Lingguizhugan Decoction, a Chinese herbal formula, improves insulin resistance in overweight/obese subjects with non-alcoholic fatty liver disease: a translational approach显示文摘Lingguizhugan Decoction(LGZG)has been investigated in basic studies, with satisfactory effects on insulin resistance in non-alcoholic fatty liver disease(NAFLD).This translational approach aimed to explore the effect and underlying mechanism of LGZG in clinical setting.A randomized,double-blinded,placebo-controlled trial was performed.A total of 243 eligible participants with NAFLD were equally allocated to receive LGZG(two groups:standard dose and low dose)or placebo for 12 weeks on the basis of lifestyle modifications. The primary efficacy variable was homeostasis model assessment of insulin resistance(HOMA-IR).Analyses were performed in two populations in accordance with body mass index(BMI;overweight/obese,BMI 24 kg/m^(2);lean,BMI<24 kg/m^(2)).For overweight/obese participants,low-dose LGZG significantly decreased their HOMA-IR level compared with placebo(0.19(1.47)versus 0.08(1.99),P=0.038).For lean subjects, neither dose of LGZG showed a superior effect compared with placebo.Methylated DNA immunoprecipitation sequencing and real-time qPCR found that the DNA N6-methyladenine modification levels of protein phosphatase 1 regulatory subunit 3A(PPP1R3A)and autophagy related 3(ATG3)significantly increased after LGZG intervention in overweight/obese population.Low-dose LGZG effectively improved insulin resistance in overweight/obese subjects with NAFLD.The underlying mechanism may be related to the regulation of DNA N6-methyladenine modification of PPP1R3A and ATG3.Lean subjects may not be a targeted population for LGZG. | Liang Dai Jingjuan Xu Baocheng Liu Yanqi Dang Ruirui Wang Lijie Zhuang Dong Li Lulu Jiao Jianying Wang Lei Zhang Linda LDZhong Wenjun Zhou Guang Ji | 2022 | Frontiers of Medicine2022,16,5: | 7

The C_(6)D_(6)detector system on the Back-n beam line of CSNS显示文摘Introduction The neutron capture cross sections are very important in the field of nuclear device design and basic physics research.Hydrogen-free liquid scintillator such as C_(6)D_(6)detectors are widely used in the neutron capture cross-sectional measurements for the low neutron sensitivity and fast time response. The Back-n white neutron source at China Spallation Neutron Source is the first spallation white neutron source in China, and it is suitable for neutron capture cross-sectional measurement.Materials and methods A C_(6)D_(6)detector system was built in the Back-n experimental station. The pulse height weighting technique was used to determine the system’s detection efficiency. The response to gamma rays of the C_(6)D_(6)detector was measured, and the energy resolution function was determined.Monte Carlo simulation with Geant4 code was carried out to get the weighting function of this C_(6)D_(6)detector system.Additionally, the systematic uncertainty of the weighting function was also determined. Conclusion According to the experimental and simulation results, this C_(6)D_(6)detector system can be used to measure neutron capture cross section. | Jie Ren Xichao Ruan Jie Bao Guangyuan Luan Wei Jiang Qi An Huaiyong Bai Ping Cao Qiping Chen Yonghao Chen Pinjing Cheng Zengqi Cui Ruirui Fan Changqing Feng Minhao Gu Fengqin Guo Changcai Han Zijie Han Guozhu He Yongcheng He Yuefeng He Hanxiong Huang Weiling Huang Xiru Huang Xiaolu Ji Xuyang Ji Haoyu Jiang Hantao Jing Ling Kang Mingtao Kang Bo Li Lun Li Qiang Li Xiao Li Yang Li Yang Li Rong Liu Shubin Liu Xingyan Liu Yinglin Ma Changjun Ning Binbin Qi Zhaohui Song Hong Sun Xiaoyang Sun Zhijia Sun Zhixin Tan Hongqing Tang Jingyu Tang Pengcheng Wang Qi Wang Taofeng Wang Yanfeng Wang Zhaohui Wang Zheng Wang Jie Wen Zhongwei Wen Qingbiao Wu Xiaoguang Wu Xuan Wu Likun Xie Yiwei Yang Han Yi Li Yu Tao Yu Yongji Yu Guohui Zhang Jing Zhang Linhao Zhang Liying Zhang Qingmin Zhang Qiwei Zhang Xianpeng Zhang Yuliang Zhang Zhiyong Zhang Yingtan Zhao Liang Zhou Zuying Zhou Danyang Zhu Kejun Zhu Peng Zhu | 2019 | Radiation Detection Technology and Methods2019,3,3: | 1

Inherited Cardiomyopathies:Genetics and Clinical Genetic Testing显示文摘Inherited cardiomyopathies are major causes of morbidity and mortality and include a group of cardiac disorders such as hypertrophic cardiomyopathy(HCM),dilated cardiomyopathy, arrhythmogenic right ventricular dysplasia/cardiomyopathy(ARVD/C),left ventricular noncompaction(LVNC),and restrictive cardiomyopathy(RCM).These diseases have a substantial genetic component and predispose to sudden cardiac death.Since the first gene was identified as a disease-causing gene for HCM over two decades ago, more than eighty genes have been identified to be associated with inherited cardiomyopathies and genetic testing has become prevalent in making clinical diagnosis. With the advent of next-generation sequencing technology, genetic panel testing of inherited cardiomyopathies has become feasible and cost efficient. In this review,we summarize the individual cardiomyopathies with the emphasis on cardiomyopathy genetics and genetic testing. | Guoliang Wang Ruirui Ji Wenxin Zou Daniel J.Penny Yuxin Fan | 2017 | Cardiovascular Innovations and Applications2017,,B02: | 1

An Adaptive Physics-Informed Neural Network with Two-Stage Learning Strategy to Solve Partial Differential Equations显示文摘Physics-Informed Neural Network(PINN)represents a new approach to solve Partial Differential Equations(PDEs).PINNs aim to solve PDEs by integrating governing equations and the initial/boundary conditions(I/BCs)into a loss function. However, the imbalance of the loss function caused by parameter settings usually makes it difficult for PINNs to converge,e.g.because they fall into local optima. In other words, the presence of balanced PDE loss, initial loss and boundary loss may be critical for the convergence. In addition, existing PINNs are not able to reveal the hidden errors caused by non-convergent boundaries and conduction errors caused by the PDE near the boundaries.Overall, these problems have made PINN-based methods of limited use on practical situations. In this paper,we propose a novel physics-informed neural network,i.e.an adaptive physics-informed neural network with a two-stage training process. Our algorithm adds spatio-temporal coefficient and PDE balance parameter to the loss function, and solve PDEs using a two-stage training process:pre-training and formal training. The pre-training step ensures the convergence of boundary loss, whereas the formal training process completes the solution of PDE by balancing various loss functions. In order to verify the performance of our method,we consider the imbalanced heat conduction and Helmholtz equations often appearing in practical situations. The Klein-Gordon equation, which is widely used to compare performance, reveals that our method is able to reduce the hidden errors.Experimental results confirm that our algorithm can effectively and accurately solve models with unbalanced loss function, hidden errors and conduction errors. The codes developed in this manuscript are publicy available at https://github.com/callmedrcom/ATPINN. | Shuyan Shi Ding Liu Ruirui Ji Yuchao Han | 2023 | Numerical Mathematics(Theory,Methods and Applications)2023,16,2: | 0
Correlation of PCSK1 with nonalcoholic fatty liver disease in a Han Chinese population:a case-control observational study显示文摘Objective: This study aimed to investigate the association between single-nucleotide polymorphisms (SNPs) ofPCSK1 (proprotein convertase subtilisin/kexin type 1) related to obesity and nonalcoholic fatty liver disease (NAFLD).Methods: In this case-control observational study, four candidate SNPs (rs6234, rs155971, rs6232, rs3811951) ofPCSK1 were genotyped in 732 NAFLD patients and 823 healthy control participants, all of whom were of ethnic Han Chinese descent. All participants came from Shanghai, China, and joined our study during 2015 to 2016. The frequencies of each allele and genotype, paired linkage disequilibrium, and haplotype were calculated on the SHEsis platform. In addition to SHEsis, five different genetic models (codominant, dominant, recessive, overdominant, and log-additive) were employed to identify the correlation between genotype frequency and NAFLD. This study was approved by the Medical Ethics Committee of Shanghai University of Traditional Chinese Medicine (approved No. 2017LCSY069).Results: In a comparison of NAFLD patients and healthy participants, none of the fourPCSK1 SNPs were significantly correlated with the occurrence of NAFLD (P>0.05), in either genotypic or allelic distribution. The recessive model of rs3811951 appeared to show a correlation (odds ratio=1.077;95% confidence interval=0.924-1.256;P=0.04), but there was no statistical significance after Bonferroni correction (Pcorr>0.0125).Conclusions: Four obesity-relatedPCSK1 SNPs (rs6234, rs155971, rs6232, rs3811951) showed no significant correlation with the development of NAFLD in a Han Chinese population. | Xiao Yang Fan Yuan Na Wu Decheng Ren Liangjie Liu Yan Bi Lei Ji Ke Han Mofan Feng Kai Su Fengping Yang Xi Wu Qing Lu Siran Yue Fengyan Jiang Ruirui Wang Guang He Baocheng Liu Xingwang Li Yi Shi | 2022 | Journal of Bio-X Research2022,5,3: | 0
