1Prevalence of Thyroid Nodules and Its Relationship with Iodine Status in Shanghai:a Population-based Study显示文摘Objective This study was designed to evaluate the prevalence of thyroid nodules(TNs) and its relationship with urine iodine concentrations(UICs) after the regional rapid economic growth and lifestyle changes. Methods A cross-sectional survey was conducted in the general population aged 15-69 years. A questionnaire regarding general and personal characteristics and relevant information was administered. Ultrasonography of the thyroid was performed, and serum triiodothyronine(T3), tetraiodothyronine(T4), serum thyroid stimulating hormone(TSH), free triiodothyronine(FT3), free tetraiodothyronine(FT4), thyroglobulin antibody(Tg Ab), thyroid peroxidase antibody(TPOAb), and TSH receptor antibody(TRAb) levels were measured for each individual subject. Results The prevalence rates of TNs in the whole population, females and males were 27.76%, 34.04%, and 21.60%, respectively. The prevalence of multiple nodules increased with age, whereas the prevalence peaks differed between males and females. The median UICs in the whole population and females with non-TNs were higher than those of subjects with TNs(P=0.0035, P=0.0068). The median UICs in subjects with a single TN were higher than those in subjects with multiple TNs(P=0.0164, P=0.0127). The result showed a U-shaped curve relationship between UIC and prevalence of TNs. The prevalence of TNs was the lowest when the UIC was 140-400 μg/L. Conclusion The prevalence of TNs was nearly 30% and increased with age. The relationship between UIC and prevalence of TNs is U-shaped, with an increase in risk when the UIC was <140 μg/L and >400 μg/L. 2Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 3Dephosphorylation of cGAS by PPP6C impairs its substrate binding activity and innate antiviral response显示文摘The cyclic GMP-AMP (cGAMP) synthase (cGAS) plays a critical role in host defense by sensing cytosollc DNA derived from microbial pathogens or mls-located cellular DNA.Upon DNA binding,cGAS utilizes GTP and ATP as substrates to synthesize cGAMP,leading to MITA-mediated innate immune response.In this study,we identified the phosphatase PPP6C as a negative regulator of cGASmediated innate immune response.PPP6C is constitutively associated with cGAS in un-stimulated cells.DNA virus infection causes rapid disassociation of PPP6C from cGAS,resulting in phosphorylation of human cGAS S435 or mouse cGAS S420 in its catalytic pocket.Mutation of this eerine residue of cGAS impairs its ability to synthesize cGAMP upon DNA virus infection.In vitro experiments indicate that S420-phosphorylated mcGAS has higher affinity to GTP and enzymatic activity.PPP6Cdeficiency promotes innate immune response to DNA virus in various cells.Our findings suggest that PPP6Cmediated dephosphorylation of a catalytic pocket serine residue of cGAS impairs its substrate binding activity and innate immune response,which provides a mechanism for keeping the DNA sensor cGAS inactive in the absence of infection to avoid autoimmune response.Mi Li Hong-Bing Shu 2020Protein & Cell2020,11,8:7
4Bioactive Study on Mixtures of Epimedin C and Icariin Monomers with Invariant Molarity to Zebrafish Osteoporosis Model显示文摘Objective To evaluate the anti-osteoporosis effect of the mixtures of epimedin C and icariin monomers with invariant molarity on zebrafish osteoporosis model. Methods The zebrafishes, fertilized 4 d ago, were exposed in 11 groups of nutrient solutions with prednisolone(25 μmol/L) as well as epimedin C and icariin(15 μmol/L) of various contents. The ratio of epimedin C and icariin in the 11 groups were as follows: A(10:0), B(9:1), C(8:2), D(7:3), E(6:4), F(5:5), G(4:6), H(3:7), I(2:8), J(1:9), and K(0:10). Meanwhile, a negative control group with prednisolone(25 μmol/L) was prepare as S. The selected zebrafish fetus was put into the 24-hole culture plate, and ensure every 5 zebrafishes in 1 hole and 2 holes as a group. They were placed in incubator at 28.5 o C, and the daily changes of fluid were investigated until they were put to death on day 8and then fixed. After dyeing with alizarin red, the segmental venter of zebrafish skulls was observed and quantitative analysis of dyed area was conducted. Results Compared with the negative control group S, the integrated optical density(IOD) values of cranial dyed area in all groups increased significantly(P < 0.05); Compared with group S, the IOD value of cranial dyed area in mixtures of epimedium monomers increased significantly(P < 0.05). The mixtures of epimedium monomers were all effective in facilitating zebrafish cranial mineralization and preventing prednisolone from inducing osteoporosis. According to mixtures of A-K groups, zebrafish cranial mineralization gradually decreased with gradually reduced content of epimedin C, with significant difference among groups(P < 0.05). 5Pharmacokinetics of H002, a novel S1PR_1 modulator, and its metabolites in rat blood using liquid chromatography–tandem mass spectrometry显示文摘A rapid and sensitive liquid chromatography–tandem mass spectrometry(LC–MS/MS) method was developed and validated for the simultaneous determination of H002 and its phosphorylated metabolite, H002-P and hydroxylated metabolite H002-M, in rat blood. H001, an analogue of H002, was used as the internal standard.Blood samples were prepared by simple protein precipitation. The analytes and internal standard were separated on a Zorbax SB-C18 column with a gradient mobile phase consisting of methanol and water containing 0.1% formic acid at a flow rate of 0.2 mL /min with an operating temperature of 20 1C. The detection was performed on a triple quadrupole tandem mass spectrometer with positive electrospray ionization in multiple-reaction monitoring mode.Linear detection responses were obtained from 0.2–100 ng/mL for H002 and H002-M, while 0.5–100 ng/mL for H002-P. The intra- and inter-day precision(RSD%) was within 11.76%, with the accuracy(RE%) ranging from –9.84% to 9.12%. 6Characteristics of Nitric-Oxide Emissions from Traditional Flame and MILD Combustion Operating in a Laboratory-Scale Furnace显示文摘This study investigated the formation and emission characteristics of nitric oxide(NO) from flameless MILD(moderate or intensive low-oxygen dilution) combustion(MILDC) versus traditional visible-flame combustion(TC) in a 30-k W furnace. Both combustion processes were experimentally operated successively in the same furnace, burning natural gas at a fixed rate of 19 k W and the equivalence ratio of 0.86. Numerical simulations of TC and MILDC were carried out to explain their distinction in the measured furnace temperature and exhaust NO emissions. Present measurements of the NO emission(XNO) versus a varying furnace wall temperature(Tw) have revealed, at the first time, that the relationship of XNO ~ Tw was exponential in both TC and MILDC. By analyzing the simulated results, the average temperature over the reaction zone was identified to be the common characteristic temperature for scaling NO emissions of both cases. Moreover, relative to TC, MILDC had a fairly uniform temperature distribution and low peak temperature, thus reducing the NO emission by over 90%. 7Synthesis of monodispersed Fe3O4@C core/shell nanoparticles显示文摘We report a facile method to synthesize dispersed Fe_3O_4@C nanoparticles(NPs). Fe_3O_4 NPs were firstly prepared via the high temperature diol thermal decomposition method. Fe_3O_4@C NPs were fabricated using glucose as a carbon source by hydrothermal process. The obtained products were characterized by X-ray diffraction(XRD), transmission electron microscopy(TEM), vibrating sample magnetometer(VSM) and Raman spectra. The results indicate that the original shapes and magnetic property of Fe_3O_4 NPs can be well preserved. The magnetic particles are well dispersed in the carbon matrix. This strategy would provide an efficient approach for existing applications in Li-ion batteries and drug delivery.
