1Preparation and activity of conjugate of monoclonal antibody HAb18 against hepatoma F( ab′ )_2 fragment and staphylococcal enterotoxin A显示文摘AIM To prepare the conjugate of staphylococcal enterotoxin A (SEA) protein which is a bacterial SAg and the F(ab')2 fragment of mAb HAbl8 against human hepatocellular carcinoma (HCC), and identify its activity in order to use SAg in the targeting therapy of HCC.METHODS MAb HAbl8 was extracted from the abdominal dropsy of Balb/ c mice, and was purified through chromatography column SP-40HR with Fast protein liquid chromatography (FPLC) system. The F(ab')2 fragment of mAb HAb18 was prepared by papainic digestion method. The conjugate of mAb HAb18 F(ab')2fragment and SEA was prepared with chemical conjugating reagent N-succinimidyl-3-( 2-pyridyldithio) propionate (SPDP) and purified through chromatography column Superose 12with FPLC system. The molecular mass and purity of each collected peak were identified with SDS-PAGE assay. The protein content was assayed by Lowry's method. The antibody activity of HAb18 F (ab')2 against HCC in the conjugate was identified by indirect immunocytochemical ABC method, and the activity of SEA in the conjugate to activate peripheral blood mononuclear cells (PBMC) was identified with MTT assay.RESULTS The lgG mAb HAb18 was extracted,and purified successfully. Immunocytochemical staining demonstrated that it reacted with most of HHCC cells of human HCC cell line. There were two peaks in the process of purification of the prepared HAb18 F(ab)2-SEA conjugate. SDS-PAGE assay demonstrated that the molecular mass of the first peak was about 130 ku, and the second peak was the mixture of about 45 ku and a little 100 ku proteins. The immunocytochemical staining was similar in HAb18 F (ab ')2-SEAconjugate and HAb18 F (ab ')2, i.e., thecytoplasm and/or cell membranes of most HHCC cells were positively stained. The MTT assay showed that the optical absorbance (A) value at 490 nm of HAb18 F (ab')2-SEA conjugate was 0.182 ± 0.012, that of negative control was 0.033± 0.009, and there was significant difference between them ( P < 0.05).CONCLUSION SPDP is a good proteinconjugating reagent and can be used in preparing protein conjugate. 3Mortality outcomes of low-dose computed tomography screening for lung cancer in urban China:a decision analysis and implications for practice显示文摘Background: Mortality outcomes in trials of low-dose computed tomography(CT) screening for lung cancer are inconsistent. This study aimed to evaluate whether CT screening in urban areas of China could reduce lung cancer mortality and to investigate the factors that associate with the screening effect.Methods: A decision tree model with three scenarios(low-dose CT screening, chest X-ray screening, and no screening) was developed to compare screening results in a simulated Chinese urban cohort(100,000 smokers aged45-80 years). Data of participant characteristics were obtained from national registries and epidemiological surveys for estimating lung cancer prevalence. The selection of other tree variables such as sensitivities and specificities of low-dose CT and chest X-ray screening were based on literature research. Differences in lung cancer mortality(primary outcome), false diagnoses, and deaths due to false diagnosis were calculated. Sensitivity analyses were performed to identify the factors that associate with the screening results and to ascertain worst and optimal screening effects considering possible ranges of the variables.Results: Among the 100,000 subjects, there were 448,541, and 591 lung cancer deaths in the low-dose CT, chest X-ray, and no screening scenarios, respectively(17.2% reduction in low-dose CT screening over chest X-ray screening and 24.2% over no screening). The costs of the two screening scenarios were 9387 and 2497 false diagnoses and 7and 2 deaths due to false diagnosis among the 100,000 persons, respectively. The factors that most influenced death reduction with low-dose CT screening over no screening were lung cancer prevalence in the screened cohort, lowdose CT sensitivity, and proportion of early-stage cancers among low-dose CT detected lung cancers. 5Implementation of regional Acute Stroke Care Map increases thrombolysis rates for acute ischaemic stroke in Chinese urban area in only 3 months显示文摘Background The rate of intravenous thrombolysis for acute ischaemic stroke remains low in China.We investigated whether the implementation of a citywide Acute Stroke Care Map(ASCaM)is associated with an improvement of acute stroke care quality in a Chinese urban area.Methods The ASCaM comprises 10 improvement strategies and has been implemented through a network consisting of 20 tertiary hospitals.We identified 7827 patients with ischaemic stroke admitted from April to October 2017,and 506 patients underwent thrombolysis were finally included for analysis.Results Compared with‘pre-ASCaM period’,we observed an increased rate of administration of tissue plasminogen activator within 4.5 hours(65.4% vs 54.5%;adjusted OR,1.724;95% CI 1.21 to 2.45;p=0.003)during‘ASCaM period’.In multivariate analysis models,‘ASCaM period’was associated with a significant reduction in onset-to door time(114.1±55.7 vs 135.7±58.4 min,p=0.0002)and onset-to needle time(ONT)(169.2±58.1 vs 195.6±59.3 min,p<0.0001).Yet no change was found in door-to needle time.Clinical outcomes such as symptomatic intracranial haemorrhage,favourable functional outcome(modified Rankin Scale≤2)and in-hospital mortality remained unchanged.Conclusion The implementation of ASCaM was significantly associated with increased rates of intravenous thrombolysis and shorter ONT.The ASCaM may,in proof-of principle,serve as a model to reduce treatment delay and increase thrombolysis rates in Chinese urban areas and possibly other highly populated Asian regions.Yi Sui Jianfeng Luo Chunyao Dong Liqiang Zheng Weijin Zhao Yao Zhang Ying Xian Huaguang Zheng Bernard Yan Mark Parsons Li Ren Ying Xiao Haoyue Zhu Lijie Ren Qi Fang Yi Yang Weidong Liu Bing Xu 2021Stroke & Vascular Neurology2021,6,1:4
6IMPACTS OF DIMENSION AND SLOPE OF SUBMERGED SPUR DIKES ON LOCAL SCOUR PROCESSES-AN EXPERIMENTAL STUDY显示文摘In this experimental study, the impacts of the slope of the upstream face of submerged spur dikes on scour patterns in the vicinity of the submerged spur dike have been investigated. Associated hydraulic parameters for scour initiation have been studied. The dependence of the criteria for scour initiation and scour patterns on the dike dimensions such as the dike length and height has been assessed. It is found that the maximum depth of a scour hole is localized upstream of the spur dike end. The location of the maximum scouring depth, the end of the spur dike, and the spot with the maximum deposition are nearly aligned and form a line approximately parallel to the flume wall. Results show that the proper selection of a sloped upstream face of the spur dike can significantly reduce the maximum scour depth and scour volume in the vicinity of the submerged spur dike. 9Defective Ni_(3)S_(2)nanowires as highly active electrocatalysts for ethanol oxidative upgrading显示文摘Electrochemical upgrading of biomass ethanol to value-added chemicals is promising for sustainable society.Here,we synthesize defective Ni_(3)S_(2) nanowires(NWs),which show high activity towards electrochemical oxidation of ethanol to acetate.The Ni_(3)S_(2) NWs are formed by the oriented attachment mechanism,and rich defects are introduced during the growth.A low onset potential of 1.31 V and high mass activity of 8,716 mA·mgNi^(-1) at 1.5 V are achieved using the synthesized Ni_(3)S_(2) NWs toward the ethanol electro-oxidation,which are better than the Ni(OH)2 NWs and the Ni_(3)S_(2) nanoparticles(NPs).And the selectivity for the acetate generation is ca.99%.The high activity of Ni_(3)S_(2) NWs is attributed to the easier oxidation of Ni(II)to the catalytically active Ni(III)species with the promotion from S component and rich defects.These results demonstrate that the defective NWs can be synthesized by the oriented attachment method and the defective Ni_(3)S_(2) NWs structure as the efficient nonnoble metal electrocatalysts for oxidative upgrading of ethanol.Yufeng Zhang Wei Zhu Jinjie Fang Zhiyuan Xu Yanrong Xue Di Liu Rui Sui Qingqing Lv Xuerui Liu Yongsheng Wang Wei Chen Zhongbin Zhuang 2022Nano Research2022,15,4:2
10Effects of Antiretroviral Therapy and HIV Exposure in Utero on Adverse Pregnancy and Infant Outcomes:A Prospective Cohort Study in Guangzhou, China显示文摘Objective This study aimed to evaluate the effects of in-utero exposure to HIV and ART on pregnancy outcome and early growth of children.Methods This cohort study enrolled 802 HIV-infected pregnant women between October 2009 and May 2018 in Guangzhou, China. The women were assigned to receive combination ART(c ART) or mono/dual ART or no treatment. The primary outcomes were the combined endpoints of any adverse pregnancy outcome [including ectopic pregnancy, spontaneous abortion, stillbirth, preterm birth, small for gestational age(SGA)] and adverse early growth outcome(including infant death, HIV infection of mother-to-child transmission, and underweight, wasting and stunting of infants at 4 weeks of age).Results Adverse pregnancy outcomes occurred in 202(35.1%) of all enrolled HIV-infected women, and121(31.3%) of all infants exhibited adverse effects on early growth at 4 weeks of age. The rates of adverse pregnancy outcomes, spontaneous abortion, ectopic pregnancy, stillbirth, infant death and perinatal HIV infection were higher among women not receiving ART, compared to those treated with c ART or mono/dual ART(P < 0.05). However, women treated with c ART had a higher rate of SGA,compared to untreated women(P < 0.05). 15Dual-responsive nanovaccine for cytosolic delivery of antigens to boost cellular immune responses and cancer immunotherapy显示文摘Cancer vaccine contributing to the success of the treatment and prevention of tumors has attracted a huge attention as a strategy for tumor immunotherapy in recent years.A major challenge of cancer vaccine is to target cytosols of dendritic cells(DCs)in the lymph nodes(LNs)to enhance efficiency of antigen cross-presentation,which elicits high levels of cytotoxic T-lymphocytes to destruct tumor cells.Here,we address this issue by conjugating ovalbumin(OVA)to PEG-PCL using disulfide bond(-ss-),and the degradable pH-responsive polymer-PEI-PCL as delivery carrier.In addition,the mol ratio of PEG-PCL to PEI-PCL in the mixed micelles was tailored to deliver the OVA to LNs.Subsequently,CpG ODN_(1826),a TLR-9 agonist,was further introduced into a mixed micelle of 30nm or less as a unique tumor vaccine.Importantly,the results demonstrated the mixed micelles with 1:1mol of PCL-PEG and PCL-PEI can effectively migrate to distal LNs where antigen were efficiently captured by DCs,meanwhile,OVA was modified to the surface of mixed micelles via disulfide bonds(-ss-)for promotion efficiency of antigen cross-presentation.More surprisingly,combination of tumor vaccine with anti-PD-1,the therapy of ectopic melanoma(B16-OVA)and lung metastasis melanoma(B16-OVA)is excellent therapeutic effect.Taken together,our works offers a novel strategy for the cytosol delivery of antigens to achieve potent cancer immunotherapy.Yang Sui Ji Li Jiqiang Qu Ting Fang Hongyan Zhang Jian Zhang Zheran Wang Mingyu Xia Yinghui Dai Dongkai Wang 2022Asian Journal of Pharmaceutical Sciences2022,17,4:1

17TRPA1 channel mediates organophosphate-induced delayed neuropathy显示文摘OBJECTIVE We want to investigate the mechanism of organophosphate-induced delayed neuropathy(OPIDN) and find appropriate therapeutic medicine.OPIDN,often leads to paresthesias,ataxia and paralysis,occurs in the late-stage of acute poisoning or after repeated exposures to organophosphate(OP) insecticides or nerve agents,and may contribute to the Gulf War Syndrome.METHODS FDSS Ca2^(+)-influx assays,single-cell calcium imaging and patch-clamp electrophysiology were the major testing techniques.Transfected HEK293 cells and dorsal root ganglion(DRG) neurons were used to evaluate the effects of compounds.Wild type and trpa1 knockout mice and adult hyline brown hens were used to evaluate the neuropathological damages caused by the OPs.Transmission electron microscopy imaging was used to observe the nerve injuries ultrastructurally.High-throughput screen for TRPA1 inhibitors was accomplished by Ion Works Barracuda(IWB) automated electrophysiology assay.RESULTS TRPA1(Transient receptor potential cation channel,member A1) channel mediates OPIDN.A variety of OPs,exemplified by malathion,activates TRPA1 but not other neuronal TRP channels.Malathion increases the intracellular calcium levels and upregulates the excitability of mouse DRG neurons in vitro.Mice with repeated exposures to malathion also develop local tissue nerve injuries and pain-related behaviors,which resembles the early symptoms of OPIDN.Both the neuropathological changes and the nocifensive behaviors can be attenuated by treatment of TRPA1 antagonist HC030031 or abolished by knockout of Trpa1 gene.In the classic hens OPIDN model,malathion causes nerve injuries and ataxia to a similar level as the positive inducer tri-ortho-cresyl phosphate(TOCP),which also activates TRPA1 channel.Treatment with HC030031 reduces the damages caused by malathion or TOCP.Duloxetine and Ketotifen,two commercially available drugs exhibiting TRPA1 inhibitory activity,show neuroprotective effects against OPIDN and might be used in emergency situations.CONCLUSION TRPA1 is the major mediator of OPIDN and targeting TRPA1 is an effective way for the treatment of OPIDN.Qiang DING Sui FANG Xue-qin CHEN You-Xin WANG Jian LI Fu-yun TIAN Xiang XU Bernard ATTALI Xin XIE Zhao-bing GAO 2017中国药理学与毒理学杂志2017,31,10:1
18The design of HEPS global timing system显示文摘Purpose The high energy photon source(HEPS)is a 4th generation synchrotron light source under construction by the institute of high energy physics.The accelerator complex consists of a 500-MeV Linac,a full-energy synchrotron booster,a 6-GeV synchrotron storage ring(SR),and three e-beam transport lines for injection and extraction among accelerators.A global timing system(GTS)covers the timing needs for all accelerator,beamline,and experiment systems.The GTS is designed to coordinate the injection processes and various measurements and protections.Most systems require that the RMS jitter of the GTS signal is less than 30 ps,while the trigger jitters for the electron gun and SR injection and extraction kickers are less than 10 ps.Method The HEPS GTS is an event-based timing system based on MicroTCA.4 hardware architecture.The MicroTCA.4300 series products from the micro-research Finland Oy are implemented in the HEPS GTS system.Results and conclusions The RMS jitter,integrated from 1 Hz to 10 MHz,of the 166.6 MHz event clock is 5.489 ps.The RMS jitter of TTL outputs is less than 30 ps.This paper reports the design of the HEPS GTS,which satisfies all of the HEPS physics requirements for timing with preliminary test results shown.Fang Liu Ge Lei Zhe Duan Cai Meng Chenyan Lu Yanhua Shao Xinpeng Ma Nan Gan Yuemei Peng Jianshe Cao Chungming Chu Yanfeng Sui Jingyi Li IHEP 2021Radiation Detection Technology and Methods2021,5,3:1
