| 1 | Analysis of genetic diversity and population structure of Chinese yak breeds (Bos grunniens) using microsatellite markers显示文摘Nine Chinese yak breeds (Maiwa, Tianzhu White, Qinghai Plateau, Sibu, Zhongdian, Pali, Tibetan High Mountain, Jiulong, and Xin-jiang) and Gayal were analyzed by means of 16 microsatellite markers to determine the level of genetic variation within populations, genetic relationship between populations, and population structure for each breed. A total of 206 microsatellite alleles were observed. Mean F-statistics (0.056) for 9 yak breeds indicated that 94.4% of the genetic variation was observed within yak breeds and 5.6% of the genetic variation existed amongst breeds. The Neighbor-Joining phylogenetic tree was constructed based on Nei's standard genetic dis-tances and two clusters were obtained. The Gayal separated from the yaks far away and formed one cluster and 9 yak breeds were grouped together. The analysis of population structure for 9 yak breeds and the Gayal showed that they resulted in four clusters; one clus-ter includes yaks from Tibet Autonomous Region and Qinghai Province, one cluster combines Zhongdian, Maiwa, and Tianzhu White, and Jiulong and Xinjiang come into the third cluster. Pali was mainly in the first cluster (90%), Jiulong was mainly in the second cluster (87.1%), Zhongdian was primarily in the third cluster (83%), and the other yak breeds were distributed in two to three clusters. | 2 | Homocysteine aggravates cerebral hemorrhage in rats: Correlation with oxygen-free radical production and cell apoptosis in tissue surrounding hematoma显示文摘BACKGROUND:Previous studies have demonstrated that homocysteine is an independent risk factor for ischemic stroke,as determined by detection of apoptosis and oxygen-free radical scavengers following cerebral ischemia.However,the mechanisms of homocysteine remain unclear.Several reports have addressed the effects of homocysteine on ischemic stroke.OBJECTIVE:To analyze the effects of homocysteine on apoptosis,intracellular superoxide dismutase(SOD) activity,and malondialdehyde content in tissue surrounding hematoma in rats with cerebral hemorrhage,and to determine the action pathway of malondialdehyde following cerebral hemorrhage.DESIGN,TIME AND SETTING:The randomized,controlled,animal experiment was performed at the Laboratory of Molecular Biology,Hospital Affiliated to Luzhou Medical College,China from April 2007 to April 2008.MATERIALS:In situ apoptosis detection kit(Roche,Mannheim,Germany),SOD detection kit and malondialdehyde detection kit(Nanjing Jiancheng Bioengineering Institute,China),and homocysteine(Sigma,St Louis,MO,USA) were used in the present study.METHODS:A total of 75 Sprague Dawley rats were equally and randomly assigned to sham surgery,model,and homocysteine groups.Autologous blood was infused into the caudate putamen of rats to establish models of cerebral hemorrhage in model and homocysteine groups.Homocysteine was injected directly into the brain through the skull at the hematoma hemisphere at 30 minutes after model induction in the homocysteine group.MAIN OUTCOME MEASURES:At 6,12,24,and 72 hours,as well as 1 week,post-surgery,neurological deficits were observed in each group.Brain water content was measured using the dry-wet weight method.Cell apoptosis in tissue surrounding the hematoma was detected utilizing terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling(TUNEL).SOD activity and malondialdehyde content in tissue surrounding the hematoma were respectively measured using the xanthine oxidase and thiobarbituric acid methods.RESULTS:Neurological function was similar between model and homocysteine groups following cerebral hemorrhage(P > 0.05).Brain water content was increased at 12 hours post-surgery,peaked at 3 days,and remained unchanged at 7 days in the model group.Brain edema was not significantly aggravated following homocysteine intervention(P > 0.05),but SOD activity significantly decreased and malondialdehyde content significantly increased(P < 0.05).The number of apoptotic cells increased in rats with cerebral hemorrhage at 12 hours(P < 0.05),and numbers peaked at 72 hours following model establishment(P < 0.05).The time of peak value was identical between model and homocysteine groups.Brain water content was negatively associated with SOD activity(rmodel group =-0.448,P < 0.05;rhomocysteine group =-0.612,P < 0.05),but was positively associated with malondialdehyde content(rmodel group = 0.542,P < 0.05;rhomocysteine group = 0.684,P < 0.05) in brain tissues surrounding the hematoma following surgery in model and homocysteine groups.CONCLUSION:Homocysteine aggravates neurological dysfunction and brain edema in rats with cerebral hemorrhage.The mechanisms of action are likely associated with production of oxygen-free radical and cellular apoptosis following cerebral hemorrhage. | Yingying Liu Xinjun Li Chun Wang Chun Chen Ping Liu Qin Feng Duanxiu Liao Xiaogang Li | 2010 | Neural Regeneration Research2010,5,13: | 3 |

| 4 | Hexafluoropropylene oxide trimer acid,a perfluorooctanoic acid alternative,induces cardiovascular toxicity in zebrafish embryos显示文摘As an increasingly used alternative to perfluorooctanoic acid(PFOA),hexafluoropropylene oxide trimer acid(HFPO-TA)has been widely detected in global water environments.However,little is known regarding its toxic effects on cardiovascular development.Here,zebrafish embryos were treated with egg water containing 0,60,120,or 240 mg/L HFPO-TA.Results showed that HFPO-TA treatment led to a significant reduction in both larval survival percentage and heart rate.Furthermore,HFPO-TA exposure caused severe pericardial edema and elongation of the sinus venous to bulbus arteriosus distance(SV-BA)in Tg(myl7:GFP)transgenic larvae,disrupting the expression of genes involved in heart development and thus causing abnormal heart looping.Obvious sprouting angiogenesis was observed in the 120 and 240 mg/L exposed Tg(fli:GFP)transgenic larvae.HFPO-TA treatment also impacted the mRNA levels of genes involved in the vascular endothelial growth factor(VEGF)pathway and embryonic vascular development.HFPO-TA exposure significantly decreased erythrocyte number in Tg(gata1:DsRed)transgenic embryos and influenced gene expression associated with the heme metabolism pathway.HFPO-TA also induced oxidative stress and altered the transcriptional levels of genes related to cell cycle and apoptosis,inhibiting cell proliferation while promoting apoptosis.Therefore,HFPO-TA exposure may induce abnormal development of the cardiovascular and hematopoietic systems in zebrafish embryos,suggesting it may not be a suitable or safe alternative for PFOA. | Sujie Sun Li Zhang Xue Li Lu Zang Ling Huang Junquan Zeng Zigang Cao Xinjun Liao Zilin Zhong Huiqiang Lu Jianjun Chen | 2024 | Journal of Environmental Sciences2024,,5: | 0 |
| 5 | Detection of genomic signatures for pig hairlessness using high-density SNP data显示文摘Hair provides thermal regulation for mammals and protects the skin from wounds,bites and ultraviolet(UV)radiation,and is important in adaptation to volatile environments.Pigs in nature are divided into hairy and hairless,which provide a good model for deciphering the molecular mechanisms of hairlessness.We conducted a genomic scan for genetically differentiated regions between hairy and hairless pigs using 60K SNP data,with the aim to better understand the genetic basis for the hairless phenotype in pigs.A total of 38405 SNPs in 498 animals from 36 diverse breeds were used to detect genomic signatures for pig hairlessness by estimating between-population(FST)values.Seven diversifying signatures between Yucatan hairless pig and hairy pigs were identified on pig chromosomes(SSC)1,3,7,8,10,11 and 16,and the biological functions of two notable genes,RGS17 and RB1,were revealed.When Mexican hairless pigs were contrasted with hairypigs,strong signatures were detected on SSC1 and SSC10,which harbor two functionally plausible genes,REV3L and BAMBI.KEGG pathway analysis showed a subset of overrepresented genes involved in the T cell receptor signaling pathway,MAPK signaling pathway and the tight junction pathways.All of these pathways may be important in local adaptability of hairless pigs.The potential mechanisms underlying the hairless phenotype in pigs are reported for the first time.RB1 and BAMBI are interesting candidate genes for the hairless phenotype in Yucatan hairless and Mexico hairless pigs,respectively.RGS17,REV3L,ICOS and RASGRP1 as well as other genes involved in the MAPK and T cell receptor signaling pathways may be important in environmental adaption by improved tolerance to UV damage in hairless pigs.These findings improve our understanding of the genetic basis for inherited hairlessness in pigs. | Ying SU Yi LONG Xinjun LIAO Huashui AI Zhiyan ZHANG Bin YANG Shijun XIAO Jianhong TANG Wenshui XIN Lusheng HUANG Jun REN Nengshui DING | 2014 | Frontiers of Agricultural Science and Engineering2014,1,4: | 0 |
