| 1 | Human natural killer cells for targeting delivery of gold nanostars and bimodal imaging directed photothermal/photodynamic therapy and immunotherapy显示文摘Objective:To construct a novel nanoplatform GNS@CaCO3/Ce6-NK by loading the CaCO3-coated gold nanostars(GNSs)with Chlorin e6 molecules(Ce6)into human peripheral blood mononuclear cells(PBMCs)-derived NK cells for tumor targeted therapy.Methods:GNS@CaCO3/Ce6 nanoparticles were prepared and characterized by TEM and UV-vis.The cell surface markers and cytokines secretion of NK cells before and after loading the GNS@CaCO3/Ce6 nanoparticles were detected by Flow Cytometry(FCM)and ELISA.Effects of the GNS@CaCO3/Ce6-NK cells on A549 cancer cells was determined by FCM and CCK-8.Intracellular fluorescent signals of GNS@CaCO3/Ce6-NK cells were detected via Confocal laser scanning microscopic(CLSM)and FCM at different time points.Intracellular ROS generation of GNS@CaCO3/Ce6-NK cells under laser irradiation were examined by FCM.The distribution of GNS@CaCO3/Ce6-NK in A549 tumor-bearing mice were observed by fluorescence imaging and PA imaging.The combination therapy of GNS@CaCO3/Ce6-NK under laser irradiation were investigated on tumor-bearing mice.Results:The coated CaC03 shell on the surface of GNSs exhibited prominent delivery and protection effect of Ce6 during the cellular uptake process.The as-prepared multifunctional GNS@CaCO3/Ce6-NK cells possessed bimodal functions of fluorescence imaging and photoacoustic imaging.The as-prepared multifunctional GNS@CaCO3/Ce6-NK cells could actively target tumor tissues with the enhanced photothermal/photodynamic therapy and immunotherapy.Conclusions:The GNS@CaCO3/Ce6-NK shows effective tumor-targeting ability and prominent therapeutic efficacy toward lung cancer A549 tumor-bearing mice.Through fully utilizing the features of GNSs and NK cells,this new nanoplatform provides a new synergistic strategy for enhanced photothermal/photodynamic therapy and immunotherapy in the field of anticancer development in the near future. | Bin Liu Wen Cao Jin Cheng Sisi Fan Shaojun Pan Lirui Wang Jiaqi Niu Yunxiang Pan Yanlei Liu Xiyang Sun Lijun Ma Jie Song Jian Ni Daxiang Cui | 2019 | Cancer Biology & Medicine2019,16,4: | 7 |

| 3 | Nitrogen-doped hierarchically porous carbon spheres for low concentration CO_(2) capture显示文摘Synthesis of spherical carbon beads with effective CO_2 capture capability is highly desirable for large scale application of CO2 sorption, but remains challenging. Herein, a facile and efficient strategy to prepare nitrogen-doped hierarchically porous carbon spheres was developed via co-pyrolyzation of poly(vinylidene chloride) and melamine in alginate gel beads. In this approach, melamine not only serves as the nitrogen precursor, but also acts as a template for the macropores structures. The nitrogen contents in the hierarchically porous carbon spheres reach a high level, ranging from 11.8 wt% to 14.7 wt%, as the melamine amount increases. | 4 | Machine learning helps improve diagnostic ability of subclinical keratoconus using Scheimpflug and OCT imaging modalities显示文摘Purpose:To develop an automated classification system using a machine learning classifier to distinguish clinically unaffected eyes in patients with keratoconus from a normal control population based on a combination of Scheimpflug camera images and ultra-high-resolution optical coherence tomography(UHR-OCT)imaging data.Methods:A total of 121 eyes from 121 participants were classified by 2 cornea experts into 3 groups:normal(50 eyes),with keratoconus(38 eyes)or with subclinical keratoconus(33 eyes).All eyes were imaged with a Scheimpflug camera and UHR-OCT.Corneal morphological features were extracted from the imaging data.A neural network was used to train a model based on these features to distinguish the eyes with subclinical keratoconus from normal eyes.Fisher’s score was used to rank the differentiable power of each feature.The receiver operating characteristic(ROC)curves were calculated to obtain the area under the ROC curves(AUCs).Results:The developed classification model used to combine all features from the Scheimpflug camera and UHR-OCT dramatically improved the differentiable power to discriminate between normal eyes and eyes with subclinical keratoconus(AUC=0.93).The variation in the thickness profile within each individual in the corneal epithelium extracted from UHR-OCT imaging ranked the highest in differentiating eyes with subclinical keratoconus from normal eyes.Conclusion:The automated classification system using machine learning based on the combination of Scheimpflug camera data and UHR-OCT imaging data showed excellent performance in discriminating eyes with subclinical keratoconus from normal eyes.The epithelial features extracted from the OCT images were the most valuable in the discrimination process.This classification system has the potential to improve the differentiable power of subclinical keratoconus and the efficiency of keratoconus screening. | Ce Shi Mengyi Wang Tiantian Zhu Ying Zhang Yufeng Ye Jun Jiang Sisi Chen Fan Lu Meixiao Shen | 2020 | Eye and Vision2020,7,1: | 3 |

| 6 | Investigation and experimental validation of curcumin-related mechanisms against hepatocellular carcinoma based on network pharmacology显示文摘Objective:To determine the potential molecular mechanisms underlying the therapeutic effect of curcumin on hepatocellular carcinoma(HCC)by network pharmacology and experimental in vitro validation.Methods:The predictive targets of curcumin or HCC were collected from several databases.the identified overlapping targets were crossed with Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses using the Database for Annotation,Visualization,and Integrated Discovery(DAVID)platform.Two of the candidate pathways were selected to conduct an experimental verification.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide tetrazolium(MTT)assay was used to determine the effect of curcumin on the viability of Hep G2 and LO2 cells.The apoptosis and autophagy of Hep G2 cells were respectively detected by flow cytometry and transmission electron microscopy.Besides,western blot and real-time polymerase chain reaction(PCR)were employed to verify the p53 apoptotic pathway and adenosine 5’-monophosphate(AMP)-activated protein kinase(AMPK)autophagy pathway.Hep G2 cells were pretreated with pifithrin-α(PFT-α)and GSK690693 for further investigation.Results:The 167 pathways analyzed by KEGG included apoptosis,autophagy,p53,and AMPK pathways.The GO enrichment analysis demonstrated that curcumin was involved in cellular response to drug,regulation of apoptotic pathway,and so on.The in vitro experiments also confirmed that curcumin can inhibit the growth of Hep G2 cells by promoting the apoptosis of p53 pathway and autophagy through the AMPK pathway.Furthermore,the protein and messenger RNA(m RNA)of the two pathways were downregulated in the inhibitor-pretreated group compared with the experimental group.The damage-regulated autophagy modulator(DRAM)in the PFT-α-pretreated group was downregulated,and p62 in the GSK690693-pretreated group was upregulated.Conclusions:Curcumin can treat HCC through the p53 apoptotic pathway and the AMPK/Unc-51-like kinase 1(ULK1)autophagy pathway,in which the mutual transformation of autophagy and apoptosis may occur through DRAM and p62. | Yang CHEN Qian LI Sisi REN Ting CHEN Bingtao ZHAI Jiangxue CHENG Xiaoyan SHI Liang SONG Yu FAN Dongyan GUO | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,8: | 3 |

| 11 | Regulation of Water Masses to Hypoxia Zones in the Changjiang Estuary显示文摘The regulating ways of different water masses affecting the locations and intensities of hypoxia zones were studied based on the time-space continuum data from August 2011 to 2013–2017.The 6-year distribution of the hypoxic area in the Changjiang Estuary(CE)and its adjacent waters show that the hypoxic area can be divided into two segments.The southern segment is out of the south branch of the CE,whereas the northern segment is in the junction zone between the South Yellow Sea and the CE.The two segments were divided along the 31.5°–32°N latitude line.The northern and southern segments were dominated by the East China Sea shelf water(ECSSW)and Kuroshio subsurface water(KSW),respectively.When the KSW(salinity>34)intrusion reached the east of 123°E and south of 31°N,hypoxia zones mainly occurred in the southern segment covered by the Changjiang Diluted Water(CDW),meanwhile the Yellow Sea cold water mass may emerge in the northeastern area.When the KSW intensely invaded westward to the region between 122°and 122.5°E and northward to 31.5°N or further north,hypoxia zones appeared in the northern segment.The strength of the KSW with low dissolved oxygen concentration is the basic driving factor for the hypoxia occurrence in the CE.Moreover,the stratification is crucial for the southern segment,whereas the organic matter decomposition is dominated for the northern segment,even with severe hypoxia across the sea surface in the study area. | YANG Xingxing FAN Haimei ZHANG Hui CHEN Sisi YANG Ying JI Huanhong JIANG Xiaoshan DENG Bangping JIA Junhe | 2023 | Journal of Ocean University of China2023,22,4: | 0 |
| 12 | Performance of Hepatitis Delta Virus(HDV)RNA Testing for the Diagnosis of Active HDV Infection:Systematic Review and Meta-analysis显示文摘Background and Aims:Hepatitis delta virus(HDV)is a defective virus and causes severe liver disease.Several HDV RNA assays have been developed,however the diagnostic efficacy remains unclear.This systematic review and metaanalysis aims to evaluate the diagnostic accuracy of HDV RNA assays to aid in the diagnosis of active hepatitis D.Methods:The PubMed,Embase,and Cochrane Library databases were systematically searched from the beginning to June 31,2022.Information on the characteristics of the literature and data on sensitivity,specificity,and area under curve(AUC)of the receiver operating characteristic(ROC)were extracted.Stata 14.0 was used for meta-analysis of the combined sensitivity,specificity,positive likelihood ratio,and negative likelihood ratio.Results:A total of 10 studies were included in the meta-analysis.The summary sensitivity,specificity,positive likelihood ratio,negative likelihood ratio,and diagnostic odds ratio of HDV RNA assays for HDV diagnosis were 0.92(95%CI:0.87-0.95),0.90(95%CI:0.86-0.93),7.74(95%CI:5.31-11.29),0.10(95%CI:0.06-0.18)and 99.90(95%CI:47.08-211.99),respectively.The AUC of the pooled ROC curve was 0.95(95%CI:0.92-0.96).Conclusions:The results show that HDV RNA assays had high diagnostic performance.However,that is limited by the number and quality of studies.Standard protocols for the development of assays by manufacturers and larger studies on the use of the assays are needed. | Sisi Chen Xiangying Zhang Ling Xu Yuan Tian Zihao Fan Yaling Cao Zhenzhen Pan Yao Gao Sujun Zheng Zhongping Duan Mei Liu Feng Ren | 2023 | Journal of Clinical and Translational Hepatology2023,11,6: | 0 |
