3Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. 4A life support-based comprehensive treatment regimen dramatically lowers the in-hospital mortality of patients with fulminant myocarditis: a multiple center study显示文摘Fulminant myocarditis(FM) has unacceptable high mortality. This study aimed to evaluate the therapeutic efficacy of a life support-based comprehensive treatment regimen(LSBCTR), a completely novel treatment regimen, for FM. A total of 169 FM patients recruited from January 2008 to December 2018 were divided into two groups: patients receiving LSBCTR(81 cases),which includes(i) mechanical life support(positive pressure respiration, intra-aortic balloon pump with or without extracorporeal membrane oxygenation),(ii) immunomodulation therapy using sufficient doses of glucocorticoids and immunoglobulins, and(iii) application of neuraminidase inhibitors, and those receiving conventional treatment(88 cases). The endpoints were in-hospital death and heart-transplantation. Of all the population, 44 patients(26.0%) died in hospitals. Inhospital mortality was 3.7%(3/81) for LSBCTR group and 46.6%(41/88) for traditional treatment(P<0.001). Early application of LSBCTR, mechanical life support, neuraminidase inhibitors, and immunomodulation therapy significantly contributed to reduction in in-hospital mortality. This study describes a novel treatment regimen for FM patients that dramatically reduces inhospital mortality. 5Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32
6The Association of Maternal Body Composition and Dietary Intake with the Risk of Gestational Diabetes Mellitus during the Second Trimester in a Cohort of Chinese Pregnant Women显示文摘Objective To investigate the association of maternal body composition and dietary intake with the risk of gestational diabetes mellitus(GDM). Methods A total 154 GDM subjects and 981 controls were enrolled in a prospective cohort study in 11 hospitals from May 20, 2012 to December 31, 2013. Bioelectrical impedance analysis and dietary surveys were used to determine body composition and to evaluate the intake of nutrients in subjects at 21-24 weeks' gestation(WG). Logistic regression analysis was applied to explore the relationships of maternal body composition and dietary intake with the risk of GDM morbidity. Results Age, pre-pregnant body weight(BW), and body mass index(BMI) were associated with increased risk of GDM. Fat mass(FM), fat mass percentage(FMP), extracellular water(ECW), BMI, BW, energy, protein, fat, and carbohydrates at 21-24 WG were associated with an increased risk of GDM. In contrast, fat free mass(FFM), muscular mass(MM), and intracellular water(ICW) were associated with a decreased risk of GDM. 11Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. 16The rice OsV4 encoding a novel pentatricopeptide repeat protein is required for chloroplast development during the early leaf stage under cold stress显示文摘Pentatricopeptide repeat(PPR)proteins,characterized by tandem arrays of a 35 amino acid motif,have been suggested to play central and broad roles in modulating the expression of organelle genes in plants.However,the molecular mechanisms of most rice PPR genes remains unclear.In this paper,we isolated and characterized a temperature-conditional virescent mutant,OsV4,in rice(Oryza sativa cultivar Jiahua1(WT,japonica rice variety)).The mutant displays albino phenotype and abnormal chloroplasts at the three leaf stage,which gradually turns green after the four leaf stage at a low temperature(20°C).But the mutant always develops green leaves and well-developed chloroplasts at a high temperature(32°C).Genetic and molecular analyses uncovered that OsV4 encodes a novel chloroplast-targeted PPR protein including four PPR motifs.Further investigations show that the mutant phenotype is associated with changes in chlorophyll content and chloroplast development.The OsV4transcripts only accumulate to high levels in young leaves,indicating that its expression is tissue-specific.In addition,transcript levels of some ribosomal components and plastid-encoded polymerase-dependent genes are dramatically reduced in the albino mutants grown at 20°C.These findings suggest that OsV4 plays an important role during early chloroplast development under cold stress in rice.Xiaodi Gong Qianqian Su Dongzhi Lin Quan Jiang Jianlong Xu Jianhui Zhang Sheng Teng Yanjun Dong 2014Journal of Integrative Plant Biology2014,56,4:19
17Exogenous NADPH ameliorates myocardial ischemia–reperfusion injury in rats through activating AMPK/mTOR pathway显示文摘Our previous study shows that nicotinamide adenine dinucleotide phosphate(NADPH)plays an important role in protecting against cerebral ischemia injury.In this study we investigated whether NADPH exerted cardioprotection against myocardial ischemia/reperfusion(I/R)injury.To induce myocardial I/R injury,rats were subjected to ligation of the left anterior descending branch of coronary artery for 30 min followed by reperfusion for 2 h.At the onset of reperfusion,NADPH(4,8,16 mg·kg?1·d?1,iv)was administered to the rats.We found that NADPH concentrations in plasma and heart were signi?cantly increased at 4 h after intravenous administration.Exogenous NADPH(8?16 mg/kg)signi?cantly decreased myocardial infarct size and reduced serum levels of lactate dehydrogenase(LDH)and cardiac troponin I(cTn-I).Exogenous NADPH signi?cantly decreased the apoptotic rate of cardiomyocytes,and reduced the cleavage of PARP and caspase-3.In addition,exogenous NADPH reduced mitochondrial vacuolation and increased mitochondrial membrane protein COXIV and TOM20,decreased BNIP3L and increased Bcl-2 to protect mitochondrial function.We conducted in vitro experiments in neonatal rat cardiomyocytes(NRCM)subjected to oxygen–glucose deprivation/restoration(OGD/R).Pretreatment with NADPH(60,500 nM)signi?cantly rescued the cell viability and inhibited OGD/R-induced apoptosis.Pretreatment with NADPH signi?cantly increased the phosphorylation of AMPK and downregulated the phosphorylation of mTOR in OGD/R-treated NRCM.Compound C,an AMPK inhibitor,abolished NADPH-induced AMPK phosphorylation and cardioprotection in OGD/R-treated NRCM.In conclusion,exogenous NADPH exerts cardioprotection against myocardial I/R injury through the activation of AMPK/mTOR pathway and inhibiting mitochondrial damage and cardiomyocyte apoptosis.NADPH may be a potential candidate for the prevention and treatment of myocardial ischemic diseases.Jiang Zhu Yi-fei Wang Xiao-ming Chai Ke Qian Ling-wei Zhang Peng Peng Pei-min Chen Jian-fang Cao Zheng-hong Qin Rui Sheng Hong Xie 2020Acta Pharmacologica Sinica2020,41,4:19
18Antiviral enantiomers of a bisindole alkaloid with a new carbon skeleton from the roots of Isatis indigotica显示文摘A pair of indole alkaloid enantiomers with a novel bisindolylacetamide skeleton, insatindibisindolamides A and B(1a and 1b), was isolated from an aqueous extract of Isatis indigotica roots. The enantiomers were separated by chiral HPLC. 194-Hydroxybenzyl-substituted amino acid derivatives from Gastrodia elata显示文摘Seven new 4-hydroxybenzyl-substituted amino acid derivatives(1-7), together with 11 known compounds, were isolated from an aqueous extract of the rhizomes of Gastrodia elata Blume.Their structures were determined by spectroscopic and chemical methods. Compounds 1-3 are pyroglutamate derivatives containing 4-hydroxybenzyl units at the N atom and 4-7 are the first examples of natural products with the 4-hydroxybenzyl unit linked via a thioether bond to 2-hydroxy-3-mercaptopropanoic acid(4-6) and 2-hydroxy-4-mercaptobutanoic acid(7), which would be biogenetically derived from cysteine and homocysteine, respectively. The structures of 1 and 2 were verified by synthesis, while the absolute configurations of 4, 5 and 7 were assigned using Mosher's method based on the MPA determination rule of ΔδRS values. 20Energy-Optimal and Delay-Bounded Computation Offloading in Mobile Edge Computing with Heterogeneous Clouds显示文摘By Mobile Edge Computing(MEC), computation-intensive tasks are offloaded from mobile devices to cloud servers, and thus the energy consumption of mobile devices can be notably reduced. In this paper, we study task offloading in multi-user MEC systems with heterogeneous clouds, including edge clouds and remote clouds. Tasks are forwarded from mobile devices to edge clouds via wireless channels, and they can be further forwarded to remote clouds via the Internet. Our objective is to minimize the total energy consumption of multiple mobile devices, subject to bounded-delay requirements of tasks. Based on dynamic programming, we propose an algorithm that minimizes the energy consumption, by jointly allocating bandwidth and computational resources to mobile devices. The algorithm is of pseudo-polynomial complexity. To further reduce the complexity, we propose an approximation algorithm with energy discretization, and its total energy consumption is proved to be within a bounded gap from the optimum.
