1A BAC clone of MDV strain GX0101 with REV-LTR integration retained its pathogenicity显示文摘The complete genome of Marek's disease virus (MDV) strain GX0101, which was integrated with the LTR sequences of REV, was cloned in Escherichia coli as a bacterial artificial chromosome (BAC). BAC vector sequences were introduced into the US2 locus of the MDV genome by homologous recombination. The viral DNA containing the BAC vector was used to transform Escherichia coli strain of DH10B. Then the recombinant virus was successfully rescued by transfection of the recombinant BAC DNA into primary chicken embryo fibroblast (CEF). This BAC viral clone was named bac-GX0101. When the reconstituted virus was inoculated into 1-day-old birds, visceral tumors could be detected as early as 62 d post infection. There was no difference in growth ability and pathogenicity to birds between the BAC derived virus and its parental virus. The BAC derived virus maintained its oncogenicity and immunosuppressive effects. In conclusion, the complete genome of GX0101 strain was successfully cloned into BAC and the infectious clone was rescued. 2Effect of composite supports on the methanation activity of Co-Mo-based sulphur-resistant catalysts显示文摘The effects of composite supports of CeO2-Al2O3,MgO-Al2O3,TiO2-Al2O3 or ZrO2-Al2O3 on the methanation activity of supported Co-Mo-based sulphur-resistant catalysts were investigated.The catalysts were further characterized by nitrogen adsorption measurement,X-ray diffraction and X-ray photoelectron spectroscopy.The catalyst of 5%CoO-15%MoO3 supported on CeO2-Al2O3,MgO-Al2O3,TiO2-Al2O3 or ZrO2-Al2O3 composite oxides,respectively,showed different catalytic performances of syngas methanation in the presence of hydrogen sulphide as compared with that of the 5%CoO-15%MoO3/Al2O3 catalyst.The Co-Mo/CeO2-Al2O3 catalyst shows the highest methanation activity among the tested catalysts.The enhanced methanation activity may be attributed to the improvement of the dispersion of active metal species and the inhibition of the formation of S6+.Haiyang Wang Zhenhua Li Erdong Wang Can Lin Yuguang Shang Guozhong Ding Xinbin Ma Shaodong Qin Qi Sun 2012Journal of Natural Gas Chemistry2012,21,6:15
3Experimental studies on the P-T stability conditions and influencing factors of gas hydrate in different systems显示文摘The P-T stability conditions of gas hydrate in different systems (i.e., solution, silica sand, and marine sediment) were studied using multi-step decomposition method with our experimental equipment. The effects of different ions with various concentrations and sediment grains on the P-T stability conditions of gas hydrate were investigated. The results show that different ions have different influences on the phase equilibrium of gas hydrate. However, the influence of ions is in a similar trend: the larger the concentration, the bigger the P-T curve shifts to the left. For the silica sand, the influence of pore capillarity of coarse particles (> 460 μm) can be negligible. The P-T curve measured in coarse silica is in agreement with that in pure water. However, the influence of pore capillarity of fine particles (< 35 μm) is significant. The maximum reduction value of temperature is 1.5 K for methane hydrate under stable state. The sediment from the South China Sea significantly affects the P-T stability conditions of methane hydrate, with an average reduction value of 1.9 K within the experimental conditions. This is mainly the result of both the pore water salinity and the pore capillarity of sediment. 4Influence of foraminifera on formation and occurrence characteristics of natural gas hydrates in fine-grained sediments from Shenhu area, South China Sea显示文摘Marine gas hydrates accumulate primarily in coarse-grained, high-permeability layers; however, highly saturated natural gas hydrates have been discovered in the fine-grained sediments of Shenhu area, South China Sea(SCS). This may be explained by key factors, such as the great abundance of foraminifera shells. In this paper, by analyzing the SCS foraminifera structure and performing hydrate formation experiments in the foraminifera shells, the contribution of foraminifera to hydrate accumulation in the SCS was investigated from a microscopic point of view. Simulations of hydrate formation were carried out in both pure SCS foraminifera shells and the host sediments. Pore structures in typical foraminifera were studied by use of micro-focus X-ray computed tomography(CT) and scanning electron microscopy(SEM). Hydrate growth and occurrence characteristics in the foraminifera shells were observed in-situ. The results showed that the presence of foraminifera significantly enhanced the effective porosity of the SCS sediments. Moreover, while the hydrates grew preferentially in the chambers of the coarse-grained foraminifera by adhering to the inner walls of the foraminifera shells, no apparent hydrate accumulation was observed in the fine-grained or argillaceous matrix. 5Simplified mathematical model of inter-turn short circuit of field windings in hydro-generators and its application显示文摘Inter-turn short circuit of field windings is a common electrical fault of generators.Simulation is an important method of investigating the fault and providing data support for fault monitoring.However,huge numbers of pole pairs and damper loops in large hydro-generators would lead to lengthy calculation time,hindering scientific research and engineering application.To deal with this problem,we analyze a theoretical basis for a damper winding simplified model and then propose an equivalent treatment method.Through the analysis of steady-state current harmonic characteristics of generators with different stator winding configurations during the fault,the simplified models suitable for steady-state calculation are derived from two aspects,namely,additional rotor harmonic current frequency characteristics and the relationship of the amplitude as well as the phase of each branch current of the stator.The calculation and experimental results of the two simplified models are then compared to verify the models' correctness.A calculation example of the Three Gorges left bank VGS generator shows few deviations between the calculation results of the simplified model and the original model.Moreover,the calculation time using the simplified model is 1/1500 that using the original model,which provides a more effective tool for on-line fault monitoring.Finally,the sensitivity-verification application of the fault-monitoring scheme based on the stator steady-state unbalanced current RMS is depicted.The result shows that the scheme can monitor two-turn short circuits of field windings in the Three Gorges generator and provide high sensitivity.HAO LiangLiang WU JunYong SUN YuGuang WANG XiangHeng 2013Science China(Technological Sciences)2013,56,4:5
6Numerical aerodynamic analysis of bluff bodies at a high Reynolds number with three-dimensional CFD modeling显示文摘This paper focuses on numerical simulations of bluff body aerodynamics with three-dimensional CFD(computational fluid dynamics) modeling,where a computational scheme for fluid-structure interactions is implemented.The choice of an appropriate turbulence model for the computational modeling of bluff body aerodynamics using both two-dimensional and three-dimensional CFD numerical simulations is also considered.An efficient mesh control method which employs the mesh deformation technique is proposed to achieve better simulation results.Several long-span deck sections are chosen as examples which were stationary and pitching at a high Reynolds number.With the proposed CFD method and turbulence models,the force coefficients and flutter derivatives thus obtained are compared with the experimental measurement results and computed values completely from commercial software.Finally,a discussion on the effects of oscillation amplitude on the flutter instability of a bluff body is carried out with extended numerical simulations.These numerical analysis results demonstrate that the proposed three-dimensional CFD method,with proper turbulence modeling,has good accuracy and significant benefits for aerodynamic analysis and computational FSI studies of bluff bodies.BAI YuGuang YANG Kai SUN DongKe ZHANG YuGuang KENNEDY David WILLIAMS Fred GAO XiaoWei 2013Science China(Physics,Mechanics & Astronomy)2013,56,2:4
7P-T stability conditions of methane hydrate in sediment from South China Sea显示文摘For reasonable assessment and safe exploitation of marine gas hydrate resource, it is important to determine the stability conditions of gas hydrates in marine sediment. In this paper, the seafloor water sample and sediment sample (saturated with pore water) from Shenhu Area of South China Sea were used to synthesize methane hydrates, and the stability conditions of methane hydrates were investigated by multi-step heating dissociation method. Preliminary experimental results show that the dissociation temperature of methane hydrate both in seafloor water and marine sediment, under any given pressure, is depressed by approximately -1.4 K relative to the pure water system. 9Conditioned medium-electrospun fiber biomaterials for skin regeneration显示文摘Conditioned medium(CM)contains variety of factors secreted by cells,which directly regulate cellular processes,showing tremendous potential in regenerative medicine.Here,for the first time,we proposed a novel regenerative therapy mediated by biodegradable micro-nano electrospun fibers loaded with highly active conditioned medium of adipose-derived stem cells(ADSC-CM).ADSC-CM was successfully loaded into the nanofibers with biological protection and controllable sustained-release properties by emulsion electrospinning and protein freeze-drying technologies.In vitro,ADSC-CM released by the fibers accelerated the migration rate of fibroblasts;inhibited the over proliferation of fibroblasts by inducing apoptosis and damaging cell membrane;in addition,ADSC-CM inhibited the transformation of fibroblasts into myofibroblasts and suppressed excessive production of extracellular matrix(ECM).In vivo,the application of CM-biomaterials significantly accelerated wound closure and improved regeneration outcome,showing superior pro-regenerative performance.This study pioneered the application of CM-biomaterials in regenerative medicine,and confirmed the practicability and significant biological effects of this innovative biomaterials.Lu Chen Liying Cheng Zhen Wang Jianming Zhang Xiyuan Mao Zhimo Liu Yuguang Zhang Wenguo Cui Xiaoming Sun 2021Bioactive Materials2021,6,2:2
10Pore capillary pressure and saturation of methane hydrate bearing sediments显示文摘To better understand the relationship between the pore capillary pressure and hydrate saturation in sediments, a new method was proposed. First, the phase equilibria of methane hydrate in fine-grained silica sands were measured. As to the equilibrium data, the pore capillary pressure and saturation of methane hydrate were calculated. The results showed that the phase equilibria of methane hydrates in fine-grained silica sands changed due to the depressed activity of pore water caused by the surface group and negatively charged characteristic of silica particles as well as the capillary pressure in small pores together. The capillary pressure increased with the increase of methane hydrate saturation due to the decrease of the available pore space. 16Programmable immune activating electrospun fibers for skin regeneration显示文摘Immune cells play a crucial regulatory role in inflammatory phase and proliferative phase during skin healing.How to programmatically activate sequential immune responses is the key for scarless skin regeneration.In this study,an“Inner-Outer”IL-10-loaded electrospun fiber with cascade release behavior was constructed.During the inflammatory phase,the electrospun fiber released a lower concentration of IL-10 within the wound,inhibiting excessive recruitment of inflammatory cells and polarizing macrophages into anti-inflammatory phenotype“M2c”to suppress excessive inflammation response.During the proliferative phase,a higher concentration of IL-10 released by the fiber and the anti-fibrotic cytokines secreted by polarized“M2c”directly acted on dermal fibroblasts to simultaneously inhibit extracellular matrix overdeposition and promote fibroblast migration.The“Inner-Outer”IL-10-loaded electrospun fiber programmatically activated the sequential immune responses during wound healing and led to scarless skin regeneration,which is a promising immunomodulatory biomaterial with great potential for promoting complete tissue regeneration.Lu Chen Liucheng Zhang Hongbo Zhang Xiaoming Sun Dan Liu Jianming Zhang Yuguang Zhang Liying Cheng Hélder A.Santos Wenguo Cui 2021Bioactive Materials2021,6,10:1

19Occurrence Characteristics of Saline-Lacustrine Shale-Oil in the Qianjiang Depression,Jianghan Basin,Central China显示文摘The amount of shale oil and its characterization are key issues in the study of shale oil.At present,many scholars use a variety of methods to evaluate the amount of shale oil,and use the calculated amount of hydrocarbons to analyze its influencing factors;however,there is lack of detailed research on the storage space for shale-oil and it’s influencing factors.In view of this issue,gas chromatography(GC),gas chromatography-mass spectrometry(GC-MS),soxhlet extraction(SE),fieldemission scanning electron microscopy(FE-SEM),low-temperature nitrogen adsorption(LTNA),highpressure mercury intrusion(HPMI),and X-ray diffraction(XRD)were used to analyze and compare samples from two wells located in different deposition locations.The unconventional Well BYY2 and BX7 s were drilled in the depocenter and distal area of the Qianjiang Formation,respectively.Controlled by differences in the organic matter type and sedimentary environment,the organic matter in Well BYY2,which was found to be characterized by laminar shale,mainly originated from aquatic algae.The results showed that as the total organic carbon(TOC)content increased,the amount of shaleoil in the pores increased.Shale-oil was mostly stored in mesopores and macropores that had been preserved by dolomite minerals of a biogenic origin and also occurred in pores sized 5–200 nm and>1μm.Dolomite minerals of a biogenic origin and clay minerals contributed to the occurrence of shaleoil.In comparison,the organic matter in Well BX7 has been greatly influenced by terrestrial organic matter.Pores in the massive mudstones from Well BX7 were determined to be mainly mesopores preserved by clay minerals and quartz,and the shale-oil was mostly stored within the pores of<40 nm.When the TOC content was~2.5 wt.%,the generated shale-oil reached saturation.Clay minerals contributed to the occurrence of shale-oil,whereas quartz only contributed to the occurrence of shale-oil in macropores.Zhongliang Sun Zhiliang He Furong Wang Yuanjia Han Sheng He Yuguang Hou Jing Luo Youheng Zheng Shiqiang Wu 2022Journal of Earth Science2022,33,4:1
