1Exosomes: roles and therapeutic potential in osteoarthritis显示文摘Exosomes participate in many physiological and pathological processes by regulating cell–cell communication, which are involved in numerous diseases, including osteoarthritis(OA). Exosomes are detectable in the human articular cavity and were observed to change with OA progression. Several joint cells, including chondrocytes, synovial fibroblasts, osteoblasts, and tenocytes, can produce and secrete exosomes that influence the biological effects of targeted cells. 2Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. 4Genome sequencing revealed the red-flower trait candidate gene of a peach landrace显示文摘Peach(Prunus persica)is an economically important fruit crop globally and an excellent material for genomic studies.While considerable progress has been made in unveiling trait-associated genes within cultivars and wild relatives,certain novel genes controlling valuable traits in peach landraces,such as the red-flowering gene,remained unclear.In this study,we sequenced and assembled the diploid genome of the red-flower landrace‘Yingzui’(abbreviated as‘RedY’).Multi-omics profiling of red petals of‘RedY’revealed the intensified red coloration associated with anthocyanins accumulation and concurrent decline in f lavonols.This phenomenon is likely attributed to a natural variant of Flavonol Synthase(FLS)harboring a 9-bp exonic insertion.Intriguingly,the homozygous allelic configurations of this FLS variant were only observed in red-flowered peaches.Furthermore,the 9-bp sequence variation tightly associated with pink/red petal color in genome-wide association studies(GWAS)of collected peach germplasm resources.Functional analyses of the FLS variant,purified from procaryotic expression system,demonstrated its diminished enzymatic activity in f lavonols biosynthesis,impeccably aligning with the cardinal trait of red flowers.Therefore,the natural FLS variant was proposed as the best candidate gene for red-f lowering trait in peach.The pioneering unveiling of the red-flowered peach genome,coupled with the identification of the candidate gene,expanded the knowledge boundaries of the genetic basis of peach traits and provided valuable insights for future peach breeding efforts.Ping Zhou Siru Lei Xiaodan Zhang Yinghao Wang Rui Guo Shaobin Yan Guang Jin Xingtan Zhang 2023Horticulture Research2023,10,11:0
5Impact of a retrogressive thaw slump on surrounding vegetation communities in the Fenghuoshan mountains,Qinghai-Tibet Plateau显示文摘Under global warming,permafrost around the world is experiencing degradation which is especially so on the Third Pole,the Qinghai-Tibet Plateau(QTP),China.Retrogressive thaw slump(RTS)is one of the thermokarst features caused by rapid degradation of ice rich permafrost,which transforms landforms and threatens infrastructures,and even affects the terrestrial carbon cycle.In this work,vegetation communities surrounding a RTS in the Fenghuoshan Mountains of the interior portion of the Qinghai-Tibet Plateau have been investigated to examine the impact from RTS.This investigation indicates that the occurrence of RTS influences the vegetation community by altering their habitats,especially the soil water content,which forces the vegetation community to evolve in order to adapt to the alterations.In the interior part of RTS where it has been disturbed tremendously,alterations have produced a wider niche and richer plant species.This favors species of a wet environment in a habitat where it was a relatively dry environment of alpine steppe prior to the occurrence of RTS.This study adds to limited observations regarding the impact of RTS to vegetation community on the QTP and helps us to reach a broader understanding of the effects of permafrost degradation as well as global warming.Gang Wei LaJia Weisai ZiJie Zhou XinNing Wu SiRu Gao ZiTeng Fu QingBai Wu GuanLi Jiang 2023Research in Cold and Arid Regions2023,15,1:0
6Human thermal comfort under lateral radiant asymmetries显示文摘Occupants’thermal comfort in buildings may be affected by the cool wall and warm wall,which is attributed to the effect of asymmetric radiation.However,the previous majority of the researches on asymmetric radiation were mainly about the comfort limits under thermally neutral condition within 1∼1.5 h but had not considered the effect of exposure duration and the condition beyond neutral.To investigate the human thermal comfort under an asymmetric environment caused by the cool wall and warm wall,forty-four subjects were exposed to neutral air temperature with lateral radiant asymmetries in winter and summer for 3 h.The results indicated that the cool wall caused thermal discomfort easier than the warm wall because the thermal sensation decreased and deviated from neutral with time.Subjects’sensitivity of local parts to asymmetric radiation was affected in the conditions beyond neutral,thus their acceptability to asymmetric radiation decreased.The currently used limits of radiant temperature asymmetry tended to underestimate the local discomfort due to the walls.For the conditions tested,The limits of 5%dissatisfaction in radiant temperature asymmetry were 4.4°C(180 min)and 1.8°C(60 min and 120 min)for the warm wall,and 1.8°C at 60 min for the cool wall.Siru Gao Liu Yang Mingyang Shi Shengkai Zhao Xiang Zhou Yongchao Zhai 2023Energy and Built Environment2023,4,4:0
7Genome-Wide Identification and Expression Analysis of the Phytocyanin Gene Family in Nicotiana tabacum显示文摘Phytocyanin(PC)is a class of plant-specific blue copper proteins involved in electron transport,plant growth,development,and stress resistance.However,PC proteins have not been systematically evaluated in tobacco plants.We determined the whole-genome sequences of the PC family in the tobacco cultivar‘K326.’The transcriptome data were used to analyze the expression of the NtPC family at different development stages and tissue-specific genes.Real-time fluorescence quantitative analysis was used to analyze the expression of the NtPC gene family under low temperature and methyl jasmonate stress.The tobacco NtPC family contained 110 members and was divided into four subfamilies:early nodulin-like protein(NtENODL),uclacyanin-like protein,stellacyanin1-like protein,and plantacyanin-like protein.According to phylogenetic and structural analyses,the NtPC family could be divided into eight structural types.Fifty-three NtPCs were randomly distributed on 22 of 24 tobacco chromosomes.Collinearity analysis revealed 33 pairs of genes belonging to the NtPC family.Gene ontology analysis showed that the PC genes are components of the plasma membrane and may participate in plasma membrane-related functions.The NtPC family contained numerous elements related to hormonal and abiotic stress responses and was specifically expressed in the tobacco prosperous,maturation,and budding periods.Tissue-specific expression analysis showed that some genes were tissue specific.The expression of NtENODL58 and other genes was significantly induced by low-temperature and methyl jasmonate stress.Thus,the NtPC gene family plays an important role in plant stress response.Peiling Wang Xiaohong Xu Yong Li Hecui Zhang Xuejie Zhang Siru Zhou Yimei Liu Yunyan Feng Tonghong Zuo Liquan Zhu 2023Phyton-International Journal of Experimental Botany2023,92,5:0
