| 2 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 3 | Antimony smelting process generating solid wastes and dust:Characterization and leaching behaviors显示文摘A large amount of solid waste has been produced by the antimony smelting process in the'World Capital of Antimony', Xikuangshan area in China. This study comprehensively investigated the physical and chemical characteristics of the various solid wastes, as well as the leaching behavior of the solid wastes, which included water-quenched slag,arsenic-alkali residue, desulfurized slag and blast furnace dust. These four types of waste were enriched in a variety of heavy metals and metalloids and more specifically with As and Sb levels up to 8.6 × 104 and 3.16 × 105mg/kg, respectively, in arsenic-alkali residue. For desulfurized slag and water-quenched slag, the leaching concentration of Sb significantly exceeded the acceptable limits during the leaching tests using the toxicity characteristic leaching procedure and the synthetic precipitation leaching procedure. In addition, As leaching in arsenic-alkali residue was extraordinarily hazardous, being three orders of magnitude higher than the regulatory level of As. | 4 | GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://bigd.big.ac.cn/gsa or http://gsa.big.ac.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 5 | Hydrogen Sulfide Positively Regulates Abscisic Acid Signaling through Persulf idation of SnRK2.6 in Guard Cells显示文摘The phytohormone abscisic acid(ABA)plays pivotal roles in triggering stomatal closure and facilitating adaptation of plants to drought stress.Hydrogen sulfide(H2S),a small signaling gas molecule,is involved in ABA-dependent stomatal closure.However,how H2S regulates ABA signaling remains largely unclear.Here,we show that ABA induces the production of H2S catalyzed by L-CYSTEINE DESULFHYDRASE1(DES1)in guard cells,and H2S in turn positively regulates ABA signaling through persulfidation of Open Stomata 1(OST1)/SNF1-RELATED PROTEIN KINASE2.6(SnRK2.6).Two cysteine(Cys)sites,Cys131 and Cys137,which are exposed on the surface of SnRK2.6 and close to the activation loop,were identified to be persulfidated,which promotes the activity of SnRK2.6 and its interaction with ABA response element-binding factor 2(ABF2),a transcription factor acting downstream of ABA signaling.When Cys131,Cys137,or both residues in SnRK2.6 were substituted with serine(S),H2S_induced SnRK2_6 activity and SnRK2.6-ABF2 interaction were partially(SnRK2.6c131s and SnRK2.6c137S)or completely(SnRK2.6c131sc137S)compromised.Introduction of SnRK2.6c131s,SnRK2.6c137S,or SnRK2.6c131sc137S into the ost1-3 mutant could not rescue the mutant phenotype:less sensitivity to ABA-and H2S-induced stomatal closure and Ca2+influx as well as increased water loss and decreased drought tolerance.Taken together,our study reveals a novel post-translational regulatory mechanism of ABA signaling whereby H2S persulfidates SnRK2.6 to promote ABA signaling and ABA-induced stomatal closure. | Sisi Chen Honglei Jia Xiaofeng Wang Cong Shi Xiao Wang Peiyun Ma Juan Wang Meijuan Ren Jisheng Li | 2020 | Molecular Plant2020,13,5: | 13 |

| 7 | Efficient microwave-assisted photocatalytic degradation of endocrine disruptor dimethyl phthalate over composite catalyst ZrO_x /ZnO显示文摘A highly active ZrO x /ZnO catalyst for microwave-assisted photocatalytic (MW/PC) degradation of endocrine disruptor dimethyl phthalate (DMP) has been prepared via cetyltrimethylammonium assisted hydrothermal method. The ZrO x /ZnO was characterized by XPS, XRD, UV-Vis, BET and SEM techniques. The XPS result showed that Zr oxides with different valences (+2, +3, +4) co-existed in ZrO x /ZnO. By using the ZrO x /ZnO (0.1 g), the TOC removal efficiency of DMP (100 mL of 50 mg/L) was 88% after 30 min reaction, which was about 15% higher than P25 TiO 2 . It was found that the removal process of DMP by MW/PC followed pseudo first-order kinetics in all cases, and ZrO x /ZnO significantly accelerated the degradation of DMP. The degradation half-life time of DMP was shortened 45% compared with P25 TiO 2 . A possible catalytic mechanism was proposed based on microwave response and interfacial charge transfer. ZrO x /ZnO could be reused for six times without obvious decrease in catalytic activity. | 8 | Betaine affects muscle lipid metabolism via regulating the fatty acid uptake and oxidation in finishing pig显示文摘Background: Betaine affects fat metabolism in animals, but the specific mechanism is still not clear. The purpose of this study was to investigate possible mechanisms of betaine in altering lipid metabolism in muscle tissue in finishing pigs.Methods: A total of 120 crossbred gilts(Landrace × Yorkshire × Duroc) with an average initial body weight of 70.1 kg were randomly allotted to three dietary treatments. The treatments included a corn–soybean meal basal diet supplemented with 0, 1250 or 2500 mg/kg betaine. The feeding experiment lasted 42 d.Results: Betaine addition to the diet significantly increased the concentration of free fatty acids(FFA) in muscle(P < 0.05). Furthermore, the levels of serum cholesterol and high-density lipoprotein cholesterol were decreased(P < 0.05) and total cholesterol content was increased in muscle(P < 0.05) of betaine fed pigs. Experiments on genes involved in fatty acid transport showed that betaine increased expression of lipoprotein lipase(LPL), fatty acid translocase/cluster of differentiation(FAT/CD36), fatty acid binding protein(FABP3) and fatty acid transport protein(FATP1)(P < 0.05). The abundance of fatty acid transport protein and fatty acid binding protein were also increased by betaine(P < 0.05). As for the key factors involved in fatty acid oxidation, although betaine supplementation didn't affect the level of carnitine and malonyl-CoA, betaine increased mR NA and protein abundance of carnitine palmitransferase-1(CPT1)and phosphorylated-AMPK(P < 0.05).Conclusions: The results suggested that betaine may promoted muscle fatty acid uptake via up-regulating the genes related to fatty acid transporter including FAT/CD36, FATP1 and FABP3. On the other hand, betaine activated AMPK and up-regulated genes related to fatty acid oxidation including PPARα and CPT1. | 11 | Hypoxia-stressed cardiomyocytes promote early cardiac differentiation of cardiac stem cells through HIF-1α/Jagged1/Notch1 signaling显示文摘Hypoxia is beneficial for the differentiation of stem cells transplanted for myocardial injury,but mechanisms underlying this benefit remain unsolved. Here, we report the impact of hypoxia-induced Jagged1 expression in cardiomyocytes(CMs) for driving the differentiation of cardiac stem cells(CSCs).Forced hypoxia-inducible factor 1α(HIF-1α) expression and physical hypoxia(5% O_2) treatment could induce Jagged1 expression in neonatal rat CMs. Pharmacological inhibition of HIF-1α by YC-1 attenuated hypoxia-promoted Jagged1 expression in CMs. An ERK inhibitor(PD98059), but not inhibitors of JNK(SP600125), Notch(DAPT), NF-κB(PTDC), JAK(AG490), or STAT3(Stattic) suppressed hypoxiainduced Jagged1 protein expression in CMs. c-Kit^+ CSCs isolated from neonatal rat hearts using a magnetic-activated cell sorting method expressed GATA4, SM22α or vWF, but not Nkx2.5 and cTnI.Moreover, 87.3% of freshly isolated CSCs displayed Notch1 receptor expression. Direct co-culture of CMs with BrdU-labeled CSCs enhanced CSCs differentiation, as evidenced by an increased number of BrdU^+/Nkx2.5^+ cells, while intermittent hypoxia for 21 days promoted co-culture-triggered differentiation of CSCs into CM-like cells. | 12 | Urban Expansion in China Based on Remote Sensing Technology:A Review显示文摘Urban areas and its evolution are important anthropogenic indicators and human ecological footprints, and play decisive roles in environmental change analysis, global geo-conditional monitoring, and sustainable development. China has the highest rate of urban expansion and has emerged as an urban expansion hotspot worldwide. In this paper, the progress of studies on Chinese urban expansion based on remote sensing technology are summarized and analyzed from the aspects of urban area definition, remotely sensed imagery applied in urban expansion, monitoring methods of urban expansion, and urban expansion applications. Existing issues and future directions of Chinese urban expansion are discussed and proposed. | 16 | Synthesis and evaluation of methionine and folate co-decorated chitosan self-assembly polymeric micelles as a potential hydrophobic drug-delivery system显示文摘In this study, an amphiphilic copolymer folate-succinyl-methionine-chitosan-octyl (FSMCO) was successfully synthesized step by step for self-assembling polymeric micelles. The copolymers formed micelle-like nanoparticles by their amphiphilic characteristics and structures were examined by UV-Vis absorption and Fourier transform spectroscopy. The sizes of blank and ICG derivativeloaded micelles measured by dynamic light scattering were about 170 and 140 nm, respectively, which were spherical in shape with an average zeta potential of 10 mV. Further studies on the stability showed that the micellar solutions maintain their sizes at room temperature for 1 month without distinct aggregation or dissociation. ICG derivative was much better photostable after being entrapped by the new carrier. The prepared FSMCO micelles displayed a good drug loading content (11.7%), entrapment efficiency (66.5%) and sustained release rate for the model drug fluorescein. The copolymers demonstrated weeny cytotoxicity toward Bel-7402, L02 and A549 cells when incubated for 2 d. Ligands modified micelles endowed preferable cell targeting capability and beautiful cell inhibition of HCPT-FSMCO on Bel-7402 tumor cells. | 18 | Nitrogen-doped hierarchically porous carbon spheres for low concentration CO_(2) capture显示文摘Synthesis of spherical carbon beads with effective CO_2 capture capability is highly desirable for large scale application of CO2 sorption, but remains challenging. Herein, a facile and efficient strategy to prepare nitrogen-doped hierarchically porous carbon spheres was developed via co-pyrolyzation of poly(vinylidene chloride) and melamine in alginate gel beads. In this approach, melamine not only serves as the nitrogen precursor, but also acts as a template for the macropores structures. The nitrogen contents in the hierarchically porous carbon spheres reach a high level, ranging from 11.8 wt% to 14.7 wt%, as the melamine amount increases. | 19 | Conductive photo-thermal responsive bifunctional hydrogel system with self-actuating and self-monitoring abilities显示文摘Despite enormous efforts in actuators,most researches are only limited to various actuation behaviors and demonstrations of soft materials.It has not yet been reported to capture and monitor its movement status in an invisible environment.Therefore,it is of great significance to develop a self-sensing and self-actuating dual-function hydrogel actuator system to realize real-time monitoring.Here,we report a bifunctional hydrogel system with self-actuating and self-monitoring abilities,which combines the functions of photothermal actuation and electrical resistance sensing into a single material.The bilayer tough conductive hydrogel synthesized by unconventional complementary concentration recombination and cryogenic freezing technique presents a dense conductive network and high-porosity structure,achieving high toughness at 190.3 kPa of tensile strength,high stretchability(164.3%strain),and the toughness dramatically(1,471.4 kJ·m^(−3)).The working mechanism of the monitoring and self-sensing system is accomplished through the integrated monitoring device of surface temperature–bending angle–electron current,to solve the problem of not apperceiving actuator motion state when encountering obstacles in an invisible environment.We demonstrated for the first time a photothermal actuator’s motion of a football player and goalkeeper to finish the penalty and a soft actuator hand,which can achieve the action of sticking to grab and release under photo-thermal actuation.When connected to the control closed circuit,the actuator realized closed-loop monitoring and sensing feedback.The development of bifunctional hydrogel systems may bring new opportunities and ideas in the fields of material science,circuit technology,sensors,and mechanical engineering. | Neng Chen Yang Zhou Yinping Liu Yuanyuan Mi Sisi Zhao Wang Yang Sai Che Hongchen liu FengJiang Chen Chong Xu Guang Ma Xue Peng Yongfeng Li | 2022 | Nano Research2022,15,8: | 3 |
| 20 | Machine learning helps improve diagnostic ability of subclinical keratoconus using Scheimpflug and OCT imaging modalities显示文摘Purpose:To develop an automated classification system using a machine learning classifier to distinguish clinically unaffected eyes in patients with keratoconus from a normal control population based on a combination of Scheimpflug camera images and ultra-high-resolution optical coherence tomography(UHR-OCT)imaging data.Methods:A total of 121 eyes from 121 participants were classified by 2 cornea experts into 3 groups:normal(50 eyes),with keratoconus(38 eyes)or with subclinical keratoconus(33 eyes).All eyes were imaged with a Scheimpflug camera and UHR-OCT.Corneal morphological features were extracted from the imaging data.A neural network was used to train a model based on these features to distinguish the eyes with subclinical keratoconus from normal eyes.Fisher’s score was used to rank the differentiable power of each feature.The receiver operating characteristic(ROC)curves were calculated to obtain the area under the ROC curves(AUCs).Results:The developed classification model used to combine all features from the Scheimpflug camera and UHR-OCT dramatically improved the differentiable power to discriminate between normal eyes and eyes with subclinical keratoconus(AUC=0.93).The variation in the thickness profile within each individual in the corneal epithelium extracted from UHR-OCT imaging ranked the highest in differentiating eyes with subclinical keratoconus from normal eyes.Conclusion:The automated classification system using machine learning based on the combination of Scheimpflug camera data and UHR-OCT imaging data showed excellent performance in discriminating eyes with subclinical keratoconus from normal eyes.The epithelial features extracted from the OCT images were the most valuable in the discrimination process.This classification system has the potential to improve the differentiable power of subclinical keratoconus and the efficiency of keratoconus screening. | Ce Shi Mengyi Wang Tiantian Zhu Ying Zhang Yufeng Ye Jun Jiang Sisi Chen Fan Lu Meixiao Shen | 2020 | Eye and Vision2020,7,1: | 3 |
