1Upregulation of RIN3 induces endosomal dysfunction in Alzheimer’s disease显示文摘Background In Alzheimer’s Disease(AD),about one-third of the risk genes identified by GWAS encode proteins that function predominantly in the endocytic pathways.Among them,the Ras and Rab Interactor 3(RIN3)is a guanine nucleotide exchange factor(GEF)for the Rab5 small GTPase family and has been implicated to be a risk factor for both late onset AD(LOAD)and sporadic early onset AD(sEOAD).However,how RIN3 is linked to AD pathogenesis is currently undefined.Methods Quantitative PCR and immunoblotting were used to measure the RIN3 expression level in mouse brain tissues and cultured basal forebrain cholinergic neuron(BFCNs).Immunostaining was used to define subcellular localization of RIN3 and to visualize endosomal changes in cultured primary BFCNs and PC12 cells.Recombinant flag-tagged RIN3 protein was purified from HEK293T cells and was used to define RIN3-interactomes by mass spectrometry.RIN3-interacting partners were validated by co-immunoprecipitation,immunofluorescence and yeast two hybrid assays.Live imaging of primary neurons was used to examine axonal transport of amyloid precursor protein(APP)andβ-secretase 1(BACE1).Immunoblotting was used to detect protein expression,processing of APP and phosphorylated forms of Tau.Results We have shown that RIN3 mRNA level was significantly increased in the hippocampus and cortex of APP/PS1 mouse brain.Basal forebrain cholinergic neurons(BFCNs)cultured from E18 APP/PS1 mouse embryos also showed increased RIN3 expression accompanied by early endosome enlargement.In addition,via its proline rich domain,RIN3 recruited BIN1(bridging integrator 1)and CD2AP(CD2 associated protein),two other AD risk factors,to early endosomes.Interestingly,overexpression of RIN3 or CD2AP promoted APP cleavage to increase its carboxyl terminal fragments(CTFs)in PC12 cells.Upregulation of RIN3 or the neuronal isoform of BIN1 increased phosphorylated Tau level.Therefore,upregulation of RIN3 expression promoted accumulation of APP CTFs and increased phosphorylated Tau.These effects by RIN3 was rescued by the expression of a dominant negative Rab5(Rab5S34N)construct.Our study has thus pointed to that RIN3 acts through Rab5 to impact endosomal trafficking and signaling.Conclusion RIN3 is significantly upregulated and correlated with endosomal dysfunction in APP/PS1 mouse.Through interacting with BIN1 and CD2AP,increased RIN3 expression alters axonal trafficking and procession of APP.Together with our previous studies,our current work has thus provided important insights into the role of RIN3 in regulating endosomal signaling and trafficking.Ruinan Shen Xiaobei Zhao Lu He Yongbo Ding Wei Xu Suzhen Lin Savannah Fang Wanlin Yang Kijung Sung Brian Spencer Robert A.Rissman Ming Lei Jianqing Ding Chengbiao Wu 2020Translational Neurodegeneration2020,9,2:1
2The mechanisms of grain growth of Mg alloys:A review显示文摘Grain growth directly influences the plasticity and strength of Mg alloys.As the grain size decreases from the microscale to the nanoscale,the plasticity of Mg alloys continually increases,whereas the strength first increases and later decreases.These trends are observed because the plastic deformation mechanism changes from dislocation–twinning dominance to grain boundary dominance.In this study,the factors influencing grain growth,such as the temperature of plastic deformation/annealing,second-phase particles and solute atoms,are examined to aid effective control of the grain size.Additionally,the mechanisms of grain growth,typically induced by strain and thermal activation,are clarified.Strain-induced grain boundary migration is attributable to the difference in the strain energy stored in adjacent grains with high-density dislocations.Heat-induced grain boundary migration is driven by the difference in the energy of the grain boundary/subgrain boundary and boundary curvature.Abnormal grain growth can be induced by anisotropy of the strain energy,anisotropy of the grain boundary mobility,depinning of the second phase and high misorientation gradient.Qinghua Chen Ruinan Chen Jian Su Qingsong He Bin Tan Chao Xu Xu Huang Qingwei Dai Jian Lu 2022Journal of Magnesium and Alloys2022,10,9:1
3Solid-state bonding process induced highly synergistic mechanical properties of 6061 Al alloy joint by shear deformation显示文摘Obtaining a highly synergistic mechanical property between joint and base metal(BM)in aluminum(Al)alloy is a chronic problem.In this work,shear bonding technology is applied to a commercial 6061 Al alloy.The results show that a flat and uniform joint interface is obtained.The joint presents a highly synergistic mechanical property compared with BM,whether after shear bonding or heat treatment.The joint coefficient reaches 95.6%after shear bonding and exceeds 100%after heat treatment,which is better than the traditional connection method of aluminum alloy.The high joint coefficient mainly originates from the well-linked joint and gradient grain structure.The gradient grain structure is beneficial to activate more slip systems to coordinate plastic deformation.Although the fine-grained structure is sacrificed after heat treatment,higher strength and joint coefficient are obtained due to the higher work hardening.This newly developed method has a large potential for application to the infinite rolling of Al alloy sheets and can also be used for Al alloy connection in automobile,aerospace,rail transportation,and other fields.The findings in this work can provide essential theoretical support and application reference for the shear connection of Al alloy.Peng Peng Jian Su Qingsong He Ruinan Chen Sensen Chai Daliang Yu Qingwei Dai Jian Lu 2023Journal of Materials Science & Technology2023,,33:0
4Very small embryonic like(VSEL) stem cells显示文摘It is generally accepted that adult bone marrow(BM) contains both hematopoietic stem cells(HSCs) and mesenchymal stem cells (MSCs). Recently, a rare population of stem cells different from HSCs and MSCs were identified in murine BM and human cord blood (CB), named as very small embryonic like(VSEL) stem cells. 5Implications of vitamin D for flesh quality of grass carp(Ctenopharyngodon idella):antioxidant ability,nutritional value,sensory quality,and myofiber characteristics显示文摘Background Muscle represents a unique and complex system with many components and comprises the major edible part of animals.Vitamin D is a critical nutrient for animals and is known to enhance calcium absorption and immune response.In recent years,dietary vitamin D supplementation in livestock has received increased attention due to biological responses including improving shear force in mammalian meat.However,the vitamin D acquisition and myofiber development processes in fish differ from those in mammals,and the effect of vitamin D on fish flesh quality is poorly understood.Here,the influence of dietary vitamin D on fillet quality,antioxidant ability,and myofiber development was examined in grass carp(Ctenopharyngodon idella).Methods A total of 540 healthy grass carp,with an initial average body weight of 257.24±0.63 g,were allotted in 6 experimental groups with 3 replicates each,and respectively fed corresponding diets with 15.2,364.3,782.5,1,167.9,1,573.8,and 1,980.1 IU/kg vitamin D for 70 d.Results Supplementation with 1,167.9 IU/kg vitamin D significantly improved nutritional value and sensory quality of fillets,enhancing crude protein,free amino acid,lipid,and collagen contents;maintaining an ideal pH;and reduc-ing lactate content,shear force,and cooking loss relative to respective values in the control(15.2 IU/kg)group.Average myofiber diameter and the frequency of myofibers>50μm in diameter increased under supplementation with 782.5–1,167.9 IU/kg vitamin D.Levels of oxidative damage biomarkers decreased,and the expression of antioxi-dant enzymes and nuclear factor erythroid 2-related factor 2 signaling molecules was upregulated in the 1,167.9 IU/kg vitamin D treatment compared to respective values in the control group.Furthermore,vitamin D supplementation activated cell differentiation by enhancing the expression of myogenic regulatory factors and myocyte enhancer factors compared to that in the control group.In addition,supplementation with 1,167.9 IU/kg vitamin D improved protein deposition associated with protein synthesis molecule(target of rapamycin)signaling and vitamin D receptor paralogs,along with inhibition of protein degradation(forkhead box protein 1)signaling.Conclusions Overall,the results demonstrated that vitamin D strengthened antioxidant ability and myofiber devel-opment,thereby enhancing nutritional value and sensory quality of fish flesh.These findings suggest that dietary vitamin D supplementation is conducive to the production of nutrient-rich,high quality aquaculture products.Yao Zhang Chaonan Li Xiaoqiu Zhou Weidan Jiang Pei Wu Yang Liu Hongmei Ren Lu Zhang Haifeng Mi Jiayong Tang Ruinan Zhang Lin Feng 2023Journal of Animal Science and Biotechnology2023,14,6:0
6Clinical features of multiple myeloma patients with isolated extramedullary relapse显示文摘This study sought to analyze the clinical features and prognosis of multiple myeloma with isolated extramedullary relapse and with the absence of systemic progression.The clinical features and outcome were retrospectively analyzed in six multiple myeloma patients.These patients had secretory multiple myeloma at diagnosis.When relapsed,the dissociation between medullary and extramedullary response was detected.The serum or urine monoclonal component was extremely low or absent.The plasma cells in bone marrow were <5%.All patients received new targeted therapies(thalidomide or bortezomib) before extramedullary relapse.It is difficult to achieve second remission for them.Even in those showing response,the duration of response was extremely short.The median of overall survival from diagnosis and from extramedullary relapse was 19 months and 6 months,respectively.The overall survival was significantly shorter compared to the patients without extramedullary involvement(84 months,P= 0.001).These patients exhibited a special and rare relapse pattern.Patients with this relapse pattern were resistant to current therapies,including novel targeted agents and associated with poor prognosis.Xiaoyan Qu Lijuan Chen Tian Tian Limin Duan Ruinan Lu Hua Lu Hanxin Wu Jianyong Li 2018The Journal of Biomedical Research2018,32,4:0
7Optical engineering of infrared PbS CQD photovoltaic cells for wireless optical power transfer systems显示文摘Infrared photovoltaic cells(IRPCs)have attracted considerable attention for potential applications in wireless optical power transfer(WOPT)systems.As an efcient fber-integrated WOPT system typically uses a 1550 nm laser beam,it is essential to tune the peak conversion efciency of IRPCs to this wavelength.However,IRPCs based on lead sulfde(PbS)colloidal quantum dots(CQDs)with an excitonic peak of 1550 nm exhibit low short circuit current(Jsc)due to insufcient absorption under monochromatic light illumination.Here,we propose comprehensive optical engineering to optimize the device structure of IRPCs based on PbS CQDs,for 1550 nm WOPT systems.The absorption by the device is enhanced by improving the transmittance of tin-doped indium oxide(ITO)in the infrared region and by utilizing the optical resonance efect in the device.Therefore,the optimized device exhibited a high short circuit current density of 37.65 mA/cm^(2)under 1 sun(AM 1.5G)solar illumination and 11.91 mA/cm^(2)under 1550 nm illumination 17.3 mW/cm^(2).Furthermore,the champion device achieved a record high power conversion efciency(PCE)of 7.17%under 1 sun illumination and 10.29%under 1550 nm illumination.The PbS CQDs IRPCs under 1550 nm illumination can even light up a liquid crystal display(LCD),demonstrating application prospects in the future.Mengqiong Zhu Yuanbo Zhang Shuaicheng Lu Zijun Wang Junbing Zhou Wenkai Ma Ruinan Zhu Guanyuan Chen Jianbing Zhang Liang Gao Jiancan Yu Pingqi Gao Jiang Tang 2023Frontiers of Optoelectronics2023,16,2:0
