| 1 | Curcumin alleviates TGF-β1-induced fibrosis in NRK-49F cells via suppression of miR-21 expression,and regulation of the TGF-β1/smad3 signaling pathway显示文摘Objective:Overexpression of microRNA-21(miR-21)has been well-recognized during fibrosis in a wide range of tissue types through regulation of fibrogenesis via diverse pathways.Curcumin exhibits antifibrotic effects in various organs.We and others have previously demonstrated that curcumin has positive effects on the protection against fibrosis.However,the effect of curcumin on miR-21 expression has not been reported.Methods:In this study,transforming growth factor-b1(TGF-b1)-stimulated rat renal fibroblast cells(NRK-49F)were transfected with miR-21 inhibitors,then the expression of Smad3,a-smooth muscle actin(a-SMA),type I collagen(COL1A1),and type III collagen(COL3A1)was determined using quantitative real-time-PCR(qRT-PCR)and western blot analysis.The effect of curcumin on production of the extracellular matrix was evaluated using immunofluorescence,qRT-PCR,and western blot analysis.Results:TGF-b1 stimulation upregulated the expression of miR-21 and induced fibrogenesis in NRK-49F cells.Transfection with miR-21 inhibitors selectively decreased smad3 activity,and noticeably reduced the expression of a-SMA,COL1A1,and COL3A1.Curcumin treatment significantly inhibited the expressions of smad3,a-SMA,COL1A1,COL3A1,as well as miR-21 expression in NRK-49F cells in a dosedependent manner.Conclusion:Curcumin exerted its anti-fibrotic effects by targeting the TGF-b1/smad3 signaling pathway and suppressing miR-21 expression,thereby provides novel insight in the protective effects of curcumin against fibrosis in various organs. | Lan Zhang Yi Liu Xiting Wang Cheng Li Mengmeng Shen Liming Di Shuangshuang He Yu Li | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 2 |
| 2 | Danlou Fang(丹蒌方)reduces microvascular obstruction through the endothelial/inducible nitric oxide synthase pathway in a rat model显示文摘OBJECTIVE:To investigate the efficacy of the Danlou Fang(DL)Traditional Chinese Medicine formula on microvascular obstruction(no-reflow)through the endothelial/inducible nitric oxide synthase(eNOS/iNOS)pathway in a rat model.METHODS:Sprague-Dawley rats were subjected to 60 min of coronary artery occlusion(or sham procedure)followed by 2 h of reperfusion and were then divided into treatment groups:sham,model,DL(500 mg/kg),DL(500 mg/kg)+eNOS inhibitor L-nitroarginine(L-NNA;7.5 mg/kg),and sodium nitroprusside(SNP;0.5 mg/kg).There were 16 per group.Areas of no-reflow were determined by thioflavin S staining of heart tissue.Cardiac function was assessed by echocardiography.Myocardial enzymes and antioxidants in serum were measured and analyzed.The relative protein expression levels of eNOS and iNOS were determined by western blotting.RESULTS:DL had a myocardial protective effect on myocardial reperfusion and reduced the area of no-reflow.The serum levels of creatine kinase(CK),myocardial CK isoenzyme CK-MB,and lactate dehydrogenase were significantly lower in the DL group than in the model(P<0.05).DL treatment also decreased the serum content of malondialdehyde and reactive oxygen species(ROS),increased the activity of superoxide dismutase and nitric oxide,and promoted eNOS expression(P<0.05)while lowering iNOS expression.CONCLUSION:DL reduced the area of no-reflow and had a myocardial protective effect that may be associated with the eNOS/iNOS pathway. | DAI Xiangdong CHEN Rui CHEN Ting YAN Haifeng WANG Yanyan ZHOU Kun ZHANG Xiting WANG Yi | 2021 | Journal of Traditional Chinese Medicine2021,41,2: | 1 |

| 6 | Activity Gel Analysis of Endopeptidases in Rose Petals显示文摘Poor resolution for the identification of endopeptidase(EP) activity in activity gel assays is a critical issue in the analysis of the postharvest physiology of rose petals. In this paper, major factors influencing EP activity gel assays were evaluated. The results showed that a phosphate(NaH_2PO_4/Na_2HPO_4) buffer favors the detection of clear EP bands, as compared to Tris–HCl buffer. Removal of salts and pigments with Sephadex G-25 columns was vital to the measurement of EP activity in rose petal extracts. For optimal resolution of bands, we show that before electrophoresis,samples should be treated for 10 min at 40 °C. Additionally, electrophoresis should be done in 12% SDS–PAGE co-polymerized with 0.15%(w/v) gelatin. After electrophoresis, the optimal incubation temperature and pH are 42 °C and 7.0, respectively. Using our optimized assay, Rh-EP1,Rh-EP2, Rh-EP3, three proteases with molecular masses of 200, 123.5, and 97.4 kD, respectively, were detected in rose petals. Experiments using EP class-specific inhibitors revealed that Rh-EP2 and Rh-EP3 were both serine proteases, while Rh-EP1 was a metalloprotease. In this study, we also measured changes in EP activity during flower opening, senescence, and water deficit stress(WDS) using our optimized activity gel assay,and found that Rh-EP3 may be more relevant to senescence in roses compared with Rh-EP1 or Rh-EP2. Changes occurring to EPs after WDS were similar to those during the period from flower opening to senescence, and Rh-EP3 activities were greatly increased by WDS treatment. | 8 | Organic Carbon Deposition on the Inner Shelf of the East China Sea Constrained by Sea Level and Climatic Changes Since the Last Deglaciation显示文摘The East China Sea(ECS),which is located in the transitional zone between land and ocean,is the main site for the burial of sedimentary organic carbon.Despite good constraints of the modern source to the sinking process of organic carbon,its fate in response to changes in climate and sea level since the last deglaciation remains poorly understood.We aim to fill this gap by presenting a high-resolution sedimentary record of core EC2005 to derive a better understanding of the evolution of the depositional environment and its control on the organic deposition since 17.3 kyr.Our results suggest that sedimentary organic carbon was deposited in a terrestrial environment before the seawater reached the study area around 13.1 kyr.This significant transition from a terrestrial environment to a marine environment is reflected by the decrease in TOC/TN and TOC/TS ratios,which is attributed to deglacial sea level rise.The sea level continued to rise until it reached its highstand at approximately 7.3 kyr when the mud depocenter was developed.Our results further indicate that the deposition of the sedimentary organic carbon could respond quickly to abrupt cold events,including the Heinrich stadial 1 and the Younger Dryas during the last deglaciation,as well as‘Bond events'during the Holocene.We propose that the rapid response of the organic deposition to those cold events in the northern hemisphere is linked to the East Asian winter monsoon.These new findings demonstrate that organic carbon deposition and burial on the inner shelf could effectively document sea level and climatic changes. | ZHANG Mingyu LIU Xiting XU Fangjian LI Anchun GU Yu CHANG Xin ZHUANG Guangchao ZHANG Kaidi BI Naishuang WANG Houjie | 2023 | Journal of Ocean University of China2023,22,5: | 0 |
