A, 2B, 2D, 3A, 3B, 4A, 4D, 5B, 6A, 6B, 7A, and 7D while those expressed under non-stress condition involved in nine chromosomes, includ-ing 1B, 1D, 2A, 2B, 3B, 4A, 5A, 5B, and 7A. The expression patterns of QTLs under HLIP condition were different from that under non-stress condition except for six loci on five chromosomes. The phenotypic variance explained by individual QTL ranged from 5.0% to 19.7% under HLIP, 8.3% to 20.8% under non-stress, and 4.9% to 20.2% for the ratios of HLIP to non-stress, respectively. | 3 | Uric acid lowering effect of Tibetan Medicine RuPeng15 powder in animal models of hyperuricemia显示文摘OBJECTIVE: To evaluate the influence of the Tibetan medicine Ru Peng15 powder(RPP15) on uric acid levels, and explore its possible mechanisms of action in hyperuricemic animal models.METHODS: Hyperuricemic mice were generated by orally administering yeast extract paste twice daily(30 g/kg) for 8 days, to mimic human hyperuricemia induced by high-protein diets. Hyperuricemic rats were generated by intraperitoneal injection of 250 mg/kg potassium oxonate to each animal 1 h before the last oral administration of test compounds, which raised the serum uric acid level by inhibiting the decomposition of uric acid. Levels of uric acid and creatinine in serum and urine were detected by the phosphotungstic acid and picric acid methods respectively, and the activity of xanthine oxidase(XOD) was assayed using a commercial test kit.RESULTS: RPP15(0.4, 0.8, 1.2 g/kg) significantly decreased the level of serum uric acid in healthy rats(P < 0.05). Furthermore, hyperuricemic rats treated with RPP15(0.4, 0.8, 1.2 g/kg) had lower serum uric acid levels(P < 0.05), accompanied by lower urine uric acid(P < 0.05). | 4 | Molecular characterization and expression analysis of Triticum aestivum squamosa-promoter binding protein-box genes involved in ear development显示文摘Wheat(Triticum aestivum L.) is one of the most important crops in the world. Squamosa-promoter binding protein(SBP)-box genes play a critical role in regulating flower and fruit development. In this study, 10 novel SBP-box genes(TaSPL genes) were isolated from wheat((Triticum aestivum L.) cultivar Yanzhan 4110). Phylogenetic analysis classified the TaSPL genes into five groups(G1–G5). The motif combinations and expression patterns of the TaSPL genes varied among the five groups with each having own distinctive characteristics: TaSPL20/21 in G1 and TaSPL17 in G2 mainly expressed in the shoot apical meristem and the young ear, and their expression levels responded to development of the ear; TaSPL6/15 belonging to G3 were upregulated and TaSPL1/23 in G4 were downregulated during grain development; the gene in G5(TaSPL3) expressed constitutively. Thus, the consistency of the phylogenetic analysis, motif compositions, and expression patterns of the TaSPL genes revealed specific gene structures and functions.On the other hand, the diverse gene structures and different expression patterns suggested that wheat SBP-box genes have a wide range of functions. | 6 | Growth and physiological responses of Agriophyllum squarrosum to sand burial stress显示文摘Agriophyllum squarrosum is an annual desert plant widely distributed on mobile and semi-mobile dunes in all the sandy deserts of China. We studied the growth and physiological properties of A. squarrosum seedlings under different sand burial depths in 2010 and 2011 at Horqin Sandy Land, Inner Mongolia to understand the ability and mechanism that A. squarrosum withstands sand burial. The results showed that A. squarrosum had a strong ability to withstand sand burial. Its survival rate, plant height and biomass increased significantly at a burial depth 25% of seedling height and decreased significantly only when the burial depth exceeded the height of the seedlings; some plants still survived even if the burial depth reached 266% of a seedling height. The malondialdehyde(MDA) content and membrane permeability of the plant did not change significantly as long as the burial depth was not greater than the seedling height; lipid peroxidation increased and cell membranes were damaged if the burial depth was increased further. When subjected to sand burial stress, superoxide dismutase(SOD) and peroxidase(POD) activities and free proline content increased in the seedlings, while the catalase(CAT) activity and soluble sugar content decreased. Sand burial did not lead to water stress. Reductions in photosynthetic area and cell membrane damage caused by sand burial may be the major mechanisms increasing mortality and inhibiting growth of the seedling. To explore the genetic basis of light responses of biomass and Researchphotosynthetic parameters in wheat(Triticum aestivum L.),a quantitative trait locus(QTL) analysis was carried out in a doubled haploid(DH) population grown in low light(LL),medium light(ML), and high light(HL), respectively. The results showed that the wheat seedlings grown in HL produced more biomass with lower total chlorophyll content(Chl), carotenoid content, and maximum photochemical efficiency of photosystem II(Fv/Fm) while the wheat seedlings grown in LL produced less biomass with higher Chl compared with those grown in ML. In total, 48 QTLs were identified to be associated with the investigated parameters in relation to growth light intensities. These QTLs were mapped to 15 chromosomes which individually explained6.3%–36.0% of the phenotypic variance, of which chromo-somes 3A, 1D, and 6B were specifically involved in LL response, 5D and 7A specifically involved in ML response,and 4B specifically involved in HL response. | 10 | DIW1 encoding a cladeⅠPP2C phosphatase negatively regulates drought tolerance by de-phosphorylating TaSnRK1.1 in wheat显示文摘Drought seriously impacts wheat production(Triticum aestivum L.),while the exploitation and utilization of genes for drought tolerance are insufficient.Leaf wilting is a direct reflection of drought tolerance in plants.Clade A PP2Cs are abscisic acid(ABA)co-receptors playing vital roles in the ABA signaling pathway,regulating drought response.However,the roles of other clade PP2Cs in drought tolerance,especially in wheat,remain largely unknown.Here,we identified a gain-of-function drought-induced wilting 1(DIW1)gene from the wheat Aikang 58 mutant library by map-based cloning,which encodes a cladeⅠprotein phosphatase 2C(TaPP2C158)with enhanced protein phosphatase activity.Phenotypic analysis of overexpression and CRISPR/Cas9 mutant lines demonstrated that DIW1/TaPP2C158 is a negative regulator responsible for drought resistance.We found that TaPP2C158 directly interacts with TaSnRK1.1 and de-phosphorylates it,thus inactivating the TaSnRK1.1–Ta AREB3 pathway.TaPP2C158 protein phosphatase activity is negatively correlated with ABA signaling.Association analysis suggested that C-terminal variation of TaPP2C158 changing protein phosphatase activity is highly correlated with the canopy temperature,and seedling survival rate under drought stress.Our data suggest that the favorable allele with lower phosphatase activity of TaPP2C158 has been positively selected in Chinese breeding history.This work benefits us in understanding the molecular mechanism of wheat drought tolerance,and provides elite genetic resources and molecular markers for improving wheat drought tolerance. | Jingyi Wang Chaonan Li Long Li Lifeng Gao Ge Hu Yanfei Zhang Matthew P.Reynolds Xueyong Zhang Jizeng Jia Xinguo Mao Ruilian Jing | 2023 | Journal of Integrative Plant Biology2023,65,8: | 1 |

| 15 | Effects of grazing and climate change on sandy grassland ecosystems in Inner Mongolia显示文摘To understand the effects of grazing activities and climate change on sandy grassland ecosystems in northern China, a livestock field grazing and enclosure experiment was conducted from 1992 to 2006 in Horqin Sand Land, Inner Mongolia. The results showed that sustained heavy grazing resulted in serious degradation of the vegetation; moderate grazing can maintain vegetation stabilization; and light grazing can promote rapid restoration of degraded vegetation. The livestock productivity was the highest in the moderate grazing grassland, and sustained heavy grazing resulted in rapid decrease of the livestock productivity. Heavy graz- ing can cause a retrogressive succession of grassland vegetation, whereas moderate and light grazing may promote progressive succession of plant species. The effects of changing climate on succession processes were not significant in the short term; a warm-humid climate is favorable to restoration of degraded vegetation, whereas a sustained warm-drought climate may result in degradation of grassland vegetation.
