1The effect of a secretion-enhanced heavy chain on improving intein-based dual-vector co-delivery of a full-length factor Ⅷ gene显示文摘Treatment of hemophilia A by gene therapy is adversely affected by inefficient FVIII secretion and the large FVIII gene,which is difficult to package in the promising adeno-associated virus (AAV) vectors.Inhibited secretion of FVIII is caused mainly by inefficient secretion of its heavy chain.Previously,we have employed a protein splicing-based dual-vector to co-transfer a B-domain-deleted FVIII (BDD-FVIII) gene,suggesting that the light chain,covalently ligated to a co-expressed heavy chain can improve the secretion of spliced BDD-FVIII.However,its level of secretion was affected by inefficient secretion the heavy chain.Here,we studied the effect of a mutant heavy chain with L303E/F309S substitutions,which enhance FVIII secretion on the heavy chain itself and spliced FVIII when using a protein splicing-based split-delivery of a full-length FVIII gene.Eukaryotic vectors expressing Ssp DnaB intein-fused mutant heavy and light chains were transiently co-transfected into cultured COS-7 cells.A spliced FVIII protein was seen in co-transfected cells by Western blot analysis.The heavy chain was secreted by cells transfected with the mutant heavy chain gene alone at (39±11) ng/mL and this secretion increased to (123±13) ng/mL when cells were co-transfected with the light chain gene,which was greater than the secretion of wild-type heavy chain.The amount of spliced FVIII in the culture supernatant of co-transfected cells was (86±14) ng/mL,with an activity of (0.61±0.08) IU/mL,which was greater than that of wild-type FVIII co-transfected cells.Spliced FVIII and bioactivity were also detected in the combined culture supernatant of cells individually transfected with mutant heavy and light chain gene at a higher level than that of combined wild-type heavy and light chain transfections.This suggested that the heavy chain with improved secretion markedly increased the efficacy of protein splicing-based split delivery of the full-length FVIII gene using a dual-vector.These results encourage the transfer of this technology to an animal model using a dual-AAV vector.ZHU FuXiang YANG ShuDe LIU ZeLong MIAO Jing QU HuiGe CHI XiaoYan 2011Chinese Science Bulletin2011,56,2:8
2Prediction of Large Structure Welding Residual Stress by Similitude Principles显示文摘On basis of the similitude principles, the conception of virtual simulative component and the auxiliary value of welding residual stress, which is deduced by the welding conduction theory, the relation of the welding residual stress between the simulative component and the practical component was attained. In order to verify the correctness of the relation, the investigation was done from the view of the welding experiment and the numerical simulation about the simulative component and the practical component. The results show that the distribution of welding residual stress of the simulative component is the same as that of the practical component. The ratio of welding residual stress attained by the experiment or the simulation method between the practical runner and the simulative component was compared with the ratio obtained by the similitude principles. Moreover, the error is less than 10%. 3Effect of Welding Technologies on Decreasing Welding Residual Stress of Francis Turbine Runner显示文摘On basis of the subsection welding and the local heating s reasonable technological parameters obtained by plane experiment,the welding residual stress field of some Francis turbine runner is regulated and controlled.For the experimental plane with a thickness of 16 mm,the effect of subsection welding on decreasing welding residual stress of the prior welding section is obvious when the distance is less than 50 mm away from the end of latter welding section.For the local heating,the best position,where the effect on decreasing the welding residual stress of plane is best,is 60 mm or so away from the edge of heating area.The experimental results of runner show that the subsection welding can make the residual compressive stress engender in the blade dangerous area while the stress value is greatly influenced by the length of the welding section of blade outlet.The local heating can further decrease the value of residual stress near blade outlet,while the local heating processed after heat treatment is better than that processed before heat treatment.Shude Ji Liguo Zhang Xuesong Liu Jianguo Yang 2010Journal of Materials Science & Technology2010,26,10:3

5Effects of nitrite on phosphate uptake in anaerobic-oxic process显示文摘An anaerobic-oxic(A/O)biological phosphorus removal reactor was operated to study the effect of nitrite on phosphate uptake.The phosphorus uptake profile was determined under different operating conditions.The results indicated that in addition to oxygen and nitrate(DPB_(Na),nitrate denitrifying phosphorus removal),to some extent,nitrite could also serve as an electron acceptor to achieve nitrite denitrifying phosphorus removal(DPB_(Ni)).The quantity and rate of phosphorus uptake of DPBNi,however,were evidently lower than that of DPBNa.The experiment results revealed that nitrite would bring toxic action to phosphate-accumulating organisms(PAOs)when NO_(2)^(−)-Ni93.7 mg/L.The nitrite existing in the anoxic reactor made no difference to the quantity and rate of denitrifying phosphorus removal,but it could reduce the consumption of nitrate.Moreover,the data showed that the aerobic phosphate uptake of DPBNi was lower than that of anaerobic phosphorus-released sludge in a traditional A/O process.However,there was not much difference between these two kinds of sludge in terms of the total phosphorus uptake quantity and the effluent quality.LI Jie XIONG Biyong ZHANG Shude YANG Hong ZHANG Jie 2007Frontiers of Environmental Science & Engineering2007,1,1:1
6Immunomodulatory effects and mechanisms of distraction osteogenesis显示文摘Distraction osteogenesis(DO) is widely used for bone tissue engineering technology. Immune regulations play important roles in the process of DO like other bone regeneration mechanisms. Compared with others, the immune regulation processes of DO have their distinct features. In this review, we summarized the immune-related events including changes in and effects of immune cells, immune-related cytokines, and signaling pathways at different periods in the process of DO. We aim to elucidated our understanding and unknowns about the immunomodulatory role of DO. 7A Novel Friction Stir Spot Riveting of Al/Cu Dissimilar Materials显示文摘In order to eliminate the disadvantages of the keyhole in conventional friction stir spot welding joint and attain the highstrength lap joint of Al/Cu dissimilar metals,a novel welding technique,named as friction stir spot riveting(FSSR),was proposed.A pinless tool and an extra filling stud were employed.The Al/Cu spot joints without keyhole defect were achieved by the FSSR.A Cu anchor-like structure was formed,which greatly increased the mechanical interlocking between the upper Al sheet and lower Cu sheet.The thin intermetallic compounds containing CuAl2 and CuAl at the Al/Cu interface strengthened the joining interface between the Al sheet and the Cu stud.Increasing rotating velocity increased frictional heat and plastic deformation and then eliminated the interfacial joining defects.The FSSR joint with the maximum tensile shear load of 3.50 kN was achieved at a rotating velocity of 1800 rpm and a dwell time of 20 s,whose fracture path passed through the softened region of upper Al sheet.In summary,the novel FSSR technique has the advantages of strong mechanical interlocking and metallurgical bonding between dissimilar materials,thereby attaining the high-strength spot joint.Jinglin Liu Qi Song Lihui Song Shude Ji Mingshen Li Zhen Jia Kang Yang 2021Acta Metallurgica Sinica(English Letters)2021,34,1:0
8Magnesium alloys for orthopedic applications:A review on the mechanisms driving bone healing显示文摘Magnesium(Mg) alloys have attracted a wealth of attention in orthopedic fields for their superior mechanical properties, degradability,and excellent biocompatibility. Consistently, to resolve the issues on rapid degradation, more studies are dedicated to the researches on the composition design, preparation and processing, surface modification, the degradation modes of Mg alloys. Nevertheless, the mechanisms by which Mg alloys promote bone healing remain elusive. This review gives an account of specific mechanisms on Mg alloys promoting bone healing from four aspects, immunomodulatory, angiogenesis, osteogenesis and regulation of osteoclast function. We highlight the regulation of Mg alloys on the functional status and interactions of numerous cells that are involved in bone healing, including immune cells, osteogenicrelated cells, osteoclasts, endothelial cells(ECs), nerve cells, etc., and summarize the signaling pathways involved, with the aim to provide the basis and support on future investigation on mechanisms on Mg alloys driving bone regeneration.
