1Th e role of ferroptosis in ionizing radiation-induced cell death and tumor suppression显示文摘Ferroptosis,a form of regulated cell death caused by lipid peroxidation,was recently identified as a natural tumor suppression mechanism.Here,we show that ionizing radiation(IR)induces ferroptosis in cancer cells.Mechanistically,IR induces not only reactive oxygen species(ROS)but also the expression of ACSL4,a lipid metabolism enzyme required for ferroptosis,resulting in elevated lipid peroxidation and ferroptosis.ACSL4 ablation largely abolishes IR-induced ferroptosis and promotes radioresistance.IR also induces the expression of ferroptosis inhibitors,including SLC7A11 and GPX4;as an adaptive response.IR-or KEAP1 deficiencyinduced SLC7A11 expression promotes radioresistance through inhibiting ferroptosis.Inactivating SLC7A11 or GPX4 with ferroptosis inducers(FINs)sensitizes radioresistant cancer cells and xenograft tumors to IR.Furthermore,radiotherapy induces ferroptosis in cancer patients,and increased ferroptosis correlates with better response and longer survival to radiotherapy in cancer patients.Our study reveals a previously unrecognized link between IR and ferroptosis and indicates that further exploration of the combination of radiotherapy and FINs in cancer treatment is warranted.Guang Lei Yilei Zhang Pranavi Koppula Xiaoguang Liu Jie Zhang Steven HLin Jaffer AAjani Qin Xiao Zhongxing Liao Hui Wang Boyi Gan 2020Cell Research2020,30,2:88

4A Survey of Unbalanced Flow Diagnostics and Their Application显示文摘This paper presents an extensive survey of the most commonly used tools for diagnosing unbalanced flow in the atmosphere, namely the Lagrangian Rossby number, Psi vector, divergence equation, nonlinear balance equation, generalized omega-equation, and departure from fields obtained by potential vorticity (PV) inversion. The basic thoery, assumptions as well as implementation and limitations for each of the tools are all discussed. These tools are applied to high-resolution mesoscale model data to assess the role of unbalanced dynamics in the generation of a mesoscale gravity wave event over the East Coast of the United States. 5Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years,primarily by curbing end-of-pipe emissions.However,reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action.Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies.We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies.However,the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health.Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He 2021National Science Review2021,8,12:23
6PM_(2.5)-bound polycyclic aromatic hydrocarbons(PAHs) in Beijing:Seasonal variations,sources,and risk assessment显示文摘Polycyclic aromatic hydrocarbons(PAHs) have been of health concern due to its carcinogenesis and mutagenesis. In this study, we aimed to assess the variations, sources, and lifetime excessive cancer risk(ECR) attributable to PAHs bound to ambient particulate matters with aerodynamic diameter less than 2.5 μm(PM_(2.5)) in metropolitan Beijing, China. We collected24-hour integrated PM_(2.5) samples on daily basis between November 2014 and June 2015 across both central heating(cold months) and non-heating(warm months) seasons, and further analyzed the PAH components in these daily PM_(2.5) samples. Our results showed that total concentrations of PM_(2.5)-bound PAHs varied between(88.6 ± 75.4) ng/m^3 in the cold months and(11.0 ± 5.9) ng/m^3 in the warm months. Benzo[a]pyrene(Ba P), the carcinogenic marker of PAHs,averaged at 5.7 and 0.4 ng/m^3 in the cold and warm months, respectively. Source apportionment analyses illustrated that gasoline, biomass burning, diesel, coal combustion and cooking were the major contributors, accounting for 12.9%, 17.8%, 24.7%, 24.3% and 6.4% of PM_(2.5)-bound PAHs, respectively. The BaP equivalent lifetime ECR from inhalation of PM_(2.5)-bound PAHs was 16.2 cases per million habitants. Our results suggested that ambient particulate reduction from energy reconstruction and adaption of clean fuels would result in reductions PM_(2.5)-bound PAHs and its associated cancer risks. 7Minimally invasive surgery for gastric cancer: A comparison between robotic, laparoscopic and open surgery显示文摘AIM To investigate the role of minimally invasive surgery for gastric cancer and determine surgical, clinical, and oncological outcomes.METHODS This is a propensity score-matched case-control study, comparing three treatment arms: robotic gastrectomy(RG), laparoscopic gastrectomy(LG), open gastrectomy(OG). Data collection started after sharing a specific study protocol. Data were recorded through a tailored and protected web-based system. Primary outcomes: harvested lymph nodes, estimated blood loss, hospital stay, complications rate. Among the secondary outcomes, there are: operative time, R0 resections, POD of mobilization, POD of starting liquid diet and soft solid diet. The analysis includes the evaluation of type and grade of postoperative complications. Detailed information of anastomotic leakages is also provided.RESULTS The present analysis was carried out of 1026 gastrectomies. To guarantee homogenous distribution of cases, patients in the RG, LG and OG groups were 1:1:2 matched using a propensity score analysis with a caliper = 0.2. The successful matching resulted in a total sample of 604 patients(RG = 151; LG = 151; OG = 302). The three groups showed no differences in all baseline patients characteristics, type of surgery(P = 0.42) and stage of the disease(P = 0.16). Intraoperative blood loss was significantly lower in the LG(95.93 ± 119.22) and RG(117.91 ± 68.11) groups compared to the OG(127.26 ± 79.50, P = 0.002). The mean number of retrieved lymph nodes was similar between the RG(27.78 ± 11.45), LG(24.58 ± 13.56) and OG(25.82 ± 12.07) approach. A benefit in favor of the minimally invasive approaches was found in the length of hospital stay(P < 0.0001). A similar complications rate was found(P = 0.13). The leakage rate was not different(P = 0.78) between groups.CONCLUSION Laparoscopic and robotic surgery can be safely performed and proposed as possible alternative to open surgery. 8Global methane and nitrous oxide emissions from terrestrial ecosystems due to multiple environmental changes显示文摘Greenhouse gas(GHG)-induced climate change is among the most pressing sustainability challenges facing humanity today,posing serious risks for ecosystem health.Methane(CH_(4))and nitrous oxide(N_(2)O)are the two most important GHGs after carbon dioxide(CO_(2)),but their regional and global budgets are not well known.In this study,we applied a process-based coupled biogeochemical model to concurrently estimate the magnitude and spatial and temporal patterns of CH_(4)and N_(2)O fluxes as driven by multiple environmental changes,including climate variability,rising atmospheric CO_(2),increasing nitrogen deposition,tropospheric ozone pollution,land use change,and nitrogen fertilizer use.The estimated CH_(4)and N_(2)O emissions from global land ecosystems during 1981-2010 were 144.39±12.90 Tg C/yr(mean 62 SE;1 Tg=1012 g)and 12.52±0.74 Tg N/yr,respectively.Our simulations indicated a significant(P,0.01)annually increasing trend for CH_(4)(0.43±0.06 Tg C/yr)and N_(2)O(0.14±0.02 Tg N/yr)in the study period.CH_(4)and N_(2)O emissions increased significantly in most climatic zones and continents,especially in the tropical regions and Asia.The most rapid increase in CH_(4)emission was found in natural wetlands and rice fields due to increased rice cultivation area and climate warming.N_(2)O emission increased substantially in all the biome types and the largest increase occurred in upland crops due to increasing air temperature and nitrogen fertilizer use.Clearly,the three major GHGs(CH_(4),N_(2)O,and CO_(2))should be simultaneously considered when evaluating if a policy is effective to mitigate climate change.Hanqin Tian Guangsheng Chen Chaoqun Lu Xiaofeng Xu Wei Ren Bowen Zhang Kamaljit Banger Bo Tao Shufen Pan Mingliang Liu Chi Zhang Lori Bruhwiler Steven Wofsy 2015Ecosystem Health and Sustainability2015,1,1:7

16Effect of Preplant Fertilizer on Agronomic and Physiological Traits of Soybean Cultivars from Different Breeding Programs显示文摘Understanding the changes in agronomic and physiological traits associated with yield genetic gain is important for soybean production and future breeding strategy. The objective of this study was to compare the older and modern cultivars to learn whether the yield improvements depend on preplant fertilizer or the plant productivity improvement. A set of older cultivars, with their modern counterparts derived from breeding programs in Liaoning and Ohio were evaluated for their agronomic and physiological traits under different fertilizer levels from 2004 to 2006. There was no improvement of response to N and P preplant fertilizer for genotypes. After more than 70 yr breeding, soybean breeders made some improvements in agronomic and physiological traits that contribute to yield increase. When compared to older cultivar, modern Liaoning and Ohio cultivars were shorter and more resistance to lodging, had greater leaf density, higher harvest index, more leaf area per plant, and greater photosynthetic rate, transpiration rate and stomatal conductance at the beginning of seed development. 18Development of a CLDN18.2-targeting immuno-PET probe for non-invasive imaging in gastrointestinal tumors显示文摘Claudin18.2(CLDN18.2)is a tight junction protein that is overexpressed in a variety of solid tumors such as gastrointestinal cancer and oesophageal cancer.It has been identified as a promising target and a potential biomarker to diagnose tumor,evaluate efficacy,and determine patient prognosis.TST001 is a recombinant humanized CLDN18.2 antibody that selectively binds to the extracellular loop of human Claudin18.2.In this study,we constructed a solid target radionuclide zirconium-^(89)(^(89)Zr)labled-TST001 to detect the expression of in the human stomach cancer BGC823CLDN18.2 cell lines.The[^(89)Zr]Zr-desferrioxamine(DFO)-TST001 showed high radiochemical purity(RCP,>99%)and specific activity(24.15±1.34 GBq/mmol),and was stable in 5%human serum albumin,and phosphate buffer saline(>85%RCP at 96 h).The EC_(50) values of TST001 and DFO-TST001 were as high as 0.413±0.055 and 0.361±0.058 nM(P>0.05),respectively.The radiotracer had a significantly higher average standard uptake values in CLDN18.2-positive tumors than in CLDN18.2-negative tumors(1.11±0.02 vs.0.49±0.03,P=0.0016)2 days post injection(p.i.).BGC823CLDN18.2 mice models showed high tumor/muscle ratios 96 h p.i.with[^(89)Zr]Zr-DFO-TST001 was much higher than those of the other imaging groups.Immunohistochemistry results showed that BGC823CLDN18.2 tumors were highly positive(+++)for CLDN18.2,while those in the BGC823 group did not express CLDN18.2().The results of ex vivo biodistribution studies showed that there was a higher distribution in the BGC823CLDN18.2 tumor bearing mice(2.05±0.16%ID/g)than BGC823 mice(0.69±0.02%ID/g)and blocking group(0.72±0.02%ID/g).A dosimetry estimation study showed that the effective dose of[^(89)Zr]Zr-DFO-TST001 was 0.0705 mSv/MBq,which is within the range of acceptable doses for nuclear medicine research.Taken together,these results suggest that Good Manufacturing Practices produced by this immuno-positron emission tomography probe can detect CLDN18.2-overexpressing tumors.Yan Chen Xingguo Hou Dapeng Li Jin Ding Jiayue Liu Zilei Wang Fei Teng Hongjun Li Fan Zhang Yi Gu Steven Yu Xueming Qian Zhi Yang Hua Zhu 2023Journal of Pharmaceutical Analysis2023,13,4:2
