1Efficacy of Recombinant Adenoviral Human p53 Gene in Treatment of Malignant Pleural or Peritoneal Effusions显示文摘Background and objective Once the malignant pleural or peritoneal effusion is developed it is difficult to control.This report presents a new method for controlling the malignant effusions.Methods Forty-eight patients,29 males and 19 females with an average age of 61.2 years old,who were satisfied with the study inclusion criteria,were recruited in this study.Twenty-seven and 21 patients had a malignant pleural and peritoneal effusion,respectively.After draining most of fluids,these patients received intra-cavity infusion of rAd-p53 once per week for 4 weeks,at dose of 2×1012 viral particles(VP) diluted into 200 mL of saline solution for pleural effusions,and 4×1012 VP diluted into 500 mL of saline solution for peritoneal effusions.Results Participants were followed up for a median time of 13.6 month.A total of 11 cases,7 with pleural effusions and 4 with peritoneal effusions achieved a complete response(CR),and 20 cases(12 pleural effusions and 8 peritoneal effusions) had a partial response(PR).The overall response rate is 64.6%.Patients' quality of life,assessed by using Karnofsky performance scale(KPS) scores,was improved by an average of 26.4.The one-year of overall survival rate was 54.2% with a median survival time of 12.5 months.There were no serious side effects observed except for self-limited fever found in 79.8% of the cases.Conclusions Intra-cavity infusion of rAd-p53 is an effective and safe treatment for the patients with malignant pleural or peritoneal effusions,especially for those patients who can't tolerate the standard treatments.Xin ZHANG Yi HU Jinliang WANG Sujie ZHANG Haitao TAO Sun JING Baishou WU 2013中国肺癌杂志2013,16,3:5
2Host Interferon-Stimulated Gene 20 Inhibits Pseudorabies Virus Proliferation显示文摘Host interferon-stimulated gene 20(ISG20)exerts antiviral effects on viruses by degrading viral RNA or by enhancing IFN signaling.Here,we examined the role of ISG20 during pseudorabies virus(PRV)proliferation.We found that ISG20 modulates PRV replication by enhancing IFN signaling.Further,ISG20 expression was upregulated following PRV infection and poly(I:C)treatment.Ectopic expression of ISG20 inhibited PRV proliferation in PK15 cells,whereas knockdown of ISG20 promoted PRV proliferation.In addition,ISG20 expression upregulated IFN-βexpression and enhanced IFN downstream signaling during PRV infection.Notably,PRV UL24 suppressed the transcription of ISG20,thus antagonizing its antiviral effect.Further domain mapping analysis showed that the N terminus(amino acids 1-90)of UL24 was responsible for the inhibition of ISG20 transcription.Collectively,these findings characterize the role of ISG20 in suppressing PRV replication and increase the understanding of host-PRV interplay.Xiaoyong Chen Dage Sun Sujie Dong Huanjie Zhai Ning Kong Hao Zheng Wu Tong Guoxin Li Tongling Shan Guangzhi Tong 2021Virologica Sinica2021,36,5:4

4“Top-down” and “bottom-up” strategies for wafer-scaled miniaturized gas sensors design and fabrication显示文摘Manufacture of large-scale patterned nanomaterials via top-down techniques,such as printing and slurry coating,have been used for fabrication of miniaturized gas sensors.However,the reproducibility and uniformity of the sensors in wafer-scale fabrication are still a challenge.In this work,a“top-down”and“bottom-up”combined strategy was proposed to manufacture wafer-scaled miniaturized gas sensors with high-throughput by in-situ growth of Ni(OH)2 nanowalls at specific locations.First,the micro-hotplate based sensor chips were fabricated on a two-inch(2″)silicon wafer by micro-electro-mechanical-system(MEMS)fabrication techniques(“top-down”strategy).Then a template-guided controllable de-wetting method was used to assemble a porous thermoplastic elastomer(TPE)thin film with uniform micro-sized holes(relative standard deviation(RSD)of the size of micro-holes<3.5%,n>300),which serves as the patterned mask for in-situ growing Ni(OH)2 nanowalls at the micro-hole areas(“bottom-up”strategy).The obtained gas microsensors based on this strategy showed great reproducibility of electric properties(RSD<0.8%,n=8)and sensing response toward real-time H _(2)S detection(RSD<3.5%,n=8).Lin Liu Yingyi Wang Fuqin Sun Yanbing Dai Shuqi Wang Yuanyuan Bai Lianhui Li Tie Li Ting Zhang Sujie Qin 2020Microsystems & Nanoengineering2020,6,1:1

8Hexafluoropropylene oxide trimer acid,a perfluorooctanoic acid alternative,induces cardiovascular toxicity in zebrafish embryos显示文摘As an increasingly used alternative to perfluorooctanoic acid(PFOA),hexafluoropropylene oxide trimer acid(HFPO-TA)has been widely detected in global water environments.However,little is known regarding its toxic effects on cardiovascular development.Here,zebrafish embryos were treated with egg water containing 0,60,120,or 240 mg/L HFPO-TA.Results showed that HFPO-TA treatment led to a significant reduction in both larval survival percentage and heart rate.Furthermore,HFPO-TA exposure caused severe pericardial edema and elongation of the sinus venous to bulbus arteriosus distance(SV-BA)in Tg(myl7:GFP)transgenic larvae,disrupting the expression of genes involved in heart development and thus causing abnormal heart looping.Obvious sprouting angiogenesis was observed in the 120 and 240 mg/L exposed Tg(fli:GFP)transgenic larvae.HFPO-TA treatment also impacted the mRNA levels of genes involved in the vascular endothelial growth factor(VEGF)pathway and embryonic vascular development.HFPO-TA exposure significantly decreased erythrocyte number in Tg(gata1:DsRed)transgenic embryos and influenced gene expression associated with the heme metabolism pathway.HFPO-TA also induced oxidative stress and altered the transcriptional levels of genes related to cell cycle and apoptosis,inhibiting cell proliferation while promoting apoptosis.Therefore,HFPO-TA exposure may induce abnormal development of the cardiovascular and hematopoietic systems in zebrafish embryos,suggesting it may not be a suitable or safe alternative for PFOA.Sujie Sun Li Zhang Xue Li Lu Zang Ling Huang Junquan Zeng Zigang Cao Xinjun Liao Zilin Zhong Huiqiang Lu Jianjun Chen 2024Journal of Environmental Sciences2024,,5:0
9PNKP Knockdown by RNA Interference Inhibits Herpes Simplex Virus-1 Replication in Astrocytes显示文摘Herpes simplex virus-1 (HSV-1) is a major pathogen that causes various central nervous system (CNS) diseases,including herpes simplex encephalitis and meningitis.According to recent studies,PNKP significantly affects the proliferation of HSV-1 in astrocytes.Here,we used viral proliferation curves to confirm the significant inhibitory effects of PNKP on HSV-1 proliferation.PNKP downregulation was also confirmed by analyzing the transcription of viral genes.We found that PNKP downregulation affects the viral DNA copy number.This study preliminarily confirms that PNKP affects viral proliferation by affecting HSV-1 genome cyclization.These results also suggest that astrocytes play a specific role in preventing HSV-1 infection.Lei Yue Sujie Guo Xia Cao Ying Zhang Le Sun Longding Liu Min Yan Qihan Li 2013Virologica Sinica2013,28,6:0
