5HOXC10 up-regulation promotes gastric cancer cell proliferation and metastasis through MAPK pathway显示文摘Objective: As an important regulator of embryonic morphogenesis,homeodomain-containing gene 10(HOXC10) has been found to promote progression of human cancers and its expression indicates poor survival outcome.However,very few studies are available on the role of HOXC10 in gastric carcinoma.Therefore,the aim of this study was to determine the role of HOXC10 in gastric cancer and the potential mechanism underlying its function for cancer biology.Methods: A primary gastric cancer mouse model was obtained via intra-gastric wall injection of gastric cancer cells and was used to evaluate the function of HOXC10 during gastric cancer progression in vivo.Immunohistochemistry was performed to visualize and measure HOXC10 protein expression in gastric cancer tissue.Cells were transfected with plasmids to increase the expression of HOXC10,and si RNA transfection was performed to suppress HOXC10 expression.Reverse transcription polymerase chain reaction(RT-PCR) and western blotting were utilized to measure m RNA and protein expression,respectively.Proliferation,migration,and invasion were investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay,wound healing assay,and matrigel invasion assay in vitro,respectively.Results: HOXC10 expression was significantly increased in gastric cancer tissues compared to matched normal tissues.HOXC10 up-regulation significantly increased tumor volumes in nude mice.Plasmid transfection significantly increased HOXC10 protein and m RNA expressions and effectively promoted cell proliferation.Moreover,HOXC10 up-regulation significantly promoted migration and invasion of gastric cancer cells.Mechanistic investigation showed that HOXC10 up-regulation significantly increased m RNA and protein expression of mitogen-activated protein kinase(MAPK) signaling related genes,including c-myc,c-jun and p53,while also modulating the phosphorylation of c-Jun N-terminal kinase(JNK),extracellular signal-regulated kinase(ERK) and P38 but not their total protein levels.Conclusions: This study demonstrated the tight link between HOXC10 and gastric cancer cell proliferation and metastasis via involvement of the MAPK pathway.Chen Guo Jianing Hou Sheng Ao Xingming Deng Guoqing Lyu 2017Chinese Journal of Cancer Research2017,29,6:10

7Geochemical characteristics and applications of 18α(H)-neohopanes and 17α(H)-diahopanes显示文摘Distributions and geochemical significances of 18α(H)-neohopanes and of 17α(H)-diahopanes in oils from west Xialiaohe depression of the Bohai Bay Basin have been studied. Relative abundance of 18α(H)-neohopanes and of 17α(H)-diahopanes can be effectively used as maturity parameters, in a range of medium-high mature stage, based on consideration on sedimentary-organic facies of source rocks. With a dual geochemical significance of maturity and sedimentary-organic facies, neohopane and diahopane parameters can be well used in oil-source correlation and in petroleum population division. Formation, primary concentration and preservation of precursors of rearranged hopanes are closely related to depositional conditions such as Eh-pH and clays content. 9Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 10Ectomycorrhizal Fungi in Jiangsu Province,China显示文摘A survey was conducted for about 3 years to study the abundance and diversity of ectomycorrhizal fungi (EMF) in Jiangsu Province, China. The identification of the fungal species was based on the microscopic and macroscopic characteristics of their fruiting bodies. About 126 species of EMF were found in Jiangsu Province. 11Adsorption of rare earth ions using carbonized polydopamine nano carbon shells显示文摘Herein we reported the structure effects of carbon nano-shells prepared by the carbonization of polydopamine for the adsorption of rare earth elements(REEs) for the first time. Solid carbon spheres, 60 nm carbon shells and 500 nm carbon shells were prepared and evaluated for adsorption and desorption of REEs. The adsorption performance of carbon nano-shells for REEs was far superior to the solid carbon spheres. In addition, the effect of acidity on the adsorption and desorption properties was discussed. 14Ultra-sensitive Nanoprobe Modified with Tumor Cell Membrane for UCL/MRI/PET Multimodality Precise Imaging of Triple-Negative Breast Cancer显示文摘Triple-negative breast cancer(TNBC)is a subtype of breast cancer in which the estrogen receptor and progesterone receptor are not expressed,and human epidermal growth factor receptor 2 is not amplified or overexpressed either,which make the clinical diagnosis and treatment very challenging.Molecular imaging can provide an effective way to diagnose TNBC.Upconversion nanoparticles(UCNPs),are a promising new generation of molecular imaging probes.However,UCNPs still need to be improved for tumor-targeting ability and biocompatibility.This study describes a novel probe based on cancer cell membrane-coated upconversion nanoparticles(CCm-UCNPs),owing to the low immunogenicity and homologous-targeting ability of cancer cell membranes,and modified multifunctional UCNPs.This probe exhibits excellent performance in breast cancer molecular classification and TNBC diagnosis through UCL/MRI/PET tri-modality imaging in vivo.By using this probe,MDA-MB-231 was successfully differentiated between MCF-7 tumor models in vivo.Based on the tumor imaging and molecular classification results,the probe is also expected to be modified for drug delivery in the future,contributing to the treatment of TNBC.The combination of nanoparticles with biomimetic cell membranes has the potential for multiple clinical applications.Hanyi Fang Mengting Li Qingyao Liu Yongkang Gai Lujie Yuan Sheng Wang Xiao Zhang Min Ye Yongxue Zhang Mingyuan Gao Yi Hou Xiaoli Lan 2020Nano-Micro Letters2020,12,5:5
15A transceptor-channel complex couples nitrate sensing to calcium signaling in Ambidopsis显示文摘Nitrate-induced Ca^(2+) signaling is crucial for the primary nitrate response in plants.However,the molecular mechanism underlying the generation of the nitrate-specific calcium signature remains unknown.We report here that a cyclic nucleotide-gated channel(CNGC)protein,CNGC15,and the nitrate transceptor(NRT1.1)constitute a molecular switch that controls calcium influx depending on nitrate levels.The expression of CNGC15 is induced by nitrate,and its protein is localized at the plasma membrane after establishment of young seedlings.We found that disruption of CNGC15 results in the loss of the nitrate-induced Ca^(2+) signature(primary nitrate response)and retards root growth,reminiscent of the phenotype observed in the nrt1.1 mutant.We further showed that CNGC15 is an active Ca^(2+)-permeable channel that physically interacts with the NRT1.1 protein in the plasma membrane.Importantly,we discovered that CNGC15-NRT1.1 interaction silences the channel activity of the heterocomplex,which dissociates upon a rise in nitrate levels,leading to reactivation of the CNGC15 channel.The dynamic interactions between CNGC15 and NRT1.1 therefore control the channel activity and Ca^(2+) influx in a nitrate-dependent manner.Our study reveals a new nutrient-sensing mechanism that utilizes a nutrient transceptor-channel complex assembly to couple nutrient status to a specific Ca^(2+) signature.Xiaohan Wang Changxin Feng LiLi Tian Congcong Hou Wang Tian Bin Hu Qian Zhang Zhijie Ren Qi Niu Jiali Song Dongdong Kong Liangyu Liu Yikun He Ligeng Ma Chengcai Chu Sheng Luan Legong Li 2021Molecular Plant2021,14,5:5
16BmSd gene regulates the silkworm wing size by affecting the Hippo pathway显示文摘Insect wings are developed from the wing disc during metamorphosis.Bombyx mori,a model lepidopteran insect,loses flight ability after long-term domestication from the wild silkworm,Bombyx mandarina.The mw mutant(ul 1 strain)shows minute wings compared to wild type(e.g.,p50 strain)wings.RNA sequencing analysis previously revealed differential Hippo-pathway-related gene expression between the ull and p50strains.The Hippo pathway is an evolutionarily conserved signaling cascade that controls organ size during development in animals.In this study,the function of BmSd which has been characterized as one of the Hippo-pathway-related genes was analyzed for silkworm wing development.We found that mats,warts,and hippo expression levels were higher in u11 compared to p50 wing discs.BmSd(scalloped)expression,which encodes a prominent transcriptional partner to Yorkie(Yki),gradually decreased during the wandering stage in ull,but exhibited the opposite expression pattern in p50.When BmSd was knocked down by small interfering RNA during the wandering stage in the p50 strain,57.9%of the individuals showed minute wings.Additionally,ex,kibras and wingless expression levels decreased in the BmSd knockdown mutant.Further,BmSd deletion mediated by clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9 induced 50%of individuals with minute wings,a phenotype similar to the mw mutant.This result demonstrates that BmSd plays pivotal roles in silkworm wing development.Our results show that the Hippo signaling pathway participates and plays crucial roles in the regulation of silkworm wing development,and our findings provide a basis for further research on B.mori wing development.Jin Yin Jing Zhang Tao Li Xia Sun Sheng Qin Cheng-Xiang Hou Guo-Zheng Zhang Mu-Wang Li 2020Insect Science2020,27,4:5
17An ecosystem-based understanding and analysis for SENCE toward sustainable development显示文摘There is a need to find a comprehensive approach focusing on the conflicts between economical growth and environmental protection. Chinese scholars advocate a comprehensive ecosystem viewpoint named social\|economic\|natural complex ecosystem(SENCE). The kernel of the concept lies in the hierarchical structure of SENCE, through which methods from ecological network can be useful to the compound system. 18The Stereocomplex Formation and Phase Separation of PLLA/PDLA Blends with Different Optical Purities and Molecular Weights显示文摘In this study, the poly(L-lactide)/poly(D-lactide)(PLLA/PDLA) blends with different optical purities of PLLA and various molecular weights of PDLA are prepared by solution mixing, and the stereocomplex formation and phase separation behaviors of these blends are investigated. Results reveal that optical purity and molecular weight do not vary the crystal structure of PLA stereocomplex(sc) and homochiral crystallites(hc). As the optical purity increasing in the blends, the melting temperature of sc(Tsc) and the content of sc(ΔHsc) increased, while the melting temperature of hc(Thm) hardly changes, although the content of hc(ΔHhm) decreased gradually. The Tsc and ΔHsc are also enhanced as the molecular weight of PDLA reduces, and the ΔHhm reduces rapidly even though the Thm does not vary apparently. With lower optical purities of PLLA and higher molecular weights of PDLA, three types of crystals form in the blends, i.e., PLA sc, PLLA hc and PDLA hc. As molecular weight decreases and optical purity enhances, the crystal phase decreases to two(sc and PDLA hc), and one(sc) finally. 20Unsteady cooperative flow type in the axial compressor显示文摘For increasing the performance of the axial compressor, a method for realizing unsteady cooperative flow type is proposed in this paper as a critical objective in the new generation of the axial compressor design system. Unsteady excitations were utilized to trigger the transformation from the unsteady natural flow pattern into the unsteady cooperative flow pattern, resulting in increment of aerodynamic performances of axial compressor. Numerical simulations show that distinct positive effect can be obtained for the 2D cascade in a wide range of subsonic working conditions. No positive effect can be observed under the 2D supersonic working conditions and unsteady excitations have little influence on the flow field space-time structure. However, positive effect can be obtained under the 3D transsonic working conditions.
